A high-performance liquid chromatographic method for the determination in human plasma of the recent noradrenergic and specific serotonergic antidepressant (NaSSA) mirtazapine and its two main metabolites, N-desmethylmirtazapine and 8-hydroxymirtazapine, has been developed. Fluorescence detection was used, exciting at λ = 290 nm and monitoring emission at λ = 370 nm. Separation was obtained by using a reversed-phase column (C8, 250 mm × 4.6 mm I.D., 5 μm) and a mobile phase composed of 75% aqueous phosphate buffer containing triethylamine at pH 3.0 and 25% acetonitrile. Melatonin was used as the internal standard. A careful pre-treatment of plasma samples was developed, using solid-phase extraction with phenyl cartridges (100 mg, 1 mL). The calibration curves were linear over a working range of 5-150 ng mL−1 for mirtazapine and of 2.5-75.0 ng mL−1 for N-desmethylmirtazapine and 8-hydroxymirtazapine. The limit of quantitation (LOQ) was 2.5 ng mL−1 and the limit of detection (LOD) was 1.25 ng mL−1 for all analytes. The method was applied with success to plasma samples from depressed patients undergoing treatment with mirtazapine. Precision data, as well as accuracy results, were satisfactory and no interference from other drugs was found. Hence the method is suitable for therapeutic drug monitoring of mirtazapine and its metabolites in depressed patients’ plasma.  相似文献

A new procedure was described with multiwalled carbon nanotubes as solid phase extraction packing material for the trace analysis of nicosulfuron, thifensulfuron and metsulfuron-methyl in water samples. The possible parameters influencing the enrichment were optimized and the optimal conditions were as followed: eluent, sample pH, flow rate and sample volume were acetonitrile containing 1% acetic acid, pH 3, 8 mL min−1 and 500 mL, respectively. Under the optimal chromatographic separation and SPE conditions, the linear range, detection limit (S/N = 3) and precision (R.S.D., n = 6) were 0.04-40 ng mL−1, 6.8 ng L−1 and 2.5% for nicosulfuron, 0.04-40 ng mL−1, 11.2 ng L−1 and 5.4% for thifensulfuron, 0.02-20 ng mL−1, 5.9 ng L−1, 2.1% for metsulfuron-methyl, respectively. The established method was well employed to determine nicosulfuron, thifensulfuron and metsulfuron-methyl in tap water, seawater, reservoir water and well water samples, and satisfactory results were obtained, the spiked recoveries in the range of 87.2-100.7%, 96.5-105.6% and 83.7-111.1% for them each, respectively.  相似文献

In the present study a cloud point extraction process using mixed micelle of the cationic surfactant cetyl-pyridinium chloride (CPC) and non-ionic surfactant Triton X-114 for extraction of beryllium from aqueous solutions is developed. The extraction of analyte from aqueous samples was performed in the presence of 1,8-dihydroxyanthrone as chelating agent in buffer media of pH 9.5. After phase separation, the surfactant-rich phase was diluted with 0.4 mL of a 60:40 methanol-water mixture containing 0.03 mL HNO3. Then, the enriched analyte in the surfactant-rich phase was determined by inductively coupled plasma-atomic emission spectrometry (ICP-AES). The different variables affecting the complexation and extraction conditions were optimized. Under the optimum conditions (i.e. 1.6 × 10−4 mol L−1 1,8-dihydroxyanthrone, 1.2 × 10−4 mol L−1 CPC, 0.15% (v/v) Triton X-114, 50 °C equilibrium temperature) the calibration graph was linear in the range of 0.006-80 ng mL−1 with detection limit of 0.001 ng mL−1 and the precision (R.S.D.%) for five replicate determinations at 18 ng mL−1 of Be(II) was better than 2.9%. In this manner the preconcentration and enrichment factors were 16.7 and 24.8, respectively. Under the presence of foreign ions no significant interference was observed. Finally, the proposed method was successfully utilized for the determination of this cation in water samples.  相似文献

A simple, rapid and sensitive high-performance liquid chromatography method was developed for the analysis of the sesquiterpene lactone 15-deoxygoyazensolide (LAC15-D) in rat plasma samples. The chromatographic separation was achieved on a LiChrospher® RP18 column using methanol:water (50:50, v/v) containing 0.6% acetic acid as mobile phase, at a flow rate of 0.7 mL min−1. UV detection was carried out at 270 nm. Phenytoin was used as internal standard. Prior to the analysis, the rat plasma samples were submitted to liquid-liquid extraction with dichloromethane. The mean absolute recoveries were 73% with R.S.D. values lower than 3.5. The method was linear over the 6.0-2000 ng mL−1 concentration range and the quantification limit was 6.0 ng mL−1. Within-day and between-day assay precision and accuracy were studied at three concentration levels (15, 300 and 480 ng mL−1) and were lower than 15%. The validated method was used to measure the plasmatic concentration of LAC15-D in rats that received a single intraperitoneal dose of 30 mg kg−1.  相似文献

Amitriptyline, doxepin and chlorpromazine are often used as psychotropic drugs in treatment of the various mental diseases, and are also partly excreted by kidney. This work developed a simple, selective and sensitive method for their simultaneous monitoring in human urine using capillary electrophoresis coupled with electrochemiluminescence (ECL) detection based on end-column ECL reaction of tris-(2,2′-bipyridyl)ruthenium(II) with aliphatic tertiary amino moieties. Acetone was used as an additive to the running buffer to obtain their absolute separation. Under optimized conditions the proposed method displayed a linear range from 5.0 to 800 ng mL−1 for the three drugs with the correlation coefficients more than 0.995 (n = 8). Their limits of detection were 0.8 ng mL−1 (3.6 fg), 1.0 ng mL−1 (4.5 fg) and 1.5 ng mL−1 (6.8 fg) at a signal to noise ratio of 3, respectively. The relative standard deviations for five determinations of 20 ng mL−1 amitriptyline, doxepin and chlorpromazine were 1.7%, 4.2% and 3.6%, respectively. For practical application an extract step with 90:10 heptane/ethyl acetate (v/v) was performed to eliminate the influence of ionic strength in sample. The recoveries of amitriptyline, doxepin and chlorpromazine at different levels in human urine were between 83% and 93%, which showed that the method was valuable in clinical and biochemical laboratories for monitoring amitriptyline, doxepin and chlorpromazine.  相似文献

A new polymeric coating consisting of a dual-phase, polydimethylsiloxane (PDMS) and polypyrrole (PPY) was developed for the stir bar sorptive extraction (SBSE) of antidepressants (mirtazapine, citalopram, paroxetine, duloxetine, fluoxetine and sertraline) from plasma samples, followed by liquid chromatography analysis (SBSE/LC-UV). The extractions were based on both adsorption (PPY) and sorption (PDMS) mechanisms. SBSE variables, such as extraction time, temperature, pH of the matrix, and desorption time were optimized, in order to achieve suitable analytical sensitivity in a short time period. The PDMS/PPY coated stir bar showed high extraction efficiency (sensitivity and selectivity) toward the target analytes. The quantification limits (LOQ) of the SBSE/LC-UV method ranged from 20 ng mL−1 to 50 ng mL−1, and the linear range was from LOQ to 500 ng mL−1, with a determination coefficient higher than 0.99. The inter-day precision of the SBSE/LC-UV method presented a variation coefficient lower than 15%. The efficiency of the SBSE/LC-UV method was proved by analysis of plasma samples from elderly depressed patients.  相似文献

A new liquid chromatographic tandem mass spectrometric method for the determination of mirtazapine and demethylmirtazapine in human plasma has been developed and fully validated. The article describes in detail the bioanalytical procedure and summarizes the validation results obtained. The samples were extracted using liquid-liquid extraction with a mixture of 1-chlorobutane/isopropanol/ethyl acetate (88:2:10, (v/v/v)). The chromatographic separation was performed on a reversed-phase XTerrra MS C8 column ( i.d.; 3.5 μm particle size) using a mobile phase consisting of 0.010 M ammonium formate (pH 7.8) and acetonitrile (35:65, (v/v)), pumped at a flow rate of 0.80 ml min−1. The analytes were detected using a Finnigan LCQ advantage ion-trap mass spectrometer with positive electrospray ionization in selected reaction monitoring (SRM) mode. Tandem mass spectrometric detection enabled the quantitation of both compounds down to 0.10 ng ml−1. Calibration graphs were linear (r better than 0.990, n=11), in concentration ranges 0.10 to 200 ng ml−1 for mirtazapine demethylmirtazapine. The intra- and inter-day R.S.D. values were less than 14.8 and 16.6% for mirtazapine and demethylmirtazapine, respectively. The method was successfully applied to a kinetic study in order to assess the main pharmacokinetic parameters of mirtazapine and demethylmirtazapine.  相似文献

A simple and sensitive method with a fast sample preparation procedure is proposed for the determination of mercury species in plasma/serum. The method combines online high-performance liquid chromatography separation, Hg cold-vapor formation and inductively coupled plasma mass spectrometry detection. Prior to analysis, plasma (250 μL) was accurately pipetted into 15 mL conical tubes. Then, an extractant solution containing mercaptoethanol, L-cysteine and HCl was added to the samples following sonication for 10 min. Quantitative mercury extraction was achieved with the proposed procedure. Separation of mercury species was accomplished in less than 8 min on a C8 reverse phase column with a mobile phase containing 3% v/v methanol + 97% v/v (0.5% v/v 2-mercaptoethanol + 0.05% v/v formic acid). The method detection limits were found to be 12 ng L−1, 5 ng L−1 and 4 ng L−1 for inorganic mercury, ethylmercury and methylmercury, respectively. Method accuracy is traceable to Standard Reference Material (SRM) 966 Toxic Metals in Bovine Blood from NIST. Additional validation was provided by the analysis of a secondary reference serum sample from the INSQ-Canada. Finally, the method was successfully applied for the speciation of mercury in plasma samples collected from volunteers exposed to methylmercury through fish consumption. For the first time to our knowledge, levels of different species of Hg in plasma samples from riverside populations exposed to MeHg were determined.  相似文献

A sensitive analytical method for the determination of tetrodotoxin (TTX) in urine and plasma matrices was developed using double solid phase extraction (C18 and hydrophilic interaction liquid chromatography) and subsequent analysis by HPLC coupled with tandem mass spectrometry. The double SPE sample cleanup efficiently reduced matrix and ion suppression effects. Together with the use of ion pair reagent in the mobile phase, isocratic elution became possible which enabled a shorter analysis time of 5.5 min per sample. The assay results were linear up to 500 ng mL−1 for urine and 20 ng mL−1 for plasma. The limit of detection and limit of quantification were 0.13 ng mL−1 and 2.5 ng mL−1, respectively, for both biological matrices. Recoveries were in the range of 75-81%. To eliminate the effect of dehydration and variations in urinary output, urinary creatinine-adjustment was made. TTX was quantified in eight urine samples and seven plasma samples from eight patients suspected of having TTX poisoning. TTX was detected in all urine samples, with concentrations ranging from 17.6 to 460.5 ng mL−1, but was not detected in any of the plasma samples. The creatinine-adjusted TTX concentration in urine (ranging from 7.4 to 41.1 ng μmol−1 creatinine) correlated well with the degree of poisoning as observed from clinical symptoms.  相似文献

This paper presents the development of a new flow-injection system combined with solid-surface fluorescence detection for the determination of the widely used fungicide thiabendazole. Nylon powder was probed as a novel solid support for building the optosensor. The method is based on the on-line immobilization of thiabendazole onto nylon in a continuous flow system, followed by the measurement of its native fluorescence. Aqueous samples are directly injected in a water carrier, resulting in a very simple and economical method. The analytical figures of merit obtained using 1500 μL of sample and 75% methanol (v/v) as eluting solution were: linear calibration range from 8 to 120 ng mL−1 (the lowest value corresponds to the quantitation limit), relative standard deviation, 0.9% (n = 5) at a level of 64 ng mL−1, limit of detection calculated according to 1995 IUPAC recommendations is to 2.8 ng mL−1, and sampling rate of 14 samples h−1. The potential interference from other agrochemicals, metal ions and common anions, and the viability of determining thiabendazole in real water samples were also evaluated.  相似文献

A comparison of direct immersion solid-phase microextraction (DI-SPME) and stir bar sorptive extraction (SBSE) coupled to liquid chromatography (HPLC) with fluorimetric detection for the rapid analysis of resveratrol isomers is described. For DI-SPME, a polar Carbowax-template resin (CW/TPR) 50 μm fiber was the most efficient and optimum extraction conditions were 40 °C and an extraction time of 30 min, stirring in the presence of 5% (m/v) sodium chloride and 0.07 M acetate/acetic acid buffer (pH 6). Desorption was carried out using the static mode for 10 min. Linearity was obtained in the 5-150 and 2-150 ng mL−1 ranges for trans- and cis-resveratrol, with detection limits of 2 and 0.5 ng mL−1, respectively. When using SBSE, a polydimethylsiloxane (PDMS) twister provided best extraction by means of a derivatization reaction in the presence of acetic anhydride and potassium carbonate. The same time and temperature were used for the extraction step in the presence of 2.5% (m/v) sodium chloride, and liquid desorption was performed with 150 μL of a 50/50 (v/v) acetonitrile/1% (v/v) acetic acid solution in a desorption time of 15 min. Linearity was now between 0.5 and 50 ng mL−1 for trans-resveratrol with a detection limit of 0.1 ng mL−1, while cis-resveratrol could not be extracted. The proposed methods were successfully applied to determining the resveratrol isomer content of wine, must and fruit juices.  相似文献

High-performance liquid chromatography (HPLC) enantioseparation of terazosin (TER) was accomplished on the immobilised-type Chiralpak IC chiral stationary phase (CSP) under both polar organic and reversed-phase modes. A simple analytical method was validated using a mixture of methanol–water–DEA 95:5:0.1 (v/v/v) as a mobile phase. Under reversed-phase conditions good linearities were obtained over the concentration range 8.76–26.28 μg mL−1 for both enantiomers. The limits of detection and quantification were 10 and 30 ng mL−1, respectively. The intra- and inter-day assay precision was less than 1.66% (RSD%). The optimised conditions also allowed to resolve chiral and achiral impurities from the enantiomers of TER. The proposed HPLC method supports pharmacological studies on the biological effects of the both forms of TER and analytical investigations of potential drug formulations based on a single enantiomer. At the semipreparative scale, 5.3 mg of racemic sample were resolved with elution times less than 12 min using a mobile phase consisting of methanol–DEA 100:0.1 (v/v) and both enantiomers were isolated with a purity of ≥99% enantiomeric excess (ee). The absolute configuration of TER enantiomers was assigned by comparison of the measured specific rotations with those reported in the literature.  相似文献

A highly selective, sensitive and rapid high performance liquid chromatographic method has been developed and validated to quantify gemifloxacin in human plasma. The gemifloxacin and internal standard (ciprofloxacin) were extracted by ultrafiltration technique followed by injection into chromatographic system. Chromatographic separation was achieved on a reversed phase C18 column with a mobile phase of acetonitrile:0.1% trifluoroacetic acid (20:80, v/v) using isocratic elution (at flow rate 1 mL min−1). The analytes were detected at 269 and 393 nm for excitation and emission, respectively. The assay exhibited a linear range of 25-5000 ng mL−1 for gemifloxacin in human plasma. The lower limit of detection was 10 ng mL−1. The method was statistically validated for linearity, accuracy, precision and selectivity following FDA guidelines. The intra- and inter-assay coefficients of variation did not exceed 7.6% deviation of the nominal concentration. The recovery of gemifloxacin from plasma was greater than 97.0%. Stability of gemifloxacin in plasma was excellent with no evidence of degradation during sample processing (auto-sampler) and at least 3 months storage in a freezer at −70 °C. This validation method is applied for clinical study of the gemifloxacin in human volunteers.  相似文献

A method for simultaneous determination of seven benzodiazepines (BZPs) (flunitrazepam, clonazepam, oxazepam, lorazepam, chlordiazepoxide, nordiazepam and diazepam using N-desalkylflurazepam as internal standard) in human plasma using liquid-liquid and solid-phase extractions followed by high-performance liquid chromatography (HPLC) is described. The analytes were separated employing a LC-18 DB column (250 mm × 4.6 mm, 5 μm) at 35 °C under isocratic conditions using 5 mM KH2PO4 buffer solution pH 6.0:methanol:diethyl ether (55:40:5, v/v/v) as mobile phase at a flow rate of 0.8 mL min−1. UV detection was carried out at 245 nm. Employing LLE, the best conditions were achieved with double extraction of 0.5 mL plasma using ethyl acetate and Na2HPO4 pH 9.5 for pH adjusting. Employing SPE, the best conditions were achieved with 0.5 mL plasma plus 3 mL 0.1 M borate buffer pH 9.5, which were then passed through a C18 cartridge previously conditioned, washed for 3 times with these solvents: 3 mL 0.1 M borate buffer pH 9.5, 4 mL Milli-Q water and 1 mL acetonitrile 5%, finally the BZPs elution was carried with diethyl ether:n-hexane:methanol (50:30:20). In both methods the solvent was evaporated at 40 °C under nitrogen flow. The validation parameters obtained in LLE were linearity range of 50-1200 ng mL−1 plasma (r ≥ 0.9927), limits of quantification of 50 ng mL−1 plasma, within-day and between-day CV% and E% for precision and accuracy lower than 15%, and recovery above 65% for all BZPs. In SPE, the parameter obtained were linearity range of 30-1200 ng mL−1 plasma (r ≥ 0.9900), limits of quantification of 30 ng mL−1 plasma, within-day and between-day CV% and E% for precision and accuracy lower than 15% and recovery above 55% for all BZPs. These extracting procedures followed by HPLC analysis showed their suitable applicability in order to examine one or more BZPs in human plasma. Moreover, it could be suggested that these procedures might be employed in various analytical applications, in special for toxicological/forensic analysis.  相似文献

A simple, fast, and sensitive liquid–liquid extraction method followed by nonaqueous capillary electrophoresis (LLE/NACE) was developed and validated for simultaneous determination of four antidepressants (fluoxetine, sertraline, citalopram and paroxetine) in human plasma. Several experimental separation conditions using aqueous and nonaqueous media separation were tested by varying the electrolyte pH value (for aqueous medium) and the ionic strength concentration considering the similar mobility of the compounds. High-resolution separation was achieved with a mixture of 1.25 mol L−1 of phosphoric acid in acetonitrile. The quantification limits of the LLE/CE method varied between 15 and 30 ng mL−1, with a relative standard deviation (RSD) lower than 10.3%. The method was successfully applied in therapeutic drug monitoring and should be employed in the evaluation of plasma levels in urgent toxicological analysis.  相似文献

A rapid and sensitive method for the determination of carbendazim (methyl benzimidazole-2-ylcarbamate, MBC) and thiabendazole (TBZ) in water and soil samples was developed by using dispersive liquid-liquid microextraction (DLLME) coupled with high performance liquid chromatography with fluorescence detection. The water samples were directly used for the DLLME extraction. For soil samples, the target analytes were first extracted by 0.1 mol L−1 HCl. Then, the pH of the extract was adjusted to 7.0 with 2 mol L−1 NaOH before the DLLME extraction. In the DLLME extraction method, chloroform (CHCl3) was used as extraction solvent and tetrahydrofuran (THF) as dispersive solvent. Under the optimum conditions, the enrichment factors for MBC and TBZ were ranged between 149 and 210, and the extraction recoveries were between 50.8 and 70.9%, respectively. The linearity of the method was obtained in the range of 5-800 ng mL−1 for water sample analysis, and 10-1000 ng g−1 for soil samples, respectively. The correlation coefficients (r) ranged from 0.9987 to 0.9997. The limits of detection were 0.5-1.0 ng mL−1 for water samples, and 1.0-1.6 ng g−1 for soil samples. The relative standard deviations (RSDs) varied from 3.5 to 6.8% (n = 5). The recoveries of the method for MBC and TBZ from water samples at spiking levels of 5 and 20 ng mL−1 were 84.0-94.0% and 86.0-92.5%, respectively. The recoveries for soil samples at spiking levels of 10 and 100 ng g−1 varied between 82.0 and 93.4%.  相似文献

Liquid chromatography (LC)/tandem mass spectrometry (MS/MS) after supramolecular solvent-based microextraction (SUSME) was firstly used in this work for the enantioselective determination of chiral pesticides in natural waters. The method developed for the quantitation of the R- and S-enantiomers of mecoprop (MCPP) and dichlorprop (DCPP) involved the extraction of the herbicides in a supramolecular solvent (SUPRAS) made up of reverse aggregates of dodecanoic acid (DoA), analyte re-extraction in acetate buffer (pH = 5.0), separation of the target enantiomers on a chiral column of permethylated α-cyclodextrin under isocratic conditions, and detection of the daughter ions (m/z = 140.9 and 160.6 for MCPP and DCPP, respectively) using a hybrid triple quadrupole mass spectrometer equipped with an electrospray source operating in the negative ion mode. Similar recoveries (ca. 75%) and actual concentration factors (ca. 94) were obtained for both phenoxypropanoic acids (PPAs). The quantitation limits were 1 ng L−1 for R- and S-MCPP, and 4 ng L−1 for R- and S-DCPP, and the precision, expressed as relative standard deviation (n = 6) was in the ranges 2.4–2.7% ([R-MCPP] = [S-MCPP] = 5 ng L−1 and [R-DCPP] = [S-DCPP] = 15 ng L−1) and 1.6–1.8% (100 ng L−1 of each enantiomer). The SUSME-LC–MS/MS method was successfully applied to the determination of the enantiomers of MCPP and DCPP in river and underground waters, fortified at concentrations between 15 and 180 ng L−1 at variable enantiomeric ratios (ER = 1–9).  相似文献

Arsine trapping on resistively heated tungsten coil was investigated and an analytical method for ultratrace arsenic determination in environmental samples was established. Several chemical modifiers, including Re, Pt, Mo, Ta and Rh, were explored as permanent chemical modifiers for tungsten coil on-line trapping and Rh gave the best performance. Arsine was on-line trapped on Rh-coated tungsten coil at 640 °C, then released at 1930 °C and subsequently delivered to an atomic fluorescence spectrometer (AFS) by a mixture of Ar and H2 for measurement. In the medium of 2% (v/v) HCl and 3% (m/v) KBH4, arsine can be selectively generated from As(III). Total inorganic arsenic was determined after pre-reduction of As(V) to As(III) in 0.5% (m/v) thiourea-0.5% (m/v) ascorbic acid solution. The concentration of As(V) was calculated by difference between the total inorganic arsenic and As(III), and inorganic arsenic speciation was thus achieved. With 8 min on-line trapping, the limit of detection was 10 ng L−1 for As(III) and 9 ng L−1 for total As; and the precision was found to be <5% R.S.D. (n = 7) for 0.2 ng mL−1 As. The proposed method was successfully applied in total arsenic determination of several standard reference materials and inorganic arsenic speciation analysis of nature water samples.  相似文献

Sulfasalazine is an antibiotic used in the treatment of inflammatory bowel diseases. For the assessment of sulfasalazine in several biological matrices, an Enzyme-Linked Immunosorbent Assay (ELISA) method based on polyclonal antibodies was developed and characterized.The immunoassay showed a high sensitivity (IC50 = 0.51 ng mL−1) and specificity, a detection limit of 0.02 ng mL−1 and a dynamic range of 0.06-3.75 ng mL−1 (80-20% inhibition). The immunoassay performed well when it was applied to spiked plasma samples (from 0.5 to 2.0 ng mL−1) previously cleaned up by protein precipitation with methanol. Recoveries ranged from 83 to 119%, with a mean value of 99% (CV = 13%).Since sulfasalazine remaining of a treatment reaches the systemic circulation in unchanged form, the immunoassay can be applied to the determination of this pharmaceutical in human plasma in order to facilitate the control of the patients through the application of personal doses.  相似文献

Organophosphate triesters are common flame retardants used in a wide variety of consumer products from which they can migrate and pollute the indoor environment. Humans may thus be continuously exposed to several organophosphate triesters which might be a risk for human health. An analytical method based on direct injection of 5 μL urine into an ultra performance liquid chromatography system coupled to a time-of-flight mass spectrometry has been developed and validated to monitor exposure to organophosphate triesters through their respective dialkyl and diaryl phosphate metabolites (DAPs). The targeted analytes were: di-n-butyl phosphate (DNBP), diphenyl phosphate (DPHP), bis(2-butoxyethyl) phosphate (BBOEP), bis(2-chloroethyl) phosphate (BCEP), bis(1-chloro-2-propyl) phosphate (BCPP) and bis(1,3-dichloro-2-propyl) phosphate (BDCIPP). Separation was achieved in less than 3 min on a short column with narrow diameter and small particle size (50 mm × 2.1 mm × 1.7 μm). Different mobile phases were explored to obtain optimal sensitivity. Acetonitrile/water buffered with 5 mM of ammonium hydroxide/ammonium formate (pH 9.2) was the preferred mobile phase. Quantification of DAPs was performed using deuterated analogues as internal standards in synthetic urine (averaged DAP accuracy was 101%; RSD 3%). Low method limits of quantification (MLQ) were obtained for DNBP (0.40 ng mL−1), DPHP (0.10 ng mL−1), BDCIPP (0.40 ng mL−1) and BBOEP (0.60 ng mL−1), but not for the most polar DAPs, BCEP (∼12 ng mL−1) and BCPP (∼25 ng mL−1). The feasibility of the method was tested on 84 morning urine samples from 42 mother and child pairs. Only DPHP was found above the MLQ in the urine samples with geometric mean (GM) concentrations of 1.1 ng mL−1 and 0.57 ng mL−1 for mothers and children respectively. BDCIPP was however, detected above the method limit of detection (MLD) with GM of 0.13 ng mL−1 and 0.20 ng mL−1. While occasionally detected, the GM of DNBP and BBOEP were below MLD in both groups.  相似文献
