壳聚糖-精氨酸树脂固定化胰凝乳蛋白酶及其性质   总被引:1,自引:0,他引:1  
以具柔性亲水手臂的壳聚糖-精氨酸树脂为载体,用戊二醛交联胰凝乳蛋白酶,获得壳聚糖-精氨酸树脂固定化胰凝乳蛋白酶. 最佳固定化条件为:m(酶)∶ m(载体)=20∶ 1 000、戊二醛体积分数为1.0%、pH=5.20、30 ℃交联60 min. 固定化酶活力达850 U/g,Km为1.83 mmol/L,比游离酶增大33.6%,比交联壳聚糖固定化酶低24.0%. 壳聚糖-精氨酸树脂固定化胰凝乳蛋白酶水解时间进程曲线与游离酶基本一致,均在反应30 min达到最大速率,最适温度为70 ℃,比游离酶升高10 ℃;在75 ℃时的半衰期可达6.0 h,比游离酶提高约4.3倍;最适pH值为5.92,比游离酶向酸性偏移2pH单位. 4 ℃贮存半衰期为49 d.  相似文献

报道了醇脱氢酶(ADH)的固定化和酶学性质研究。以壳聚糖作为载体,戊二醛作为交联剂。固定化ADH的最适条件为:以6%戊二醛将壳聚糖交联2 h,与ADH反应2.5 h。对游离和固定化ADH酶学性质的研究表明:酶促反应的最适pH均为8.2,最适温度分别为37℃和40℃,对乙醇的表观米氏常数Km分别为33.9 mmol/L和46.2 mmol/L。与游离酶相比,固定化酶具有良好的操作稳定性。  相似文献

以磁性壳聚糖作为载体,戊二醛作为交联剂,对乳酸脱氢酶(LDH)进行固定化.固定化的最适条件为:戊二醛浓度6%,pH值7.5,酶的偶联时间2 h.对游离及固定化LDH酶学性质的研究表明,酶促反应的最适pH值为9.2,最适温度分别为37℃和50℃,对乳酸的表观米氏常数分别为1.6 mmol/L和0.9 mmol/L.游离酶和固定化酶在40℃放置150 min后,其活力分别为最初的56.5%和76.1%.固定化酶在4℃贮存4周后,活力仍保留50%以上.固定化酶在室温下与底物重复反应6次后,活力仍保留60%以上,说明固定化酶具有较好的热稳定性、贮存稳定性和复用性.  相似文献

在异维生素C钠(D-VcNa)的合成过程中,以SO42-/S iO2-TiO2复合固体超强酸作为酯化反应催化剂,2-酮基-D-葡萄糖酸(2-KGA)和甲醇为原料,采用精馏脱水酯化工艺,不断除去酯化过程中产生的水。催化剂的最佳焙烧温度为550℃,催化剂用量为2.5%;反应时间3~4 h,产品收率为88.6%,催化剂的重复使用性较好,催化体系优于硫酸法。  相似文献

以壳聚糖为载体对醇脱氢酶(ADH)和脲酶(UASE)进行固定化。固定化的最适条件:ADH和UASE的偶联时间分别为2.5h和1.5h,交联剂戊二醛浓度为0.6%和0.5%,偶联pH值为6.8和5.0。对游离酶及固定化酶的性质研究表明,ADH酶促反应的最适宜pH为8.0和8.4,最适宜温度为33℃和35℃,米氏常数为48mmol/L和13mmol/L;UASE酶促反应的最适宜pH均为7.0,最适宜温度为60℃和72℃,米氏常数为0.4mmol/L和2mmol/L。固定化酶与游离酶相比在复用性上具有优势,用固定化酶测定了试样中尿素含量以及模拟样中银离子含量。  相似文献

以十六烷基三甲基溴化铵(CATB)-辛烷-己醇反胶束体系对乳酸脱氢酶(LDH)进行固定化,试验了含水量、CTAB和己醇浓度对LDH固定化的影响。对游离酶和固定化酶的催化动力学性质研究表明:酶促反应的最适pH值分别为9.2和9.5,最适温度分别是37℃和43℃,对乳酸的米氏常数Km分别为1.4 mmol/L和2.3mmol/L。30℃时,游离酶存放2 h,约失活50%,而固定化酶仅失活10%,表明反胶束固定化LDH具有较好的热稳定性。  相似文献

采用淀粉酶、糖化酶、纤维素酶和蛋白酶对茶皂素进行纯化水解,探讨了温度、p H、时间等因素对茶皂素纯化效果的影响。结果表明,通过酶法可以提高茶皂素的纯度,当温度50℃、p H=7、水解时间24 h的条件下,酶可使茶皂素的纯度由67.5%提高到90%以上。  相似文献

以油茶饼粕为原材料,采用微波预处理复合酶法,通过单因素试验研究了液固比、温度、时间、pH值和复合酶用量对油茶饼粕中茶皂素提取率的影响,采用正交试验确定最佳提取工艺条件。结果表明在时间为1.5 h、液固比为9∶1、pH值为6、浸提温度为55℃、复合酶用量为0.45%的条件下,茶皂素的提取率为13.35%,产品纯度为58.47%。  相似文献

采用超临界CO2萃取蝼蛄总脂肪酸,对影响萃取效果的各个因素应用正交设计试验,优化了各萃取参数.各个因素的影响顺序为压力>温度>时间>流量.萃取的优化条件为压力35 MPa,温度50℃,时间1 h,流量45 kg/h.运用该萃取条件改善了萃取物的物理性状,脂肪酸的纯度提高,总脂肪酸的含量可达83.29%;而采用常规的有机溶剂提取法,脂肪酸的纯度仅为60%左右.甲酯化采用气相色谱-质谱分析技术对其中的16个成分进行了鉴定,并测定了相对含量,其中主要有油酸甲酯(30.40%)、十六烷酸甲酯(14.19%)、9,12-十八碳二烯酸甲酯(11.99%)、油酸乙酯(11.75%)、9-十六碳烯酸甲酯(7.77%)、十六烷酸乙酯(5.97%)、9,12-十八碳二烯酸乙酯(4.53%)及9-十六碳烯酸乙酯(2.87%)等.  相似文献

采用交联酶聚体(CLEAs)技术制得了木瓜蛋白酶CLEAs, 优化了制备条件. 以纯乙醇为蛋白沉淀剂, 质量分数40%戊二醛为交联剂, 于4 ℃下对酶沉淀聚体交联16 h; 所得木瓜蛋白酶CLEAs的最适pH为6.0(游离酶最适pH=7.0), 最适温度范围由游离酶的80 ℃拓宽为50~80 ℃, 热稳定性和溶液稳定性亦明显提高; 微观形貌分析证明木瓜蛋白酶CLEAs优良的催化效能及稳定性来自于CLEAs单元所具有的高比表面积及单元内部多点共价固定的结合方式.  相似文献

用溶胶-凝胶法以磷钼酸(MPA)的镍盐溶液水解钛酸四丁酯制备了NiPMo/TiO2催化剂.使用ICP、 XRD、 TG-DTA、 IR、 TPD-MS和微反应技术研究了催化剂的化学组成、热稳定性、化学吸附性质和催化反应性能.杂多钼酸盐与TiO2通过O2-在TiO2表面发生了键合.在623 K下,杂多阴离子仍保持原有的Keggin结构.CO2在Lewis酸位Ni(Ⅱ)和Lewis碱位Ni-O-Mo的桥氧协同作用下生成CO2卧式吸附态Ni(Ⅱ)←O-(CO)←(O--Ni).丙烯有多种吸附态在催化剂上吸附.在563 K、 1 MPa和空速1500 h-1的反应条件下,丙烯的摩尔转化率为3.2%,产物MAA选择性为95%.  相似文献

In the context of the preparation of camptothecin and luotonin A analogs, the synthesis of some key keto-precursors and their use in Friedländer condensation are described. This paper also focuses on the stability of these keto intermediates and emphasizes the major differences between indolizinones and pyrroloquinazolinones series. The Langevin paramagnetic theory can’t describe the relation between magnetization of ferrofluids and applied magnetic field. The structuralization of ferrofluids, which is considered the main influence factor of the magnetization, is regarded. The part of magnetization works is deposited when the structure is forming. This action influences the magnetization of ferrofluids directly or indirectly. On the base of the “compressing” model, the Langevin function that usually describes the magnetization of ferrofluid is modified, and a well-fitted curve is obtained. An equation of the relation between the equivalent volume fraction after being “compressed” and the intensity of magnetic field is discovered, which approximately describes the process of magnetization. The relation between the approximate initial susceptibility and the volume fraction can be obtained from modified formula.  相似文献

The highly regioselective Buchwald–Hartwig amination at C-2 of the cheap and readily accessible reagent, 2,4-dichloropyridine with a range of anilines and heterocyclic amines is described. This new methodology is robust and provides a facile access to 4-chloro-N-phenylpyridin-2-amines on 0.25 mol scale. These intermediates undergo a further Buchwald–Hartwig amination at higher temperature to enable rapid exploration of the chemical space at C-4 and to provide a library of 2,4-bisaminopyridines.  相似文献

KMnO4-mediated oxidative CN bond cleavage of tertiary amines producing secondary amine was introduced, which was trapped by electrophiles (acyl chloride and sulfonyl chloride) to form amides and sulfonamides. A general synthesis of previously unknown semicarbazone-based α-amidoalkylating reagents, 4-(tosylmethyl)semicarbazones, has been developed. The synthesis involved three-component condensation of semicarbazones of aliphatic or aromatic aldehydes with the same or other aldehydes and p-toluenesulfinic acid. The scope and limitations of this reaction were investigated. The compounds obtained were demonstrated to be an efficient α-(4-semicarbazono)alkylating agents. They were reacted with H- (sodium borohydride), O- (sodium methylate), S- (sodium phenylthiolate), N- (pyrrolidine, sodium succinimide), P- (trialkyl phosphites), and C-nucleophiles (sodium diethyl malonate) to give the corresponding products of the tosyl group substitution, 4-substituted semicarbazones, including analogues of nitrofurazone. Among the prepared compounds tested in vitro for antibacterial and antifungal activity, three nitrofuryl-containing semicarbazones exhibited high biological activities with minimum inhibitory concentration (MIC) values of 8–32 μg/mL.  相似文献

Regiospecific and direct imidation of the methyl C(sp3)–H bond of thioanisoles is realized under mild and metal-free conditions with N-fluorobis(benzenesulfonyl)imide as an oxidant and nitrogen source. Proposed mechanism suggests that thionium ion intermediates and a Pummerer-type reaction are involved. The imidation has advantages such as high step-economy, excellent functionality tolerance, and regiospecificity, giving structurally diverse imidation products.  相似文献

A small library of new chiral bidentate hydroxyalkyl-imidazolium salts 1 is conveniently synthesized on multi-gram scale from inexpensive and commercially available chiral pool amino acids. The corresponding carbenes, generated by deprotonation of imidazolium salts 1, in combination with palladium(II) chloride were tested in the Mizoroki–Heck coupling reaction. The most significant results in terms of yields and reactivities were achieved with low catalyst loading. The catalytic activities of these imidazolium salts were also investigated in the asymmetric addition of diethylzinc to benzaldehyde. The use of MgO nanoparticles as an additive in conjunction with these ligands played a crucial role in increasing the efficiency of these reactions.  相似文献
