Asarones (alpha-asarone and beta-asarone) are the active components in the traditional Chinese medicine (TCM) of Acorus tatarinowii Schott, which has been used to treat epilepsy for several thousand years. To perform the pharmacokinetics (PK) study of alpha- and beta-asarone from the TCM essential oil, a simple, rapid and sensitive method was developed for the determination of asarones from the TCM in rabbit plasma, based on headspace solid-phase microextraction (HS-SPME) followed by gas chromatography/mass spectrometry (GC/MS) with electron ionization (EI). The extraction parameters of headspace volume, fiber coating, sample temperature, extraction time, stirring rate and ion strength were systemically optimized. Furthermore, the method linearity, detection limit and precision were also investigated. It was shown that the proposed method provided a good linearity (0.02-20 microg/mL, R(2) > 0.99), low detection limit (<2.0 ng/mL) and good precision (RSD < 7.0%). Finally, HS-SPME followed by GC/MS was applied to fast determination of alpha- and beta-asarone in rabbit plasma at different time points after oral adminstration of the essential oil from A. tatarinowii. The experimental results suggest that the proposed method provides an alternative approach to the PK studies of volatile compounds in TCMs. 相似文献

A new method is described for the qualitative and quantitative analysis of midazolam, a short-acting 1,4-imidazole benzodiazepine, in human plasma. It involves a plasma deproteinization step, solid-phase microextraction (SPME) of midazolam using an 85-microm polyacrylate fiber, and its detection by gas chromatography/mass spectrometry (GC/MS) in selected ion monitoring (SIM) mode, using pinazepam as internal standard. The assay is linear over a midazolam plasma range of 1.5-300 ng/mL, relative intra- and inter-assay standard deviations at 5 ng/mL are below 7%, and the limit of detection is 1 ng/mL. The method is simple, fast and sufficiently sensitive to be applied in clinical and forensic toxicology as well as for purposes of therapeutic drug monitoring. 相似文献

Analysis of amino acids in blood samples is an important tool for the diagnosis of neonatal amino acid metabolism disorders. In the work, a novel, rapid and sensitive method was developed for the determination of amino acids in neonatal blood samples, which was based on microwave-assisted silylation followed by gas chromatography/mass spectrometry (GC/MS). The amino acids were derivatized with N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) under microwave irradiation. The controlled reaction was carried out employing BSTFA under conventional heating at 120 degrees C for 30 min. Experimental results show that microwave irradiation can accelerate the derivatization reaction of amino acids with BSFTA, and much shorten analysis time. The method validations (linear range, detection limit, precision and recovery) were studied. Finally, the method was tested by determination of amino acids in neonatal blood by the measurement of their trimethylsilyl derivatives by GC/MS in electron impact (EI) mode. Two biomarkers of L-phenylalanine and L-tyrosine in phenylketonuria (PKU)-positive blood and control blood were quantitatively analyzed by the proposed method. The results demonstrated that microwave-assisted silylation followed by GC/MS is a rapid, simple and sensitive method for amino acid analysis and is also a potential tool for fast screening of neonatal aminoacidurias. 相似文献

In the current work, a simple, rapid, accurate and inexpensive method was developed for the determination of acetone in human blood. The proposed method is based on derivatization with O-(2,3,4,5,6-pentafluorobenzyl)hydroxylamine hydrochloride (PFBHA), followed by headspace liquid-phase microextraction (HS-LPME) and gas chromatography/mass spectrometry (GC/MS). In the present method, acetone in blood samples was derivatized with PFBHA and acetone oxime formed in several seconds. The formed oxime was enriched by HS-LPME using the organic solvent film (OSF) formed in a microsyringe barrel as extraction interface. Finally, the enriched oxime was analyzed by GC/MS in electron ionization (EI) mode. HS-LPME parameters including solvent, syringe plunger withdrawal rate, sampling volume, and extraction cycle were optimized and the method reproducibility, linearity, recovery and detection limit were studied. The proposed method was applied to determination of acetone in diabetes blood and normal blood. It has been shown that derivatization with HS-LPME and GC/MS is an alternative method for determination of the diabetes biomarker, acetone, in blood samples. 相似文献

The purpose of this study was to develop a simple, rapid and sensitive analytical method for determination of amino acids in neonatal blood samples. The developed method involves the employment of derivatization and a solid-phase microextraction (SPME) technique together with gas chromatography/mass spectrometry (GC/MS). Amino acids in blood samples were derivatized by a mixture of isobutyl chloroformate, methanol and pyridine, and the N(O,S)-alkoxycarbonyl alkyl esters thus formed were headspace extracted by a SPME fiber. Finally, the extracted analytes on the fiber were desorbed and detected by GC/MS in electron impact (EI) mode. L-Valine, L-leucine, L-isoleucine, L-phenylanaline and L-tyrosine in blood samples were quantitatively analyzed by measurement of the corresponding N(O,S)-alkoxycarbonyl alkyl esters using an external standard method. SPME conditions were optimized, and the method was validated. The method was applied to diagnosis of neonatal phenylkenuria (PKU) and maple syrup urine disease (MSUD) by the analyses of five amino acids in blood samples. The results showed that the proposed method is a potentially powerful tool for simultaneous screening for neonatal PKU and MSUD. 相似文献

A method for the extraction and gas chromatographic determination of methylmercury in biological matrices is presented. By combining the advantages of two extraction techniques-microwave-assisted extraction (MAE) and solid-phase microextraction (SPME)--the separation of methylmercury from biological samples is possible. Specifically, the procedure involves microwave extraction with 3 M hydrochloric acid, followed by aqueous-phase derivatization with sodium tetraphenylborate and headspace SPME with a silica fibre coated with polydimethylsiloxane (PDMS). For optimization of the derivatization-SPME procedure, a central composite experimental design with alpha = 1.682 and two central points was used to model gas-chromatographic peak areas as functions of pH, extraction temperature and sorption time. A desirability function was then used for the simultaneous optimization for methylmercury and Hg(II). The optimal derivatization-SPME conditions identified were close to pH 5, temperature 100 degrees C, and sorption time 15 min. The identification and quantification of the extracted methylmercury is carried out by gas chromatography with microwave-induced plasma atomic emission spectrometry detection. The validity of the new procedure is shown by the results of analyses of certified reference materials. 相似文献

Paeonol is the active component in the traditional Chinese medicines (TCMs), such as Cynanchum paniculatum, which has been used to treat many diseases, such as eczema. In this work, a simple, rapid and sensitive method was developed for the determination of paeonol in rabbit plasma, which was based on headspace solid-phase microextraction (HS-SPME) followed by gas chromatography-mass spectrometry (GC-MS). The extraction parameters of fiber coating, sample temperature, extraction time, stirring rate and ion strength were systemically optimized; the method linearity, detection limit and precision were also investigated. It was shown that the proposed method provided a good linearity (0.02-20 μg mL−1, R2 > 0.990), low detection limit (2.0 ng mL−1) and good precision (R.S.D. value less than 8%). Finally, GC/MS following HS-SPME was applied to fast determination of paeonol in rabbit plasma at different time point after oral demonstration of Cynanchum paniculatum essential oil. The experimental results suggest that the proposed method provided an alternative and novel approach to the pharmacokinetics study of paeonol in the TCMs. 相似文献

Aldehydes are considered potential markers for enhanced oxidative stress and have been proposed as a diagnostic measure of cancer status. Do to their volatility and activity, it is very difficult to accurately measure aldehydes in human blood. In the present work, gas chromatography/mass spectrometry (GC/MS) and solid-phase microextraction (SPME) with on-fiber derivatization was developed for determination of aldehydes in human blood. O-(2,3,4,5,6-Pentafluorobenzyl)hydroxylamine hydrochloride (PFBHA) in aqueous solution was first adsorbed by a SPME fiber, and then the aldehydes in blood samples were headspace extracted by the SPME fiber and rapidly derivatized with PFBHA on the SPME fiber. Finally, the oximes formed were desorbed and detected by GC/MS in electron ionization (EI) mode. Validation of the present method was carried out, and the method was applied to quantitative analysis of the aldehydes in lung cancer blood. The results demonstrated that GC/MS and SPME with on-fiber derivatization is a simple, rapid, sensitive and solvent-free method for the determination of aldehydes in human blood. 相似文献

A rapid and simple method of using solid-phase microextraction was developed for determination of 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) in water samples. In this method, the target analyte is extracted from the sample into the polymeric coating of the fused-silica fiber. After exposure, the fiber is thermally desorbed in the heated injection port of the gas chromatograph, and a chromatographic analysis is performed by using low-resolution tandem mass spectrometry. Parameters that may affect the extension of the microextraction process, such as sampling mode, sample volume, temperature, agitation, and sampling time, were studied. Extraction efficiencies for 3 coating fibers were investigated: 100 microm poly(dimethylsiloxane) (PDMS), 65 microm PDMS-divinylbenzene, and 75 microm carboxen-PDMS. Linearity was evaluated (R = 0.999) for a 250-fold concentration range from the fg/mL to the pg/mL level. The 2,3,7,8-TCDD was detected at the fg/mL level when the headspace over the water sample was sampled for 60 min; the limit of detection obtained was better than that of Method 8280B of the U.S. Environmental Protection Agency. The proposed method performed well when applied to the analysis of tap water, lake water, and seawater samples. 相似文献

A sensitive procedure for the determination of five ultraviolet (UV) absorbers, belonging to the benzotriazole class, in environmental
water samples is proposed. Analytes were first extracted and concentrated from the matrix and then selectively determined
by gas chromatography in combination with tandem mass spectrometry detection. The high lipophilic character of some of the
investigated species resulted in a strong trend to remain sorbed on solid surfaces, even after addition of considerable percentages
of methanol (up to 30%) to water. Thus, minimizing sample handling during the enrichment step is mandatory in order to obtain
acceptable accuracy and precision. Solid-phase microextraction (SPME), as sample preparation approach, fulfilled the above
requirement and provided acceptable figures of merit for the determination of target species in environmental water samples,
including raw wastewater. Optimization of SPME conditions showed that the combination of headspace extraction, with a sample
temperature of 100 °C and addition of 15 mg of NaCl per milliliter of sample rendered the best compromise in terms of extraction
efficiency for all species. Considering a sampling time of 30 min with a poly(dimethylsiloxane)–divinylbenzene-coated SPME
fiber, limits of quantification below 2 ng l−1 and relative standard deviations between 5% and 12% were achieved. Three of the five species included in this research were

A simple and sensitive method for determination of phosphoric acid triesters at trace levels in human plasma sample is described. In this work, solid-phase microextraction (SPME) is employed as a sample preparation procedure for extraction and pre-concentration of alkyl and aryl phosphates followed by gas chromatography coupled to inductively coupled plasma mass spectrometry (GC-ICP-MS) for phosphorus-specific and very sensitive determination of these compounds in human plasma. The detection limits from blood plasma were 50 ngL(-1) (tripropyl phosphate), 17 ngL(-1) (tributyl phosphate), 240 ngL(-1) (tris(2-chloroethyl) phosphate) and 24 ngL(-1) (triphenyl phosphate). Sample preparation involves plasma deproteinization followed by direct immersion SPME with 65 microm poly(dimethylsiloxane/divinylbenzene) fiber. Extraction was performed at 40 degrees C for 30 min and at pH 7.0 in 10 mM sodium carbonate buffer. The reported method, to our knowledge, describes the first application of SPME with element-specific detection for analysis of phosphoric acid esters. Application of the method to the plasma samples, previously stored in poly(vinyl chloride) plasma bags revealed the presence of triphenyl phosphate, which was further confirmed by SPME GC time-of-flight high-resolution mass spectrometry. 相似文献

The original solid-phase microextraction (SPME) fibers use an epoxy resin adhesive that releases bisphenol A (BPA) during thermal desorption of the fiber. This adversely affects the method detection limit and accuracy when these products are used for the determination of BPA. In this work, 5 new metal alloy SPME fibers that do not use epoxy resins were compared for the extraction of BPA in water. The performance of the optimum SPME fiber with 60 microm carbowax-polyethylene glycol coating for the headspace SPME of BPA in water was investigated systematically under different extraction conditions. Salt was found to increase the partitioning of BPA from water into the headspace until saturation was reached. Partitioning of BPA from water into the headspace also increased at higher extraction temperatures, as did longer extraction times. However, extraction of BPA from water onto the SPME fiber was not improved for solutions adjusted to pH 2 compared to the unadjusted neutral solutions. The new BPA method showed good linearity over the concentration range of 2.5 to 40 microg/L [correlation coefficient (r2) = 0.995] .The method detection limit for BPA was 0.5 microg/L, while the instrument detection limit was as low as 0.05 microg/L. Good repeatability was observed for BPA at levels of 5 and 20 microg/L with relative standard deviation values < 10%. The automated headspace SPME method developed in this work was used to investigate migration of BPA from polycarbonate bottles into water, and levels of BPA in water ranged from 1.7 to 4.1 microg/L. 相似文献

A new method is described for the qualitative and quantitative analysis of cyanide, a very short-acting and powerful toxic agent, in human whole blood. It involves the conversion of cyanide into hydrogen cyanide and its subsequent headspace solid-phase microextraction (HS-SPME) and detection by gas chromatography/mass spectrometry (GC/MS) in selected ion monitoring (SIM) mode. Optimizing the conditions for the GC/MS (type of column, injection conditions, temperature program) and SPME (choice of SPME fiber, effect of salts, adsorption and desorption times, adsorption temperature) led to the choice of a 75-microm carboxen/polydimethylsiloxane SPME fiber, with D3-acetonitrile as internal standard, and a capillary GC column with a polar stationary phase. Method validation was carried out in terms of linearity, precision and accuracy in both aqueous solutions and blood. The limit of detection (LOD) and limit of quantitation (LOQ) were determined only in aqueous solutions. The assay is linear over three orders of magnitude (water 0.01-10, blood 0.05-10 microg/mL); and the LOD and LOQ in water were 0.006 and 0.01 microg/mL, respectively. Good intra- and inter-assay precision was obtained, always <8%. The method is simple, fast and sensitive enough for the rapid diagnosis of cyanide intoxication in clinical and forensic toxicology. 相似文献

In this work, a novel technique of aqueous-phase derivatization followed by headspace solid-phase microextraction and gas chromatography-mass spectrometry was developed for the determination of organic acids in urine. The analytical procedure involves derivatization of organic acids to their ethyl esters with diethyl sulfate, headspace sampling, and GC/MS analysis. The proposed method was applied to the determination of methylmalonic acid and glutaric acid in urine. The experimental parameters and method validation were studied. Optimal conditions were obtained: PDMS fiber, extraction temperature 55 degrees C, extraction time 30 min, and 60 microL of diethyl sulfate as derivatization reagent with 2 mg of the ion pairing agent tetrabutylammonium hydrogensulfate. The method was linear over three orders of magnitude, and detection limits were 21 nM for methylmalonic acid and 34 nM for glutaric acid, respectively. Consequently, in-situ derivatization/HS-SPME/GC/MS is an alternative and powerful method for determination of organic acids as biomarkers in biological fluids. 相似文献

A simple, sensitive and rapid method for the determination of diisopropylfluorophosphate (DFP) in rat plasma and brain tissue using headspace solid-phase microextraction (HS-SPME) and gas chromatography/mass spectrometry (GC/MS) is presented. A 65 microm polydimethylsiloxane/divinylbenzene (PDMS/DVB) fiber was selected for sampling. The main parameters affecting the SPME process such as extraction and desorption temperature, extraction and desorption time, salt addition, and fiber preheating time were optimized in each matrix to enhance the extraction efficiency of the method. The lower limits of quantitation for DFP in plasma and brain tissue were 1 ng/mL and 3 ng/g, respectively. The method showed good linearity over the range from 1-100 ng/mL in plasma and 3-300 ng/g in brain tissue with correlation coefficient (R(2)) values higher than 0.995. The precision and accuracy for intra-day and inter-day were less than 10%. The relative recoveries in plasma and brain for DFP were greater than 50%. Stability tests including autosampler and freeze and thaw were also investigated. Determination of acetic acid in aqueous samples, by water-phase derivatisation, solid-phase microextraction and gas chromatography 总被引:3,自引:0,他引:3
The direct derivatisation of acetic acid with n-hexyl chloroformate and with benzyl bromide in water was evaluated. With n-hexyl chloroformate, acetic acid did not give the n-hexyl acetate derivative, but the reaction of acetic acid with benzyl bromide in aqueous solution resulted in the formation of benzyl acetate. The derivatisation of acetic acid with benzyl bromide and the headspace solid-phase microextraction (SPME) of benzyl acetate were optimised. Under optimum conditions, the limit of detection for acetic acid was 260 nM, and the relative standard deviation of the overall procedure at 1.10(-4) M acetic acid was 15.6% (n = 10). A linear response was obtained in the 1 x 10(-4) to 5 x 10(-6) M concentration range (R2 = 0.993, n = 6). Although Carbowax-divinylbenzene (CW-DVB)-coated fibres exhibited a higher extraction capacity for benzyl acetate, polyacrylate (PA) was selected, because its mechanical stability was better than that of CW-DVB fibres. Moreover, the relative standard deviation of the SPME was better with PA (1.5%, n = 10 at 1 x 10(-5) M) than with CW-DVB-coated fibres (8.0%, n = 10 at 1 x 10(-5) M). Thus, a new analytical method for the quantitative determination of micromolar concentrations of acetic acid in the aqueous phase was developed. This method is based on water-phase derivatisation with benzyl bromide, headspace SPME with PA fibres and GC-FID. It was observed experimentally that benzyl alcohol formed by hydrolysis of the reagent affected the fibre-gas phase partitioning of benzyl acetate. 相似文献

Angelica sinensis (danggui in Chinese) is a common traditional Chinese medicine (TCM), and its essential oil has been used for the treatment of many diseases such as hepatic fibrosis. Z-Ligustilide has been found to be an important active component in the TCM essential oil. In this work, for the first time, headspace single-drop microextraction (HS-SDME) followed by gas chromatography-mass spectrometry (GC-MS) was developed for the determination of Z-ligustilide in rabbit plasma after oral administration of essential oil of danggui. The extraction parameters of solvent selection, solvent volume, sample temperature, extraction time, stirring rate, and ion strength were systemically optimized. Furthermore, the method linearity, detection limit, and precision were also investigated. It was shown that the proposed method provided good linearity (0.02-20 microg/mL, R2 = 0.997), low detection limit (10 ng/mL), and good precision (RSD value less than 9%). Finally, HS-SDME followed by GC/MS was used for fast determination of Z-ligustilide in rabbit plasma at different time intervals after oral administration of danggui essential oil. The experimental results suggest that HS-SDME followed by GC/MS is a simple, sensitive, and low-cost method for the determination of Z-ligustilide in plasma, and a low-cost approach to pharmacokinetics studies of active components in TCMs. 相似文献

The Dufour's gland content of workers of two ant species of the genus Messor has been analyzed by solid-phase microextraction and gas chromatography with mass spectrometry (GC/MS). The structures of the compounds in the pheromonal mixtures have been determined. In both cases only one intact gland, inserted in a properly dimensioned capillary vial, is sufficient to produce a clean and fully interpretable GC/MS profile. It is worth noting that, for the first time in Messor ants, farnesol has been detected as a minor component of glandular secretion in Messor capitatus. 相似文献

An in situ derivatization solid-phase microextraction (SPME) method has been developed for the determination of the trichloroethylene (TCE) metabolites, trichloroacetic acid (TCA), dichloroacetic acid (DCA) and trichloroethanol (TCOH), in rat blood. The analytical procedure involves derivatization of TCA and DCA to their ethyl esters with acidic ethanol, headspace sampling using SPME, and gas chromatography/negative chemical ionization mass spectrometry (GC/NCI-MS) determination. Parameters affecting both derivatization efficiency and the headspace SPME procedure, such as the concentration of sulfuric acid, amount of ethanol, derivatization-extraction temperature and time, sample preheating time, agitator speed and desorption conditions, were optimized. The method showed good linearity over the range of 1-1000 ng/mL in rat blood for each metabolite with correlation coefficients (R(2)) higher than 0.99. The intra-day and inter-day precision and accuracy were less than 10%. The relative recoveries for all analytes were greater than 84%. Validation results demonstrated that selected ion monitoring of the (35)Cl and (37)Cl isotopes using NCI resulted in reliable and sensitive quantitation of all three TCE metabolites. This validated method was successfully applied to study the toxicokinetic behavior of TCE metabolites following a 1 mg/kg oral dose of TCE. 相似文献

The combined use of an automatic solid-phase extraction (SPE) apparatus with Oasis MCX cartridges and gas chromatography/mass spectrometry (GC/MS) to rapidly quantify quinine in biological samples with cyproheptadine as the internal standard is described. The selected ion monitoring mode, with the quantification ions m/z 136 and 287 (qualifier ions: m/z 261, 381 and 215, 96), allows the estimation of quinine levels, respectively. Separation was completed within 12.7 min. Excellent linearity was found up to 10 000 microg/L of plasma. The limit of detection (LOD) was 12.2 microg/L and the limit of quantification (LOQ) was 40.6 microg/L. High reproducibility (intra-assay CV range 1.9-4.3%, inter-assay CV range 2.2-11.3%) and accuracy values (intra-assay range 83.2-103.7%, inter-assay range 86.8-103.7%) were obtained. Recoveries were concentration-independent (97.2% and 89.8% for 4000 and 10 000 microg/L, respectively). This sensitive, simple assay for quinine in various matrices meets the current requirements for bioanalytical assays and may be used to monitor quinine levels in patients developing severe malaria with acute renal failure during hemofiltration. The optimal quinine dose in this situation is not really established and to improve clinical care, quinine concentrations might be explored to improve efficacy and minimise potential toxicity. 相似文献
