Stereoselective diazotization of (S)-2-amino-2-phenyl acetic acid (L-phenyl glycine) (4) with NaNO2 in 6% H2SO4 in a mixture of acetone and water gave optically pure (S)-2-hydroxy-2-phenyl acetic acid (L-mandelic acid) (5). Esterification, gave (S)-2-hydroxy-2-phenyl acetic acid esters (6). The latter was treated with chloroacetyl chloride in the presence of triethylamine (TEA) in dichloromethane (DCM) to yield (S)-2-chloroacetyloxy phenyl acetic acid ester (2). In another sequence, the reaction of 2-(chloromethyl)-3-arylquinazolin-4(3H)-one (9) treated with N-Boc piperazine, followed by deprotection of the Boc group, to obtain 3-aryl-2-((piperazin-1-yl)methyl) quinazolin-4(3H)-one (3). Reaction of 2 with 3 in the presence of K2CO3 and KI gave the title compound, 2-(2-(4-((3,4-dihydro-4-oxo-3-arylquinazolin-2-yl)methyl)piperazin-1-yl) acetoyloxy)-2-phenyl acetic acid esters (1). The structures of all the new compounds obtained in the present work are supported by spectral and analytical data.  相似文献

Abstract

Treatment of aminoalkanols 1 with phosphorous acid and formaldehyde in presence of conc. hydrochloric acid gave mixtures of [(2-hydroxy alkyl)imino] dimethylene diphosphonic acids 3 and 4-(phosphonomethyl)-2-hydroxy-2-oxo-1,4,2-oxazaphosphorinanes 2 from which 2 were isolated as crystalline solids. Similar treatment of 2-amino-2-methyl-1,3-propanediol 8 gave a complex mixture from which dimethylene diphosphonic acid of 5-amino-5-methyl-1,3-dioxane 9 was isolated. 2-Aminoethanethiol, when subjected to phosphonomethylation. gave an unexpected novel quarternary nitrogen product 11. N-Alkylaminoalkanols 4 on phosphonomethylation gave 3:1 mixtures of [N-alkyl-N-(2-hydroxyalkyl)amino] methane phosphonic acid 6 and N-alkyl-2-hydroxy-2-oxo-1,4,2-oxazaphosphorinane 5. Treatment of the crude mixtures of 5 and 6 with aqueous sodium hydroxide gave disodium salts of [N-alkyl-N-(2-hydroxyalkyl)amino] methanephosphonic acid 7. The ratio of the cyclic to the open chain structures obtained as well as the formation of any unexpected novel products is dependent on the structure of the aminoalkanol that is phosphonomethylated. The 1H, 13C and 31P spectra are reported for all new compounds.  相似文献

Abstract

of 1,4-dihydro-4-oxo-1,8-naphthyridine-3-carbohydrazide (4) with substituted phenyl isothiocyanates (5) in ethanol under reflux for 30 min gave thiosemicarbazide derivatives 6, which on cyclization in 2N NaOH under refluxing conditions for 1 h resulted in 3-(5-mercapto- 4-phenyl-4H-1,2,4-triazol-3-yl)-1,8-naphthyridin-4(1H)-one (7). Alternatively, 7 could also be prepared from following sequence of reactions, i.e., 4 → 8 → 7. In another sequence of reactions, condensation of 7 with chloroacetic acid in dimethylformamide (DMF) and K2CO3 as a mild base at 120 °C for 2 h resulted in 2-((5-(1,4-dihydro-4-oxo-1,8-naphthyridin-3-yl)-4-phenyl-4H-1,2,4-triazol-3-yl)sulfanyl)acetic acid (10). The latter, on reaction with substituted o-phenylenediamine (11) in 6N HCl for 4 h yielded 3-(5-((1H-benzo[d]imidazol-2-yl)methylthio)-4-phenyl-4H-1,2,4-triazol-3-yl)-1,8-naphthyridin-4(1H)-one (12). Alternatively, 12 could also be prepared by reacting 7 with 13 in DMF and K2CO3 as a mild base at 120 °C for 2 h, followed by oxidation with H2O2 resulting in the corresponding sulfonyl derivatives 14.  相似文献

An efficient domino protocol for the synthesis of 1,2,4-oxadiazole derivatives from readily available 1-(2-oxo-2-arylethyl)pyridin-1-iums and amidine hydrochlorides was developed. In this practical approach, N-acyl amidine precursors were formed firstly via a simple nucleophilic substitution, without the purification of N-acylamidine intermediates, and the following intramolecularly dehydrative cyclization gave 1,2,4-oxadiazole derivatives in the presence of I2/K2CO3/DMSO, which exhibited excellent functional group tolerance and proceeded under simple experimental conditions.  相似文献

It has been found that malonodinitrile and 2-(6-R1-oxo-3,4-dihydro-2-quinazolyl)acetonitrile in the presence of triethylamine undergo hetarylation by 5,6-dichloro-2,3-pyrazinedicarbonitrile at the active methylene group to give the triethylammonium salt of 2-(3-chloro-5,6-dicyano-2-pyrazinyl)malononitrile or 5-chloro-6-cyano(6-R1-4-oxo-1,2,3,4-tetrahydro-2-quinazolylidene)methyl-2,3-pyrazinedicarbonitriles. Reaction of these with primary amines leads to annelation of the pyrrole ring at the pyrazine [b] edge to give 6-amino-5-R-5H-pyrrolo[2,3-b]pyrazine-2,3,7-tricarbonitriles and 6-amino-5-R2-7-(6-R1-4-oxo-3,4-dihydro-2-quinazolyl)-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles respectively.  相似文献

3-Hydroxy-3-(2-oxoethyl)-6-phenyl-2,3-dihydropyridazin-4(1H)-ones were obtained by the reaction of methyl 3-oxo-5-phenylfuran-2(3H)-ylideneacetate or 2-[2-(4-chlorophenyl)-2-oxoethylidene]-5-phenylfuran-3(2H)-one with benzoic or p-nitrobenzoic isopropylidenehydrazides. Abstract

The ligand 2-(2-pyridyl)benzothiazole (L) can act both as an N-N and an N-S chelating donor. The latter coordination mode is expected to be preferred when it is involved in coordination to Ru(II) which is a soft acceptor centre However, in the title compound, chlorobis(acetonitrile)triphenylphosphino-2-(2-pyridyl)benzothiazole-N,N-ruthenium(II) chlride, [Ru(L)(PPh3(CH3CN)2Cl]Cl, the ligand acts in N,N-bidentate manner and the Ru(II) ion is found to be present in an N4PCl coordination environment. PPh3 and Cl are trans to each other and the two CH3CN ligands occupy cis positions facing the NN donor atoms of ligand L.  相似文献

The reactions of 2-cyanomethylbenzimidazole with polyfluorobenzoyl chlorides afforded 2-(1,3-dihydrobenzimidazol-2-ylidene)-3-oxo-3-polyfluorophenylpropionitriles. Refluxing of the latter compounds in acetonitrile in the presence of DBU or in dimethylformamide in the presence of amines gave rise to fluorine-containing derivatives of benzimidazo[1,2-a]quinolone.  相似文献

In the reaction with carbon disulfide, 2-amino-7-methyl-5-oxo-5H-1,3,4-thiadiazolo[3,2-a]pyrimidine (1) forms the alkaline salts of substituted dithiocarbamic or iminodithiocarbonic acids depending on the molar ratio between1, CS2, and the alkali. The alkylation of these salts leads to the esters ofN-(7-methyl-5-oxo-5H-1,3,4-thiadiazolo[3,2-a]pyrimidine-2-yl)dithiocarbamic (2) and diesters of (7-methyl-5-oxo-5H-1,3,4-thiadiazolo[3,2-a]pyrimidine-2-yl)iminodithiocarbonic acids (3). 为了寻找高效低毒的非甾体抗炎药物, 将5-氯-2-吲哚酮与氯甲酸苯酯经酯化及水解制得1-苯氧羰基-5-氯-2-吲哚酮, 在4-N,N-二甲氨基吡啶作用下与对甲磺酰基苯甲酰氯反应, 经酸化得1-苯氧羰基-3-[羟基-(p-甲磺酰基)苯甲撑基]-5-氯-2-吲哚酮(II1), 最后与相应的胺(氨)反应, 经盐酸中和制得未见文献报道的目标化合物II2II15, 其结构经IR, 1H NMR, MS和元素分析确证. 二甲苯致小鼠耳肿胀模型测试显示II10, II11, II15具有明显的抗炎活性; 角叉菜胶致大鼠足跖肿胀模型试验结果表明, II10, II11抗炎活性与双氯芬酸钠和替尼达普钠相当(P>0.05); 其中II11的胃肠道副作用显著小于双氯芬酸钠(P<0.05)和替尼达普钠(P<0.01).  相似文献

The interaction of 4-oxo-3,4-dihydroquinazolinyl- and benzimidazolylacetonitriles with 2,6-dihalobenzaldehydes leads to 3-(2,6-dihalophenyl)-2-(4-oxo-3,4-dihydro-2-quinazolinyl)acrylonitriles and 2-(1H-benzo[d]imidazol-2-yl)-3-(2,6-dihalophenyl)acrylonitriles respectively. As a result of intramolecular cyclization of these nitriles 4-halo-12-oxo-12H-quino[2,1-b]quinazoline-6-carbonitriles and 4-halobenzo[4,5]imidazo[1,2-a]quinoline-6-carbonitriles respectively are formed.  相似文献

N‐Alkylation of 10H‐9‐fluoropyrido[3,2‐b][1,4]‐benzothiazine 5a, 10H‐7‐fluoropyrido[3,2‐b][1,4]‐benzothiazine 5b, and 10H‐7‐chloropyrido[3,2‐b][1,4]‐benzothiazine 5c with different N‐(bromoalkyl)phthalimides using anhydrous K2CO3 and tetrabutylammonium bromide (TBAB) under dry conditions with microwave irradiation leads to the formation of 10‐(phthalimidoalkyl)‐halosubstitutedpyrido[3,2‐b][1,4]‐benzothiazine (6af) along with some unidentified product. Compound 5a is a new azaphenothiazine derivative and was obtained from hitherto unknown 2‐acetylamino‐3‐fluorophenyl‐3′‐nitro‐2′‐pyridylsulfide 4a via Smiles rearrangement. Compound 4a is required for the synthesis and has been prepared starting from 2‐amino‐3‐fluorobenzenethiol 1a in three steps.  相似文献

N,N′-1,2-phenylenebis(2-thienylideneimine) (PTI) can be obtained via the condensation of o-phenylenediamine and thiophene-2-carboxaldhyde in cyclohexane. It isomerized to its cyclic form 1-(2-thienylmethyl)-2-(2-thienyl)benzimidazole (TMTBI) readily in the presence of Fe2(CO)9. Coordination reactions of PTI with cuprous iodide and silver nitrate in acetonitrile resulted in the formation of [Cu(TMTBI)I]2 and [Ag(TMTBI)2]NO3, respectively, in which the PTI ligand had been isomerized to TMTBI. Both PTI, TMTBI, and complexes were characterized by NMR, IR, and Mass spectroscopies as well as elemental analysis. The crystal structures of TMTBI and [Cu(TMTBI)I]2 were determined by single-crystal X-ray diffraction. Crystallographic details for X-ray structures are as follows. TMTBI is crystallized in the monoclinic space group P21/n with a = 8.953(3), b = 9.150(1), c = 17.436(2) Å, β = 93.73(2)°, and Z = 4. The final R factor was 0.050 (Rw = 0.066) for 1337 observed reflections. Complex [Cu(TMTBI)I]2 is crystallized in the triclinic space group $ {\rm P}\bar 1 $ with a = 9.961 (2), b = 10.825(2), c = 9.184(2) Å, α = 112.98(2), β = 110.78(2), γ = 88.71 (2)°, and Z = 2. The final R factor was 0.056 (Rw = 0.057) for 2363 observed reflections.  相似文献

The corresponding 1-(2-quinoxalyl)-, 1-[3,5-di(trifluoromethyl)phenyl]-, and 1-ethoxycarbonyl-3-methyl-4-oxo-4,5,6,7-tetrahydroindazoles have been obtained from reactions of 2-acetyl-1,3-cyclohexanedione, its 5,5-dimethyl and 5-(2-furyl) derivatives, with 2-hydrazinoquinoxaline, 3,5-di(trifluoromethyl)phenylhydrazine, and ethoxycarbonylhydrazine. On interaction with ethoxycarbonylhydrazine the intermediate 2-[1-(-ethoxycarbonyl)hydrazino]ethylidene-1,3-cyclohexanediones were also isolated. From the potassium salt of 2-formyldimedone and 2-carboxyphenylhydrazine hydrochloride, 2-(2-carboxyphenyl)hydrazinomethylene-5,5-dimethyl-1,3-cyclohexanedione was obtained, the cyclization of which in ethanol in the presence of HCl led to 1-(2-carboxyphenyl)- and 1-(2-ethoxycarbonylphenyl)-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazole.  相似文献

The reactions of 3-[N-chloracetylamino-N-(4-nitrophenyl)]-2-formylindole (1a) with 2-(N, N-dialkylamino)ethylamines afford complex condensation products 7b,c consisting of two similar but not identical diazepinoindole fragments. For the reaction of compound 1a with 3-(N,N-diethylamino)propylamine, the process occurs in a different manner, and the predominant product is 4-ethylaminopropyl-1-(4-nitrophenyl)-2-oxo-1,2,3,4,5,6-hexahydro[1,4]diazepino[6,5-b]indole hydrocloride (14). Two routes of these unexpected transformations were proposed. The structures of the synthesized products were proved by the 1sH and 13C NMR, HMBC, and HSQC (direct proton-carbon correlation) spectra. The reactions of 2-amino-5-mercapto-(or 2,5-dimercapto)-1,3,4-thiadiazoles with 2-bromo-7-methyl-5-oxo-5H-1, 3,4-thiadiazolo[3, 2-a]pyrimidine to give the corresponding sulfides have been studied. The possibility of S-alkylation and addition of quinone at the free mercapto group in the 1,3,4-thiadiazole ring has been shown. The reactions at the amino group with benzoyl chloride and chloroformates have been investigated. The reaction of [2-(dimethylaminomethyl)]cymantrenyl]lithium with ytterbium diiodide affords [2-(dimethylaminomethyl)cymantrenyl]ytterbium iodide or bis[2-(dimethylaminomethyl)cymantrenyl]ytterbium depending on the ratio of the starting reactants. The reaction of ethyl 4-chloro-1-methyl-2-oxo-1,2-dihydroquinoline-3-carboxylate with p-toluene-sulfonylhydrazide at room temperature in the system DMSO/K2CO3 gives 5-methyl-2-(toluene-4-sulfonyl)-1,2-dihydro-5H-pyrazolo[4,3-c]quinoline-3,4-dione, alkylation of which using ethyl iodide gives the 1N-substituted derivative. 通过3-氯-6-溴甲基哒嗪与各种取代的苯酚在K2CO3/DMF体系中反应, 合成了一系列3-取代苯氧甲基-6-氯哒嗪化合物. 它们的结构均经1H NMR, IR和元素分析确证. 在浓度为100 μg•mL-1时, 初步的生物活性测试结果表明, 所合成的化合物对油菜和稗草均有一定的抑制作用. 并讨论了其结构与除草活性的关系.  相似文献
