A procedure is described for the rapid determination of benzoyl metronidazole in an oral suspension which is based on measurement of the change in absorbance at 276 nm during alkaline hydrolysis of the compound in 2 mol dm-3 NaOH. The change in absorbance follows a linear relationship with concentration in the range 3-18 micrograms ml-1. Results of the determination of benzoyl metronidazole in an oral suspension and of the recovery experiments performed on this formulation, and also on various individual excipients and other additives, confirmed the applicability of the proposed method to complex formulations.  相似文献

The use of spectrofluorimetry to determine the fluorescent derivative of the insecticide promecarb, following hydrolysis to the corresponding phenol in basic media and subsequent coupling with the labelling agent dansyl chloride, is described and discussed. A study of media of different basicity and of different temperatures for both reactions gave optimum conditions of 20 min for the hydrolysis reaction and 10 min for the labelling reaction at 55 degrees C in 0.05 mol dm-3 sodium hydrogen carbonate solution with a reagent to insecticide ratio of 12:1. The effect of the solvent on the formation of the dansyl derivative and on the extraction process was studied using nine and seven solvents, respectively. The use of a mixture of acetone and water (50 + 50, v/v) and an extraction into cyclohexane gave the best results. The minimum detectable concentration of promecarb in the experimental assays was 100 ng ml-1. The error and relative standard deviation at a concentration level of 0.6 micrograms ml-1 were 9.7 and 10.9%, respectively. Air samples containing promecarb at different concentration levels were analysed.  相似文献

The absorption spectra of the praseodymium complex with 1-cyclopropyl-6-fluoro-1,4-dihydro-7-(4-ethyl-1-piperazinyl)-4-oxo-3- quinoline carboxylic acid hydrochloride (NNFX) has been studied by normal and derivative spectrophotometry. The complex showed maximum absorption at 350 nm at pH 6.0. The stoichiometry of the Pr-NNFX complex was calculated by the molar ratio and continuous variation methods. The ratio of Pr to NNFX was 1:3. The absorption bands of the 4f electron transitions of the praseodymium complex with NNFX are enhanced markedly, especially the wavelength at 481 nm. Using the third-derivative spectrum, the calibration graph is linear over the range 2.5 x 10(-5)-3.5 x 10(-4) mol dm-3 for praseodymium. The detection limits (signal-to-noise ratio of 2) is 1.4 x 10(-6) mol dm-3. The relative standard deviation is 1.2% for 7.0 x 10(-5) mol dm-3 of praseodymium. A method for the direct determination of praseodymium in rare earth mixtures with good accuracy and selectivity is described.  相似文献

This paper describes a novel and sensitive pre-column derivatisation method for the detection and quantitation of beta-lactams and their biosynthetic precursors at trace levels in fermentation media. Filtered broths from fermentations of strains of Penicillium chrysogenum and Cephalosporium acremonium, after deproteination and centrifugation, were incubated with 9-fluorenylmethylchloroformate for 5 min at 20 degrees C in 0.2 M borate buffer at pH 7.7. Following two-fold pentane extraction of the reagent hydrolysis product, the aqueous layer was injected directly onto a C18 reversed-phase column, and products were detected spectrofluorimetrically with excitation and emission wavelengths of 260 and 313 nm, respectively. Detection limits of 0.01 and 0.05 micrograms ml-1 were achieved for both 6-aminopenicillanic acid (6-APA) and isopenicillin N in borate buffer and filtered fermentation broths, respectively, using a 10-microliter injection volume. A linear calibration for 6-APA in fermentation broth was obtained for a very wide concentration range (0.05-100 micrograms ml-1). Detection limits for solutions of cephalosporin C, deacetylcephalosporin C and deacetoxycephalosporin C in broth were all 0.25 micrograms ml-1. The detection limit for the beta-lactam precursor delta-(L-aminoadipyl)-L-alpha-cysteinyl-D-valine (ACV) dimer in borate buffer was 0.5 microgram ml-1. The cephalosporins and ACV dimer gave linear plots in the ranges 3-25 and 1-100 micrograms ml-1, respectively. Repeated analysis of 6-APA at a concentration of 10 micrograms ml-1 in filtered broth gave a mean peak area of 2.5.10(6) with a standard deviation of 2.6.10(5) using a 10-microliter injection volume. Ampicillin spiked into deproteinated blood serum gave a linear calibration in the concentration range 2-100 micrograms ml-1.  相似文献

For the first time, Crystal Violet (CV) was used to determine nucleic acid concentrations using the resonance light-scattering (RLS) technique. Based on the enhancement of the RLS of CV by nucleic acids, a new quantitative determination method for nucleic acids in aqueous solutions has been developed. At pH 5.03 and ionic strength 0.005 mol kg-1, the interaction of CV with nucleic acids results in three characteristic RLS peaks at 344.0, 483.0 and 666.0 nm. With 4.0 x 10(-5) mol l-1 of CV, linear relationships were found between the enhanced intensity of RLS at 666.0 nm and the concentration of nucleic acids in the range 0-2.5 micrograms ml-1 for herring sperm DNA, 0-4.0 micrograms ml-1 for calf thymus DNA and 0-4.5 micrograms ml-1 for yeast RNA. The limits of determination were 13.8 ng ml-1 for herring sperm DNA, 36.8 ng ml-1 for calf thymus DNA and 69.0 ng ml-1 for yeast RNA. The assay is convenient, rapid, inexpensive and simple.  相似文献

Two rapid assay procedures based on high-performance liquid chromatography (HPLC) and derivative ultraviolet (UV) spectrophotometry have been developed for the simultaneous determination of amoxycillin and dicloxacillin in two-component capsule formulations. The HPLC determination was carried out on a reversed-phase C8 column with use of a mobile phase consisting of methanol-0.02 mol dm-3 ammonium acetate (pH 5) (50 + 50) at a flow rate of 1.0 cm3 min-1, with UV detection at 230 nm. The fourth-derivative spectrophotometric procedure depends on the measurement of the derivative amplitudes, in 0.1 mol dm-3 NaOH, at 308.5 and 275 nm for amoxycillin and dicloxacillin, respectively. For both procedures, the calibration graphs were linear in the ranges 20-200 and 20-140 micrograms cm-3 for the HPLC and UV derivative methods, respectively, with an almost zero intercept and a correlation coefficient of 0.999. Commercial capsules and laboratory-prepared mixtures containing both penicillins in different proportions were assayed by the developed procedures. The results were of comparable accuracy as indicated by a statistical analysis of the data, using both t- and F-tests.  相似文献

The chelating poly(dithiocarbamate) resin with macroreticular support is shown to be effective for the preconcentration of cadmium and lead from non-saline waters. The retained ions can be eluted efficiently in 5 ml of 8 mol dm-3 nitric acid from 1-20 cm columns of resin. Poly(dithiocarbamate) resin is used for preconcentration in the determination of cadmium and lead by flame atomic absorption spectrometry. The common ions present in non-saline waters do not interfere. The sensitivity was 0.10 micrograms l-1 of Cd and 0.73 micrograms l-1 of Pb. The detection limits were 0.03 and 0.34 micrograms l-1 for Cd and Pb, respectively. The cadmium and lead concentration rages, determined in mineral and tap waters, were between 0.1 and 0.8 micrograms l-1 and between 0.7 and 16.9 micrograms l-1, respectively.  相似文献

A simple, sensitive and accurate colorimetric method is described for the quantitative determination of pindolol either in the pure form or in its tablets. The method is based on the oxidation of the indole moiety in pindolol with potassium persulphate or hydrogen peroxide-in acid medium-to give a highly coloured product that exhibits maximum absorption at 535 nm and 570 nm, respectively. Beer's law is obeyed over concentrations ranging from 0.07-0.35 mg/10 ml for potassium persulphate and 0.1-0.5 mg/10 ml for hydrogen peroxide with mean recoveries of 99.6+/-0.6 and 99.8+/-0.9%, respectively. The method is applied for the assay of pindolol tablets without any interference due to tablet fillers.  相似文献

以中压汞灯为光源,采用吸收光谱法,观察了UO2(NO3)2•6H2O在DMF(N,N-二甲基甲酰胺)溶剂中光化还原时的吸收光谱随照射时间、铀浓度及溶液酸度的变化.经短时间光照后,在UO2(NO3)2-DMF溶液中有铀(Ⅴ)形成,其吸收峰在755和635nm处.结果表明,它们分别是UO2+-DMF和UOOH2+-DMF的特征吸收,溶液中铀(Ⅴ)的岐化反应与H+浓度密切相关,UOOH2+是岐化反应的中间体.  相似文献

A new simple, accurate and precise spectrophotometric method for the determination of six phenothiazine drugs in pure form and in dosage forms is described. The method is based on the oxidation of the studied drugs by a known excess of Chloramine-T in hydrochloric acid medium and subsequent determination of the unreacted oxidant by reacting it with indigocarmine in the same acid medium. The reacted oxidant corresponds to the drug content. The colored species exhibits maximum absorption at 610 nm. The apparent molar absorptivity values and Sandell sensitivity values are in the range 1.53 x 10(4)-2.96 x 10(4) l mol-1 cm-1 and 13.75-37.15 ng cm-2, respectively. The method is highly sensitive and suitable for 1-15 micrograms ml-1 concentrations with the detection limits being in the range, 0.0651-0.1724 microgram ml-1. The method was successfully applied to the studied drugs in their dosage forms. The results are reproducible within +/- 1% and compare favorably with those obtained by the procedures of the British Pharmacopeia.  相似文献

Reversed-phase high-performance liquid chromatography with ultraviolet detection was optimized for the simultaneous separation and quantification of nine organic mercury compounds: methyl-, ethyl-, phenyl-, methoxyethyl-, ethoxyethyl-, benzoic and tolylmercury, mersalylic acid and nitromersol. The nine compounds were successfully separated on octadecylsilane columns (200 x 3 mm i.d.) by gradient elution with a methanol-water mixture ranging from 30 to 50% v/v. The detection limits for the various compounds are in the range 7.0-95.1 micrograms dm-3. For the extraction of five organomercurials from spiked soils, eight different extraction solutions were tested to differentiate between the total content and the available/soluble fraction of the analytes. Ammonium acetate solutions (1 mol dm-3) and water proved to be suitable agents for the estimation of the available and soluble fractions of methyl-, ethyl-, benzoic, methoxyethyl- and ethoxyethylmercury. For the determination of the total content of methyl- and benzoic mercury in soils, solutions of potassium iodide (1 mol dm-3)-ascorbic acid (0.1 mol dm-3) and oxalic acid (1 mol dm-3) provided recoveries in the ranges 53-81%. None of the solutions tested is suitable for the extraction of ethyl-, methoxyethyl- and ethoxyethylmercury.  相似文献

A simple, rapid and accurate spectrophotometric method has been developed for the determination of sorbic acid in various food samples based on the oxidation of sorbic acid by iron(III) at 100 degrees C to malonaldehyde, which then reacts with 2-thiobarbituric acid to form a reddish brown product. The optimum experimental conditions for colour development have been assessed. Absorbance measurements were made at 529 nm in the presence of 0.4% m/V citric acid. The calibration graph was linear for 0-6 micrograms ml-1 of sorbic acid with a slope of 0.131 A micrograms-1 ml. The recoveries of sorbic acid at concentrations of 164-557 micrograms ml-1 ranged from 96 to 103%. The relative standard deviations of ten replicate determinations of sorbic acid in a synthetic cream soda sample spiked with 573 micrograms ml-1 of sorbic acid and in an onion juice sample containing 82 micrograms ml-1 of sorbic acid were 1.6 and 1.9%, respectively. Interferences from several common food additives can be minimised by extracting sorbic acid with diethyl ether and then back-extracting the acid with sodium hydrogen carbonate. The method has been applied successfully to the determination of sorbic acid in a wide range of food samples including beverages, cake, cake mate, garlic bread sprinkle, onion juice, oyster flavoured sauce and grenadine syrup.  相似文献

采用循环伏安法和差分脉冲伏安法研究了咖啡酸苯乙酯(CAPE)在玻碳电极表面上的电化学行为。在0.05 mol.dm-3磷酸盐缓冲溶液(PBS,pH=7.6)中,CAPE在0.229 V和0.214 V处呈现一对明显的氧化还原峰。考察了溶液pH值和扫描速率等因素对CAPE电化学行为的影响,结果表明,电化学过程中CAPE的电子转移数为2,参与反应的质子数也为2,且为吸附控制过程。在0.05 mol.dm-3磷酸盐缓冲溶液(PBS,pH=7.6)中,CAPE的氧化峰电流与其浓度在0.34~3.40μmol.dm-3范围内成线性关系,其响应灵敏度为0.88μA/μmol.dm-3,检出限为60 nmol.dm-3(S/N=3)。本研究建立了一种简单、快速、可灵敏测定CAPE的方法。同时对CAPE在玻碳电极上的电化学行为进行了较为详细的研究。  相似文献

The construction and performance characteristics of an ion-selective electrode for fentanyl-drug cation, based on an ion-pair complex with tetrakis[3,5-bis-(trifluoromethyl)phenyl]borate anion in a PVC matrix were studied. A linear response for 1 x 10(-5) mol dm-3 to 1 x 10(-2) mol dm-3 drug with a slope of 57.9 +/- 0.5 mV/decade was established. The optimum pH range was 2 to 6. The lower detection limit was 6.29 x 10(-6) mol dm-3 fentanyl citrate (2.1165 micrograms cm-3 fentanyl). There were negligible interferences from a number of inorganic cations, structural analogues, and some common drug additives in injections. The electrode proposed has been successfully applied to determine fentanyl citrate in injections. The results correlated well with those obtained by the United States Pharmacopoeia standard procedure.  相似文献

Ethopropazine hydrochloride (EPH) has been proposed as a sensitive reagent for the spectrophotometric determination of tungsten(VI). The method is based on the formation of a chloroform-soluble yellow-colored ternary complex by the interaction of EPH and thiocyanate with tungsten(V). The complex exhibits the absorption maximum at 404 nm with Sandell's sensitivity value of 20.03 ng cm-2. The complex obeyed Beer's law in the concentration range of 1-15 micrograms ml-1 with an optimum concentration range of 2.3-12.9 micrograms ml-1. The effects of foreign ions in the determination of tungsten(VI) were investigated. The method has also been successfully applied to the analysis of alloy steels.  相似文献

A sensitive colorimetric method for the quantitative estimation of 11 phenothiazine drugs was developed. The method was based on the interaction of phenothiazine compounds with diphenylamine in presence of N-bromosuccinimide and sulfuric acid. Most of studied phenothiazines yielded bluish green products with two absorption maxima, one in the range of 392-396 nm with higher molar absorptivity and the other in the range of 770-780 nm with lower molar absorptivity. Phenothiazine base, mepazine HCl and pericyazine yielded blue products with only one maximum at 655, 775 and 778 nm, respectively. The color was stable for at least 1 h. The reproducibility and recovery of the method were excellent. The method was applied successfully to the determination of some commercially available phenothiazines in different dosage forms. Results were comparable to those obtained by official and reported methods.  相似文献

Simple, sensitive, and accurate visible spectrophotometric methods are described for the determination of paroxetine hydrochloride (PA) in tablets. Among them, the first 3 methods are based on the ion-pair complexes of PA formed with bromothymol blue (BTB), bromophenol blue (BPB), and bromocresol green (BCG) in aqueous acidic buffers. The complex species extracted into chloroform were quantitatively measured at 414 nm with BTB and BCG and at 412 nm with BPB. Beer's law was obeyed over the concentration ranges of 2-20, 2-16, and 2-16 microg/mL, respectively. The fourth method described is based on a coupling reaction between PA and 7-chloro-4-nitrobenzofurazon (NBD-Cl) in borate buffer, pH 8.5, in which a yellow reaction product that was measured at 478 nm was formed. The Beer's law range for this method was 2-10 microg/mL. The last method developed describes the interaction of PA base, as an n-electron donor, with 7,7,8,8-tetracyanoquinodimethane (TCNQ), as a pi-acceptor, in acetonitrile to give blue-colored TCNQ- radical anion with absorption maxima at 750 and 845 nm. Measured at 845 nm, the absorbance-concentration plot was rectilinear over the range of 1.5-15 microg/mL. The new methods developed were successfully applied to the determination of PA in tablets without any interference from common tablet excipients. The results of the methods were in good agreement with those obtained with an official liquid chromatographic method. This report describes first colorimetric methods for the determination of PA.  相似文献

A high-performance liquid chromatographic (HPLC) method for the determination of sulbactam in human and rat plasma and urine has been developed. Sulbactam was reacted with 1,2,4-triazole to yield a product having an ultraviolet absorption maximum at 326 nm. The product was separated using reversed-phase HPLC from the regular components of plasma and urine with an ion-pair buffer at 50 degrees C and detected at the ultraviolet maximum. The limits of accurate determination were 0.2 and 1.0 micrograms/ml in plasma and urine, respectively. The coefficients of variation of inter- and intra-assays in human plasma spiked at 4.0 micrograms/ml (n = 5) were 1.02 and 3.05%, respectively. Coexisting cefoperazone, penicillins, or the alkaline degradation product(s) of sulbactam did not interfere in the sulbactam assay. The pharmacokinetic behaviour of sulbactam and cefoperazone coadministered to rats was estimated by moment analysis.  相似文献

An indirect method for the determination of sulfate by atomic absorption spectrometry (AAS) is described. Sulfate forms a stable ion-association complex, [Cu(neocuproine)2]2+(SO4(2-)), in neutral medium, which can be extracted into isobutyl methyl ketone in the presence of a polar medium (methanol) with an efficiency higher than 98.0% and the extract can be analysed directly for copper (and hence indirectly for sulfate) by AAS. Measurement of the copper atomic absorption signal from the organic phase allows the indirect determination of 0.14-1.12 micrograms ml-1 of sulfate, giving a 450-fold increase in sensitivity over the conventional method of precipitation with barium. The limit of detection (3 sigma) is 3.2 ng ml-1 which is better than that of ion chromatography (0.15 micrograms ml-1). Indirect AAS allows the accurate assay of inorganic sulfate anion in biological fluids and tissues. The sulfate concentration determined by the proposed method in human blood serum (n = 6 in each instance) was 35.4-43.3 micrograms ml-1 in normal persons, 50.3-62.5 micrograms ml-1 in jaundice patients and 83.3-155.6 micrograms ml-1 in diabetic patients. A good correlation between measured sulfate and the sulfate added to blood serum was obtained.  相似文献

A new, simple, sensitive and selective catalytic method is developed for the determination of vanadium in natural and sea waters. The method is based on the catalytic effect of V(V) and/or V(IV) on the bromate oxidative-coupling reaction of metol with 2,3,4-trihydroxybenzoic acid (THBA). The reaction is followed spectrophotometrically by tracing the oxidation product at 380 and/or 570 nm after 5 min of mixing the reagents. The optimum reaction conditions are 6.4 x 10(-3) mol l-1 of metol, 2.0 x 10(-3) mol l-1 of THBA and 0.16 mol l-1 of bromate at 35 degrees C and in the presence of an activator-buffer solution of 1 x 10(-2) mol l-1 of tartrate (pH = 3.10). Following the recommended procedure, V(V) and/or V(IV) can be determined with linear calibration graphs up to 0.75 ng ml-1 and detection limits, based on the 3Sb criterion, of 0.008 and 0.018 ng ml-1 at 380 and 570 nm, respectively. The developed method was successfully applied, without any separation or preconcentration processes, to the determination of vanadium in natural and seawaters following the direct calibration and standard addition techniques, respectively.  相似文献
