3-Methylmercapto-5-oxo-6-vinyl- and 2-methyl-3-mercapto-5-oxo-6-vinyl-2,5-dihydro-1,2,4-triazines were readily converted into the corresponding thieno[2,3-e]-1,2,4-triazine in one step by the action of phosphorus pentasulfide in pyridine. 4-Methyl-3-mercapto-5-oxo-4,5-dihydro-1,2,4-triazine is only converted into the 5-thioxo analog and no thiophene ring product was obtained under the same conditions. Thieno[2,3-e]-1,2,4-triazines were also more efficiently obtained by the action of phosphorus petnasulfide in pyridine on the appropriate 6-acylmethyl-3-mercapto-5-oxo-2,5-dihydro-1,2,4-triazine.  相似文献

A number of 2-aryl-substituted pyrido[3,2-e] and [4,3-e][1,2,4]triazolo[1,5-c]pyrimidines and [1,2,4]triazolo[1,5-c]pteridines 11,12a,b,e , their corresponding 5-carbonyl derivatives 7,8a,b,e and some pyrimido[5,4-e][1,2,4]triazolo[1,5-c]pyrimidin-5-ones 7,8c,d have been synthesized, according to different pathways. The amination of 5-R- and 6-R-3-X-1,2,4-triazines (R = C6H5, t-C4H9, X = SCH3, SO2CH3, N+ (CH3)3, Cl) by potassium amide in liquid ammonia has been studied. In all reactions the formation of the corresponding 3-amino-1,2,4-triazines takes place; in some reactions by-products were found: from 5-phenyl- and 5-t-butyl-3-(methylthio)-1,2,4-triazine a ring contracted product i.e. 5-phenyl and 5-t-butyl-3-(methylthio)-1,2,4-triazole, from 6-phenyl-3-(methylthio)-1,2,4-triazine the dimer 3,3′-bis-(methylthio)-6,6′-bisphenyl-5,5′-bi-1,2,4-triazine and from 5-t-butyl-3-(trimethylammonio)-1,2,4-triazine chloride compound bis-(5-t-butyl-1,2,4-triazin-3-yl)- amine. Furthermore the conversion of 5-phenyl- and 5-t-butyl-1,2,4-triazin-3-one into the corresponding 3-amino compound by treatment with phenyl phosphorodiamidate (PPDA) was studied. A 15N study of these aminations showed that nearly all compounds undergo substitution according to both SN(AE) and SN(ANRORC) processes. The contribution of each of the competitive mechanisms to the amination is strongly influenced by the character of the leaving group.  相似文献

The cyclocondensation of 4,6-dichloro-2-methylthiopyridine-5-carbaldehyde and heterocyclic thiones such as 1,2,4-triazole-5-thione, imidazolidine-2-thione, imidazole-2-thione, and 4,5-diphenylimidazole-2-thione in dimethylformamide gave tricyclic heterocycles containing a pyrimido[5,4-e][1,3]thiazine fragment. Cyclization of azo compounds, synthesized from 5-diazoimidazoles and 2-naphthol or p-substituted phenols, into naphth[2,1-e]imidazo[5,1-c]-1,2,4-triazines and benz[e]imidazo[5,1-c]-1,2,4-triazines occurs only in the presence of p-toluenesulfonic acid. Condensation of 4-amino-2-methylthiothiazolium salts with 2,4,6-trichloro-5-formylpyrimidine or 3-R-2,4-dioxo-6-chloro-5-formyl-1,2,3,4-tetrahydropyrimidines yielded mesoionic derivatives of a new heterocyclic system — thiazolo[3,4-a]pyrimido[5,4-e]-pyrimidine — which can be used for the synthesis of polymethine dyes. The absorption maxima of these dyes are bathochromically shifted by 10–20 nm relative to the maxima of the thiazolopyrimidine analogs. Substituted 2,5-dihydro-3[3-methyI(or phenyl)-5-aminopyrazolyl]-1,2,4-triazin-5-ones reacted with orthoesters to yield exclusively 1,2,4-triazino[2,3-e]pyrazolo[1,5-a]-1,3,5-triazin-5-ones. The structure elucidation was supported by independent synthesis of the isomeric 1,2,3-triazino-[4,3-e]pyrazolo[1,5-α]-1,3,5-triazin-7-one as well as by spectroscopical methods.  相似文献

The Hofmann reaction on 6-methylpyridazine-3,4-dicarboxamide (1) gave a mixture of 3-methylpyrimido[4,5-c]pyridazine-5,7-dione (2), 3-methylpyrimido[5,4-c]pyridazine-6,8-dione (3) and an acid (4) of unknown structure. The Hofmann reaction on pyridazine-3,4-dicarboxamide (9) gave a mixture of pyrimido[4,5-c]pyridazine-5,7-dione ( 10 ) and an acid ( 11 ) of unknown structure. The reaction of 3-amino-6-methylpyridazine-4-carboxamide ( 18 ) with ethyl orthoformate gave 3-methylpyrimido[4,5-c]pyridazin-5-one ( 21 ). 4-Aminopyridazine-3-carboxamide ( 36 ) upon fusion with urea gave pyrimido[5,4-c]pyridazine-6,8-dione ( 37 ) while with ethyl orthoformate 36 gave pyrimido[5,4-c]pyridazin-8-one ( 38 ). Pyrimido[5,4-c]-pyridazine-8-thione ( 39 ) was obtained by the action of phosphorus pentasulfide on 38. 4-Amino-3-cyanopyridazine ( 16 ) when treated with formamide produced 8-aminopyrimido[5,4-c]-pyridazine ( 41 ). The synthesis of 4-aminopyridazine-3-carboxamide ( 36 ) and 4-amino-3-cyanopyridazine ( 16 ), both key intermediates in the synthesis of the novel pyrimido[5,4-c]pyridazine ring system was accomplished by the Reissert reaction of 4-aminopyridazine-2-oxides and subsequent conversion of the nitrile to the amide.  相似文献

A new synthesis of certain pyrimido[5,4-e]-as-triazine 4-oxides and their ring transformation to pyrrolo-[3,2-d]pyrimidines by the 1,3-dipolar cycloaddition reaction is described. Thus, reaction of 6-hydrazino-1,3-dimethyluracil ( 1 ) with triethyl orthoformate gave 6-ethoxymethylenehydrazino-1,3-dimethyluracil ( 2 ). Treatment of 2 with arylamines gave 6-arylaminomethylenehydrazino-1,3-dimethyluracils ( 3a-e ). Nitrosative cyclization of 3a-e with sodium nitrite afforded 3-arylaminofervenulin 4-oxides ( 6a-e ). Reaction of 6a-e with acetylenic esters yielded 7-alkoxycarbonyl-6-arylamino-1,3-dimethylpyrrolo[3,2-d]pyrimidine-2,4(1H,3H-diones ( 15a-e and 16 ).  相似文献
