Abstract

To obtain new polyfunctional inhibitors of radical-chain oxidative processes, C- and N-benzylation of a number of phosphorylacetic acid hydrazide derivatives is realized in their reactions with 3,5-di-tert-butyl-4-hydroxybenzyl acetate. N-benzylation of mixtures of Z and E isomers of hydrazones of phosphorylacetic acid derivatives proceeds stereoselectively with the formation of only EC=N isomers.  相似文献

Terbinafine, a widely used antifungal drug, is a challenging molecule for quantitative bioanalysis due to certain factors contributing assay variability. Despite previous attempts at human plasma determination of terbinafine, exhaustive stability of the drug or an internal standard was lacking. Internal standard stability with negligible variation throughout the analysis is an indicator of a reliable bioanalytical method as the majority of LC–MS/MS assays are based on analyte/IS response ratios for quantitation. A newly developed high‐throughput simple LC‐MS/MS method is described for human plasma determination of terbinafine using naftifine internal standard and eluting all compounds within 2 min. A solid‐phase extraction of terbinafine achieving mean recovery of 84.3% (CV < 4%) without compromising sensitivity (limit of quantitation 5.11 ng/mL) or linearity (5.11–3014.19 ng/mL) is delineated in this paper. A heated nebulizer in positive multiple reaction monitoring mode was employed with transitions m/z 292.2 →141.1 and 288.2 →117.0 for terbinafine and naftifine, respectively, resulting in excellent chromatographic separation on a Hypurity Advance (50 × 4.6 mm, 5 µm) column. The developed method was successfully applied to clinical samples and for the first time demonstrated marked improved extraction efficiency and reliable long‐term plasma stability results without any internal standard response variation during the entire course of study. We report two different synthetic approaches for the stereoselective synthesis of (2E,4E)-4-(4,4-dimethylpent-2-ynylidene)-N1,N5-dimethyl-N1,N5-bis(naphthalen-1-ylmethyl)pent-2-ene-1,5-diamine 1, an important impurity-reference standard for the chemical characterization of terbinafine. The diamine 1 was obtained in only six steps with a good overall yield starting from commercially available glutaconic acid.  相似文献

█tl="American"█The synthesis of the three N,N′-di(4-coumaroyl)tetramines, i.e., of (E,E)-N-{3-[(2-aminoethyl)amino]propyl}-3,3′-bis(4-hydroxyphenyl)-N,N′-(ethane-1,2-diyl)bis[prop-2-enamide] ( 1a ), (E,E)-N-{4-[(2-aminoethyl)amino]butyl}-3,3′-bis(4-hydroxyphenyl)-N,N′-(ethane-1,2-diyl)bis[prop-2-enamide] ( 1b ), and (E,E)-N-{6-[(2-aminoethyl)amino]hexyl}-3,3′-bis(4-hydroxyphenyl)-N,N′-(ethane-1,2-diyl)bis[prop-2-enamide] ( 1c ), is described. It proceeds through stepwise construction of the symmetric polyamine backbone including protection and deprotection steps of the amino functions. Their behavior on TLC in comparison with that of 1,4-di(4-coumaroyl)spermine (=(E,E)-N-{4-[(3-aminopropyl)amino]butyl}-3,3′-bis(4-hydroxyphenyl)-N,N′-(propane-1,3-diyl)bis[prop-2-enamide]; 2 ) is discussed.  相似文献

New optically active C 2-symmetric salen-type ligands were synthesized on the basis of (4S,5S)-4,5-bis(aminomethyl)-2,2-dimethyl-1,3-dioxolane. These ligands were used to obtain cationic (trifluoromethanesulfonate) and neutral (chloride) rhodium(I) complexes with [(4S,5S)-2,2-dimethyl-5-{[(E)-pyridin-2-ylmethylidene]aminomethyl}-1,3-dioxolan-4-yl]-N-[(E)-pyridin-2-ylmethylidene]methanamine and [2,2-dimethyl-5-{[(E)-quinolin-2-ylmethylidene]aminomethyl}-1,3-dioxolan-4-yl]-N-[(E)-quinolin-2-ylmethylidene] methanamine. The latter complex ensured preparation of (S)-2-phenylethanol with an optical yield of 34.8% by transfer hydrogenation of acetophenone.  相似文献

Carbon-13 NMR spectra of 24 new N-carboxyalkyl- and N-alkoxycarbonylalkylsubstituted derivatives of (E)-4-styrylpyridine and (E)-4-o(m- or p)hydroxystyrylpyridine bromides have been calculated and fully assigned. The Ai and AiK empirical parameters of the N-carboxyalkyl, N-alkoxycarbonylalkyl, N-carboxyalkyl-4-vinylpyridyl and N-alkoxycarbonylalkyl-4-vinylpyridyl groups were calculated for the prediction of the chemical shifts of substituted stytylpyridines.  相似文献

The N-phenylbenzo[g]indazole derivatives, 3-(4-chlorophenyl)-3,3a,4,5-tetrahydro-N-phenylbenzo[g]indazole-2-carbothioamide (4CLPBIC), 3-(4-bromophenyl)-3,3a,4,5-tetrahydro-N-phenylbenzo[g]indazole-2-carbothioamide (4BRPBIC), and 3-(3-bromophenyl)-3,3a,4,5-tetrahydro-N-phenylbenzo[g]indazole-2-carbothioamide (3BRPBIC), were synthesized by the one-pot green amalgamation of solvent-free granulating methodology procedure at room temperature. The synthesized crystals were characterized by single-crystal X-ray diffraction (SC-XRD), FT-IR, FT-Raman, NMR, and UV–Vis techniques. The molecular geometries from XRD experimental values of synthesized compounds 4CLPBIC, 4BRPBIC, and 3BRPBIC in the ground state are compared theoretically by applying the density functional theory (DFT), a method with the B3LYP/6-311G(d,p) basis set using Gaussian 09 software. The vibrational assignments of the synthesized compounds were studied based on potential energy distribution (PED) by the VEDA4 program. The scaled DFT/B3LYP/6-311G(d,p) results show the best agreement with the experimental values. Computational 1H and 13C NMR were acquired by utilizing gauge-independent atomic orbital (GIAO) procedure, and chemical shift results are in good agreement with the experimental values. A web-based theoretical investigation was performed to understand the drug-likeness and ADMET properties of the compounds. Molecular docking studies were carried out against bacterial cholesterol inhibitor block and inhibitor of lanosterol-14α-demethylase CYP51 used in the treatment of topical and systemic mycoses in fungal to understand the inhibitory activity of synthesized compounds. The synthesized molecules were also tested for antibacterial and antifungal activities.  相似文献

In this study, new benzoylthiourea derivatives, (E)-N-[(2-benzamidomethyleneamino)ethylcarbamothioyl]benzamide H3L′, N-(1-(3-benzoylthioureido)propan-2-ylcarbamothioyl)-benzamide H4L″, (E)-N-[4-(benzamidomethyleneamino)phenylcarbamothioyl]benzamide H3L‴, were synthesized. Structures of the compounds were identified by spectroscopic techniques. In addition, all synthesized compounds were evaluated for in vitro antibacterial and antifungal activity. Summary A new, rapid, and precise liquid chromatographic method has been developed for simultaneous identification and quantification of amphetamine,N-methyl-3,4-methylenedioxymetamphetamine,N-ethyl-3,4-methylenedioxymetamphetamine, andN-methyl-1-(3,4-methyl-enedioxyphenyl)-2-butanamine in the presence of other constituents. The compounds were separated on a monolithic column with a gradient prepared from acetonitrile and 20mm monobasic potassium buffer at a flow rate of 1.5 mL min−1. Quantitation was performed with metoclopramide as internal standard. Use of different flow rates was investigated and enabled reduction of the separation time from 11 to 3.5 min for seven substances. The method was then applied to ten seized tablets to identify and quantify their active ingredients.  相似文献

Oxidation of N-(N-arylsulfonylimidoyl)-4-aminophenols gave the corresponding N-[N-arylsulfonylimidoyl)-1,4-benzoquinonimines, derivatives of N-aroyl- and N-acetyl-1,4-benzoquinonimines. The structure of the products was studied by the X-ray diffraction method and 1H and 13C NMR spectroscopy. N-(N-Arylsulfonylimidoyl)-1,4-benzoquinonimines were found to undergo fast (on the NMR time scale) Z E isomerization about the CÍN bond in the quinonimine fragment. N-(N-Arylsulfonylacetimidoyl)-1,4-benzoquinonimines in solution give rise to dynamic Z E-isomerization with respect to the CÍN bond in the N-arylsulfonylacetimidoyl fragment.  相似文献

Condensation of 2-[(E)-pent-3-en-2-yl]-4-, 2-[(E)-pent-2-en-2-yl]-4-, or 2-(cyclopent-1-enyl)-6-methylaniline with 1-chloro-1-(N-methylimino)- or 1-chloro-1-(N-phenylimino)ethane affords the corresponding amidines. In order to characterize the structural elements that might play a role in non-covalent DNA binding by the antitumor antibiotic leinamycin, we have solved the crystal structures of the two leinamycin analogs, methyl (R)-5-{2-[1-(tert-butoxy­carbonyl­amino)­ethyl]­thia­zol-4-yl}penta-(E,E)-2,4-dienoate, C16H22N2O4S, (II), and 2-methyl-8-oxa-16-thia-3,17-di­aza­bicyclo­[12.2.1]­heptadeca-(Z,E)-1(17),10,12,14-tetraene-4,9-di­one, C14H16N2O3S, (III). The penta-2,4-dienone moiety in both of these analogs adopts a conformation close to planarity, with the thia­zole ring twisted out of the plane by 12.9 (2)° in (II) and by 21.4 (4)° in (III).  相似文献

The polar 1, 4-cycloaddition of phenylsulfene (generated in situ from phenylmethanesulfony] chloride and triethylamine) to N, N-disubstituted (E)-2-aminomethylenecyclohexanones I gave in general a mixture of N, N-disubstituted cis- and trans-4-amino-3, 4, 5, 6, 7, 8-hexahydro-3-phenyl-1, 2-benzoxathiin 2, 2-dioxides III and IV, which were separated by column chromatography and whose structural and conformational features were determined from uv, ir and nmr spectral data. Knoevenagel condensation of N-hetaryl-substituted cyanoacetamides with aldehydes gave the corresponding (E)-N-hetaryl-2-cyanoacrylamides which were converted into (E)-N-alkyl-N-hetaryl-2-cyanoacrylamides and 6-amino-2-oxo-4-phenyl-1-(pyridin-2-yl)-1,2-dihydropyridine-3,5-dicarbonitriles. In addition to two ceramides 1-O-β-D-glucopyranosyl-(2S,3R)-N-(2'-hydroxyhexadecanoyl)-octadeca-4E,8Esphingenine (2) and (2S,3S,4R)-N-(2'-hydroxytetracosanoyl)-octadecasphingenine (3), which separated for the first time, a new ceramide 1-O-β-D-glucopyranosyl-(2S,3R)-N-(2'-hydroxyhexacosanoyl)octadecasphingenine (1) was isolated from the traditional Chinese medicinal herb Isatis indigotica. Two new phenylpentadienamides isolated from the culture filtrate of Streptomyces sp. were assigned the structures 5-(4-aminophenyl)penta-2,4-dienamide ( 2 ) and N2-[5-(4-aminophenyl)penta-2,4-dienoyl]-L -glutamine ( 3 ). In addition, 5-(4-aminophenyl)penta-2,4-dienoic acid ( 1 ) has been isolated, and its spectroscopic characteristics are reported for the first time. Compounds 1–3 exist in both the (2E,4E)- and (2E,4Z)-configurations. 1-(6-Bromohexyloxy)-4-propargyloxybenzene upon quaternization with 3-dimethylamino-1-propanol and N,N-dimethyldodecylamine produced two new inhibitor molecules: N-[6-(4-Propargyloxyphenoxy)hexyl]-N,N-dimethyl-N-(3-hydroxypropyl)ammonium bromide (PHAB) and N-[6-(4-Propargyloxyphenoxy)hexyl]-N,N-dimethyl-N-dodecylammonium bromide (PDAB), respectively, in excellent yields. The inhibitor molecules were characterized by elemental analysis, Fourier transform infrared spectroscopy, 1H NMR, and 13C NMR spectroscopy. The inhibitors were evaluated for X-60 mild steel corrosion in 15 wt.% HCl using different electrochemical and gravimetric techniques. The potentiodynamic polarization confirms both the inhibitors as mixed-type corrosion inhibitors. A low concentration (15 ppm) of PDAB has demonstrated excellent corrosion inhibition efficiencies of 97%, 98%, and 86% at 25 °C, 50 °C, and 70 °C, respectively, for 24 h exposure time. SEM and EDX spectra reveal that the adsorptions of corrosion inhibitors on X-60 mild steel create a protective film that serves as a barrier to mitigate the corrosion process. The X-ray photoelectron spectroscopy confirmed the chemical interaction between the corrosion inhibitors and mild steel, which was predicted by the Langmuir adsorption model. Assembly of inhibitive motifs of the alkyne, π-electron-rich aromatic, quaternary ammonium and C12 alkyl chain hydrophobe in PDAB has augmented its inhibiting action.  相似文献
