Steroidal propargylic ester 6 and the propiolic ester 7 were prepared in a few steps from the deoxycholic acid. The Huisgen cycloaddition reaction of alkynes 6 and 7 as dipolarophiles with a 1,3-dipolar compound, the azide 5, afforded the dimeric compounds 8 and 9, respectively, in good yield. The dimers of deoxycholic acid derivatives linked by 1,4-disubstituted 1,2,3-triazole were characterized by spectroscopic methods.  相似文献

We have developed an efficient method for the synthesis of functionalized C-glycosyl 1,2,3-triazoles through a Cu(I)-promoted azide–alkyne 1,3-dipolar cycloaddition between a TMS-protected C-alkynyl-glycoside and organic azides. The reaction was accelerated by ultrasound irradiation and the addition of a base was not necessary to obtain the 1,2,3-triazole product. Moreover, further manipulation of the products led to chiral molecules with a C-glycoside linkage.  相似文献

Abstract

Novel helicenoid based 1,2,3-triazole tweezer 4 was synthesized using a multistep synthetic protocol with high yields from 2,7-Dihydroxynaphtlene 3 as a precursor. This helicenoid-based bis-1,2,3-triazole tweezer 4 selectively causes non-covalent interaction with iodide anion. The UV-vis absorption exhibited enhancement while fluorescence spectra exhibited significant quenching. 1H NMR titration showed shift of 1,2,3-triazole C-H signal with an increase in iodide concentration. Association constant of 3.818?×?104 M?1 was recorded for the host interaction with iodide ions. This value of association constant for iodide sensing using 1,2,3-triazole is the best reported so far for hosts with 1,2,3-triazole moiety and suggests that the helicenoid geometry is responsible for this remarkable behavior.  相似文献

A new 4-(benzotriazol-1-yl)-1,2,3-triazole structure was obtained by the diazotization reaction of either of 1-(2-aminophenyl)-4-carboxamido-5-amino-1H-1,2,3-triazole ( 1c ) or of the corresponding Dimroth isomer 1d . It underwent some common reactions to evaluate its chemical behaviour and structure. An analogous reaction sequence was carried out from the 2-nitro-4-methylphenyl azide, to assign the structure to the nitro derivatives prepared. The structure of the new compounds prepared was confirmed by chemical and spectroscopic methods.  相似文献

The interesting bioactivities of 2(5H)-furanone, 1,2,3-triazole, and amino acid derivatives have promoted their combination into one multifunctional molecule. The symmetrical bis-1,2,3-triazoles and mono-1,2,3-triazoles with one free azide group are synthesized respectively by controlling the molar ratio of reactants, N-[5-alkoxy-2(5H)-furanonyl] amino acid propargyl ester and 1,4-diazidobutane. The unsymmetrical bis-1,2,3-triazoles are afforded by the subsequent reaction of mono-1,2,3-triazoles with other terminal alkynes with good to excellent yields in a short time under the same mild “click” reaction conditions. The 32 new compounds obtained in the reactions are characterized by Fourier transform infrared, 1H NMR, 13C NMR, mass spectrometry, and elemental analysis. Because of the diversity of four or five basic units in molecule, this methodology provides easy access to different chiral 2(5H)-furanone compounds with polyheterocyclic structure, especially with unsymmetrical bis-1,2,3-triazole moiety. Importantly, a simple approach is provided for the synthesis of unsymmetrical bis-1,2,3-triazoles using common diazides.  相似文献

Reaction of 3-aryl-2-bromopropanoic acids esters, products of the Meerwein arylation, with sodium azide afforded alkyl 2-azido-3-arylpropanoates. The latter react with ethyl acetoacetate and phenylacetylene to form 1,2,3-triazole derivatives. A one-pot method for the synthesis of 3-aryl-2-(4-phenyl-1H-1,2,3-triazol-1-yl)-propanoic acid esters via a tricomponent reaction of alkyl 2-bromo-3-arylpropanoates, sodium azide, and phenylacetylene in the presence of CuI was developed.  相似文献

A novel method for the synthesis of cholic acid derivatives has been developed using ‘click chemistry’. Intermolecular 1,3-dipolar cycloaddition of the propargyl ester and azide groups of 3α-azidoacetoxy-7α,12α-diformyloxy-5β-cholan-24-oate gave a new dimer and oligomer linked by a 1,2,3-triazole ring. The structures of the products were confirmed by spectral (1H NMR, 13C NMR, and FT-IR) analysis, mass spectrometry and PM5 semiempirical methods. Estimation of the pharmacotherapeutic potential has been accomplished for the synthesized compounds on the basis of Prediction of Activity Spectra for Substances (PASSs).  相似文献

The synthesis of 1,2,3-triazoles was developed employing sequential copper azide-alkyne cycloaddition, tosylation, sodium azide, and copper azide-alkyne steps. This approach allowed the synthesis of two and three 1,2,3-triazole rings. A preliminary study to gain further insight into the reaction was performed using in situ ReactIR technology.  相似文献

This paper mainly studied Pd-catalyzed regioselective chlorination of disubstituted 1,2,3-triazoles directed by the 1,2,3-triazole ring. A series of regioselective chlorinated products were synthesized in 47–86% yields using Pd(OAc)2 as a catalyst and CuCl2 as a chlorinated reagent. This method provides a new mean for the synthesis of 1,2,3-triazole halides which combines the formation of C-X bond with C-H activation.  相似文献

This study develops a new efficient pathway for synthesis of 2,4-disubstituted 1,2,3-triazoles through regioselective direct arylation between 2-aryl-1,2,3-triazole N-oxides and Ar-B(OH)2. The reaction proceeds smoothly at room temperature and exhibits good yield and high C5 position selectivity. The possible pathway of oxidative Suzuki coupling is also discussed. This simple methodology can be used to construct 2,4-disubstituted 1,2,3-triazole moiety.  相似文献

1,2,3-Triazoles, significant five-membered ring N-heterocycles, are main structural moieties in well-designed materials, pharmaceutical agents, bioactive products, and synthetic intermediates. In the research of life sciences and pharmaceuticals, by seeing the spacious applications of 1,2,3-triazoles, the progress of metal-free method is exceedingly desirable to evade the heterocyclic product metal contamination. Moreover, on a larger scale, the toxicity and explosiveness of azides makes azides discommode and hard to hold, to synthesize 1,2,3-triazoles. The need to steer the development of the synthesis of 1,2,3-triazoles toward more maintainable synthesis is a pressing issue. There are rare methods to construct 1,2,3-triazoles under azide-free and metal-free environments. These rare methods are compiled in this review. The afford of the collection and compilation of azide-free and metal-free synthesis methodologies of 1,2,3-triazole in single podium is supportive and crucial for synthetic chemist to extend the diversity of the synthesis of 1,2,3-trizoles through green protocol.  相似文献

Montmorillonite KSF clay supported CuO nanoparticles efficiently catalyzes one-pot aromatic azidonation of aryl boronic acids followed by regioselective azide–alkyne 1,3-dipolar cycloaddition (CuAAC) reaction producing corresponding 1-aryl-1,2,3-triazole derivatives at room temperature in excellent yields without use of any additives. Investigations on mechanism of CuAAC revealed that sodium azide, which is used as azidonating reagent in one-pot protocol reduces Cu(II) to click-active Cu(I). The catalytic efficiency of another Cu(II) source CuSO4 in combination with NaN3 for this one-pot CuAAC protocol, further supported our mechanism. This is the first report for use of Cu(II)/NaN3 catalytic system for CuAAC protocol. The clay–Cu(II) catalyst being ligand-free, leaching-free, easy to synthesize from inexpensive commercially available precursors, recyclable, and environmentally friendly will be highly useful for economical synthesis of 1,4-disubstituted 1,2,3-triazoles.  相似文献

In this work, we report the synthesis of a novel carbohydrate-lithocholic acid conjugate linked through of 1,2,3-triazole rings and its derivatives in good to excellent yields. The conjugate was synthesized via copper-catalyzed azide?alkyne cycloaddition (CuAAC) from methyl 4,6-O-benzylidene-2,3-di-O-propargyl-α-d-glucopyranoside and methyl 3-azidolithocholate. The structures of all new compounds were properly characterized by infrared (IR), high-resolution mass spectroscopy (HRMS) and one- and two-dimensional nuclear magnetic resonance (NMR).  相似文献

The boom in growth of 1,4-disubstituted triazole products, in particular, since the early 2000’s, can be largely attributed to the birth of click chemistry and the discovery of the CuI-catalyzed azide–alkyne cycloaddition (CuAAC). Yet the synthesis of relatively simple, albeit important, 1-substituted-1,2,3-triazoles has been surprisingly more challenging. Reported here is a straightforward and scalable click-inspired protocol for the synthesis of 1-substituted-1,2,3-triazoles from organic azides and the bench stable acetylene surrogate ethenesulfonyl fluoride (ESF). The new transformation tolerates a wide selection of substrates and proceeds smoothly under metal-free conditions to give the products in excellent yield. Under controlled acidic conditions, the 1-substituted-1,2,3-triazole products undergo a Michael addition reaction with a second equivalent of ESF to give the unprecedented 1-substituted triazolium sulfonyl fluoride salts.  相似文献

An efficient two step synthetic methodology of new 5(4)-aryl-4(5)-[3(5)-(2-hydroxyphenyl)-1H-pyrazol-5(3)-yl]-1H-1,2,3-triazole dyads was established. The reaction of (E)-2-styryl-4H-chromen-4-ones, used as building blocks, with sodium azide, gave 4(5)-aryl-5(4)-(chromon-2-yl)-1H-1,2,3-triazoles bearing either electron-donating or electron-withdrawing substituents in their styryl moiety which upon reaction with hydrazine hydrate afforded the expected pyrazoles in moderate to very good yields.  相似文献

Per-O-acetylated β-d-glucopyranosyl azide was transformed into an intermediate iminophosphorane by PMe3 which was then acylated to N-acyl-β-d-glucopyranosylamines. The same azide and substituted acetylenes gave 1-(β-d-glucopyranosyl)-4-substituted-1,2,3-triazoles in Cu(I)-catalyzed azide–alkyne cycloadditions. Deprotection of these products by the Zemplén method furnished β-d-Glcp-NHCO-R derivatives as well as 1-(β-d-Glcp)-4-R-1,2,3-triazoles which were evaluated as inhibitors of rabbit muscle glycogen phosphorylase b. Pairs of amides versus triazoles with the same R group displayed similar inhibition constants. X-ray crystallographic studies on the enzyme–inhibitor complexes revealed high similarities in the binding of pairs with R = 2-naphthyl and hydroxymethyl, while for the R = Ph and 1-naphthyl compounds a different orientation of the aromatic part and changes in the conformation of the 280s loop were observed. By this study new examples of amide-1,2,3-triazole bioisosteric relationship have been provided.  相似文献

Reactions of 4,5-dibromo-1,2,3-triazole, 1H-1,2,3-benzotriazole, and 2-phenyl-2H-1,2,3-triazole-4-carbonyl chloride with trifluoromethanesulfonyl chloride and trifluoromethanesulfonic anhydride were studied. 4,5-Dibromo-1,2,3-triazole sodium salt reacted with CF3SO2Cl in tetrahydrofuran to give 4,5-dibromo-2-(2-tetrahydrofuryl)-2H-1,2,3-triazole rather than expected 4,5-dibromo-2-trifluoromethylsulfonyl-2H-1,2,3-triazole. The latter was synthesized by treatment of 4,5-dibromo-1,2,3-triazole sodium salt with trifluoromethanesulfonic anhydride. The reaction of benzotriazole with (CF3SO2)2O afforded 1-trifluoromethylsulfonyl-1H-1,2,3-benzotriazole and 1,2,3-benzotriazolium trifluoromethanesulfonate. 2-Phenyl-2H-1,2,3-triazole-4-carbonyl chloride reacted with trifluoromethanesulfonamide sodium salt in DMF, yielding N-(dimethylaminomethylene)trifluoromethanesulfonamide. Possible ways for formation of the unexpected products were proposed.  相似文献
