A smooth one-step synthesis of 2-(2-bromo-5-methoxyphenyl)-5-(3-arylidene)-1,3-thiazolo[3,2-b]-1,2,4-triazol-6-(5H)-ones (4a–n) is described. The newly prepared compounds are characterized by analytical and IR, 1 H NMR, 13 C NMR, and FABMS spectral analysis. A few compounds are screened for anticonvulsant activity. Compounds 4i and 4n exhibit promising anticonvulsant activity and are recommended for further studies.  相似文献

ABSTRACT

-The reaction of 3-(1-adamantyl)-4-methyl-1,2,4-triazole-5-thiol 1 with certain 2-aminoethyl chlorides in alkaline medium yielded a separable mixture of the S-(2-aminoethyl) derivatives 2 and the N-(2-aminoethyl) derivatives 3. Meanwhile, alkylation of 2-(1-adamantyl)-1,3,4-oxadiazole-5-thiol 4 with 2-aminoethyl chlorides under the same conditions yielded only the S-alkyl derivatives 5. Interaction of 4 with primary or secondary amines and formaldehyde solution yielded the corresponding N-aminomethyl derivatives in high yields.  相似文献

A short and efficient synthesis of 1-(3-tert-butyl-1-phenyl-1H-pyrazol-5-yl)-3-(5-(2-morpholinoethoxy)-2H-chromen-8-yl) urea derivatives (1ac), a novel type of p38 MAPK inhibitors, is described. The Claisen thermal rearrangement of arylpropargyl ethers was employd as a key step to synthesize the chromene core. The solvent effect on the ratio of the resultant two isomers of Claisen thermal rearrangement, namely 2-methylbenzofuran and 2H-chromen, was also investigated.  相似文献

N-(2-Azido-5-nitrobenzylidene)-3,5-di-tert-butyl-4-hydroxyaniline (3a) and N-(2-azido-5-nitrobenzilidene) aniline (3b), when heated in dimethylformamide yielded 2-(3,5-di-tert-butyl-4-hydroxyphenyl)-5-nitroindazole (4a) and 2-phenyl-5-nitroindazole (4b), respectively. The structure of4b was confirmed by X-ray analysis. A stable phenoxyl radical was shown to originate from the oxidation of4a with lead (IV) dioxide.Translated fromIzvestiya Akademii Nauk. Two new complexes, trans-[MnL2(NCS)2] (1) and trans-[CoL2(H2O)(EtOH)](ClO4)2?·?H2O (2) with asymmetrical triaryltriazole ligands [L?=?3-(p-chlorophenyl)-4-(p-methylphenyl)-5-(2-pyridyl)-1,2,4-triazole], have been synthesized and characterized by elemental analysis, FT-IR, ESI-MS, and single-crystal X-ray diffraction. In the complexes each L adopts a chelating bidentate mode via the nitrogen of pyridyl and triazole. Both complexes have a similar distorted octahedral core with two NCS? ions in the trans position in 1, while one H2O and one EtOH are present in the axial sites in 2.  相似文献

The quantification of 3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (Mutagen X or MX) in drinking water is difficult due to the low concentration of MX in drinking water, its high sensitivity to pH change, and matrix effects that interfere with the derivatization and analysis. Typically, the quantification of MX involves derivatization by methylation. We present a one-step derivatization procedure for MX using N-methyl-bis-trifluoroacetamide (MBTFA) and analysis by ion trap GC/MS/MS. The new method resulted in a significant reduction in analysis time, and improved detection limits. The abundant and selective ions in the mass spectrum of the trifluoroacylated MX (trifluoroacetic acid-4-chloro-3-dichloromethyl-5-oxo-2-hydro-furan-2-yl ester) allowed for a clear identification and quantification of the compound, with a method detection limit of 7.7 ng L−1, and a limit of quantitation of 24.4 ng L−1. The trifluoroacylated MX was shown to be stable for 30 days in an excess of the derivatization reagent. The new method was applied for the determination of MX in several drinking water samples, with a concentration range from not-detected to 517 ng L−1; these values are comparable to those obtained in previous studies. The development of this new simplified analytical method for MX is an important step forward in the field of disinfection by-product (DBP) research, particularly in light of the recent scientific recognition of halogenated furanones as emerging drinking water contaminants. Increased analytical ability may well be a decisive factor in the monitoring of these disinfection by-products.  相似文献

This article describes the reaction of 3,6-diphenyl-thieno[3,2-b]furan-2,5-dione 1 with different amino acids 2a–j in glacial acetic acid which afforded the 2-(2-(3-hydroxy-5-oxo-4-phenylthiophen-2(5H)-ylidene)-2-phenylacetamido)propanoic acid analogues 3a–j and also describes the reaction of 3,6-diphenyl-thieno[3,2-b]thiophene-2,5-dione 4 with different amino acids 5a–e in acidic medium to give 2-(2-(3-mercapto-5-oxo-4-phenylthiophen-2(5H)-ylidene)-2-phenylacetamido)propanoic acid analogues 6a–e. All the compounds have been screened for their anti-inflammatory activity against the carrageenan induced rat paw edema in albino rats. In the primary screening, some of the compounds exhibited appreciable activity.

Abstract

An expeditious, one-pot multicomponent reaction has been developed for the synthesis of (E)-2-(benzylideneamino)-5-mercapto-4H-1,2,4-triazol-3-yl)-2,3-dihydrophthalazine-1,4-dione derivatives. Condensation of 4-amino-5-hydrazino-4H-1,2,4-triazole-3-thiol with phthalic anhydride and aromatic aldehyde afforded the (E)-2-(benzylideneamino)-5-mercapto-4H-1,2,4-triazol-3-yl)-2,3-dihydrophthalazine-1,4-diones in acetic acid medium with excellent yields. All the synthesized compounds were characterized by their analytical and spectral data.  相似文献

4H-Chromenyl-tetrahydro-2H-pyrancarboxylates 3a–k have been conveniently prepared by the reaction of 3-formylchromones 1a–k and ethyl 4,4,4-trifluoro-3-oxobutanoate 2a with good yields. Thus obtained 4H-chromenyl-tetrahydro-2H-pyrancarboxylates 3a–k were reacted with amines 4a–e to provide series of 5-(2-hydroxybenzoyl)-2-(trifluoromethyl)-1,2-dihydropyridine-3-carboxylates 5a–o. The reaction proceeded via Michael addition (C-N bond formation) and followed by cleavage of chromone and pyran rings (C-O bond cleavage) in one pot.  相似文献

Treatment of 2-aminopyridine ( 1 ) with chloroacetyl chloride in dry benzene gave 2-chloro-N-(pyridin-2-yl)acetamide ( 3 ), which on further reaction with potassium thiocyanate gave 2-imino-3-(pyridin-2-yl)thiazolidin-4-one ( 4 ) as an intermediate compound for the synthesis of pyridin-2-yl substituted 2-imino-thiazolidine-4-one derivatives. Cyclocondensation reaction of ( 4 ) with a series of aromatic aldehydes gave 5-arylidene derivatives of pyridin-2-yl substituted 2-imino-thiazolidine-4-ones 5a–j . 1 H and 13C NMR spectroscopy, as well as elemental analyses, were used for the identification of these new compounds.  相似文献

Summary 3-Aryl-, 3-benzyl-, and 3H-5-(2-aryl-2-oxoethyl)-2-thioxo-4-oxo-1,3-thiazolidines3a–h react with bromine in acetic acid solution to give mixtures of the respective 5-aroylmethylene (E) and (Z) diastereomeric derivatives5 and6. They contain more than 85% of the (E)-diastereomers along with some pure isomers. The intermediacy of the 5-bromo derivatives4 is proven and a plausible route of the reaction is presented. Structures of compounds3–6 are evidenced by analytical and spectral data.

Zusammenfassung 3-Aryl-, 3-Benzyl- und 3H-5-(2-Aryl-2-oxoethyl)-2-thioxo-4-oxo-1,3-thiazolidine3a–h reagieren mit Brom in essigsaurer Lösung zu Gemischen der entsprechenden diastereomeren 5-Arylmethylen-Derivate ((E) und (Z))5 und6. Sie enthalten mehr als 85% des (E)-Diastereomeren. Die intermediäre Natur der 5-Brom-Derivate4 wird bewiesen; ein Reaktionsweg wird vorgeschlagen. Die Strukturen der Verbindungen3–6 werden durch analytische und spektroskopische Daten abgesichert.

We have synthesized a new series of polyphenylene compounds with a pyridine ring, 2-(4-n-alkoxyphenyl)-5-methylpyridines (CH3C5H3NC6H4OC i H2i1) (1a-i ; the carbon number, i, of the alkoxy group is 1-9), and studied their phase transitions and mesogenicity using DSC and polarizing microscopy. In the homologous series of 1a-i , no mesomorphic phase appeared for i=1-5 and only a monotropic nematic phase was observed for i=6-9. The mesomorphic potential is relatively weaker for the 1a-i than for the 2a-i and 3a-i homologues, which are pyridine-containing three- and four-ring similar systems. This lowering of the mesogenicity may be due to the lack of a phenyl ring between the pyridine ring and the methyl group in the 1a-i homologues.  相似文献

(Z)- and (E)-3-(2-chloropyridin-5-ylmethyl)oximino-(22E,24R)-ergosta-4,7,22-trienes (56) were synthesized by chemical transformation of ergosterol. Several oxidative transformations of them were studied. It was found that oxidation of these compounds by chromium(VI) oxide formed the corresponding O-substituted 3-ketoximes of the mycosteroid (22E,24R)-ergosta-4,7,22-trien-3,6-dione (7) and (8), which contained α-chloropyridine fragments characteristic of biologically active neonicotinoids. It was shown that oxidation of 5 and 6 by selenium dioxide occurred with formation of the corresponding 9α-hydroxy derivatives 9 and 10.  相似文献

利用三苯基氯化锡和现场生成的5-氯水杨醛缩-α-氨基酸钾反应合成了6种三苯基锡N-(5-氯亚水杨基)-α-氨基酸酯16, 通过元素分析、IR和NMR对其结构进行了表征. 利用X射线单晶衍射测定了三苯基锡N-(5-氯亚水杨基)缬氨酸酯(3)和三苯基锡N-(5-氯亚水杨基)异亮氨酸酯(5)的晶体结构. 结果表明锡原子是五配位的, 通过单齿羧基氧原子和相邻分子的酚氧原子对锡的配位形成了具有反式-O2SnC3三角双锥结构的一维链状聚合物.  相似文献

By cycloaddition of arylazides to acetylacetone are obtained derivatives of 1,2,3-triazole. In the reaction of 1-[5-methyl-1-(R-phenyl)-1H-1,2,3-triazol-4-yl] ethanones (IIa–IIe) and 1-[4-(4-R-5-methyl-1H-1,2,3-triazol-1-yl)phenyl] ethanones (VIIa-VIIe) with isatin are obtained 2-[1-(R-phenyl)-5-methyl-1H-1,2,3-triazol-4-yl]-4-quinolinecarboxylic acids (IIIa–IIIe) and 2-[4-(4-R-5-methyl-1H-1,2,3-triazol-1-yl)phenyl] -4-quinolinecarboxylic acids (IXa, IXb), respectively. We found that 1-[5-methyl-1-(R-phenyl)-1H-1,2,3-triazol-4-yl] ethanones (IIa–IIe) readily transform into [5-methyl-1-(R-phenyl)-1H-1,2,3-triazol-4-yl] acetic acids (IVa–IVc) by the method of Wilgerodt-Kindler. The (5-methyl-1-phenyl-1H-1,2,3-triazol-4-yl)acetic acid reacts with 5-phenyl-4-amino-4H-1,2,4-triazol-3-thiol affording 6-[(5-methyl-1-phenyl-1H-1,2,3-triazol-4-yl) methyl]-3-phenyl[1,2,4] triazolo[3,4-b] [1,3,4] thiadiazole (VI). 5-氨基-N,N'-双(2,3-二羟丙基)-2,4,6-三碘-1,3-苯二甲酰胺(2)在N,N-二甲基乙酰胺中可直接与乙酰氧基乙酰氯反应, 产物再经碱性水解得5-羟乙酰氨基-N,N'-双(2,3-二羟丙基)-2,4,6-三碘-1,3-苯二甲酰胺(3), 后者再与氯乙醇反应生成5-(N-2-羟乙基)羟乙酰胺基-N,N'-双(2,3-二羟丙基)-2,4,6-三碘-1,3-苯二甲酰胺(1), 经乙二醇甲醚/正丁醇重结晶, 纯度高于99% (HPLC), 反应总收率由39.3% (文献值)提高到55.1%.  相似文献
