Racemic (E)-α-bisabolene (E)(1) was synthetized starting from 4-methyl-3-cyclohexenecarboxylic acid (3) by a reaction sequence involving the Pd(0)-catalyzed cross-coupling reaction between the (E)-2-methyl-1-alkenyltrimethylstannane 8 and 3-methyl-2-buten-1-yl acetate (9). Three different procedures, in which a common precursor was used as key intermediate, were tested for the synthesis of racemic (Z)-α-bisabolene (Z)(1). The best one, which involved the reaction between bromide 18 and lithium dialkenylcuprate 19, afforded a mixture of (Z)- and (E)-1 in a 93 : 7 molar ratio, respectively. Finally, racemic β-bisabolene (2) was synthetized by a simple reaction sequence involving the Zr-promoted methylenation of ketone 22 prepared from 3.  相似文献

Summary A synthesis of (+)-cis-disparlure is described starting from racemic 2-hydroxy-dodecannitrile (2), which is transformed into the O-protected enantiomerically pure cyanohydrine4. Grignardreaction followed by reduction with BH3 · (CH3)2S yields the correspondingthreo-configurated secondary alcohol10. After tosylation and cleavage of theMBE-protective-group (+)-cis-disparlure (14) is obtained by treatment with KOH. Mating disruption field tests exhibited a significantly increased effectiveness of (+)-cis-disparlure as compared to the racemic product.

 The sterospecificities of hydantoinases and N-carbamoyl amino acid amidohydrolases (N-carbamoylases) from different microbial sources were investigated for the stereoselective syntheses of the unnatural silicon-containing amino acids D- and L-3-trimethylsilyl-alanine (3) from the respective racemic hydantoin D,L-1. In a preparative biotransformation, whole resting cells of Agrobacterium sp. IP I 671, immobilized in a Ca-alginate matrix, were used for the synthesis of amino acid D-3 in 88% yield and 95% enantiomeric excess. Since the purified D-N-carbamoylase from Agrobacterium sp. IP I 671 was shown to be 100%D-selective, the enantiomeric purity of 95% of D-3 arising from the transformation with the immobilized cells must be explained by the participation of a further, L-selective N-carbamoylase or, which is more likely, by racemization of the final hydrolysis product by the action of an amino acid racemase. Isolated hydantoinases from Bacillus thermoglucosidasius, Thermus sp., Arthrobacter aurescens DSM 3745, and Arthrobacter crystallopoietes DSM 20117 turned out to be stereospecific for the conversion of the D-form of hydantoin D,L-1. The enantiomerically pure L-form of 3 was also prepared. It was synthesized from racemic N-carbamoyl amino acid D,L-2 by enantiomer-specific hydrolysis of the L-form in presence of L-N-carbamoylase from Arthrobacter aurescens DSM 3747.  相似文献

Partial reduction of racemic mixture of S-alkyl-S-aryl-p-tosyl sulfilimines with L(–)cysteine affords optically active recovered sulfilimine. In the case of S-methyl-S-p-tolyl-N-p-tosylsuifilimine the absolute configuration and the optical yield were determined.  相似文献

Based on the results obtained in the racemic series (part I), (—)-17β-hydroxy-des-A-androst-9-en-5-one has been synthesized, starting with (S)-(—)-5-heptanolide. The key step, viz. the condensation of (S)-(—)-7-hydroxy-1-nonen-3-one (or its amine adduct) with 2-methyl-cyclopentane-1, 3-dione involves an asymmetric induction. Model experiments with (R)-(+)-5-decanolide leading to the enantiomeric homolog of the BCD-tricyclic compound are also described.  相似文献

(S)‐(+)‐3‐Octanol (S)‐1 was prepared in high enantiomeric excess through catalyzed acetylation of racemic alcohol 1 by using lipase from Candida antarctica (Chirazyme L‐2) in the presence of vinyl acetate in toluene at 30°C. The pure (S)‐1 was obtained in 73% isolated yield with 62% conversion. Moreover, acetate (R)‐2 was converted to (S)‐1 via mesylation and followed by hydrolysis using sodium bicarbonate solution in 44% yield.  相似文献

The molecules of racemic 3‐benzoylmethyl‐3‐hydroxyindolin‐2‐one, C16H13NO3, (I), are linked by a combination of N—H...O and O—H...O hydrogen bonds into a chain of centrosymmetric edge‐fused R22(10) and R44(12) rings. Five monosubstituted analogues of (I), namely racemic 3‐hydroxy‐3‐[(4‐methylbenzoyl)methyl]indolin‐2‐one, C17H15NO3, (II), racemic 3‐[(4‐fluorobenzoyl)methyl]‐3‐hydroxyindolin‐2‐one, C16H12FNO3, (III), racemic 3‐[(4‐chlorobenzoyl)methyl]‐3‐hydroxyindolin‐2‐one, C16H12ClNO3, (IV), racemic 3‐[(4‐bromobenzoyl)methyl]‐3‐hydroxyindolin‐2‐one, C16H12BrNO3, (V), and racemic 3‐hydroxy‐3‐[(4‐nitrobenzoyl)methyl]indolin‐2‐one, C16H12N2O5, (VI), are isomorphous in space group P. In each of compounds (II)–(VI), a combination of N—H...O and O—H...O hydrogen bonds generates a chain of centrosymmetric edge‐fused R22(8) and R22(10) rings, and these chains are linked into sheets by an aromatic π–π stacking interaction. No two of the structures of (II)–(VI) exhibit the same combination of weak hydrogen bonds of C—H...O and C—H...π(arene) types. The molecules of racemic 3‐hydroxy‐3‐(2‐thienylcarbonylmethyl)indolin‐2‐one, C14H11NO3S, (VII), form hydrogen‐bonded chains very similar to those in (II)–(VI), but here the sheet formation depends upon a weak π–π stacking interaction between thienyl rings. Comparisons are drawn between the crystal structures of compounds (I)–(VII) and those of some recently reported analogues having no aromatic group in the side chain.  相似文献

A novel anthracene-derived α-aminophosphonate has been synthesized through a classical addition reaction of dimethyl phosphite to a Schiff base and via a microwave assisted Kabachnik–Fields reaction. The compound has been characterized by elemental analysis, TLC, IR, NMR, and fluorescent spectra. The X-ray analysis showed that the compound crystallizes in the orthorhombic space group Pbca (N° 61) with one molecule per asymmetric unit. The enantioseparation of two racemic aminophosphonates (5 and 6) into enantiomers (5a, 5b and 6a, 6b, respectively) has been performed. Data about genotoxicity and antiproliferative activity in vivo of the racemic compounds and their enantiomeric forms is also presented. The studied racemic α-aminophosphonates 5 and 6 and their enantiomers have weak genotoxic effect. Both pairs of enantiomers did not show a significant inhibition of cell dividing processes in the bone marrow cells compared to 5 and 6. The hematopoietic function of the bone marrow will not be discontinued after exposure of the tested compounds.  相似文献

Abstract

The absolute configuration of 4-α-D-glucopyranosyl-myo-inositol, one of the enzymatic transglycosylation products from cyclodextrin to myo-inositol, was confirmed by chemical synthesis. The glycosylation was shown to proceed in a highly diastereoselective manner by HPLC analysis of the chemically synthesized racemic glycoside.  相似文献

The enzymatic resolutions of two racemic ethyl hydroxyalkane(P-phenyl)phosphinates were performed by both esterification and hydrolysis approaches. The first reaction was performed in anhydrous diisopropyl ether with triethylamine or pyridine as additives by using lipases from three different sources (Candida cylindracea, Aspergillus niger, and Mucor javanicus). The increase in enantioselectivity was observed when NEt3 was applied. The second reaction—lipase-catalysed hydrolysis of ethyl butyryloxyalkane(P-phenyl)phosphinates—was carried out by Candida cylindracea lipase in diisopropyl ether saturated with water or in aqueous solutions containing MgCl2, LiCl, or Triton X-100. The usefulness of biphasic systems consisting of diisopropyl ether and water or aqueous solution of MgCl2, LiCl, or Triton X-100 also were tested. The use of biphasic system in the presence of Triton X-100 resulted in the higher conversion of the substrates.  相似文献

Oxidation of three di-O-isopropylidene derivatives 2a4a, newly derived from (+)-epi-quercitol (1), with acetic anhydride in DMSO gave the corresponding ketones 57, which underwent aldol-type condensation with nitromethane under basic conditions to give selectively the protected derivatives 8a10a of C-nitromethyl-1,2,3,4,5-cyclohexanepentols, respectively. On treatment with diazomethane in DMSO, the ketones 6 and 7 gave single spiro epoxides 11 and 12, the structures of which were confirmed by converting them into new C-(azidomethyl)cyclohexanepentols 16 and 17. The nitro compounds were hydrogenated in the presence of Raney nickel to give the amines isolated as the N-acetyl derivatives. Deprotection gave three new 1- and 3-C-aminomethyldeoxyinositols 15c17c. The aminocyclitols obtained and their N-acetyl derivatives were assayed for inhibitory activity against examples of glycosidases.  相似文献

Abstract

Enantiomers have generally been reported mostly for racemic mixtures with a 1:1 ratio, as in that case there were weak Cotton effects in the ECD spectrum and negligible optical rotations. A furofuran lignan (sesamin), with a remarkable rotation and significant Cotton effects, was isolated from Juglans mandshurica Maxim. Subsequently, sesamin was resolved by chiral HPLC to afford a pair of enantiomers, (+)-sesamin (a) and (?)-sesamin (b), in a ratio of approximately 1:3. Their absolute configurations were determined by computational analysis of their electronic circular dichroism (ECD) spectrum. In addition, the pair of enantiomers were evaluated for the inhibition of self-induced Aβ aggregation. Interestingly, (+)-sesamin (a) (67.7%) and (?)-sesamin (b) (80.6%) exhibited different degrees of anti-Aβ aggregation activity. The different inhibition profiles were further explained by molecular dynamics and docking simulation study.  相似文献

The asymmetric unit of the racemic form of the title compound, C12H15NOS, contains four crystallographically independent molecules. The olefinic bond connecting the 2‐thienyl and 1‐azabicyclo[2.2.2]octan‐3‐ol moieties has Z geometry. Strong hydrogen bonding occurs in a directed co‐operative O—H...O—H...O—H...O—H R44(8) pattern that influences the conformation of the molecules. Co‐operative C—H...π interactions between thienyl rings are also present. The average dihedral angle between adjacent thienyl rings is 87.09 (4)°.  相似文献

Four previously undescribed isochromanes were isolated from the fermentation broth of an endophytic fungus Aspergillus fumigatus, which was obtained from the fruiting body of Cordyceps sinensis. Their structures were elucidated through extensive spectroscopic analyses. One racemic isochromane was further purified by chiral HPLC to yield a pair of enantiomers and their absolute configurations were determined by quantum chemical ECD calculations. These isolated compounds were evaluated for cytotoxicity against two cell lines (MV4-11 and MDA-ME-231) and the result showed that compounds 1a and 2 exhibited moderate growth inhibition against MV4-11 cell line.  相似文献

Summary The reaction of 3-iodo-4-methoxy-2(1H)-quinolinone (1) and 3-iodo-4,6,8-trimethoxy-2(1H)-quinolinone (2) with 2-methyl-3-butyn-2-ol under modified Heck-conditions gave the 2-substituted derivatives 2-(1-hydroxy-1-methylethyl)-4-methoxyfuro[2,3-b]-quinoline (3) and 2-(1-hydroxy-1-methylethyl-4,6,8-trimethoxyfuro[2,3-b]-quinoline (4). By a subsequent hydrogenation-reaction with a homogeneous catalyst (PtO2/Rh2O3), the furoquinoline-derivatives yielded the dihydrofuro-[2,3-b]quinolines, identified as 2-(1-hydroxy-1-methylethyl-4-methoxy-2,3-dihydrofuro[2,3-b]quinoline (5) (racemic platydesmine) and 2-(1-hydroxy-1-methylethyl)-4,6,8-trimethoxy-2,3-dihydrofuro-[2,3-b]quinoline (6) (racemic precursor of O4-methylptelefolonium salt).

Abstract

Herein, we report the synthesis of thioridazine-VLA-4 hybrid epimers. These hybrid molecules were obtained by means of a click reaction involving N-propargyl northioridazine enantiomers and an azide containing VLA-4 antagonist. A synthesis of northioridazine enantiomers from racemic thioridazine was developed and the absolute stereochemistry was confirmed by X-ray crystallography. Access to N-substituted thioridazine analogs may reveal that this phenothiazine core has therapeutic potential in other disease avenues.  相似文献

A series of chiral 1,3‐dioxolanes, 3 – 12 , with >99% ee values, have been synthesized. This is the first study of chiral ketalization reaction starting from ketones with aryl, monosubstituted aryl, and long alkyl chains (C11—AC13). Their ee values were determined by chiral high‐performance liquid chromatography (HPLC) on Chiralcel OD column, using their racemic 1,3‐dioxolanes rac‐ 3 – 12 , which were also synthesized for the first time. These chiral and racemic 1,3‐dioxolanes were characterizated by infrared, NMR (1H, 13C), mass spectrometry, elemental analysis, optical rotation, and chiral HPLC.  相似文献
