In the present work we analyze the dynamics of fibronectin (FN) adsorption on two different stable titanium oxides, with varied surface roughness, and chemically similar to those used in clinical practice. The two types of titanium oxide surfaces used were TiO2 sputtered on Si (TiO2 sp) and TiO2 formed on commercially pure titanium after immersion in H2O2 (TiO2 cp). Surface characterization was previously carried out using different techniques (Sousa, S. R.; Moradas-Ferreira, P.; Melo, L. V.; Saramago, B.; Barbosa, M. A. Langmuir 2004, 20 (22), 9745-9754). Imaging and roughness analysis before and after FN adsorption used atomic force microscopy (AFM) in tapping mode, in air, and in magnetic alternating current mode, in liquid (water). FN adsorption as a function of time was followed by X-ray photoelectron spectroscopy (XPS), by radiolabeling of FN with 125I (125I-FN), and by ellipsometry. Exchangeability studies were performed using FN and HSA. AFM roughness analysis revealed that, before FN adsorption, both TiO2 surfaces exhibited a lower root-mean-square (Rq) and maximum peak with the depth of the maximum valley (Rmax) roughness in air than in water, due to TiO2 hydration. After protein adsorption, the same behavior was observed for the TiO2 sp substrate, while Rq and Rmax roughness values in air and in water were similar in the case of the TiO2 cp substrate, for the higher FN concentration used. Surface roughness was always significantly higher on the TiO2 cp surfaces. AFM led to direct visualization of adsorbed FN on both surfaces tested, indicating that after 10 min of FN incubation the TiO2 sp surface was partially covered by FN. The adsorbed protein seems to form globular aggregates or ellipsoids, and FN aggregates coalesce, forming clusters as the time of adsorption and the concentration increase. Radiolabeling of FN revealed that a rapid adsorption occurs on both surfaces and the amount adsorbed increased with time, reaching a maximum after 60 min of incubation. Time dependence is also observed for the evolution of the atomic (%) of N determined by XPS and by the increase of the thickness by ellipsometry. TiO2 cp adsorbs more FN than the TiO2 sp surfaces, after 60 min of adsorption, as shown by the radiolabeling data. FN molecules are also more strongly attached to the former surface as indicated by the exchangeability studies. The overall results provide novel evidence that FN spontaneously adsorbs as a self-assembly at TiO2 surfaces as a function of time. The aggregate structure is an intermediate feature shared by some protein fibrillar assemblies at interfaces, which is believed to promote cell adhesion and cytoskeleton organization (Pellenc, D.; Berry, H.; Gallet, O. We study the wetting by water of complex "hydrophobic-hydrophilic" surfaces made of a hydrophobic substrate covered by a hydrophilic polymer brush. Polystyrene (PS) substrates covered with polystyrene- block-poly(acrylic acid) PS- b-PAA diblock copolymer layers were fabricated by Langmuir-Schaefer depositions and analyzed by atomic force microscopy (AFM) and ellipsometry. On bare PS substrate, we measured advancing angles theta A = 93 +/- 1 degrees and receding angles theta R = 81 +/- 1 degrees . On PS covered with poorly anchored PS- b-PAA layers, we observed large contact angle hysteresis, theta A approximately 90 degrees and theta R approximately 0 degrees , that we attributed to nanometric scale dewetting of the PS- b-PAA layers. On well-anchored PS- b-PAA layers that form homogeneous PAA brushes, a wetting transition from partial to total wetting occurs versus the amount deposited: both theta A and theta R decrease close to zero. A model is proposed, based on the Young-Dupre equation, that takes into account the interfacial pressure of the brush Pi, which was determined experimentally, and the free energy of hydration of the polyelectrolyte monomers Delta G PAA (hyd), which is the only fitting parameter. With Delta G PAA (hyd) approximately -1300 J/mol, the model renders the wetting transition for all samples and explains why the wetting transition depends mainly on the average thickness of the brush and weakly on the length of PAA chains.  相似文献

Monte Carlo simulations within the primitive model of calcium-mediated adsorption of linear and comb polyelectrolytes onto like-charged surfaces are described, focusing on the effect of calcium and polyion concentrations as well as on the ion pairing between polymers and calcium ions. We use a combination of Monte Carlo simulations and experimental data from titration and calcium binding to quantify the ion pairing. The polymer adsorption is shown to occur as a result of surface overcharging by Ca(2+) and ion pairing between charged monomers and Ca(2+). In agreement with experimental observations, the simulations predict that the polymer adsorption isotherm goes through a maximum as the calcium or the polymer concentration is increased. The non-Langmuir isotherms are rationalized in terms of charge-charge correlations.  相似文献

A CO stretching frequency analysis is presented for the adsorption of CO on various Au(110) surfaces from density functional theory calculations. The structure sensitivity of the adsorption has been studied by considering the unreconstructed (1 x 1) surface, the missing-row reconstructed (1 x 2) surface, the vicinal stepped (20) surface, and the adsorption on adatoms deposited on the (110)-(1 x 2) surface. The calculated CO stretching frequencies are compared with infrared reflection-absorption spectroscopy (IRAS) measurements carried out at room temperature and pressure below 1 atm. The overall stability of the systems is discussed within the calculations of surface free energies at various coverages. At room temperature, the adsorption of CO on the ridge of the missing-row reconstructed surface competes in the high pressure regime with more complex adsorption structures where the molecule coadsorbs on the ridge and on adatoms located along the empty troughs of the reconstruction. This result is supported by the CO stretching frequency analysis.  相似文献

Molecular dynamics simulations are carried out to study the adsorption of three lipids, namely, DOPC, DOPS, and DMTAP, on TiO2(110) rutile surfaces and the influence of the interface on their conformational properties. Three types of rutile (110) surfaces, characterized by a different degree of hydroxylation (the neutral nonhydroxylated and hydroxylated surfaces and a partially hydroxylated surface with charge density corresponding to physiological pH) are investigated using force fields derived from ab initio calculations and experimental data. It is found that the stability of the adsorbate and the strength of the attachment are strictly connected with the nature of both the lipid and the surface. Direct coordination of the phosphate or carbonyl oxygens of the lipids with available titanium sites, observed in the case of partially or nonhydroxylated layers, determines stronger adsorption and, as a consequence, reduced dynamics. For a given hydration state of the surface, the adsorption strengths are in the order DOPS > DOPC > DMTAP, in agreement with experimental data according to which the presence of DOPS units inside lipid bilayers favors stronger adsorption and lower mobility. The adsorption geometry, the hydration state of the lipid headgroups, and the dynamical processes (detachment, diffusion, etc.) occurring at the lipid/oxide interface are analyzed in detail, putting on a roughly quantitative basis time scales and energy barriers of the latter processes.  相似文献

The behavior of polymers adsorbed on a single surface or bridging two surfaces is fairly well understood when thermodynamic equilibrium conditions prevail. The work of Nobel prize winner, P.G. de Gennes, to whom the papers in this issue of Advances in Colloid and Interface Science are dedicated, has greatly contributed to this field. Useful as this knowledge is, in practise many situations are encountered in which polymer adsorption is not thermodynamically but kinetically controlled. In this paper we discuss various aspects of non-equilibrium polymer and polyelectrolyte adsorption, based on recent experimental findings. The discussion of these aspects is mainly qualitative, since we are still far from a detailed quantitative theory of the kinetics of polymer adsorption. Many of the examples are relevant to the chemistry of papermaking.  相似文献

The adsorption of polyelectrolyte complexes, PEC, made from the cationic poly (diallyldimethylammonium) chloride (PDADMAC) and the anionic maleic acid-co-propene copolymer (MA-P) on a Si-wafer surface has been studied. The application of highly diluted colloidally dispersed PEC solutions led to the deposition of single PEC particles onto the surface of the Si-wafer. The interaction forces of the heterogeneously covered surface were monitored by direct force measurements with an atomic force microscope (AFM) in the force volume mode. On the surface of a single PEC particle drastic changes in the interaction forces were found in comparison with the unmodified Si-wafer: in all force vs. distance curves a strong increase of the adhesion was measured that can be attributed to the formation of electrostatic bonds between the negatively charged Si3N4-tip and the cationic excess charge of the PEC. Additionally, the behavior during approach of both surfaces has been distinct: at pH 6.1 we see a long range electrostatic attraction between the tip and the PEC particle. The attraction becomes even stronger at pH 4.1, because of an increased positive net charge. Generally, a heterogeneous surface with a wide variety of interaction features can be created by the adsorption of PEC particles.  相似文献

This work describes the immobilization of beta-galactosidase onto polyelectrolyte multilayer assemblies of the polyanion poly[1-[4-(3-carboxy-4-hydroxyphenylazo)benzenesulfonamido]-1,2-ethanediyl, sodium salt] (PAZO) and the polycation poly(ethylenimine) (PEI) constructed by electrostatic self-assembly (ESA). A single layer of beta-galactosidase was deposited over a precursor film comprising up to five bilayers of the PEI/PAZO polyelectrolyte pair. The enzyme was deposited on both the polycationic (PEI) and the polyanionic (PAZO) surfaces. Quartz crystal microbalance with dissipation monitoring (QCM-D), single-wavelength ellipsometry, and UV-visible absorption spectroscopy revealed differences in both the amount of beta-galactosidase incorporated in each of the multilayer assemblies and the resulting enzyme packing density in the films. The enzymatic films were immersed in a reaction solution containing o-nitrophenyl-beta-d-galactopyranoside (ONPG), and absorbance measurements were used to monitor the concentration of o-nitrophenyl (ONP), the product of the beta-galactosidase catalyzed by hydrolysis of ONPG. Although our data indicate that comparable amounts of beta-galactosidase are incorporated onto both surfaces, enzymatic activity is substantially inhibited when the beta-galactosidase is immobilized on the polyanionic surface compared to the enzyme on the polycationic surface. The difference in catalytic activities reflects the different abilities of the two polyelectrolytes to screen the protein's active site from the substrate environment. In both assemblies, the protein interpenetrated the PEI/PAZO multilayer, disrupting the J-aggregated state of the PAZO chromophores. This work demonstrates that the charge, conformation, and composition of underlying polyelectrolyte cushions have a significant effect on the structure and function of an immobilized protein within functional nanoassemblies.  相似文献

Adsorption phenomena are relevant in a wide variety of subjects, from biophysics to technological applications. Different aspects, such as molecular recognition, multilayer deposition, and dynamics of polymer adsorption have been addressed. The methodologies used range from analytical and numerical methods to molecular dynamics or Monte Carlo simulations. In this work, a coarse‐grained model is used to explore the adsorption of charged backbones to oppositely charged regions of a surface. These regions encompass those small enough to prevent complete adsorption, but extend to surfaces sufficiently large to promote adsorption with minimal effect on the three‐dimensional conformation in bulk. Apart from the different surface areas explored, variations on the surface charge density, polyelectrolyte chain length, and chain stiffness were also considered. The degree of compaction of the polyelectrolyte, on adsorption, is different from that found in the bulk. Also, results indicate an nonuniform adsorption pattern on regularly charged surfaces. © 2013 Wiley Periodicals, Inc.  相似文献

The effects of pH and ultraviolet (UV) light with ligated formic acid on mesoporous TiO2 were characterized by transmission Fourier transform infrared (FTIR) spectroscopy and compared with adsorbed formate complexes. Surface-modified anatase thin films were prepared from acidic aqueous nanoparticulate anatase suspensions diluted with methanol and ethanol. Bands assigned to carboxylic acid groups displayed unique bonding character in the ligated formic acid on the anatase surface. For increased proton concentrations in the films, separation in -COO stretching bands (delta nu) for formic acid increased (increase in frequency for nuC=O and decrease in frequency for nuC-O). With UV exposure, surface-bound organics were rapidly removed by photocatalytic oxidation at 40 degrees C and 40% relative humidity (RH). In addition, the delta nu of the formic acid bands decreased as organics were mineralized to carbonates and CO2 with UV light. Aqueous formic acid adsorption experiments showed a distinctly different bonding environment lacking carbonate, and the delta nu for the carboxylic groups indicated a bridging bidentate coordination. The delta nu of the bands increased with increasing proton concentration, with both bands shifting to higher wavenumbers. The shifts may be ascribed to the influence of protonation on surface charge and the effect of that charge on the electronegativity of carboxylate groups bound to the surface. As alcohols are used in the mesoporous TiO2 solar cell preparation, implications of these surface modifications to dye-sensitized photovoltaics are discussed.  相似文献

We present an accurate comparative analysis of N 2 adsorption at 77 K on nonporous silica and the pore wall surface of MCM-41 materials. The analysis shows that in the low-pressure region of N 2 adsorption obeys a peculiar mechanism governed by short-ranged forces, which makes the surface curvature effect on the N 2 adsorption in mesopores nearly negligible. We used this observation to define more exactly compared to the BET technique the specific surface area of the reference adsorption isotherm on nonporous silica basing on XRD data and linear sections of t-plots. Calculation of the capillary evaporation and condensation pressures seems to confirm our previous finding that the capillary condensation pressure corresponds to the equilibrium transition rather than spinodal condensation at least for pore sizes less than 7 nm. It allowed us to provide more reliable pore size distribution (PSD) analysis of mesoporous silica materials. For example, the PSDs of MCM-41 samples do not show artificial peaks in the micropore range that we obtained in our earlier publications.  相似文献

Undesired adsorption of proteins brings big troubles to marine structures.The settled proteins change the physical and chemical properties of the surfaces,which allow marine fouling organisms to settle down on the structures.Therefore,to understand the adsorption mechanism of proteins is very helpful to find an environment-friendly solution against biofouling.Many approaches have been developed to study protein adsorption,but most of them are insufficient to give the chemical interaction information between proteins and surfaces.Fourier transform infrared spectroscopy with attenuated total reflection(FTIR-ATR)is an efficient,fast and non-destructive method for in situ surface measurement,which greatly minimizes the interference of water to infra red spectra,because of the very small depth of penetration of the evanescent wave.In this paper,an in situ FTIR-ATR technology was used to investigate the adsorption process of trypsin on a bare ZnSe surface and on a TiO2 coated ZnSe surface,and the effect of calcium cation strength and ultraviolet light irradiation on the secondary structure of trypsin were also evaluated.FTIR spectra of trypsin showed that Amide I band red shift and AmideⅡband blue shift in aqueous environment on both surfaces compared with the dry trypsin powder,and the addition of calcium cations further changed the Amide bands position,which indicated that the change of the secondary structure could be interfered by the environment.The hydrogen bond formation between water and trypsin,the interaction between surface and trypsin,the interaction between hydrated calcium cations and trypsin,are major facto rs to change the secondary structure of trypsin,and UV light irradiation also showed its influence for the secondary structure.  相似文献

The adsorption of single polyelectrolyte molecules onto surfaces decorated with periodic arrays of charged patches was studied using Brownian dynamics simulations. A free-draining, freely jointed bead-rod chain was used to model the polyelectrolyte, and electrostatic interactions were incorporated using a screened Coulombic potential with the excluded volume accounted for by a hard-sphere potential. The simulations predicted that the polyelectrolyte lies close to the adsorbing surface if the patch length, surface charge density, and screening length are sufficiently large. Chain conformations were found to be very sensitive to patch length, patch spacing, and the nature of the charge on adjacent patches. This is due both to the size of the polymer relative to patch length and spacing and to the structure of the electric field near the surface. In some cases, the component of the radius of gyration parallel to the surface can be made smaller than its free-solution value, which is contrary to what is observed for a uniformly charged surface. Isolated charged patches were also considered, and significant adsorption was observed above a critical surface charge density. The results demonstrate how polyelectrolyte conformations can be controlled by the design of the charged patches and may be useful for applications in which adsorbed polyelectrolyte films play a key role.  相似文献

The effect of patterned surface topography on the adsorption of single polyelectrolyte molecules is explored using Brownian dynamics simulations. The polyelectrolyte is modeled as a free-draining, freely jointed bead-rod chain, and electrostatic interactions are incorporated using a screened Coulombic potential with excluded volume interactions accounted for by the repulsive part of a Lennard-Jones potential. Topography consisting of periodically spaced valleys of square cross section separated by flat hills is considered. Chain conformations are characterized for a wide range of valley widths, depths, and spacings as well as for several different types of surface charge distributions. Depending on the parameter values describing the topography, the chains are found to adopt conformations ranging from flat and extended to those associated with bridge-, brush-, or semi-bridge-like structures. The formation of these structures is rationalized on the basis of a free-energy model that takes into account the increase in free energy due to entropic confinement, excluded volume interactions, and chain stretching as well as the decrease in free energy due to bead-surface electrostatic attraction. The results of this work are expected to be useful in designing patterned surface topography to control the conformations of adsorbed polyelectrolyte molecules.  相似文献

CO2 adsorption properties on Mg modified silica mesoporous materials were investigated. By using the methods of co-condensation, dispersion and ion-exchange, Mg2+ was introduced into SBA-15 and MCM-41, and transformed into MgO in the calcination process. The basic MgO can provide active sites to enhance the acidic CO2 adsorption capacity. To improve the amount and the dispersion state of the loading MgO, the optimized modification conditions were also investigated. The XRD and TEM characteristic results, as well as the CO2 adsorption performance showed that the CO2 adsorption capacity not only depended on the pore structures of MCM-41 and SBA-15, but also on the improvement of the dispersion state of MgO by modification. Among various Mg modified silica mesoporous materials, the CO2 adsorption capacity increased from 0.42 mmol g−1 of pure silica SBA-15 to 1.35 mmol g−1 of Mg–Al–SBA-15-I1 by the ion-exchange method enhanced with Al3+ synergism. Moreover, it also increased from 0.67 mmol g−1 of pure silica MCM-41 to 1.32 mmol g−1 of Mg–EDA–MCM-41-D10 by the dispersion method enhanced with the incorporation of ethane diamine. The stability test by 10 CO2 adsorption/desorption cycles showed Mg–urea–MCM-41-D10 possessed quite good recyclability.  相似文献

Mean-field theory is used to derive criteria for the adsorption of a weakly charged polyelectrolyte molecule from salt solution onto surfaces patterned with charge and topography. For flat surfaces patterned with periodic arrays of charged patches, the adsorbed layer thickness predicted using mean-field theory and that found by Brownian dynamics simulations are in quantitative agreement in the strong-adsorption regime, which corresponds to sufficiently small kappa or sufficiently large |sigma(eff)q|, where kappa is the inverse Debye screening length, sigma(eff) is an effective surface charge density, and q is the charge on each segment of the polyelectrolyte. Qualitative agreement is obtained in the weak-adsorption regime, and for the case where surfaces are patterned with both charge and topography. For uniformly charged, sinusoidally corrugated surfaces, the theory predicts that the critical temperature required for adsorption can be greater than or less than the corresponding value for a flat surface depending on the relative values of kappa and the corrugation wave number. If the surface charge is also allowed to vary sinusoidally, then adsorption is predicted to occur only when the topography crests have a surface charge opposite to that of the polyelectrolyte. Surfaces patterned with rectangular indentations having charged bottoms which are separated by flat charged plateaus are investigated as well. Adsorption is predicted to occur even when the net surface charge is zero, provided that the plateaus have a charge opposite to that of the polyelectrolyte. If the charge on the plateaus and polyelectrolyte is the same, adsorption may still occur if electrostatic attraction from the indentation bottoms is sufficiently strong.  相似文献
