用毛细管电泳电化学检测法测定了马钱子中的士的宁和马钱子碱.以BR缓冲溶液(35 mmol/L混酸用180 mmol/L NaOH调节至pH 6.50)为电泳液,分离电压为6 kV,检测电位为0.95 V,士的宁和马钱子碱在30 min内得到了良好分离.士的宁和马钱子碱浓度分别在0.35~35.00 μg/mL和0.40(40.00 μg/mL内具有良好的线性关系,检测限分别为0.08μg/mL和0.15μg/mL.应用于马钱子样品的测定.  相似文献

A new capillary zone electrophoresis (CZE) method was developed for simultaneous assay of four chalcones, hydroxysafflor yellow A, safflor yellow A, safflamin C, and safflamin A, in the Chinese herbal extract from Carthamus tinctorius L. The optimum buffer system was 30 mM borate buffer (Na2B407/HCl, pH 9.00) with 10% (v/v) methanol. The voltage was 15 kV and detection was at 270 nm. Regression equations revealed linear relationships (correlation coefficients: 0.9973, 0.9992, 0.9989, and 0.9996) between the peak area of each compound and its concentration. The within-day relative standard deviations of migration times and peak areas were < 1.53 and 4.14%, respectively. The effects of several CE parameters on the resolution were studied systematically. The contents of four chalcones in Carthamus tinctorius L. were successfully determined with satisfactory repeatability and recovery. The possibilities of using this method for the determination of chalcones in Chinese medicinal preparation was also tested.  相似文献

An on-line sample preconcentration method by two-step stacking i.e., sweeping and micelle to solvent stacking, in capillary zone electrophoresis (CZE) has been developed for the determination of strychnine and brucine in traditional Chinese herbal medicines. After experimental optimizations, the best separation was achieved by using 75 mM phosphate buffer (pH 2.5) with 30% methanol (v/v). Compared with normal CZE injection, 51- and 38-fold improvement in concentration sensitivity was achieved for strychnine and brucine, respectively. The calibration curve was linear in the range of 0.1–5.0 μg mL−1 for both strychnine and brucine, with the correlation coefficients of 0.9998 and 0.9997, respectively. The limits of detection (S/N = 3) for both alkaloids were 0.01 μg mL−1. The inter-day (n = 8) and intra-day (n = 5) reproducibilities expressed as the relative standard deviations for corrected peak area were less than 9.5%. The method was applied to determine strychnine and brucine in two Chinese herbal medicines, with recoveries ranging from 94.2% to 105.4%. The results indicated that the method is simple, rapid, reliable, and can be applied to determine strychnos alkaloids in traditional Chinese herbal medicines.  相似文献

A capillary zone electrophoresis (CZE) method has been developed for investigating the physicochemical characteristics of five Strychnos alkaloids in Strychnos nux-vomica L. Firstly, the dissociation constants of the five Strychnos alkaloids were determined, based on the relation between the effective mobility of the solutes and the buffer pH. The mathematical relationship was strictly deduced from the fundamental electrophoretic theory and the dissociation equilibrium. Secondly, an equation describing the relation between the migration time of alkaloids of similar structure and their molecular weights was developed and used to predict the migration order and to calculate the electrosomotic velocity. The results predicted by the theory agreed with those from experiments.  相似文献

A novel micelle to solvent stacking on-line sample preconcentration technique in capillary zone electrophoresis(MSS-CZE) has been developed to determine the strychnine and brucine in traditional Chinese medicine preparations.The optimal running buffer was 30 mmol/L H3PO4 containing 20%acetonitrile at pH 4.0.The sample matrix was 8 mmol/L H3PO4 containing 5 mmol/L sodium dodecyl sulfate(SDS) at pH 3.0.The established MSS-CZE method afforded more than 50-fold improvements in concentration sensitivity compared with typical CZE-UV analysis.The calibration curve was linear in the range from 0.2 to 15.0μg/ mL for both strychnine and brucine,with correlation coefficients of 0.9984 and 0.9976,respectively.The limits of detection(5/ N = 3:1) for strychnine and brucine were 0.02 and 0.05μg/mL,respectively.The MSS-CZE method has been successfully applied to the analysis of strychnine and brucine in Chinese medicinal preparations.  相似文献

Summary The potential of capillary zone electrophoresis has been investigated for the separation and quantitative determination of some quinolone antibiotics. The influence of different conditions, such as the nature and concentration of the electrophoretic electrolyte, on migration time, peak symmetry, efficiency and resolution was studied. A buffer consisting of 100mm HEPES adjusted to pH 8.5 containing 10% (v/v) acetonitrile was found to furnish a very efficient and stable electrophoretic system for the separation of exoxacin, ciprofloxacin, ofloxacin, oxolinic acid, nalidixic acid and pipemedic acid. A linear relationship between concentration and peak area for each compound was obtained in the concentration range 0.25–40 μg mL−1; detection limits were approximately 0.25 ng mL−1. It was demonstrated that the method can be used for the simultaneous determination of these six antibiotics in serum and urine samples.  相似文献

Cation determination is important for quality control of beverage products. To determine a large group simultaneously, a capillary electrophoresis procedure is developed with indirect UV at 214 nm in a three-complex buffer system (10 mM N,N-dimethylbenzylamine (DBA), 8 mM lactic acid and 2 mM 18-crown-6) with good mobility matching with desired cations. Under optimized conditions with pH adjusted to 4.65, a baseline separation is achieved for 14 cations (Rb(+), NH(4)(+), K(+), Ca(2+), Na(+), Mg(2+), Mn(2+), Co(2+), Fe(2+), Cd(2+), Cr(3+), Ni(2+), Zn(2+) and Cu(2+)) within 7 min using an uncoated silica column. To cover ng/l to mug/l range, both hydrostatic and electrokinetic sampling are studied, showing working ranges within (0.05-50)/(0.005-2) microg/l and detection limits (13-78)/(1.4-10) ng/l, respectively with satisfactory repeatability (RSD 0.31-0.47% for migration time, and 3.0-4.0% for peak height measurement). Agreeable results with established inductively coupled plasma-atomic emission spectrometry method have been obtained for orange juice and tea samples.  相似文献

Capillary zone electrophoresis (CZE), using a 20 mmol/L borate buffer at pH 10.5, was developed for the identification and determination of three coumarins--7-hydroxy-coumarin (HC), 7-hydroxy-8-methoxy-coumarin (HMC) and 7-O-beta-D-glucosyl-coumarin (GC)--in the extracts of the flower of Cacalia tangutica. Regression equations revealed linear relationships (correlation coefficient 0.9986-0.9990) between the corrected peak area (the ratio of peak area to migration time) of each constituent and its concentration. The relative standard deviations (RSD) of the migration time and peak area were 1.45-1.52 and 2.60-3.84% (intra-day), and 1.75-2.22 and 2.90-4.04% (inter-day), respectively. The recoveries of three constituents ranged between 94.5 and 105.6%. The effects of pH value, buffer concentration and applied voltage on the migration behavior of HC, HMC and GC were investigated. The contents of the three active constituents in the flower of Cacalia tangutica were successfully determined within 6 min under the optimum conditions chosen. Moreover, the dissociation constants for three coumarins were also determined by CE.  相似文献

Tetracycline was separated from its main impurities 4-epitetracycline, anhydrotetracycline, 4-epianhydrotetracycline, chlortetracycline, and demethyltetracycline by capillary electrophoresis. Systematic method development was performed in which following parameters were consecutively optimized: type and pH of the buffer, buffer concentration, type and concentration of organic modifier, voltage and temperature. 采用区带毛细管电泳法分离测定洋金花中3种莨菪烷类生物碱—阿托品、山莨菪碱和东莨菪碱。以57 cm×50μm熔融石英毛细管为分离通道,工作电压25 kV,温度25℃,200 mmol/L Tris-H3PO4(pH=8.3)为背景电解质。经优化分离条件,3种莨菪烷类生物碱达到基线分离,此方法快速、简便,可作为中药材洋金花中有效成分分离的方法和质量控制方法。  相似文献

Separation conditions suitable to a rapid resolution of a group of eight nitrophenols by capillary zone electrophoresis (CZE) were found. Required differences in their effective mobilities were achieved via host-guest complexation of -cyclodextrin combined with intermolecular interactions involved by polyvinylpyrrolidone. When both additives were present in the carrier electrolyte at pH=9.1 nitrophenols could be separated in the column of a, 300 m I.D. and 180 mm in the length within 8–9 minutes. It is shown that the column of such an I.D. providing enhanced sample load capacity, can operate with high separation efficiencies as maintaining zone dispersions due to Joule heating on a tolerable level. CZE on-line coupled with isotachophoretic sample pretreatment is shown to provide the concentration limits of detection at low ppb concentrations by using an on-column photometric detector operating at 254 and 405 nm detection wavelengths.  相似文献

The use of capillary zone electrophoresis (CZE) and capillary zone electrophoresis/mass spectrometry (CZE/MS) has been demonstrated, in principle, for the separation of nicotine and nicotine metabolites. The buffer system developed for separation and detection by CZE/UV was modified for use in CZE/MS analysis. A simple, rapid, sensitive and low‐cost method using capillary electrophoresis (CE) coupled with field‐amplified sample stacking (FASS) has been developed and validated for the simultaneous determination of strychnine and brucine residues in human urine. Before sample loading, a water plug (3.5 kPa, 3 s) was injected to contain sample cations and to permit FASS. Electrokinetic injection at a voltage (20 kV, 25 s) was then used to introduce cations. Separation was performed using 20 mM acetate buffer (pH 3.8) with an applied voltage of 20 kV. The calibration curves were linear over a range of 8.00–2.56 ∞ 102 ng/mL (r = 0.9995) for strychnine and 10.0–3.20 × 102 ng/mL (r = 0.9999) for brucine. Extraction recoveries in urine were greater than 79.6 and 82.8% for strychnine and brucine, respectively, with an RSD of less than 4.9%. The detection limits (signal‐to‐noise ratio 3) for strychnine and brucine were 2.00 and 2.50 ng/mL, respectively. A urine sample from one healthy female volunteer (26 years old, 50 kg) was pretreated and analyzed. Strychnine and brucine levels in urine could be detected 24 h after administration. On these grounds, this method was feasible for application to preliminary screening of trace levels of abused drugs for both doping control and forensic analysis. An accurate capillary electrophoresis method was developed for the determination of dissociation constants of five Strychnos alkaloids from Strychnos nux-vomica L. The method relies on measuring the effective mobility of the solute as a function of the buffer pH. The mathematical relationship was strictly derived from the fundamental electrophoresis theory and the dissociation equilibrium of a weak base without any simplifications. Careful optimization of the running buffer permitted base-line resolution of the five structurally similar alkaloids.  相似文献

In this paper, capillary zone electrophoresis with amperometric detection (CZE-AD) was firstly applied to the simultaneous separation and determination of nitroaniline positional isomers. The three analytes could be perfectly analyzed by using the buffer of extreme pH. The effects of several important factors were investigated to find optimum conditions. A carbon-disk electrode was used as working electrode. The optimal conditions were 40 mmol/L tartaric acid-sodium tartrate (pH 1.2) as running buffer, 17 kV as separation voltage and 1.10 V (versus saturated calomel reference electrode, SCE) as detection potential. Under the optimum conditions, o-, m- and p-nitroaniline were separated successfully and good linearity, reproducibility and recovery results were obtained. The detection limit for m-nitroaniline was as low as at 9.06 × 10−9 mol/L. This proposed method demonstrated long-term stability and reproducibility with relative standard deviations of less than 1.8% for migration time and 1.1% for peak areas. The utility of this method was demonstrated by monitoring dyestuff wastewater and the assay results were satisfactory.  相似文献

A method of capillary electrophoresis with wall-jet amperometric detection (AD) has been developed for separation and determination of l-tyrosine (Tyr) and its metabolites, such as Tyramine (TA), p-hydroxyphenylpyruvic (pHPP), homogentisic acid (HGA) and some dipeptides containing Tyr, such as Tyr-Gly-Gly (YGG), Tyr-Arg (YR) and Tyr-d-Arg (Y-d-R). A carbon disk electrode was used as the working electrode and the optimal detection potential was 1.00 V (versus Ag/AgCl). At 18 kV of applied voltage, the seven compounds were completely separated within 20 min in 110 × 10−3 mol/L Na2HPO4-NaH2PO4 buffer (pH 7.10) containing 3 × 10−3 mol/L β-cyclodextrin (β-CD). Good linear relationship was obtained for all analytes and the detection limits of seven analytes were in the range of 0.95-4.25 ng/mL. The proposed method has been applied to examine the metabolic process of l-tyrosine in rabbit's urine.  相似文献

A simple and rapid capillary electrophoretic method was developed for the simultaneous determination of noradrenaline (NA) and dopamine (DA) in Portulaca oleracea L. The buffer solution used in this method was 40 mM tris (hydroxymethyl) aminomethane (Tris)-H3PO4 at pH 2.00 containing 15% methanol. The effects of pH value, organic modifier, and applied voltage were investigated. The linear ranges of NA and DA were 0.5-100 microg/mL (r=0.9952) and 6.25-200 microg/mL (r=0.9992), respectively. The relative standard deviations of the corrected peak area were 6.73% and 4.26%, respectively. NA and DA in Portulaca oleracea L. were simultaneous determined successfully within 5.6 min. In this way, the contents of NA and DA in different parts (stem, leaves, and seeds) of P. oleracea L. and in different extracts of leaves with different solvents (distilled water, 50% methanol, and methanol) were studied.  相似文献

We describe a method for the simultaneous determination of the five fibrinopeptide forms derived from the thrombin-promoted activation of human fibrinogen by capillary zone electrophoresis (CZE). The fibrinopeptide mixture was first desalted by a solid-phase extraction (SPE) step. The analysis was performed in reversed polarity in a highly cross-linked polyethylene glycol (PEG)-coated capillary with UV-light absorption detection at 200 nm. Several parameters including buffer concentration and pH, presence of an organic modifier, temperature, and applied voltage, have been tested. The best separations were obtained within 20 min, utilizing a 20 mM sodium phosphate buffer without organic modifier, in the narrow 6.1-6.2 pH range, at 25 degrees C, with an applied voltage of 20 kV. Quantitative analysis is made possible by the use of sheep fibrinopeptide A as an internal standard to correct for both extraction and injection errors.  相似文献

The effects of pH on separation parameters such as migration mobility, resolution, sensitivity, column efficiency and peak shape were emphatically studied. Better separation of magnolol and honokiol using capillary zone electrophoresis was achieved by optimizing pH in the range 5.0–11.7. The influences of applied voltage and temperature were also investigated. We adopted a better sample extraction procedure by which higher contents of honokiol and magnolol with sample compositions unchanged were obtained. The analysis was performed with direct UV detection using a 10 mM borate-10 mM phosphate buffer at pH of 11.6. The method was successfully applied to the simultaneous determination of magnolol and honokiol inMagnolia officinalis bark within 9 min.  相似文献

A new capillary electrophoresis procedure with field-enhanced stacking concentration for the analysis of strychnine and brucine is established. After optimization of the separation and concentration conditions, the two alkaloids can be separated within 5 min and quantified with high sensitivity (The detection limits were 1.0 ng mL(-1) for strychnine and 1.4 ng mL(-1) for brucine). The method was useful for qualitative and quantitative analysis of strychnine and brucine in Strychnos nux-vomica L with recovery of 105.1% for strychnine and 98.4% for brucine.  相似文献
