An inductively coupled plasma/MS method was developed for the simultaneous determination of Cr, Se, and Mo in infant formulas and other nutritional products. All samples were digested using a closed vessel microwave oven system, together with Ni and Te internal standards. The practical quantitation limits for Cr, Se, and Mo were 0.4, 0.2, and 0.4 ng/mL, respectively; dilution factors were 250 for powders and 50 for liquids. The Cr, Se, and Mo concentrations in 10 nutritional products were within specification limits, and within-day and day-to-day (6 independent days) precision values were <5% RSD. For two control samples, the observed precision was < or = 2% RSD over 10 independent days. In three National Institute of Standards and Technology standard reference materials, Cr, Se, and Mo results were within the certified limits. The average sample spike recoveries for 10 nutritional products ranged from 93 to 107%. Robustness studies showed a minimal effect from concomitant easily ionized element concentrations. However, the choice of internal standard and matrix matching carbon content were critical to obtaining accurate Se results. All indications are that this method would be a suitable candidate as a global reference method for the determination of these trace elements in nutritional products.  相似文献

An enzyme-assisted extraction used to extract all species of selenium in rice sample and a sensitive analytical method for the determination of ultratrace Se(VI), Se(IV), SeCys2 (selenocystine) and SeMet (selenomethionine) with capillary electrophoresis-inductively coupled plasma mass spectrometry were firstly described in this study. The extraction method is simple, effective and can be used to extract trace selenium compounds in rice with high extraction efficiency and no altering its species. The analytical method has a detection limit of 0.1-0.9 ng Se/mL, and can be used to determine trace Se(VI), Se(IV), SeCys2 and SeMet in rice directly without any derivatization and pre-concentration. With the help of above methods, we have successfully determined Se(VI), Se(IV), SeCys2 and SeMet in selenium-enriched rice within 18 min with a recovery of 90-103% and a RSD (relative standard deviation, n = 6) of 3-7%. Our results indicated that selenium-enriched rice contained only one species of selenium, SeMet, and its concentration is in range of 0.136-0.143 μg Se/g dried weight. The proposed method providing a realistic approach for the nutritional and toxical evaluation of different selenium compounds in nutritional supplements.  相似文献

A new method for speciation analysis of two inorganic selenium species was developed by on-line coupling of capillary electrophoresis (CE) with hydride generation-atomic fluorescence spectrometry (HG-AFS) and on-line conversion of Se(VI) to Se(IV). Baseline separation of Se(VI) and Se(IV) was achieved by CE in a 50 cm x 75 microm inside diameter (ID) fused-silica capillary at -20 kV using a mixture of 15 mmol.L(-1) NaH2PO4 and 0.5 mmol.L(-1) cetyltrimethylammonium bromide (pH 7.5) as electrolyte buffer. Se(VI) was on-line reduced to Se(IV) by mixing the CE effluent with concentrated HCl. The precision (relative standard deviation, RSD, n=7) ranged from 0.7 to 1.3% for migration time, 6.4 to 3.7% for peak height response, and 5.9 to 6.1% for peak area for the two selenium species at the 500 microg.L(-1) (as Se) level. The detection limits were 33 and 25 microg.L(-1) (as Se) for Se(VI) and Se(IV), respectively. The recoveries of the two selenium species in five locally collected water samples ranged from 88 to 114%. The developed method was applied to speciation analysis of inorganic selenium species in spiked natural water samples.  相似文献

Values of the total selenium and selenomethionine (Semet) content of four wheat-based reference materials have been obtained by gas chromatography-stable isotope dilution mass spectrometry methods. The total Se method is an established one, and the results obtained with it are consistent with previously-assigned values. The Semet method (previously reported by our laboratory) is based on reaction with CNBr. Our data indicate that the four wheat samples (wheat gluten, durum wheat, hard red spring wheat, and soft winter wheat), though having a 30-fold range in total Se content, all have about 45% of their total Se values in the form of selenomethionine. Investigation of the CNBr-based method suggests that additional experiments are needed to verify that all selenomethionine in the wheat samples is accounted for, but also indicates that the values obtained are within 15% of the true values. As the form in which Se occurs in foods and dietary supplements is important from a nutritional perspective, adding information about Se speciation to total Se values in appropriate reference materials makes these materials more valuable in relevant analytical work.  相似文献

A novel, fast, and cheap nonchromatographic method for direct speciation of dissolved inorganic and organic selenium species in environmental and biological samples was developed by flow injection (FI) dual-column preconcentration/separation on-line coupled with ICP-MS determination. In the developed technique, the first column packed with nanometer-sized Al(2)O(3) could selectively adsorb the inorganic selenium [Se(IV), Se(VI)], and the retained inorganic selenium could be eluted by 0.2 mol l(-1) NaOH, while the organic Se [selenocystine (SeCys(2)) and selenomethionine (Se-Met)] was not retained. On the other hand, the second column packed with mesoporous TiO(2) chemically modified by dimercaptosuccinic acid (DMSA) could selectively adsorb Se(IV) and SeCys(2) and barely adsorb Se(VI) and Se-Met. When the sample solution was passed through the column 1, separation of inorganic selenium and organic selenium could be achieved first. Then, the effluent from column 1 was successively introduced into the column 2 and the speciation of organic selenium could be attained due to the different adsorption behaviors of Se-Met and SeCys(2) on DMSA modified TiO(2). After that, the eluent from column 1 contained Se(IV), and Se(VI) was adjusted to desired pH and injected into column 2, and the speciation of Se(IV) and Se(VI) could also be realized thanks to their different retention on column 2. The parameters affecting the separation were investigated systematically and the optimal separation conditions were established. The detection limits obtained for Se(IV), Se(VI), Se-Met and SeCys(2) were 45-210 ng l(-1) with precisions of 3.6-9.7%. The proposed method has been successfully applied for the speciation of dissolved inorganic and organic selenium in environmental and biological samples. In order to validate the methodology, the developed method was also applied to the speciation of selenium in certified reference material of SELM-1 yeast, and the determined values were in good agreement with the certified values.  相似文献

A single-laboratory validation by inductively coupled plasma-mass spectrometry was developed for the determination total iodine (m/z 127) in infant formula and adult nutritional products. All samples were digested in nitric acid using a closed vessel microwave oven system; Te (m/z 130) was used as an internal standard. To prevent loss of iodine, ammonium hydroxide solution was added to the samples immediately after digestion. The method quantitation limit for total iodine was 0.3 ng/mL, but a practical LOQ was used at 1.0 ng/mL, a concentration at which there was a negligible bias due to nonlinearity. The total iodine concentrations (112-1900 ng/g) in 14 out of 15 nutritional products were within specification limits. Within-day and day-to-day (6 independent days) precision values were < 10% RSD. The observed precision for the overall mean (18 independent days) of a control sample was approximately 4% RSD. In two National Institute of Standards and Technology standard reference materials, total iodine results were within certified limits. Sample spike recoveries for all 15 nutritional products were 92-105%. The data show that a conventional microwave oven digestion procedure can be used to prepare samples for iodine determination. Therefore, this technique is very compatible with other methods being proposed as modern official methods for the analysis of minerals in nutritional products.  相似文献

Abstract

Trace amounts of iodine in thirty-eight cereal grain samples cultivated at different locations in Austria were determined for the first time in this study by radiochemical neutron activation analysis. For the dissolution of cereal grain samples and standard reference materials, two different procedures, alkaline and acidic dissolution, were applied in the presence of an iodine carrier. Rapid and simple dissolution procedure with acidic solution was demonstrated in this study. The analytical values in the cereal grain as well as in the standard reference materials obtained by the different dissolution procedures were in good agreement within one standard deviation. The iodine in cereal grains and the standard reference materials ranged from 0.002 to 0.03 μg g?-1 and 0.0015 to 0.30 μg g?-1, respectively. The distribution of relative standard deviation (RSD) for iodine concentration below 0.01 μg g?-1 were 21% and 24% of all data for the range 1–10% RSD and 11–20% RSD, respectively. LC/MS/MS was developed to determine the residues of bithionol (BTN), bromofen (BMF), nitroxynil (NTX), oxyclozanide (OCZ), and tribromsalan (TBS) in milk. Samples were extracted with ethyl acetate and cleaned up by liquid-liquid separation with acetonitrile and n-hexane. The compounds were determined by RP-LC using a C18 column with 0.1% formic acid-methanol. Mass spectral acquisition was performed in the negative mode by applying selected-reaction monitoring. The method was validated in milk spiked with these compounds at 5-600 microg/kg; average recoveries were in the range 83.8-97.1%, with RSD values of 1.4-8.0%. The interassay RSDs were less than 11%. The LODs of these compounds in milk were 0.1 microg/kg. The method was applied to 24 raw milk samples. The concentration of these compounds in all samples was lower than the Japanese maximum residue limits. The method is rapid, sensitive, and specific for monitoring residues of BTN, BMF, NTX, OCZ, and TBS in milk.  相似文献

Hydrophilic oxygen radical absorbance capacity (H-ORAC) is a method for evaluating antioxidant capacities of solutions of hydrophilic compounds. In this study, we improved the original method for H-ORAC determination, and evaluated the precision of the two improved methods (methods A and B) by interlaboratory studies using 5 antioxidant solutions and 5 food extracts as test samples. An interlaboratory study of method A, in accordance with the harmonized protocol, demonstrated satisfactory performance (intermediate precision relative standard deviations (RSD(int)) ranging from 4.6 to 18.8%; the reproducibility relative standard deviations (RSD(R)) ranging from 7.0 to 21.1%, and the HorRat values ranging from 0.40 to 1.93). However, methodological problems remained, and a further improved method, method B, was thus developed. An interlaboratory study of method B by 5 participating laboratories showed better intermediate precision and reproducibility (RSD(int) and RSD(R) ranging from 1.8 to 9.4%, and from 4.4 to 13.8%, respectively), and all HorRat values for the test samples were less than 1.3, suggesting good performance for the H-ORAC measurement.  相似文献

Glutathione (GSH), glutathione disulfide (GSSG) and 2‐hydroxyethylated glutathione (HESG) are important biomarkers for exploring the genotoxicity mechanism of ethylene oxide (EO) or ethylene in vivo. A liquid chromatography–tandem mass spectrometry method was developed for simultaneous determination of GSH, GSSG and HESG in mouse lung tissues after inhalation exposure to EO. The lower limit of quantitation for all these biomarkers was 0.002 µg/mL. The linearity of the calibration curves for all analytes was >0.998. The intra‐day assay precision relative standard deviation (RSD) values for quality control samples for all analytes were ≤12.8% with accuracy values ranging from 87.2 to 113%. The inter‐day assay precision (RSD) values for all analytes were ≤13.1% with accuracy values ranging from 86.9 to 103%. This method was applied to concurrently determine the levels of GSH, GSSG and HESG in lung samples isolated from mouse after 4‐week inhalation exposure to EO at 0, 10, 50, 100 and 200 ppm. A formerly developed Se speciation method was applied to human serum. Chomatographic performance was checked regularly by measuring control standards after four sample run, each, to ensure sufficient separation of the species and sensitivity of the method even after a considerable number of serum samples. Detection limits of investigated species were similar to those reported and showed values below 1 microg/L related to Se for each Se compound. Sera of cystic fibrosis (CF) patients were investigated in comparison to sera from healthy volunteers with respect to total selenium and Se species. Generally, CF sera showed lower values of total Se, Selenocystine (SeC), and cationic/neutral Se compounds compared to serum of healthy persons. No significant gender-specific differences were found. Total Se in sera of healthy persons was determined at 102+/-12 microg/L (n = 12 individuals, mean value from male and female, age 4-38 years), whereas CF patients showed 58+/-10 microg/L (n = 31 individuals, mean value from male and female, age 3-35 years). Se-cystine showed significant differences between the CF and healthy group with a lowered SeC value in sera of CF patients by -75% (mean ca. 26 microg/L in healthy sera compared to about 6.5 microg/L (mean) in CF sera). A similar situation is seen for neutral/cationic Se compounds, which partly may comprise of Se proteins. The lowered SeC values together with lowered cationic/neutral Se compounds (probably Se enzymes) point to a Se-depleted regulated pathway combined with a reduced capability of protective functions such as protection from peroxides.  相似文献

A simple and sensitive method was developed for the simultaneous separation and determination of trace earthy-musty compounds including geosmin, 2-methylisoborneol, 2-isobutyl-3-methoxypyrazine, 2-isopropyl-3-methoxypyrazine, 2,3,4-trichloroanisole, 2,4,6-trichloroanisole, and 2,3,6-trichloroanisole in water samples. This method combined headspace solid-phase microextraction (HS-SPME) with gas chromatography-mass spectrometry and used naphthalene-d(8) as internal standard. A divinylbenzene/carboxen/polydimethylsiloxane fiber exposing at 90°C for 30 min provided effective sample enrichment in HS-SPME. These compounds were separated by a DB-1701MS capillary column and detected in selected ion monitoring mode within 12 min. The method showed a good linearity from 1 to 100 ng L(-1) and detection limits within (0.25-0.61 ng L(-1)) for all compounds. Using naphthalene-d(8) as the internal standard, the intra-day relative standard deviation (RSD) was within (2.6-3.4%), while the inter-day RSD was (3.5-4.9%). Good recoveries were obtained for tap water (80.5-90.6%), river water (81.5-92.4%), and lake water (83.5-95.2%) spiked at 10 ng L(-1). Compared with other methods using HS-SPME for determination of odor compounds in water samples, this present method had more analytes, better precision, and recovery. This method was successfully applied for analysis of earthy-musty odors in water samples from different sources.  相似文献

A novel method has been developed to simultaneously determine and quantify seven organic UV filters employing liquid (solid)-liquid extraction, derivatization with N-methyl-N-(trimethylsilyl) trifluoroacetamide (MSTFA) and gas chromatography with mass spectrometric detection in various environmental matrices. The UV filters determined were: benzophenone (BP), benzhydrol (BH), 4-hydroxybenzophenone (HBP), 2-hydroxy-4-methoxybenzophenone (HMB), 2,4-dihydroxybenzophenone (DHB), 2,2'-dihydroxy-4-methoxybenzophenone (DHMB) and 2,3,4-trihydroxylbenzophenone (THB). Under optimal conditions, the analysis required 23 min and good linearity over the range of 10-2,500 ng/L in water and 100-25,000 ng/kg in soil for each UV filter obtained. The high recovery (62-114% and 60-125% for water and soil samples, respectively) and the low RSD values (less than 13.9 and 17.2% for water and soil samples, respectively) indicated the high performance of this method. The method detection limits (MDLs) were relatively low, ranging from 5 to 100 ng/L or kg and quantification limits ranged between 25 and 500 ng/L or kg for all test compounds. This validated method was applied in the analysis of seven BP-type UV filters collecting water and soil samples in Korea, between April and May 2003. The overall concentration of UV filters in the soil sample (500-18,380 ng/kg) was highly distributed in water sample (27-204 ng/L). The established method was successfully applied to monitor the residue measurement of the BP-type UV filters in environmental water and soil samples.  相似文献

This paper describes a method for the sensitive and selective determination of glyphosate, glufosinate and aminomethylphosphonic acid (AMPA) residues in water and soil samples. The method involves a derivatization step with 9-fluorenylmethylchloroformate (FMOC) in borate buffer and detection based on liquid chromatography coupled to electrospray tandem mass spectrometry (LC-ESI-MS/MS). In the case of water samples a volume of 10 mL was derivatized and then 4.3 mL of the derivatized mixture was directly injected in an on-line solid phase extraction (SPE)-LC-MS/MS system using an OASIS HLB cartridge column and a Discovery chromatographic column. Soil samples were firstly extracted with potassium hydroxide. After that, the aqueous extract was 10-fold diluted with water and 2 mL were derivatized. Then, 50 microL of the derivatized 10-fold diluted extract were injected into the LC-MS/MS system without pre-concentration into the SPE cartridge. The method has been validated in both ground and surface water by recovery studies with samples spiked at 50 and 500 ng/L, and also in soil samples, spiked at 0.05 and 0.5 mg/kg. In water samples, the mean recovery values ranged from 89 to 106% for glyphosate (RSD <9%), from 97 to 116% for AMPA (RSD < 10%), and from 72 to 88% in the case of glufosinate (RSD < 12%). Regarding soil samples, the mean recovery values ranged from 90 to 92% for glyphosate (RSD <7%), from 88 to 89% for AMPA (RSD <5%) and from 83 to 86% for glufosinate (RSD <6%). Limits of quantification for all the three compounds were 50 ng/L and 0.05 mg/kg in water and soil, respectively, with limits of detection as low as 5 ng/L, in water, and 5 microg/kg, in soil. The use of labelled glyphosate as internal standard allowed improving the recovery and precision for glyphosate and AMPA, while it was not efficient for glufosinate, that was quantified by external standards calibration. The method developed has been applied to the determination of these compounds in real water and soil samples from different areas. All the detections were confirmed by acquiring two transitions for each compound.  相似文献

Selenium was determined in samples with high copper and iron contents by hydride generation-inductively coupled plasma optical emission spectrometry (HG-ICP-OES) after flow-injection (FI) on-line copper and iron removal and selenium(VI) reduction. A Dowex 1X-8 anion-exchange microcolumn was used for the separation of selenium from copper and iron as their chloro-complexes. Se(VI) was then reduced on-line by heating a PTFE coiled reactor (150 cm long, 0.7 mm i.d.) in a 100°C water bath. After reduction of Se(VI), a 900-μl sample was injected into the carrier stream containing hydrochloric acid and sodium tetrahydroborate to generate the hydride. A limit of detection of 0.4 μg l−1 (RSD 2.3% for 20 μg l−1 selenium) was obtained. The application of the method to Geochemical Standard Reference Samples and copper metal reference materials (MBH) demonstrated that results were in good statistical accordance with certified values.  相似文献

A method for the simultaneous analysis of methadone in urine samples by microextraction in a packed syringe online with GC-MS (MEPS-GC-MS) is described. The new method reduced the sample handling and the detection limit by two- to seven-fold compared to published methods. Using a quantitation method based on the calculation of analyte concentration by comparison to an internal standard, we were able to measure methadone levels consistent with values reported for healthy individuals. The intra-assay precisions (RSD) of the method using quality control (QC) samples at three different concentration levels were about 11-14% (n = 6). The interassay precisions (RSD) were 11-15% for methadone in urine samples (n = 18). The accuracy varied from 89 to 109% for intra-assay (n = 6), and 97 to 107% for inter-assay (n = 18). The regression correlation coefficients (r(2)) were over 0.99 in all experiments.  相似文献

For human urine beryllium (Be), each sample (500 microl) was diluted (1+1) with Nash reagent (containing 0.2% (v/v) acetylacetone and 2.0 M ammonium acetate buffer at pH 6.0) and then a 20-microl volume of Triton X-100 (0.4%, v/v) aqueous solution was added. An aliquot (10 microl) of the diluted urine mixture was introduced into a graphite cuvette and was atomized according to a temperature program. The method detection limit (MDL, 3sigma) for Be was 0.37 microg/l in the undiluted urine sample and the calibration graph was linear up to 65.0 microg/l. Calibration graphs were prepared by the standard addition method. Accuracies of 98.6-102% were obtained when testing standard reference material (SRM 2670) freeze dried human urine samples. Precision (relative standard deviation, RSD) for urine Be was < or = 2.3% (withinrun, n = 5) and was < or = 3.0% (between-run, n = 3). For human urine and serum selenium (Se), samples (100 microl) were diluted with HNO3 (0.2%, v/v) to make a (1+1) dilution for urine analysis or a (1+4) dilution for serum analysis. An additional aliquot (10 microl) of Triton X-100 (0.1%, v/v) was added to each 200 microl of (1+1) diluted urine (or 20 microl of the Triton X-100 was added to each 500 microl of (1+4) diluted serum) sample. After the diluted sample mixture (10 microl) was introduced into a graphite cuvette, the corresponding chemical modifier (10 microl, containing Ni2+ + Pd + NH4NO3 in HNO3 (0.2%, v/v)) was added to it and the mixture was atomized. The MDL (3sigma) for Se in urine and in serum was 4.4 and 21.4 microg/l in undiluted sample, respectively, and the calibration graphs were linear up to 150 and 400 microg/l. Accuracies of urine Se were 98.9 - 99.4% by testing SRM 2670 (NIST) urine standards with RSD (between-run, n = 3) within 2.9%; and that of serum Se was 97.2% when testing a certified second-generation human serum (No. 29, #664) with RSD (between-run, n = 3) of 1.4%. The proposed method can be applied easily, directly, and accurately to the measurement of Be and Se in real samples (including six urine Se and four serum Se from patients of Blackfoot Disease in Taiwan).  相似文献
