New and very easily accessible monodentate phosphoramidite ligands have been developed that lead to excellent ee's and full conversions in the hydrogenation of (E)- and (Z)-beta-dehydroamino acid derivatives with both aliphatic and aromatic side chains. Particularly, two different catalytic systems were established for (E)-beta-(acylamino)acrylates (98-99% ee) and (Z)-beta-(acylamino)acrylates (92-95% ee) based on phosphoramidites 2 and 3, respectively.  相似文献

New monodentate chiral phosphonites were synthesized from enantiomerically pure 1,1'-spirobiindane-7,7'-diol. The phosphonites 2 were efficient ligands for the Rh-catalyzed asymmetric hydrogenation of alpha- and beta-dehydroamino acid derivatives, providing the amino acids in high enantioselectivities. The study of electronic effect showed that the electron-withdrawing substitutent on the P-phenyl ring of the phosphonite ligand dramatically decreased both the reactivity and enantioselectivity of the ligand.  相似文献

[reaction: see text] A new spirocyclic diol, 9,9'-spirobixanthene-1,1'-diol, was synthesized in two steps from readily available starting material m-phenoxyanisole. Resolution of the racemic diol was achieved by cocrystallization with N-benzylcinchonidinium chloride and N-benzylquininium chloride in acetonitrile. The corresponding spiro monodentate phosphoramidite ligand has been prepared for Rh-catalyzed asymmetric hydrogenation of alpha-dehydroamino acid derivatives and itaconic acid with excellent enantioselectivities (up to >99% ee).  相似文献

A new generation of monodentate phosphoramidite ligands bearing a primary amine moiety was found to display comparable or better efficiency than bisphosphines in the Rh-catalyzed asymmetric hydrogenation of challenging substrates, such as (Z)-methyl alpha-acetoxyacrylate or (E)-beta-aryl itaconate derivatives, affording the corresponding hydrogenation products with excellent enantioselectivities (up to >99% ee). The presence of intermolecular hydrogen bonding (HB) between two monodentate ligands in the catalyst was found to be critical for excellent catalyst performance. This finding provides a basis for design and development of further catalyst systems using this type of monodentate phosphoramidite ligands.  相似文献

The relatively inexpensive chiral monodentate phosphoramidite (S)-MONOPHOS may be used in combination with pyridines to prepare iridium complexes effective for catalysis of asymmetric imine hydrogenation with comparable enantioselectivity to some of those containing more costly chiral bidentate phosphines. [Ir(cod)((S)-MONOPHOS)(L)]BArF (cod = 1,5-cyclooctadiene; L = 3-methylisoquinoline, acridine, 2,6-lutidine, acetonitrile, or 2,3,3-trimethylindolenine; BArF = tetrakis[3,5-bis(trifluoromethyl)phenyl]borate) are efficient catalysts for the asymmetric hydrogenation of 2,3,3-trimethylindolenine. An important observation is that the catalyst containing acridine is more enantioselective than the catalyst derived from 2,3,3-trimethylindolenine which suggests that the other N-donor ligands are not readily displaced by the substrate during the catalytic cycle.  相似文献

[reaction: see text] Herein we describe a new protocol for catalyst evaluation in asymmetric hydroformylation reactions where multisubstrate screening is performed in an array of parallel reactors. This method was successfully demonstrated using a mixture of styrene, allyl cyanide, and vinyl acetate. Using this screening methodology, a set of phosphite ligands was evaluated and led to the discovery of a bisphosphite ligand that gave 88% ee and unprecedented >100:1 branched:linear regioselectivity in asymmetric hydroformylation of vinyl acetate.  相似文献

Herein, we report the automated parallel synthesis of solution-phase libraries of phosphoramidite ligands for the development of enantioselective catalysts. The ligand libraries are screened in situ in the asymmetric rhodium-catalyzed addition of arylboronic acids to aldehydes and imines. It is shown that the described methodology results in the straightforward discovery of leads for highly efficient enantioselective catalysts.  相似文献

Novel chiral monodentate phosphorus ligands, SIPHOS, were conveniently synthesized from 1,1'-spirobiindane-7,7'-diol. The Rh complexes of SIPHOS can catalyze the hydrogenation of alpha-dehydroamino esters in mild conditions, providing alpha-amino acid derivatives in up to 99% ee. Enamides and beta-dehydroamino esters can also be hydrogenated in good to excellent enantioselectivities (up to 99% and 94% ee, respectively). The SIPHOS ligand with smaller alkyl groups on the N-atom afforded higher enantioselectivity. The X-ray analysis of single crystal showed that the structure of Rh/SIPHOS catalyst is [Rh(COD)((S)-SIPHOS-Me)(2)](+), which clarified the configuration of the catalyst with the monodentate chiral phosphorus ligand in Rh-catalyzed asymmetric hydrogenation. A positive nonlinear effect in the relationship of the optical purities of ligand and product was observed in the hydrogenation of dehydroamino acid derivatives. The kinetic study of hydrogenation showed that the reaction is zero order in the concentration of substrate and first order in the concentration of Rh catalyst and the hydrogenation pressure. The rate of hydrogenation decreased when the Rh/L ratio changed from 1:1 to 1:4.  相似文献

Libraries of succinamic acid derivatives resulting from the condensation of a series of succinic acid derivatives with amines are reported as putative khafrefungin analogues. A total of 480 compounds derived from the initial condensation of 8 scaffolds with 60 different amines have been synthesized using automated technology with the help of scavenger resins. A simple acetate hydrolysis of five of the above sublibraries afforded additional 300 compounds for a total of 780 compounds. Around 55% of the library members showed purities higher than 70% (HPLC-ELS-MS) thus proving the generality of this approach. Results on growth inhibition of the yeast Saccharomyces cerevisiae in the presence of selected library members are also reported as a preliminary evaluation of the antifungal activity.  相似文献

A library of 20 monodentate phosphoramidite ligands has been prepared and applied in rhodium-catalyzed asymmetric hydrogenation. This resulted in the identification of two ligands, PipPhos and MorfPhos, that afford excellent and in several cases unprecedented enantioselectivities in the hydrogenation of N-acyldehydroamino acid esters, dimethyl itaconate, acyclic N-acylenamides, and cyclic N-acylenamides. A catalytic amount of Hantzsch ester that could be regenerated in situ by Ru complexes under hydrogen gas has been employed in the biomimetic asymmetric hydrogenation of benzoxazinones with up to 99% ee in the presence of chiral phosphoric acid. A series of diphosphine ClickFerrophos ligands (CF), based on a triazoleferrocene backbone, was synthesized in a four-step sequence via click chemistry methodology. In addition to the four previously synthesized ligands CF1, CF4, CF7 and CF10, six novel CF ligands CF23 and CF58 were prepared. Hydrogenation reactions of alkenes and ketones were significantly improved upon by using CF ligands as rhodium- or ruthenium-complexes in which the % ee values can be optimized by choosing the appropriate CF ligand depending on the substrate.  相似文献
