以多孔硅胶为基质,用改进的合成方法制备了Sinopak-s-DEAE高效弱阴离子交换色谱填。考察了反应条件对填料合成的影响,并以标准蛋白为样品进行了色谱行为的研究,结果表明:所制备的填料对蛋白质的分离性能良好,且对蛋白质的非特异性吸附小。  相似文献

Capillary-channeled polymer (C-CP) fibers extruded from nylon-6 are used as the stationary phase for the ion-exchange/reversed-phase mixed-mode chromatographic separation of a three protein mixture. The nylon-6 C-CP fibers are packed collinearly in a 250 x 1.5-mm i.d. column with an interstitial fraction of approximately 0.6. The effects of four displacing salts at three different pHs are studied with regards to protein retention time, peak width, selectivity, and resolution for a synthetic mixture consisting of myoglobin, ribonuclease A, and lysozyme to determine the optimum mobile phase conditions. The net charge model is found to be inadequate in fully explaining the retention behavior, as the proteins are retained by anion and cation-exchange interactions, as well as hydrophobic interactions with the stationary phase. It is found that pH and displacing salt strength had a significant influence on the retention properties and resolution of the proteins.  相似文献

A β‐cyclodextrin (β‐CD) bonded phase with diamine‐s‐triazine moiety was prepared. The separation and retention behavior of the isomers of five aromatic carboxylic acids, including toluic acid, aminobenzoic acid, nitrobenzoic acid, hydroxybenzoic acid, and naphthoic acid were investigated by a high‐performance liquid chromatography (HPLC) using the β‐CD bonded phase prepared. The influence of mobile phase pH in the range of 2.7‐3.6 on the retention of these analytes was examined. The isomers of the aromatic carboxylic acids, with the exception of nitrobenzoic acid, were optimally and effectively separated at pH 2.7, while the three isomers of nitrobenzoic acid could be well separated at pH 3.3. Compared with the chromatographic results obtained previously on the amine‐s‐triazine‐β‐CD bonded phase, the retention factors of the isomers of aromatic carboxylic acid on the diamine‐s‐triazine‐β‐CD bonded phase increase to a relatively much greater extent. Thus, the functionality of the spacer arm of the bonded phase playing an important role in the retention of aromatic carboxylic acid isomers is demonstrated. The results also imply that the hydrogen‐bonding interaction and the mechanism of anion exchange sorption as well may contribute significantly to the retention mechanisms.  相似文献

pISep is a major new advance in low ionic strength ion exchange chromatography. It enables the formation of externally controlled pH gradients over the very broad pH range from 2 to 12. The gradients can be generated on either cationic or anionic exchangers over arbitrary pH ranges wherein the stationary phases remain totally charged. Associated pISep software makes possible the calculation of either linear, nonlinear or combined, multi-step, multi-slope pH gradients. These highly reproducible pH gradients, while separating proteins and glycoproteins in the order of their electrophoretic pIs, provide superior chromatographic resolution compared to salt. This paper also presents a statistical mechanical model for protein binding to ion exchange stationary phases enhancing the electrostatic interaction theory for the general dependence of retention factor k, on both salt and pH simultaneously. It is shown that the retention factors computed from short time isocratic salt elution data of a model protein can be used to accurately predict its salt elution concentration in varying slope salt elution gradients formed at varying isocratic pH as well as the pH at which it will be eluted from an anionic exchange column by a pISep pH gradient in the absence of salt.  相似文献

A new stationary phase Al2O3/SiO2-10 was prepared and characterized by XPS, XRD, SEM and surface analysis. The anion exchanger properties of this new stationary phase were investigated by the separation of inorganic anions in ion chromatography (IC). pH of the mobile phase, concentration and strength of the Lewis base of the elute, and the organic modifier of the mobile phase strongly affect the separation of inorganic anions, and anion exchange selectivities of the analyte on the new support are significantly different from quaternary ammonium styrene based anion exchangers. The result of separation of inorganic anions shows that the new stationary phase provides excellent column efficiency, well-defined chromatographic peaks and favorable retention times.  相似文献

A novel type of ion exchanger was prepared by multipoint covalent binding of polystyrene chains onto the surface of porous silica followed by polymer-analogous modification of the bonded layer. Both anion and cation exchangers were synthesized and examined in the separation of nucleotides and proteins. Rapid and efficient separation of basic polypeptides on strong anion exchangers and that of acidic polypeptides on strong cation exchangers could be achieved. For the separation of complete mixtures of polypeptides the application of zwitter-ionic ion exchangers can be recommended.  相似文献

Oligonucleotides are used in many research areas. Thus, there is a need for their successful separation methods. Ion-exchange chromatography is the most popular separation technique, but it has limitations for these compounds. For this reason, new stationary phases are developed in order to increase separation selectivity. This study aimed to apply a series of dendrimer anion exchangers with various bonded layers to separate oligonucleotides by using different mobile phases to find conditions that allow sufficient separation. The number of anion-exchange layers, type of salt, and pH significantly impacted the oligonucleotide analysis. The developed chromatographic method was characterized by adequate selectivity for oligonucleotides differing in sequence length. It is essential to underline that the number of bonded layers appeared to have a significant influence, and the three layers appeared optimal. Based on our results, it may be concluded that the dendrimer stationary phases can be successfully used as an alternative to commonly applied packing materials in ion-exchange chromatography.  相似文献

Several aliphatic ionenes (2-6-, 6-6-, 10-6-ionene) have been prepared as ion exchangers for the development of novel high-performance stationary phases for anion chromatography (IC). A macroporous polystyrene/divinylbenzene (PS/DVB) resin with adjusted cation exchange capacity was used as support. Therefore the immobilization of ionenes to polystyrene carriers with remaining positive surface charge became possible for the first time. Strong ion-exchange interactions, resulting in high retention times, between the stationary phase and inorganic as well as organic anionic analytes have been observed. The influence of different ionenes on the retention behaviour during the ion chromatographic separation was investigated. Additionally, partly aromatic and polar ionene backbones were prepared and their retention behaviour as anion exchanger was investigated. The highest number of theoretical plates obtained was about 90.000 per meter. The signal asymmetries were generally lower than obtained for surface functionalized anion exchangers.  相似文献

The use of a phenylalanine (Phe) functionalized tentacle-type polymer coated capillary column for protein separation by open tubular capillary electrochromatography (OTCEC) was demonstrated in this work. The tentacle-type stationary phase was prepared from silanized fused-silica capillaries of 50 microm I.D. by glycidyl methacrylate graft polymerization and subsequent Phe functionalization. Due to the amphoteric functional groups of the Phe bonded on the tentacle-type polymer stationary phase, protein separation in the prepared column can be performed under both cathodic and anodic electroosmotic flow (EOF) by varying the pH values of the mobile phase. Model proteins including ribonuclease A (RNase A), myoglobin, transferrin, insulin were baseline separated under cathodic EOF with a mobile phase of pH 8.8. Comparison between the separation result of the four proteins under conditions of OTCEC and capillary zone electrophoresis indicates that the migration behavior of the four proteins in the prepared column was the result of the interplay of chromatographic retention and electrophoretic migration. Besides, three basic proteins including RNase A, cytochrome c (Cyt-c) and lysozyme (Lys) were fully resolved under anodic EOF with an acidic running buffer (pH 2.5).  系统研究了流动相中盐的性质和浓度、溶液 pH以及竞争配体对蛋白质在金属螯合色谱中保留值的影响。导出了描述蛋白质在金属螯合色谱中保留特征的数学表达式 ,提出用洗脱强度指数ε表征盐溶液的洗脱能力。根据不同色谱条件下蛋白质的保留特性 ,发现蛋白质在金属螯合色谱中的保留是配位、静电和疏水的协同作用。对与蛋白质强结合的金属螯合柱 ,以配位作用为主 ,静电作用为辅 ;对弱结合的金属柱 ,以静电作用为主 ,配位作用为辅。在流动相中加入高浓度非成络盐 ,可增强蛋白质和固定相间的疏水作用。  相似文献

合成了分离蛋白质的乙二醇-磷霉素钠改性氧化锆高效液相色谱固定相, 通过漫反射红外光谱、元素分析等分析方法对该固定相进行了表征. 以溶菌酶、核糖核酸酶A、细胞色素C和糜蛋白酶四种标准碱性蛋白质为探针, 系统地考察了固定相的疏水相互作用色谱性能. 结果表明, 乙二醇-磷霉素改性氧化锆固定相对蛋白质有一定的保留, 表现出较高的分离选择性.  相似文献

A type of mixed‐mode chromatography was integrated with high‐performance liquid chromatography for protein analysis and separation. The chromatographic behavior was tested using bovine serum albumin and lysozyme as model proteins. For the mixed‐mode column, the silica beads were activated with γ‐(2,3‐epoxypropoxy)‐propytrimethoxysilane and coupled with 4‐mercaptopyridine as the functional ligand. The effects of pH, salt, and the organic solvent conditions of the mobile phase on the retention behavior were studied, which provided valuable clues for separation strategy. When eluted with a suitable pH gradient, salt concentration gradient, and acetonitrile content gradient, the separation behavior of bovine serum albumin and lysozyme could be controlled by altering the conditions of the mobile phase. The results indicated this type of chromatography might be a useful method for protein analysis and separation.  相似文献

发展新型高效的亲水作用色谱分离材料对于极性化合物的分离分析具有重要的意义。本文设计合成了一种新型咪唑嵌合的氨基亲水作用色谱固定相(Sil-IEASP),并分别采用傅里叶变换红外光谱仪、热重分析仪和元素分析仪对该固定相进行了表征,结果表明该固定相制备成功。以核苷和核酸碱基为样品,分别考察了流动相中的水含量、盐浓度和pH对其保留的影响,结果表明所发展的固定相具有良好的亲水作用特性;此外,缓冲盐浓度和pH几乎不影响上述物质的保留。进一步将该固定相应用于分离尿嘧啶、腺嘌呤、胞嘧啶、尿苷和3种位置异构体(邻三联苯、间三联苯和苯并菲),与常用的氨基固定相相比,本文所发展的固定相具有更好的分离效果,有望在亲水作用色谱分离领域发挥潜在的应用价值。  相似文献

The ACCELL chromatographic media is a new packing designed specifically for the isolation and purification of proteins. The anionic and cationic functional groups are bonded to an encapsulated 40μ silica particle which can be readily packed into any size column. These columns can be operated on both high performance and medium performance liquid chromatographic equipment. The optimization of the separation conditions on the anion exchange media for the preparative purification of a monoclonal antibody (Mab) from ascites will be discussed as well as a multi-step purification of the enzyme prostatic acid phosphatase (PAP) from seminal fluids on the anion and cation exchangers.  相似文献

A glutamic acid-bonded silica (Glu-silica) stationary phase with cation-exchange properties was synthesized using l-glutamic acid as ligand and silica gel as matrix. The effects of solution pH value, salt concentration and metal ion on the retention of proteins were examined. The standard protein mixture was separated with a prepared chromatographic column and an iminodiacetic acid column, and compared. The influence of the binding capacity of an immobilized metal ion on the complexation of metal chelate column was studied. The results indicate that the obtained column displays cation-exchange characteristic and better separation ability for proteins. As fixing metal ion on the Glu-silica column, retention of proteins on the column is a cooperative interaction of metal chelate and cation-exchange. The stationary phase shows the typical metal chelate properties with the increase of the sorption capacity of immobilized metal ion.  相似文献

A procedure for the preparation of a monolithic column for weak cation exchange chromatography was presented. The structure of the monolithic column was evaluated by mercury intrusion. The hydrodynamic and chromatographic properties of the monolithic column--such as back pressures at different flow rates, effects of pH on protein retention, dynamic loading capacity, recovery, and stability--were determined under conditions typical for ion-exchange chromatography. The prepared monolithic column might be used in a relatively broad pH range from 4.0 to 12.0 and exhibited an excellent separation to five proteins at the flow rates of both 1.0 and 8.0 mL/min, respectively. In addition, the prepared column was first used in the purification and simultaneous renaturation of recombinant human interferon gamma (rhIFN-gamma) in the extract solution with 7.0 mol/L guanidine hydrochloride. The purity and specific bioactivity of the purified rhIFN-gamma in only one chromatographic step were obtained to be 93% and 7.8 x 10(7) IU/mg, respectively.  相似文献

A macrocyclic polyamine, 1,5,9,13,17,21,25,29-octaazacyclodotriacontane ([32]ane-N(8)), in the bonded phase was employed as a molecular receptor for CEC separation of oligopeptides. Parameters affecting the performance of the separations were considered. Baseline separation for the mixture of angiotensin I, angiotensin II, [Sar(1), Thr(8)]-angiotensin II, beta-casomorphin bovine, beta-casomorphin human, oxytocin acetate, tocinoic acid, vasopressin, and FMRF amide could be achieved using phosphate buffer (30 mM, pH 7) as the mobile phase. Column efficiency with average theoretical plate numbers of 69 000 plates/m and RSDs of <1% (n = 6) was achieved. [Met(5)]-enkephalin and [Leu(5)]-enkephalin, which have identical pI values and similar masses could be completely separated using acetate buffer (30 mM) with pH gradient (pH 3 inlet side and pH 4 outlet side). The results suggest that the mechanism for the peptide separation was mediated by a combination of electrophoretic migration and chromatographic retention involving anion coordination and anion exchange. After long-term use, the deviation of the EOF of the column after more than 600 injections was still within 6.0% of that for a freshly prepared column.  相似文献

A high-performance liquid chromatographic (HPLC) procedure has been evaluated to establish a routine test in the clinical laboratory for measuring the profiles of estrogen and progestin receptor isoforms in human breast and endometrial tumors. This procedure will be used to determine if there is a relationship between particular isoform profiles and response to various endocrine therapies. Evaluation of various HPLC modes has shown that high-performance ion-exchange chromatography (HPIEC) with silica-based anion exchangers offers a promising approach. In this paper, we have compared HPIEC columns of different lengths (10 and 25 cm) and pore sizes (300, 500 and 1,000 A) in order to obtain an optimal separation procedure. Because of receptor lability, all investigations were performed at 4 degrees C. The mobile phase consisted of 10-500 mM phosphate buffer, supplemented with the stabilizing agent, sodium molybdate at pH 7.4. Recoveries from each of the columns were between 70-100%. The length of the column did not influence significantly the retention time and salt concentration required for elution of receptor proteins. However, pore sizes appeared to alter these parameters. With a larger pore size (1,000 A), the retention of proteins was lower (elution with 50 mM phosphate) than that observed with the 500-A pore size column (elution with 100 mM phosphate) or of the 300-A pore size column (elution with 150 mM phosphate). Based solely on recovery patterns and peak shape, we conclude that separation of receptor isoforms on a 1,000-A, 25-cm column is best suited for clinical analysis.  相似文献
