Abstract— The absolute action spectrum has been determined for photoenzymatic splitting of cyclobutadipyrimidines ("pyrimidine dimers") from natural DNA, and from the synthetic polydeoxyribonucleotides poly(dA)·poly(dT) (forming only cyclobutadithymine) and poly(dG)·poly(dC) (forming only cyclobutadicytosine). These action spectra differ strikingly from each other, even when using the same enzyme preparations. On the other hand, the action spectrum for splitting cyclobutadithymine in natural DNA containing "dimers" of only this one type closely resembles the action spectrum for splitting the total mixture of "dimer" types in natural DNA, and is entirely different from the spectrum for splitting of the same photoproduct from poly(dA)·poly(dT). These results mean that the action spectrum is not simply the absorption spectrum of a chromophore carried by the photoreactivating enzyme, nor is it solely determined by the nature of the substrate photoproduct. It is at least partly determined by the over-all polynueleotide structure (viz. exact helical dimensions, pattern of neighboring bases to the "dimers," or both), affecting a ground state interaction between the enzyme and substrate in the enzyme-substrate complex.  相似文献

Abstract— Analytical modeling that interrelates the optical properties of multilayered structures is applied to the skin. The mathematical approach is based on relations of diffuse reflectance and transmittance of a multilayered system and the diffuse reflectance and transmittance of each component layer. The formula can also be derived from the Kubelka–Munk theory of radiation transfer. Using both collimated and diffuse incident irradiance, the applicability of the model to human epidermis over the UV and visible region has been verified. The model has been applied to calculate the absorption and scattering coefficients of human epidermis in vitro , and to estimate the epidermal transmittance under simulated in vivo condition.  相似文献

Excision repair of pyrimidine dimers was examined at the genome overall in three strains of hairless ( hr/hr ) and congenic wild-type mice, as well as in the expressed H- ras gene in hairless mice. The assay used a pyrimidine dimer-specific endonuclease from Micrococcus luteus and alkaline agarose gel electrophoresis. From 0 to 25% of endonuclease-sensitive sites were removed at the genome level in either hairy or hairless mice but about 50% were removed in the H- ras gene in hairless mice by 24 h after exposure to 5.4 J/cm2 UV (290-400 nm) irradiation. No differences were observed in the repair capacity between hairy and hairless mice, thus eliminating defective DNA repair as the explanation for the greater susceptibility to UV carcinogenesis in hairless mice.  相似文献

The phage T4 denV gene, coding for the pyrimidine-dimer specific T4 endonuclease V, was transfected into human repair-proficient fibroblasts, repair-deficient xeroderma pigmentosum fibroblasts, and into wild type CHO hamster cells. Transfectants maintained denV DNA and expressed denV mRNA. Purified T4 endonuclease V encapsulated in liposomes was also used to treat repair-proficient and -deficient human cells. The denV transfected clones and liposome-treated cells showed increased unscheduled DNA synthesis and enhanced removal of pyrimidine dimers compared to controls. Both denV gene transfection and endonuclease V liposome treatment enhanced post-UV survival in xeroderma pigmentosum cells but had no effect on survival in repair-proficient human or hamster cells. The results demonstrate that an exogenous DNA repair enzyme can correct the DNA repair defect in xeroderma pigmentosum cells and enhance DNA repair in normal cells.  相似文献

Abstract— Unscheduled DNA synthesis has been measured at several times during the differentiation of cultured rat skeletal muscle cells in response to exposures to 254nm UV light. Abstract— An aqueous effluent produced during the retorting of oil shale has been shown to induce a significant genotoxic response in cultured Chinese hamster (CHO) cells following activation by near ultraviolet light (UVA). In this report the light-activated responses induced by this complex mixture were compared between two DNA excision repair deficient mutants and their parental strain, CHO-AA8-4. The mutants, UV-5 and UV-135, were hypersensitive to both the cytotoxic and mutagenic effects of concurrent exposures to the retort process water and UVA. Repair proficiency appeared to render AA8-4 relatively insensitive to low doses of UVA treatment, whereas in the two mutants a linear dose-response in the induction of 6-thioguanine resistant (6TG®) mutations was observed even at the lower UVA doses examined. Filter DNA alkaline elution methods were utilized to demonstrate that both single-strand breaks and DNA-DNA interstrand crosslinks were induced in CHO DNA following process water and UVA treatment. Results were also obtained which indicated that the inability to repair DNA-DNA crosslinks contributed significantly to the hypersensitive response seen in the excision repair mutants following the photoactivation of this complex mixture.  相似文献

Abstract— Populations of radiation sensitive spores ( Bacillus subtilis UVSSP), vegetative bacteria ( E. coli K12-AB2480) and bacteriophage ( E. coli phage T4vx) have been considered as possible biological dosimeters to integrate DNA-absorbed solar energy incident on the Earth's surface.
Irradiation of spores of B. subtilis UVSSP with monochromatic far- and near-UV radiation and solar radiation have indicated that these radiations have a similar efficiency in inducing spore photoproducts per lethal event. Action spectra for lethality taken with the three radiation sensitive biological systems show a similar pattern in each case with a broad shoulder in the 334–365 nm wavelength region. This finding indicates a relatively high susceptibility of the DNA to chemical alteration in this wavelength range. Although less sensitive to sunlight than the other biological systems tested, the B. subtilis UVSSP spore mutant has the advantage of temperature independence of inactivation, stability between irradiation and assay and a simple, reproducible irradiation and assay procedure. Field measurements have supported the utility of this mutant as a sunlight dosimeter.  相似文献

用角分布XPS法研究了MOD法制得的YBCO膜在热处理过程中膜的表面元素浓度变化以及膜与村底ZrO_2之间的原子扩散和固态化学反应。结果表明无论是薄膜(约0.1μm)和较厚的膜(约1~1.5μm),在大约530~720℃的温度范围内加热后都发生铜表面富集和钡表面浓度偏低。在800℃以上加热后铜的表面浓度显著降低,温度愈高,降低愈甚。膜与衬底之间的化学反应也随温度升高而加剧。例如薄膜在890℃加热后钡向ZrO_2衬底扩散,膜中的铜仍以 2价为主;在950℃加热后衬底表面生成了富钡层,而铜则主要以 1价的形式存在于富钡层表面。与厚膜相比,在800℃以上薄膜与衬底的原子扩散和固态化学反应对于膜超导电性的损害更显著。  相似文献

Abstract— Continuous DNA degradation and resynthesis, without a net change in cellular DNA content, were observed in buffer-held, non-irradiated E. coli B/r. This constant DNA turnover probably involves most of the genome and reflects random sites of DNA repair due to the polA-dependent excision-resynthesis repair pathway. Under these non-growth conditions, it appears that at any given time there is a minimum of one repair site per 6.5 × 106 daltons DNA, each of which is at least 160 nucleotides long.
While the amount of DNA degradation is not influenced by prior exposure to UV radiation, the synthetic activity decreases with increasing UV fluence. We suggest that when sites of DNA turnover occur opposite to cyclobutyl dipyrimidines in UV-irradiated cells, repair of the latter damage can be prevented. This implies that both beneficial and deleterious processes take place in irradiated buffer-held cells, and that cell survival depends on the delicate balance between DNA turnover and repair of UV-damage. Based on these findings, we propose a model to explain the limited repair observed during post-irradiation liquid-holding and to account for the large difference in cell survival between irradiation at low fluence rates (fluence-rate dependent recovery) and at high fluence rates followed by liquid-holding (liquid-holding recovery).  相似文献

Abstract— In stationary phase, strains of Escherichia coli deficient in excision (B/r Her) or recombination repair (K.12 AB2463) were more sensitive than a repair proficient strain (B/r) to monochromatic near-ultraviolet (365 nm) and visible (460 nm) radiations. The relative increase in sensitivity of mutants deficient in excision or recombination repair, in comparision to the wildtype, was less at 365 nm than at 254 nm. However, a strain deficient in both excision and recombination repair (K12 AB2480) showed a large, almost equal, increase in sensitivity over mutants deficient in either excision or recombination repair at 365 nm and 254 nm. All strains tested were highly resistant to 650 nm radiation. Action spectra for lethality of strains B/r and B/r Her in stationary phase reveal small peaks or shoulders in the 330–340, 400–410 and 490–510 nm wavelength ranges. The presence of 5μg/ml acriflavine (an inhibitor of repair) in the plating medium greatly increased the sensitivity of strain B/r to radiation at 254, 365 and 460 nm, while strains E. coli B/r Her and K12 AB2463 were sensitized by small amounts. At each of the wavelengths tested, acriflavine in the plating medium had at most a small effect on E. coli K.12 AB2480. Acriflavine failed to sensitize any strain tested at 650 nm. Evidence supports the interpretation that lesions induced in DNA by 365 nm and 460 nm radiations play the major role in the inactivation of E. coli by these wavelengths. Single-strand breaks (or alkali-labile bonds), but not pyrimidine dimers are candidates for the lethal DNA lesions in uvrA and repair proficient strains. At high fluences lethality may be enhanced by damage to the excision and recombination repair systems.  相似文献

Abstract. –A large difference in survival occurs between buffered suspensions of E. coli irradiated with UV radiation at a low fluence rate and those irradiated at a high fluence rate. For sufficiently large fluences, the extent of this fluence rate dependent recovery (FRR) is about two orders of magnitude greater than that which can be brought about by liquid holding recovery (LHR) following high fluence rate irradiation in most of the E. coli strains studied. LHR and FRR occur in excision resynthesis repair proficient (ERR+) but not ERR- strains of E. coli , although its observation can be masked in strains with complete repair potential upon subsequent growth on nutrient plates. Accumulation of DNA strand interruptions and excision of cyclobutyl dipyrimidine occur during LHR and FRR but are more extensive for the latter. Our data suggest mat events beyond incision and excision occur during LHR and FRR, but differences in the extent of ERR during LHR and FRR cannot account for the difference in cell survival between these two phenomena.  相似文献

Abstract— The effects of selected DNA intercalating and non-intercalating drugs on the UV excision repair process in human fibroblasts have been examined. 9-Amino acridine, acridine orange, quinacrine, doxorubicin (adriamycin), ethidium bromide and actinomycin-D all inhibited the removal of pyrimidine dimers from cellular DNA by inhibiting the incision process as monitored by the nick translation assay and by an endonuclease-sensitive site assay. These agents also partially inhibited incision by the M. luteus endonuclease in an in vitro system. This is the only class of compounds tested to date that appears to block this early step of repair in mammalian cells. The DNA topoisomerase inhibitors, m -amsacrine and VP-16 (etoposide) and the bacterial gyrase inhibitors nalidixic acid and oxolinic acid were shown not to inhibit UV repair. As shown previously, however, novobiocin does block dimer removal and we show here that it is a potent inhibitor of the M. luteus UV endonuclease. While it has recently been demonstrated that many DNA intercalating agents block the strand-passing activity of DNA topoisomerase II giving rise to protein associated DNA strand breaks, the finding that the specific inhibitors of topoisomerase, m -AMSA and VP-16, do not inhibit repair, even though they block this strand passing activity, strongly suggests that inhibition of DNA topoisomerase is not associated with inhibition of DNA repair.  相似文献

Abstract— A mutant cell line, DRP 287, sensitive to solar UV radiation and deficient in the repair of solar UV-induced nondimer DNA damage, was derived from ICR 2A frog cells. These cells were transfected with human DNA and a secondary transformant obtained in which normal solar UV sensitivity was restored and the repair defect corrected. The DNA from this secondary transformant was used to construct a genomic DNA library from which a recombinant phage was isolated containing the human gene capable of restoring normal solar UV sensitivity and correcting the repair defect in the DRP 287 cells. This represents the first human gene which has been isolated that is specifically involved in the repair of nondimer DNA damage induced by solar UV radiation. It has been designated SUVCC1 to denote solar UV cross-complementing gene number 1.  相似文献

A COMPARISON OF IN VIVO AND IN VITRO TESTING OF SUNSCREENING FORMULAS   总被引:2,自引:0,他引:2  
Abstract— Seven commercially available sunscreens were compared by three different methods. Absorbance spectra were measured for each product in isopropanol solution and also on hairless mouse epidermis. In vivo tests were performed on human volunteers using a Xe arc solar simulator. Sun Protection Factors (SPF) were calculated by each method for each product tested and the results compared. By all methods used, the combination of 7% octyl dimethyl para-aminobenzoic acid and 3% oxybenzone provided the most protection from U.V. light. While estimates of the effectiveness of all products were much too high when calculated by the isopropanol solution method, the hairless mouse epidermis technique seems to be an accurate tool for predicting product efficacy in vivo .  相似文献
