Synthesis of thiopyrano[4″,3″:4′,5′]pyrido[3′,2′:4,5]furo[3,2-d]pyrimidines

<正>Reactions of the 6-hydroxy-thiopyrano[3,4-c]pyridine-5-carbonitrile derivative 1 withα-halo-carbonyl compounds gave the ortho-substituted intermediates 2a-c which were converted into furo[2,3-b]thiopyrano[4,3-d]pyridines 3a-c by fusion of a furan moiety under basic conditions.Further cyclization of 3a-c led to a fusion of a pyrimidine ring,yielding the tetracyclic products 6,7 and 8.In addition,condensation of 6 with various aromatic aldehydes afforded the corresponding imines 9a,b.Mannich reaction of 7 gave products 10a,b.     

Synthesis and antitumor evaluation of novel tetrahydrobenzo[4′,5′]thieno[3′,2′:5,6]pyrimido[1,2-b]isoquinoline derivatives

In the present study, a novel 8,9,10,11-tetrahydro-7H,14H-benzo[4′,5′] thieno[2′,3′:4,5]-1,3-oxazino[3,2-b]isoquinoline-7,14-dione 5 was prepared by condensation of 2-amino-3-carbethoxy-4,5,6,7-tetrahydrobenzothiophene with homophthalic anhydride under microwave irradiation, followed by alkaline hydrolysis and cyclization using acetyl chloride. Compound 5 was further allowed to react with different nitrogen nucleophiles to get new tetrahydrobenzothienopyrimido isoquinolinone derivatives. The structures of the prepared compounds were elucidated by IR, ¹H-NMR, ¹³C-NMR, and mass spectroscopy. The newly prepared compounds were tested in vitro against a panel of two human tumor cell lines, namely, hepatocellular carcinoma (liver) HepG2, and mammary gland breast MCF-7. Almost all the tested compounds showed satisfactory activity.     

Colorimetric Cyanide Chemosensor Based on 1′,3,3′,4-Tetrahydrospiro[chromene-2,2′-indole]

A new class of chemosensors based on the 1′,3,3′,4-tetrahydrospiro[chromene-2,2′-indole] ring system, which detects cyanide with high specificity, is described. These chemosensors show a distinct color change when treated with cyanide in acetonitrile solution buffered with sodium phosphate, and this procedure is not affected by the presence of other common anions. The chemisensors exhibit high sensitivity to low concentrations of cyanide, meeting the European Union water quality control criterion of sensitivity below 0.05 mg L⁻¹, and show a very fast response within tens of seconds. The mechanism for detection is rationalized by the nucleophilic substitution of the phenolic oxygen atom at the indoline C-2 atom by the cyanide anion to form a stable indolylnitrile adduct and to generate the colored 4-nitrophenolate chromophore. These chemosensors can be synthesized by a simple procedure from commercially available starting materials.     

Photochromic and electrochromic properties of 1′,3′,3′-trimethyl-6-piperidino- and -morpholinospiro[indoline-2′,3-3H-naphtho[2,1-b][1,4]-oxazines]

Electrochromic properties of 1,3,3-trimethyl-6-piperidino- and-morpholinospiro[indoline-2,3-3H-naphtho[2,1-b][1,4]-oxazines] resulting from their ionization and the formation of intermediate products were studied. The characteristic features of the photochromic behavior of these compounds are positive solvatochromism for absorption spectra of a colored form and an increase in the rate constant for thermobleaching with increasing solvent polarity.Translated fromIzvestiya Akademii Nauk. Synthesis of 3,4-Biheterylthieno[2,3-b]-thiophenes. Part I: Synthesis of 3,4-Bi(1′,3′,4′-thiadiazolyl-, s-triazolyl-,1′,3′,4′-thiadiazinyl-, 1′,3′,4′-triazinyl-, thiazolyl-, 1′,3′-thiazinyl- and primidinyl)-thieno[2,3-b]thiophenes

3,4-Diamino-2,5-dicarbethoxythieno[2,3-b]thiophene ( 1 ) was allowed to react with NaNO 2 and active methylenes to afford the corresponding azo compounds 2a-c . Hydrazonyl chloride 2a was treated with carbon disulfide, phenyl isothiocyanate, benzonitrile, benzyl cyanide, malononitrile, benzalaniline, ethyl mercaptoacetate, and ethyl glycinate to give 1,3,4-thiadiazolyl-, s-triazolyl-, 1,3,4-thiadiazinyl-, 1,3,4-triazinylthieno[2,3-b]thiophenes 3-6 respectively. The reaction of 2b,c with urea, thiourea, and guanidine afforded pyrimidinyl- and thiazinylazothieno [2,3-b]thiophenes 7-10 respectively. Bithiazolylthieno[2,3-b]thiophenes 11 and 13 were synthesized by treating compound 1 with CS 2 along with halo compounds. The addition of S,S-, N,S-, and N,O-acetals to the Schiff base 14 afforded compounds 15-17 respectively.     

Electronic structure of diindeno[1,2,3-cd:1′,2′,3′-lm]perylene radical anion

The electronic structure of diindeno[1,2,3-cd:1′,2′,3′-lm]perylene (periflanthene) radical anion has been studied on the basis of the ESR, electronic absorption, and linear dichroism spectra. The observed results are interpreted in terms of the SCF MO calculations within the framework of the π-electron approximation.

Summary 8,9-Diphenylpyridazino[4,3:4,5]thieno[3,2-d]-1,2,3-triazin-4(3H)-one (2), 3-substituted 8,9-diphenylpyridazino[4,3:4,5]thieno[3,2-d]-1,2,3-triazin-4(3H)-ones (3a–c), 3,4-diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazin-8(7H)-one (4), 8-chloro-3,4-diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazine (5), 8-substituted 3,4-diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazines (6a–h) and 7-substituted 3,4-diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazin-8(7H)-ones(7a–c) were synthesized from 5-amino-3,4-diphenylthieno[2,3-c]pyridazine-6-carboxamide (1).

Zusammenfassung Folgende Verbindungen wurden ausgehend von 5-Amino-3,4-diphenylthieno[2,3-c]pyridazin-6-carboxamid (1) synthetisiert: 8,9-Diphenylpyridazino[4,3:4,5]thieno[3,2-d]-1,2,3-triazin-4(3H)-on (2), 3-substituierte 8,9-Diphenylpyridazino[4,3:4,5]thieno[3,2-d]-1,2,3-triazin-4(3H)-one (3a–c), 3,4-Diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazin-8[7H]-on (4), 8-Chlor-3,4-diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazin (5), 8-substituierte 3,4-Diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazine (6a–h) und 7-substituierte 3,4-Diphenylpyrimido[4,5:4,5]thieno[2,3-c]pyridazin-8(7H)-one (7a–c).

     

Structural and Spectroscopic Studies of p-Nitrophenolato[2,2′,6′,2′′-Terpyridyl]-Copper(II) Perchlorate

The X-ray structure and low temperature electronic spectra of the title complex, 1, are presented, It crystallized in the monoclinic space group P2₁/c, with a = 10.387(1), b = 13.103(1), c = 16.153(2)Å, β = 98.59(1)°, V =2174(2) ų, Z =4 and refined to R(R _w) = 0.039(0.054) for 1980 reflections. The structure consists of monomeric Cu(lI) complexes having nearly square planar N₃(terpy)O(phenolate) equatorial ligation and weak axial ligation to a single perchlorate oxygen atom. An electronic absorption at 26,300 cm^-1 is consistent with phenolate oxygen → Cu(lI) LMCT while a second band at ca 14,300 cm^-1 is assigned to a ligand field transition.     

New convenient synthesis of substituted 6,9-dihydropyrido[3′,2′:4,5]thieno[3,2-b]pyridines and 6,9-dihydropyrido[3′,2′:4,5]thieno[3,2-d]pyrimidines

1,4-Dihydro-3-cyano-2-pyridinethiolates react with 2-bromo-l-(4-bromophenyl)ethylidenemalononitrile or N-cyanochloracetanudine to give 6, 9-dihydropyrido[3, 2': 4, 5]thieno[3. 2-b]pyridines or 6, 9-dihydropyrido-[3.2:4.5Jthieno(3,2-d]pyrimidines, respectively.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 553–556, April, 1996.     

Dipyrrolo[1,2-a; 2′,1′-c]pyrazines. 5. Azo coupling of dipyrrolo[1,2-a;2′,1′-c]pyrazines and 5,6-dihydrodipyrrolo[1,2-a; 2′,1′-c]pyrazines

We have synthesized a series of previously unreported azo dyes via the azo coupling of alkyl substituted dipyrrolopyrazines with aryldiazonium chloride. For this type of substrate where one or both -positions of the pyrrole rings of the molecules are not occupied by substituents, electrophilic attack was found to occur initially at carbon atom C₍₃₎.For communication 4 see [1]M. Dipyrrolo[1,2-a; 2′,1′-c]pyrazines. 4. Aminomethylation of alkyl substituted dipyrrolo[1,2-a; 2′,1′-c]pyrazines and 5,6-dihydrodipyrrolo[1,2-a; 2′,1′-c]pyrazines

We have studied the aminomethylation reaction of alkyl substituted dipyrrolo[1,2-a; 2,1-c]pyrazines and their 5,6-dihydro analogs using different aminomethylating agents. Use of alkoxydialkylaminomethanes (aminoacetals) as Mannich reagents leads to the highest yields of the aminomethylated dipyrrolopyrazines. Synthesis and THF Accompanied Crystal Structure of 11H-Naphtho[1′,2′,:5,6]-pyrano[2,3-d]pyrimidin-11(10H)-one

8,9,12-Trihydro-9,9-pentamethylene-12-(3-nitrophenyl)-11H-naphtho[1′,25,6] pyrano[2,3-d]pyrimidin-11(10H)-one 1, another conversion product of Friedl(a)ndler reaction, has been synthesized by the reaction of 2-amino-4-(3-nitrophenyl)-4H benzo[f]chromene-3-carbo- nitrile 2 with cyclohexanone in the presence of Lewis acid catalysts. The crystal of the title compound 1 THF solvate, C60H62N6O10, was obtained and determined by X-ray diffraction me- thod. The crystal is of triclinic, space group P with a = 11.4633(11), b = 12.6247(12), c = 19.658(2)(A), α = 72.642(8), β = 89.045(9), γ = 68.340(5)°, V = 2509.6(4), Z = 2, Dc = 1.359 g/cm3, F(000) = 1088, Mr = 1027.16, the final R = 0.0674 and wR = 0.1869 with I > 2σ(I) and the goodness-of-fit S = 1.045 on F2. The pyrimidine ring has an envelope conformation, linked with a six-membered ring through a spiro C atom. The crystal packing of 1 THF solvate is stabilized by N-H…O hydrogen bonds, and π-π stacking interaction occurs between two adjacent nearly planar molecules of 1.     

Efficient and mild one-pot synthesis of (E)-8′-arylidene-5′,6′,7′,8′-tetrahydrospiro[oxindole-3,4′-pyrano[3,2-c]pyridin] derivatives with potential antitumor activity

A mild and efficient cyclization procedure for the synthesis of (E)-8′-arylidene-5′,6′,7′,8′-tetrahydrospiro[oxindole-3,4′-pyrano[3,2-c]pyridin] derivatives was achieved via one-pot three-component condensation of isatins, malononitrile and (E)-3-arylidene-1-methylpiperidin-4-ones using piperidine as an efficient catalyst and ethanol as an environmentally benign solvent. The in vitro antitumor activity of these compounds was evaluated in human cervical carcinoma cell line (Hela), human liver hepatocellular carcinoma cell line (HepG2), and human breast carcinoma cell line (MDA-MB-231).     

Synthesis of novel spiro-fused pyrazolo[4′,3′:5,6]pyrido[2,3-d]pyrimidines

New pyrazolo[4′,3′:5,6]pyrido[2,3-d]pyrimidines, with an associated spiro-3,3′-oxindole attachment, were prepared by three-component combinations of 5,6-dihydro-4H-pyrrolo[3,2,1-ij]quinoline-1,2-dione with a pair of reactants chosen from a pyrazol-5-one, a pyrazole-5-amine, a barbituric acid, or a 6-aminouracil.     

Molecular structure of 1′,3′,3′-trimethyl-6-trifluoromethylsulfonyl-spiro-(indoline-2,2′-benzo[b]pyran)

Photochromic 1, 3, 3 -trimethyl-6-trifluoromethylsulfonyl-spiro(indoline-2,2-ben-zo[b]pyran) (1) was studied by X-ray diffraction analysis. In compound1, the C_spiro-O bond (1.49(1) Å (average)), broken on photoexcitation, is the longest of all the indoline spiropyrans studied.Translated fromIzyestiya Akademii Nauk. Synthesis and structure of 3,7-dihydro-2h-1,2,4-triazolo-[1′,5′-a′]tripyrimido[4,5-d]benzo[bpyrans

The condensation of 5-(2-hydroxyphenyl)-6, 7-dihydro-1, 2, 4-triazolo[l, 5-a]pyrimidines with aldehydes yields derivatives of a new heterocyclic system, namely, 3,7-dihydro-2H-1,2,4-triazolo[1,5-a]pyrimido[4,5-d]-benzo[b]pyran, including the 9-methyl-2,3-diphenyl derivative, whose structure was established by x-ray diffraction structural analysis.*Deceased.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 2. pp. 240–246, February, 1996.
