The chromatographic behaviors of lanthanide chelates of acetylacetone (ACAC), trifluoroacetylacetone (TFA), thenoyltrifluoroacetone (TTA), dipivaloylmethane (THD), 6,6,7,7,8,8,8-heptafluoro-2,2-dimethyl-3,5-octanedione (FOD), and the thenoyltrifluoroacetonepyridine (TTA·Py) adduct were investigated using packed column supercritical fluid chromatography. Mobile phases consisting of neat and alcohol modified CO2 were used with a phenyl packed stationary phase. Lanthanide complexes of ACAC, THD, and FOD were shown to have better chromatographic performance compared to the corresponding chelates with TFA, TTA, and TTA·Py. In particular, TTA complexes such as Eu(TTA)3 showed characteristic thermal decomposition in the mobile phase at elevated temperature. In addition, retention behavior was found to be a temperature dependent function of volatility and solubility for all chelates studied.  相似文献

An efficient two‐step method has been developed for the separation of β‐cypermethrin stereoisomers by supercritical fluid chromatography with polysaccharide chiral stationary phases. With respect to retention, selectivity, and resolution of β‐cypermethrin, the effects of chiral stationary phases, cosolvents, mobile phases, and column temperature have been studied in detail. Through a two‐step separation, β‐cypermethrin was firstly separated by using a cellulose‐derived chiral stationary phase to obtain two stereoisomeric pairs, and further resolved on an amylose‐based chiral stationary phase to produce four enantiopure stereoisomers. The electronic circular dichroism patterns of the first‐ and the third‐eluted isomers in methanol solution showed the mirror image of each other in the wavelength range 200∼300 nm, indicating that they were a pair of enantiomers. Moreover, the second‐ and the fourth‐eluted isomers were also enantiomers. This proposed two‐step strategy showed low solvent consumption, fast separation speed, and high‐purity, which may provide an effective approach for preparative separation of compounds with multiple chiral centers and difficult‐to‐separate multicomponent samples.  相似文献

A method is described to separate α‐ from β‐arylalanines by ligand exchange chromatography on a nickel nitrilotriacetate agarose column with UV monitoring of the effluent. Separate mixtures containing an α‐ and β‐arylalanine pair (1 mg of each) were individually loaded onto the nickel resin pre‐equilibrated with the mobile phase at room temperature, and the amino acids were eluted from the column with a gradient from pH 12.0–8.0. The β‐arylalanines eluted first, followed by the α‐isomers. The four α/β‐amino acid pairs tested were well separated with baseline resolution. An aliquot of each fraction was chemically treated to derivatize the amino acids to their N‐acyl methyl ester analogs, and their identities were confirmed by GC/MS analysis. The sample recovery was quantitative (>98%), and the column matrix was very resilient, as demonstrated by consistent separation of the solutes after ~100 preparative cycles.  相似文献

Two amino acid phosphoramidates with high antiviral activity were used as model compounds and separated using supercritical fluid chromatography on an achiral Hypersil BDS cyano column (250 × 4.6 mm, 5 µm). Supercritical CO2 was used as a mobile phase with different co-solvents, including methanol, ethanol, and 2-propanol. Several key processing parameters were evaluated, including the type and concentration of alcohol modifier (5 to 15%), backpressure (100 to 200 bar), and column temperature (30 to 45 ?C) in terms of their impact on retention factor, diastereoselectivity, and resolution. The optimized chromatographic conditions allowed for a complete separation of mixture of two diastereomers with good resolution over a short time (5–9 min). At a backpressure of 150 bar and a temperature of 33°C, the diastereomers of phenylalanine methyl ester phenyl 5′-phosphoamidate of d4T (d4T-P-N-PheOMe) and alanine methyl ester phenyl 5′-phosphoamidate of d4T (d4T-P-N-AlaOMe) were separated with resolutions of 4.43 and 2.55 using 10.0% and 5.0% (v/v) methanol, respectively.  相似文献

采用超临界流体色谱法(SFC),在多糖固定相Chiralpak IA、IB、IC、ID、IE和IF上成功拆分了11种手性化合物。分离结果表明,这6支手性色谱柱对这些手性化合物具有良好的手性识别互补性,均可以在10 min之内得到良好的分离结果,具有较好的实用性。改性剂甲醇、乙醇和异丙醇对手性化合物的保留时间以及手性选择性均具有良好的调节作用,需要根据不同手性物质在手性柱上的分离情况加以区别,选择使用,并调节改性剂至合适的比例。针对键合型固定相溶剂通用性的特征,特殊改性剂的应用也有助于优化手性分离。  相似文献

A significant uptake of supercritical mobile phases into immobilized stationary phases has been observed. The phase swelling effect has been evaluated hydrodynamically for SE-30, SE-54, and cyanopropyl silicone phases with respect to the mobile-phase density of supercritical butane, column temperature, and the presence of a polar retention moderator. 建立了基于反相液相制备色谱和超临界流体制备色谱的组合方法,用于分离纯化醇提水沉后石油醚层中的海风藤。首先以甲醇作为改性剂,采用醇提水沉法去除海风藤甲醇提取物中的叶绿素,加入硅藻土后用石油醚回流富集目标成分。选用反相C18制备色谱柱将其分为18个组分,然后将组分在SFC模式下进行制备。选用酰胺色谱柱,以甲醇为改性剂,在柱温30℃、背压15.0 MPa的条件下进行分离。基于反相色谱和超临界流体色谱不同的分离选择性,最后分离得到6个高纯度化合物。该法展示了反相制备色谱和超临界流体制备色谱在海风藤分离纯化方面的优势,特别是超临界流体色谱在天然产物的分析和制备方面的巨大潜力。  相似文献

Enantiomeric separations of several β-amino alcohol drugs, i.e., phenylephrine, epinephrine, norepinephrine, synephrine, and chlorprenaline were performed by capillary electrophoresis using DM-β-CD as a chiral selector. Five test solutes were baseline resolved in six minutes. The effects of DM-β-CD concentration, pH value, ionic strength of the buffer, and the type of β-CDs on resolution were investigated. The results indicated that DM-β-CD is suitable for enantiomeric separation of β-amino alcohols containing a phenyl group on the chiral atom. Enantiorecognition mechanisms for test solutes are also discussed.  相似文献

Direct analysis of polar solutes by supercritical fluid chromatography (SFC) has been somewhat limited because the mobile phases in common use are all relatively nonpolar. Derivatization is demonstrated as a viable means for facilitating the SFC separation of sugars. Oligo- and polysaccharides containing up to 18 glucose units are completely resolved by capillary SFC.  相似文献

The study on retention behavior in supercritical fluid chromatography (SFC) is necessary to understand the mechanism of the various interactions in SFC. The retention of SFC in carboxylic acid methyl ester/polymethylsiloxane/CO2 system was studied systematically and the retention behavior of this kind of compounds under various typical operation conditions was described using the method of an alternative unified theory of chromatographic retention. The results illustrated that expression: Ink.= a + b/T + cp + dp/T + ep2/T can be used to describe quantitatively the retention behavior of carboxylic acid methyl ester/polymethylsiloxane/CO2 system in the ranges of reduced density from 0.549 to 1.411. It was also found that the entropy of solute in stationary phase is dependent on the density of supercritical fluid (SF) under typical operating conditions of SFC.  相似文献

A series of β‐bromoketones and β‐chloroketones were synthesized by the addition reactions of α,β‐unsaturated ketones under BX3 (X = Br, Cl) and ethylene glycol reaction system.
