Two polypyridyl ligands 6-fluro-3-(1H-imidazo [4,5-f] [1,10]-phenanthroline-2-yl)-4H-chromen-4-one (FIPC), 6-chloro-3-(1H-imidazo [4,5-f] [1,10]-phenanthroline-2-yl)-4H-chromen-4-one (ClIPC) polypyridyl ligands and their Ru(II) complexes [Ru(bipy)2FIPC]2+(1), [Ru(dmb)2FIPC]2+(2), [Ru(phen)2FIPC]2+(3), [Ru(bipy)2ClIPC]2+(4), [Ru(dmb)2ClIPC]2+(5) and [Ru(phen)2ClIPC]2+(6) ((bipy = 2,2′-bipyridine, dmb = 4,4′-dimethyl-2,2′-bipyridine and phen = 1,10-phenanthroline) have been synthesised and characterised by elemental analysis, Mass spectra, IR, 1H and 13C-NMR. The DNA-binding of the six complexes to calf-thymus DNA (CT-DNA) has been investigated by different spectrophotometric, fluorescence and viscosity measurements. The results suggest that 1–6 complexes bind to CT-DNA through intercalation. The variation in binding affinities of these complexes is rationalised by a consideration of electrostatic, steric factors and nature of ancillary ligands. Under irradiation at 365 nm, the three complexes have also been found to promote the photocleavage of plasmid pBR 322 DNA. Inhibitor studies suggest that singlet oxygen (1O2) plays a significant role in the cleavage mechanism of Ru(II) complexes. Thereby, under comparable experimental conditions [Ru(phen)2FIPC]2+(3), [Ru(phen)2ClIPC]2+(6) cleaves DNA more effectively than 1, 2, 4 and 5 complexes do. The Ru(II) polypyridyl complexes (1–6) have been screened for antimicrobial activities.  相似文献

Based on the a ligand BDPPZ [(9a,13a‐dihydro‐4,5,9,14‐tetraaza‐benzo[b]triphenylene‐11‐yl)‐phenyl‐methanone] (1) and its polypyridyl hetero‐ and homoleptic Ru(II) metal complexes, [Ru(bpy)2L](PF6)2 (2), [Ru(phen)2L](PF6)2 (3), [Ru(dafo)2L](PF6)2 (4), [Ru(dcbpy)2L](PF6)2 (5) and [RuL3](PF6)2 (6) (where, L = ligand, bpy = 2,2′‐bipyridine, phen = 1,10‐phenantroline, dafo = 4,5‐diazafluoren‐9‐one and dcbpy = 3,3′‐dicarboxy‐2,2′‐bipyridine), have been synthesized and characterized by elemental analysis, UV–vis, FT‐IR, 1H and 13C‐NMR spectra (for ligand), molar conductivity measurements and X‐ray powder techniques. The electrochemical parameters of the substituted ligand and its polypyridyl hetero‐ and homoleptic Ru(II) metal complexes are reported by cyclic voltammetry. UV–vis spectroscopy is used to compare the differences between the conjugated π systems in this ligand and its Ru(II) metal complexes. The polypyridyl hetero‐ and homoleptic Ru(II) metal complexes also tested as catalysts for the formation of cyclic organic carbonates from carbon dioxide and liquid epoxides which served as both reactant and solvent. The results showed that the [Ru(L)3](PF6)2 (6) complex is more efficient than the other Ru(II) complexes for the formation of cyclic organic carbonates from carbon dioxide. An intercalative ligand, ppip (ppip = {2-(4-(piperidin-1-yl)phenyl)-1H-imidazo[4,5-f][1,10]phenanthroline}), and its mononuclear Ru(II) polypyridyl complexes, [Ru(phen)2(ppip)]2+ (1) (phen=1,10-phenanthrolene), [Ru(bpy)2(ppip)]2+ (2) (bpy=2,2′-bipyridine) and [Ru(dmb)2(ppip)]2+ (3) (dmb=4,4′-dimethyl-2,2′-bipyridine), have been synthesized and characterized by elemental analysis and spectroscopic techniques such as UV–vis, IR, 1H, as well as 13C NMR and ESI-MS. The interaction of these complexes with DNA/BSA (bovine serum albumin) was investigated using absorption, emission spectroscopy, viscosity measurements and molecular docking studies. The docking study infers that the binding strength (Kb) of these complexes was in agreement with results from absorption and emission techniques. These studies reveal that these three Ru(II) polypyridyl complexes bind to DNA/BSA. The binding ability of these complexes in the presence of different ions and solvents were also reported. All complexes were effectively cleaving pBR322 DNA in different forms and follows order which is similar to absorption and emission studies. These complexes were effective exhibiting the antimicrobial activity against different microbes Bacillus subtilis, Escherichia coli and Staphylococcus aureus.  相似文献

A novel ruthenium(II) polypyridyl complex [Ru(phen)2{ipbd)](ClO4)2 (ipbd = 3-(1H-Imidazo[4,5-f][1,10]phenanthrolin-2-yl)-1-benzodioxane, phen = 1,10-phenanthroline} has been synthesized and characterized by elemental analysis, ES-MS and 1H-NMR spectra. The interaction of the complex with calf thymus DNA (CT-DNA) was investigated by absorption titration, fluorescence spectra, thermal denaturation, viscosity measurements, circular dichroism and photoinduced cleavage. The results suggest that the complex intercalates into the CT-DNA base pairs. Upon irradiation, this novel Ru(II) complex can cleave the plasmid pBR 322 DNA from the supercoiled form I to the open circular form II.  相似文献

Two new luminescent ruthenium(II) polypyridyl complexes, [Ru(bpy)2(tpt‐phen)]Cl2 ( 1 ; bpy=2,2′‐bipyridine, tpt‐phen=triptycenyl‐1,10‐phenanthroline) and [Ru(phen)2(tpt‐phen)]Cl2 ( 2 ; phen=1,10‐phenanthroline), have been developed as potential nonviral vectors for DNA delivery. Photophysical and electrochemical properties of the complexes have been investigated and corroborated with electronic structure calculations. DNA condensation by these complexes has been investigated by UV/Vis and emission spectroscopy, circular dichroism spectroscopy, atomic force microscopy, dynamic light scattering, confocal microscopy, and electrophoretic mobility studies. These complexes interact with DNA and efficiently condense DNA into globular nanoparticles that are taken up efficiently by HeLa cells. DNA cleavage inability and biocompatibility of complexes have been explored. Both complexes have good gene transfection abilities.  相似文献

A novel porphyrin–polypyridyl ruthenium(II) hybrid compound, [C4-TPP-(ip)Ru(phen)2](ClO4)2 (TPP = 5,10, 15,20-tetraphenylporphyrin, ip = imidazo[4,5-f][1,10]phenanthroline and phen = 1,10-Phenanthroline), in which a polypyridyl ruthenium(II) moiety is linked to a porphyrin moiety by a butyl chain, has been synthesized by the reaction of (C4ip)TPP with cis-Ru(phen)2Cl2 in glacial acetic acid under an Ar atmosphere and characterized by elemental analysis, i.r., 1H-n.m.r., u.v.–vis., ESI-MS and electrochemistry. Its photoinduced DNA cleavage activity was investigated preliminarily. It has been found that the hybrid compound promoted photocleavage of plasmid pBR 322 DNA upon irradiation. Singlet oxygen (1O2) is suggested to be the reactive species responsible for the cleavage.  相似文献

Three inorganic–organic hybrid materials based on Keggin-type polyoxometalates (POMs), [CuII2(phen)2(4,4′-bipy)(H4,4′-bipy)2(H2O)2][PMo12O40]2·2H2O (1), [CuII(phen)2(H4,4′bipy)][PW12O40]·H2O (2), and [CuII2(phen)2(4,4′-bipy)(BW12O40)(H2O)2](H24,4′-bipy)0.5·3H2O (3) (phen = 1,10-phenanthroline, 4,4′-bipy = 4,4′-bipyridine), were synthesized using different POMs in the hydrothermal conditions. Compounds 1–3 were characterized by single-crystal X-ray diffraction, IR spectra, elemental analyses, powder X-ray diffraction analyses, and thermogravimetric analyses. Compound 1 presents a two-dimensional (2-D) network containing the Keggin-type [PMo12O40]3? anion and dinuclear metal–organic units [CuII2(phen)2(4,4′-bipy)(H4,4′-bipy)2(H2O)2]3+. Compound 2 is a 2-D architecture constructed from a [PW12O40]3? and mononuclear metal–organic units [CuII(phen)2(H4,4′-bipy)]3+. In 3, the [BW12O40]5? anions link [CuII2(phen)2(4,4′-bipy)] units to form a one-dimensional (1-D) chain [CuII2(phen)2(4,4′-bipy)(BW12O40)(H2O)2]; the 1-D chain connects with protonated 4,4′-bipy ligands and lattice waters, yielding a 2-D layer. Fluorescence spectra, UV–vis spectra, and electrochemical properties of 1–3 have been investigated.  相似文献

DNA-binding properties of a number of ruthenium complexes with different polypyridine ligands are reported. The new polypyridine ligand BFIP (=2-benzo[b] furan-2-yl-1H-imidazo[4,5-f][1,10]phenanthroline) and its ruthenium complexes [Ru(bpy)2BFIP]2+ (bpy = 2,2′-bipyridine), [Ru(dmb)2BFIP]2+ (dmb = 4,4′-dimethyl-2,2′-bipyridine), and [Ru(phen)2BFIP]2+ (phen = 1,10-phenanthroline) have been synthesized and characterized by elemental analysis, mass spectra, IR, UV-Vis, 1H- and 13C-NMR, and cyclic voltammetry. The DNA binding of these complexes to calf-thymus DNA (CT-DNA) was investigated by spectrophotometric, fluorescence, and viscosity measurements. The results suggest that ruthenium(II) complexes bind to CT-DNA through intercalation. Photocleavage of pBR 322 DNA by these complexes was also studied, and [Ru(phen)2BFIP]2+ was found to be a much better photocleavage agent than the other two complexes.  相似文献

Abstract

Ruthenium (II) complexes of the type RuL(CO)2Cl2, [RuL(CO)2L? 2]2+ and [RuL(CO)2Cl L′]+ [L = bipyridine (bpy), phenanthroline (phen), biquinoline (biq) and L′ = pyridine (py), 4-chloropyridine (Cl-py), 4-methoxypyridine (MeO-py)] were synthesized from [Ru(CO)2Cl2]n and L, to produce the intermediate RuL(CO)2Cl2 followed by hydrolysis and reaction with L′. The catalytic activity of these complexes in epoxidation of olefins with iodosylbenzene under ambient conditions was investigated. A possible mechanism of these reactions, explaining the effects of the ligands on the reaction was explored. At least one carbonyl ligand remained bound to the metal through the reaction. The formation of an oxo intermediate was inferred from spectroscopic detection of bridged oxygen Ru—O—Ru and Ru=O species.  相似文献

This article presents recent progress in our laboratory on the interactions of Ru(II) polypyridyl complexes with calf thymus DNA (CT-DNA). Mixed polypyridyl Ru(II) complexes [Ru(L)4(AIP)]2+ and [Ru(L)4PyIP]2+, where L is 4-amino pyridine and pyridine (AIP?=?2-(9-anthryl)-1H-imidazo[4,5-f][1,10]phenanthroline; PyIP?=?2-(1-pyrenyl)-1H-imidazo[4,5-f][1,10]phenanthroline), have been synthesized and characterized by elemental analysis, and physicochemical methods such as ESI-MS, UV-Vis, IR, and NMR spectroscopic techniques. Electronic absorption titrations, fluorescence spectroscopy, viscosity measurements, and salt-dependent studies of CT-DNA in the presence of incremental amounts of all four Ru(II) complexes clearly demonstrate that all four complexes bind to DNA by intercalation. The DNA-binding affinities of these complexes follow the order [Ru(4-APy)4(PyIP)]2+?>?[Ru(Py)4PyIP]2+?>?[Ru(4-APy)4(AIP)]2+?>?[Ru(Py)4AIP]2+. Irradiation of pBR 322 DNA with these complexes results in nicking of the plasmid DNA. All four complexes were screened for antimicrobial activity. All complexes also exhibited DNA “light switch” properties. These results suggest that both ancillary ligand and intercalative ligand influence the binding of these complexes to DNA.  相似文献

The synthesis of a number of new 2,2′‐bipyridine ligands functionalized with bulky amino side groups is reported. Three homoleptic polypyridyl ruthenium (II) complexes, [Ru(L)3]2+ 2(PF6?), where L is 4,4′‐dioctylaminomethyl‐2,2′‐bipyridine (Ru4a), 4,4′‐didodecylaminomethyl‐2,2′‐bipyridine (Ru4b) and 4,4′‐dioctadodecylaminomethyl‐2,2′‐bipyridine (Ru4c), have been synthesized. These compounds were characterized and their photophysical properties examined. The electronic spectra of three complexes show pyridyl π → π* transitions in the UV region and metal‐to‐ligand charge transfer bands in the visible region. The ruthenium (II) mixed-ligand complexes cis-[Ru(bpy)2(NCS)2] (I), [Ru(bpy)2(pn)]2+ (II) and [Ru(bpy)2(phen)]2+ (III) (bpy, 2,2′-bipyridine; pn, 1,2-diaminopropane; phen, 1,10-phenanthroline) were subjected to two (for I and II), and three (for III) stepwise, one-electron reductions, and one-electron oxidations (all fully reversible); the products were studied in situ by solution UV—vis-near IR spectroscopy. The first two reductions took place in all cases on separate bpy ligands, while the third reduction of III took place on phen. Bands of the reduced species in the near IR—vis region are observed and assigned to anion radical ligands. Novel features of the LMCT bands of the oxidized species are presented and discussed.  相似文献

Aquation has been proposed as crucial chemical action step for ruthenium (Ru) complexes, but its effects on the action mechanisms remain elusive. Herein, we have demonstrated the aquation process of a potent Ru polypyridyl complex (RuBmp=[RuII(bmbp)(phen)Cl]ClO4, bmbp=2,6‐bis(6‐methylbenzimidazol‐2‐yl) pyridine, phen=phenanthroline) with a chloride ligand, and revealed that aquation of RuBmp effectively enhanced its hydrophilicity and cellular uptake, thus significantly increasing its anticancer efficacy. The aquation products (H‐RuBmp=[RuII(bmbp)(phen)Cl]ClO4, [RuII(bmbp)(phen)(H2O)]ClO4, bmbp) exhibited a much higher apoptosis‐inducing ability than the intact complex, with involvement of caspase activation, mitochondria dysfunction, and interaction with cell membrane death receptors. H‐RuBmp demonstrated a higher interaction potency with the cell membrane and induced higher levels of ROS overproduction in cancer cells to regulate the AKT, MAPK, and p53 signaling pathways. Taken together, this study could provide useful information for fine‐tuning the rational design of next‐generation metal medicines.  相似文献

In this work, we report the efficient, high stable copper(II) complexes intercalated graphene oxide (GO) used as green catalysts for copper(II) complex mediated click reaction. Copper(II) Bis(2,2′-bipyridine) [CuII (bpy)2] (C1) and Copper(II) Bis(1,10-phenanthroline) [CuII (phen)2] (C2) have synthesized for the intercalation of corresponding nanocomposites with GO, [GO@CuII (bpy)2] (GO-C1) and [GO@CuII (phen)2] (GO-C2). The noncovalent interaction of complexes supported on the surface of the GO nanosheets proves as an evident active site to facilitate the enhanced catalytic activity of copper-catalyzed alkyne azide cycloaddition (CuIIAAC) reaction for the isolation of 1,4-disubstituted-1,2,3-triazoles as click products in shorter reaction time with 80%–91% yield (five examples). The X-ray diffraction (XRD) pattern of these composites shows the enhanced interlayers d-spacing range of 1.01–1.12 nm due to the intercalation of copper(II) complexes in between the GO basal planes and characterized by X-ray photoelectron spectroscopy (XPS), Fourier-transform infrared spectroscopy (FT-IR), Raman, UV, scanning electron microscope (SEM), and thermogravimetric analysis (TGA). The as-prepared nanocomposites were employed for the typical click reactions using the substrates of azide and acetylene. These classes of composite materials can be referred to recyclable, heterogeneous, green catalysts with high atom economy and could also be used for the isolation of click products in biomolecules.  相似文献

In this work, a new, label-free, homogeneous, highly sensitive, and selective fluorescent biosensor for DNA detection is developed by using rolling-circle amplification (RCA) based single-color quantum dots–ruthenium complex (QDs–Ru) assembling dyads. This strategy includes three steps: (1) the target DNA initiates RCA reaction and generates linear RCA products; (2) the complementary DNA hybridizes with the RCA products to form long double-strand DNA (dsDNA); (3) [Ru(phen)2(dppx)]2+ (dppx = 7,8-dimethyldipyrido [3,2-a:2′,3′-c] phenanthroline) intercalates into the long dsDNA with strong fluorescence emission. Due to its strong binding propensity with the long dsDNA, [Ru(phen)2(dppx)]2+ is removed from the surface of the QDs, resulting in restoring the fluorescence of the QDs, which has been quenched by [Ru(phen)2(dppx)]2+ through a photoinduced electron transfer process and is overlaid with the fluorescence of dsDNA bonded Ru(II) polypyridyl complex (Ru-dsDNA). Thus, high fluorescence intensity is observed, and is related to the concentration of target. This sensor exhibits not only high sensitivity for hepatitis B virus (HBV) ssDNA with a low detection limit (0.5 pM), but also excellent selectivity in the complex matrix. Moreover, this strategy applies QDs–Ru assembling dyads to the detection of single-strand DNA (ssDNA) without any functionalization and separation techniques.  相似文献

A novel ligand 3‐(1H‐imidazo[4,5‐f][1,10]phenanthrolin‐2‐yl)‐4H‐1‐benzopyran‐4‐one (ipbp) and its ruthenium(II) complexes [Ru(bpy)2(ipbp)]2+ ( 1 ) and [Ru(ipbp)(phen)2]2+ ( 2 ) (bpy=2,2′‐bipyridine, phen=1,10‐phenanthroline) were synthesized and characterized by elemental analysis and mass, 1H‐NMR, and electronic‐absorption spectroscopy. The electrochemical behavior of the complexes was studied by cyclic voltammetry. The DNA‐binding behavior of the complexes was investigated by spectroscopic methods and viscosity measurements. The results indicate that complexes 1 and 2 bind with calf‐thymus DNA in an intercalative mode. In addition, 1 and 2 promote cleavage of plasmid pBR 322 DNA from the supercoil form I to the open circular form II upon irradiation.  相似文献

Experimental results indicate that the three complexes can intercalate into DNA base pairs. Photo activated cleavage of pBR‐322 DNA by the three complexes was also studied. Mononuclear copper(II) and trinuclear cobalt(II) complexes, namely [Cu(L1)]2 · CH2Cl2 and [{Co(L2)(EtOH)}2Co(H2O)] · EtOH {H2L1 = 4,6‐dichloro‐6′‐methyoxy‐2,2′‐[1,1′‐(ethylenedioxydinitrilo)dimethylidyne]diphenol and H3L2 = 6‐ethyoxy‐6′‐hydroxy‐2,2′‐[1,1′‐(ethylenedioxydinitrilo)dimethylidyne]diphenol}, were synthesized and characterized by elemental analyses, IR and UV/Vis spectroscopy, and single‐crystal X‐ray diffraction. In the CuII complex, the CuII atom is four‐coordinate, with a N2O2 coordination sphere, and has a slightly distorted square‐planar arrangement. Interestingly, the obtained trinuclear CoII complex is different from the common reported 2:3 (L:CoII) salamo‐type CoII complexes. Infinite 2D layer supramolecular structures are formed via abundant intermolecular hydrogen bonding and π ··· π stacking interactions in the CuII and CoII complexes.  相似文献

Ruthenium(II) polypyridyl complexes with long‐wavelength absorption and high singlet‐oxygen quantum yield exhibit attractive potential in photodynamic therapy. A new heteroleptic RuII polypyridyl complex, [Ru(bpy)(dpb)(dppn)]2+ (bpy=2,2′‐bipyridine, dpb=2,3‐bis(2‐pyridyl)benzoquinoxaline, dppn=4,5,9,16‐tetraaza‐dibenzo[a,c]naphthacene), is reported, which exhibits a 1MLCT (MLCT: metal‐to‐ligand charge transfer) maximum as long as 548 nm and a singlet‐oxygen quantum yield as high as 0.43. Steady/transient absorption/emission spectra indicate that the lowest‐energy MLCT state localizes on the dpb ligand, whereas the high singlet‐oxygen quantum yield results from the relatively long 3MLCT(Ru→dpb) lifetime, which in turn is the result of the equilibrium between nearly isoenergetic excited states of 3MLCT(Ru→dpb) and 3ππ*(dppn). The dppn ligand also ensures a high binding affinity of the complex towards DNA. Thus, the combination of dpb and dppn gives the complex promising photodynamic activity, fully demonstrating the modularity and versatility of heteroleptic RuII complexes. In contrast, [Ru(bpy)2(dpb)]2+ shows a long‐wavelength 1MLCT maximum (551 nm) but a very low singlet‐oxygen quantum yield (0.22), and [Ru(bpy)2(dppn)]2+ shows a high singlet‐oxygen quantum yield (0.79) but a very short wavelength 1MLCT maximum (442 nm).  相似文献

The Schiff N‐allylamine‐4‐(ethylenediamine‐5‐methylsalicylidene)‐1,8‐naphthalimide (H2L) and its copper(II) complex, [Cu(HL)2] · 0.5DMF, were synthesized and characterized. The crystal structure of the CuII complex reveals a slightly distorted square‐planar arrangement provided by two N and O donors from two deprotonated ligands. In addition, the DNA‐binding properties of the ligand and CuII complex were investigated by fluorescence spectra, electronic absorption, and viscosity measurements. The experimental studies of the DNA‐binding properties indicated that the ligand and CuII complex reacted with DNA via intercalation binding mode, and binding affinity for DNA takes the order: ligand > CuII complex. The antioxidant assay in vitro suggested that both exhibited potential intensely antioxidant properties, and the ligand is more effective than its CuII complex.  相似文献
