Dehydroalanine (Dha) residues are attractive noncanonical amino acids that occur naturally in ribosomally synthesised and post-translationally modified peptides (RiPPs). Dha residues are attractive targets for selective late-stage modification of these complex biomolecules. In this work, we show the selective photocatalytic modification of dehydroalanine residues in the antimicrobial peptide nisin and in the proteins small ubiquitin-like modifier (SUMO) and superfolder green fluorescent protein (sfGFP). For this purpose, a new water-soluble iridium(III) photoredox catalyst was used. The design and synthesis of this new photocatalyst, [Ir(dF(CF3)ppy)2(dNMe3bpy)]Cl3, is presented. In contrast to commonly used iridium photocatalysts, this complex is highly water soluble and allows peptides and proteins to be modified in water and aqueous solvents under physiologically relevant conditions, with short reaction times and with low reagent and catalyst loadings. This work suggests that photoredox catalysis using this newly designed catalyst is a promising strategy to modify dehydroalanine-containing natural products and thus could have great potential for novel bioconjugation strategies.  相似文献

Transition metal catalysis is one of the most important tools to construct carbon-carbon and carbon-heteroatom bonds in modern organic synthesis. Visible-light photoredox catalysis has recently drawn considerable attention of the scientific community owing to its unique activation modes and significance for the green synthesis. The merger of photoredox catalysis with transition metal catalysts, termed metallaphotoredox catalysis, has become a popular strategy for expanding the synthetic utility of visiblelight photocatalysis. This strategy has led to the discovery of novel asymmetric transformations, which are unfeasible or not easily accessible by a single catalytic system. This contemporary area of organic chemistry holds promise for the development of economical and environmentally friendly methods for the asymmetric synthesis of chiral compounds. In this review, the advances in the enantioselective metallaphotoredox catalysis(EMPC) are summarized.  相似文献

Ketyl radical and amino radical anions, valuable reactive intermediates for C? C bond‐forming reactions, are accessible through a C?O/C?NR umpolung. However, their utilization in catalysis remains largely underdeveloped owing to the high reduction potential of carbonyl compounds and imines. In the context of photoredox catalysis, tertiary amines are commonly employed as sacrificial co‐reducing agents. Herein, an additional role of the amine is proposed, in which it is essential for the organocatalytic substrate activation. The combination of photoredox catalysis and carbonyl/imine activation enables the reductive coupling of aldehydes, ketones, and imines under mild reaction conditions.  相似文献

Visible-light photoredox catalysis has been established as a popular and powerful tool for organic transformations owing to its inherent characterization of environmental friendliness and sustainability in the past decades. The thiol-ene/yne reactions, the direct hydrothiolation of alkenes/alkynes with thiols, represents one of the most efficient and atom-economic approaches for the carbon-sulfur bonds construction. In traditional methodologies, harsh conditions such as stoichiometric reagents or a specialized UV photo-apparatus were necessary suffering from various disadvantages. In particular, visible-light photoredox catalysis has also been demonstrated to be a greener and milder protocol for the thiol-ene/yne reactions in recent years. Additionally, unprecedented advancements have been achieved in this area during the past decade. In this review, we will summarize the recent advances in visible-light photoredox catalyzed thiol-ene/yne reactions from 2015 to 2021. Synthetic strategies, substrate scope, and proposed reaction pathways are mainly discussed.  相似文献

Visible-light copper photocatalysis has recently emerged as a viable technology for building sustainable synthetic processes. To broaden the applications of phosphine-ligated copper(I) complexes, we describe herein an effective metal-organic framework (MOF)-supported copper(I) photocatalyst for multiple iminyl radical-mediated reactions. Due to site isolation, the heterogenized copper photosensitizer has a significantly higher catalytic activity than its homogeneous counterpart. Using a hydroxamic acid linker to immobilize copper species on MOF supports affords the heterogeneous catalysts with high recyclability. The post-synthetic modification sequence on MOF surfaces allows for the preparation of previously unavailable monomeric copper species. Our findings highlight the potential of using MOF-based heterogeneous catalytic systems to address fundamental challenges in the development of synthetic methodologies and mechanistic investigations of transition-metal photoredox catalysis.  相似文献

Herein we report the development of a photocatalytic strategy for the divergent preparation of functionalized bicyclo[1.1.1]pentylamines. This approach exploits, for the first time, the ability of nitrogen‐radicals to undergo strain‐release reaction with [1.1.1]propellane. This reactivity is facilitated by the electrophilic nature of these open‐shell intermediates and the presence of strong polar effects in the transition‐state for C?N bond formation/ring‐opening. With the aid of a simple reductive quenching photoredox cycle, we have successfully harnessed this novel radical strain‐release amination as part of a multicomponent cascade compatible with several external trapping agents. Overall, this radical strategy enables the rapid construction of novel amino‐functionalized building blocks with potential application in medicinal chemistry programs as p‐substituted aniline bioisosteres.  相似文献

The creation of new bonds via C-F bond cleavage of readily available per- or oligofluorinated compounds has received growing interest. Using such a strategy, a myriad of valuable partially fluorinated products can be prepared, which otherwise are difficult to make by the conventional C-F bond formation methods. Visible light photoredox catalysis has been proven as an important and powerful tool for defluorinative reactions due to its mild, easy to handle, and environmentally benign characteristics. Compared to the classical C-F activation that proceeds via two-electron processes, radicals are the key intermediates using visible light photoredox catalysis, providing new modes for the cleavage of C-F bonds. In this review, a summary of the visible light-promoted C-F bond cleavage since 2018 was presented. The contents were classified by the fluorosubstrates, including polyfluorinated arenes, gem-difluoroalkenes, trifluoromethyl arenes, and trifluoromethyl alkenes. An emphasis is placed on the discussion of the mechanisms and limitations of these reactions. Finally, my personal perspective on the future development of this rapidly emerging field was provided.  相似文献

Visible light has been recognized as an economical and environmentally benign source of energy that enables chemoselective molecular activation of chemical reactions and hence reveal a new horizon for the design and discovery of novel chemical transformations. On the other hand, asymmetric catalysis represents an economic method to satisfy the increasing need for enantioenriched compounds in the chemical and pharmaceutical industries. Therefore, combining visible light photocatalysis with asymmetric catalysis creates a wider range of opportunities for the development of mechanistically unique reaction schemes. However, there arise two main problems like undesirable photochemical background reactions and difficulties in controlling the stereochemistry with highly reactive photochemical intermediates which can pose a serious challenge to the development of asymmetric visible light photocatalysis. In recent years, several methods have been developed to overcome these challenges. This review summarizes the recent advances in visible light‐induced enantioselective reactions. We divide our discussion into four categories: Asymmetric photoredox organocatalysis, asymmetric transition metal photoredox catalysis, asymmetric photoredox Lewis acid catalysis and asymmetric photoinduced energy transfer catalysis. Special emphasis has been given to different catalytic activation modes that enable the construction of challenging carbon‐carbon and carbon‐heteroatom bond in an enantioselective fashion. A brief analysis of substrate scope and limitation as well as reaction mechanism of these reactions has been included.  相似文献

Photocatalysis for direct C−H trifluoromethylation represents an ideal way to synthesize trifluoromethyl-containing chemical compounds, but the conventional batch processes are inefficient with limited light penetration and indispensably irradiated for a long while. Herein, we report a continuous-flow protocol for photocatalytic direct C−H trifluoromethylation of heterocycles in the presence of an organic photoredox catalyst: 2,4,6-tris(diphenylamino)-3,5-difluorobenzonitrile (3DPA2FBN). In this approach, benefiting from the merger of organic photoredox catalysis and continuous-flow techniques, a variety of trifluoromethylated heterocycles were rapidly synthesized up to 85 % yield with 80 min residence time under metal- and oxidant-free reaction conditions.  相似文献

Visible‐light‐induced radical decarboxylative functionalization of carboxylic acids and their derivatives has recently received considerable attention as a novel and efficient method to create C? C and C? X bonds. Generally, this visible‐light‐promoted decarboxylation process can smoothly occur under mild reaction conditions with a broad range of substrates and an excellent functional‐group tolerance. The radical species formed from the decarboxylation step can participate in not only single photocatalytic transformations, but also dual‐catalytic cross‐coupling reactions by combining photoredox catalysis with other catalytic processes. Alkoxy radicals have been intensively studied as versatile synthetic intermediates with well-established reactivity and selectivity. Due to the difficulties associated with the generation of these transient and high-energy radicals, the utilization of the unique reactivity of alkoxy radicals in catalytic platforms has become a rather challenging task. Recent studies on the alkoxy radical-mediated transformations have benefited tremendously from advances in modern photoredox catalysis. In this digest, we aim to highlight reported transformations that utilize alkoxy radicals in a photocatalytic fashion, in particular, underline novel catalytic platforms which we anticipate will find broad application in the near future.  相似文献

Over the past decades, organometallic complexes with precious elements, such as ruthenium and iridium, are widely used as visible-light photoredox catalysts. Recently, more and more complexes based on earth-abundant and inexpensive elements have been used as sensitizers in photochemistry. Although the photoexcited state lifetimes of iron complexes are typically shorter than those of traditional photosensitizers, the utilization of iron catalysts in photochemistry has sprung up owing to their abundance, low price, nontoxicity, and novel properties, including exhibiting ligand to metal charge transfer states. This concept focuses on recent advances in light-driven iron catalysis in organic transformations, including iron/photoredox dual catalysis, light-induced iron photoredox catalysis and light-induced generation of active iron catalysts. The development of iron complexes for the photoredox catalysis is a huge challenge. Indeed, Iron complexes can be ideal candidates due to their potential visible light absorption and redox properties but also because they are less toxic, inexpensive and environmentally friendly compared to other catalysts. In the present paper, a series of novel iron complexes have been synthesized and utilized to initiate the free radical promoted cationic polymerization of epoxides or the free radical polymerization of acrylates through photoredox catalysis processes upon exposure to near UV (385 nm) or visible violet (405 nm) light emitting diodes (LEDs). When combined with an iodonium salt and N‐vinylcarbazole, the iron complex‐based photoinitiating systems are able to generate radicals, cations, and radical cations. The initiation efficiency is investigated through real‐time Fourier transform infrared spectroscopy and a satisfactory initiating ability is found. The mechanisms for the generation of the reactive initiating species through photoredox catalysis are studied by different methods (steady state photolysis, cyclic voltammetry and electron spin resonance spin trapping techniques) and discussed in detail. © 2016 Wiley Periodicals, Inc.
