A sequential one-pot intermolecular oxy-Michael addition, intermolecular Heck coupling, and intramolecular degradation (retro-oxy-Michael addition) followed by condensation method has been developed for the synthesis of interesting 2-benzoxepin-3(1H)-ones. Significantly, the 2-benzoxepin-3(1H)-ones form the core quantum of biologically vital natural products. The initial oxy-Michael addition and Heck coupling steps involve a straight forward construction of C–O and C–C bonds, whereas, the final condensation step follows a novel mechanistic path via intramolecular degradation, double bond isomerization, and intramolecular condensation. Notably, a remarkable solvent effect has been observed in-order to promote the final intramolecular condensation.  相似文献

A simple and convenient sequential one-pot synthesis of 1,2,3,4-tetrahydrobenzo[b][1,6] naphthyridines has been developed. The reductive amination of 2-chloro-3-formylquinolines with various amines in the presence of sodium borohydride provided the corresponding secondary amines in high yields. Further, a sequential one-pot reaction involving N-allylation and intramolecular Heck type 6-exo-trig cyclization was performed on the secondary amines to afford a range of desired 1,2,3,4-tetrahydrobenzo[b][1,6]-naphthyridine derivatives in good to high yields.  相似文献

The asymmetric total synthesis of UCS1025A is accomplished by establishing a novel and efficient method for the construction of a tricyclic pyrrolizidinone skeleton based on a sequential Michael-retro Michael strategy. The key step is a one-pot six-step cascade reaction including oxidation of a primary alcohol to the corresponding carboxylic acid, a retro thio-Michael reaction, and an intramolecular oxy-Michael reaction. This newly-developed synthetic strategy inspired by “masked” electrophilic character of tricyclic pyrrolizidinone is efficient and high-yielding compared to that developed in previously-reported total syntheses.  相似文献

The total synthesis of curvulone B was achieved, and its absolute configuration unambiguously established, as had been determined by TD-DFT ECD calculations on the computed solution conformers. Key features of the synthesis were an olefin cross-metathesis of an α,β-unsaturated ketone as a common key building block with (R)-6-triethylsilyloxyhept-1-ene, and an intramolecular oxy-Michael addition of the hydroxyl enone obtained therefrom. An intermolecular metathesis of the enone with (S)-5-benzyloxyhept-ene also enabled us to prepare a proposed structure for dothiorelone B, thus leading to the confirmation of the structural revision of phomopsin A, a novel nine-membered cyclic phenol ether.  相似文献

A Pd2dba3/P(i-BuNCH2CH2)3N catalyzed one-pot synthesis of unsymmetrically substituted trans-4-N,N-diarylaminostilbenes and both symmetrically and unsymmetrically substituted N,N-diarylaminostyrene derivatives is reported. The procedure involves two or more palladium catalyzed sequential coupling reactions (an amination and an inter-molecular Heck reaction) in one-pot using the same catalyst system with two different aryl halides, including aryl chlorides and hetero aryl halides as the coupling partners.  相似文献

Bidentate phosphine ligands in palladium-catalyzed intramolecular Heck reactions of 2-chloroquinolin-3-yl-(1-homoallyl)alcohols are described to afford facile synthesis of 3-methylene-2,3-dihydro-1H-cyclopenta[b]quinolines in improved yields. We further observed using bulky aromatic phosphine ligand in Pd-catalyzed intermolecular Heck coupling reaction at olefinic centers with iodobenzene also favored exclusively Heck products in excellent yield.  相似文献

The rapid synthesis of cis-2,6-disubstituted dihydropyrans is achieved in a three-component, one-pot cascade reaction. BiBr3-mediated addition of ketene silyl acetals or silyl enol ethers to β,γ-unsaturated cis-4-trimethylsilyl-3-butenal provides a Mukaiyama aldol adduct containing a vinylsilane moiety tethered to a silyl ether. Addition of a second aldehyde initiates a domino sequence involving intermolecular addition followed by an intramolecular silyl-modified Sakurai (ISMS) reaction. Isolated yields of this one-pot reaction vary from 44 to 80% and all compounds were isolated as the cis-diastereomers (10 examples).  相似文献

A palladium-catalyzed Heck reaction of in-situ generated benzylic iodides and styrenes has been achieved. The reaction proceeds in a one-pot manner through 1) addition perfluoroalkyl iodides to styrenes to give benzylic iodides, 2) followed by a palladium-catalyzed Heck coupling of the resulting benzylic iodides with the same alkenes in high regio- and stereoselectivity. A variety of perfluoroalkylated alkenes were obtained in moderate to excellent yields.  相似文献

A chiral scandium-bipyridine complex catalyzes the highly enantioselective addition of phenylselenol to aromatic meso-epoxides and furnishes 1,2-seleno alcohols in good yields and up to 94% ee. The first total synthesis of Goniothalesdiol A, isolated from the stems of Goniothalamus amuyon (Annonaceae) is reported. The C2 stereocentre and C3/C4 syn diol were created by a Sharpless kinetic resolution followed by acetonide formation. Alkyl amines have become available for the synthesis of diverse N-alkyl carbazoles through twofold SNAr aminations of dibenzothiophene dioxides by using alkali metal bases. Of particular importance is the choice of counter cations on alkali metal bases, that is, i) the use of Li base for the efficient intermolecular reaction and ii) the sequential addition of heavier alkali metal bases (Na, K, or Cs) to promote intramolecular cyclization in a one-pot manner. This protocol also enables the cascade synthesis of N-H-carbazoles by using 2-phenylethylamine by removal of the 2-phenethyl group from N-(2-phenethyl) carbazoles in a single operation.  相似文献

A new class of conformationally rigid phosphino-oxazoline ligands 3 were synthesized via an efficient ortho-substitution of phenyl glycinol as the key step. Divergent synthetic routes for easy ligand modulation, as well as a procedure suitable for scale-up synthesis, were established. The catalytic potential of ligands 3 has been demonstrated in the highly enantioselective Ir-catalyzed hydrogenation of alkenes and Pd-catalyzed allylic substitution and intermolecular Heck reactions.  相似文献

Both a three-step and a five-step syntheses of monoterpene alkaloid (±)-oxerine from alcohol 6 have been accomplished. In the second approach, the synthetic efficiency was enhanced by implementing a one-pot protocol (deprotonation/silylation/ alkylation/desilylation). The construction of the cyclopenta[c]pyridine framework was realized by an intramolecular Heck reaction, which should be adaptable for the synthesis of other related monoterpene pyridine alkaloids.  相似文献

A simple and straightforward methodology of general utility to construct sterically encumbered furo[3,2-b]furanone scaffolds present in a diverse range of bioactive natural products is delineated.
