Electrospray ionization (ESI) of peptides and proteins produces a series of multiply charged ions with a mass/charge (m/z) ratio between 500 and 2000. The resulting mass spectra are crowded by these multiple charge values for each molecular mass and an isotopic cluster for each nominal m/z value. Here, we report a new algorithm simultaneously to deconvolute and deisotope ESI mass spectra from complex peptide samples based on their mass-dependent isotopic mean pattern. All signals corresponding to one peptide in the sample were reduced to one singly charged monoisotopic peak, thereby significantly reducing the number of signals, increasing the signal intensity and improving the signal-to-noise ratio. The mass list produced could be used directly for database searching. The developed algorithm also simplified interpretation of fragment ion spectra of multiply charged parent ions.  相似文献

Collisional cooling in radio frequency (RF) ion guides has been used in mass spectrometry as an intermediate step during the transport of ions from high pressure regions of an ion source into high vacuum regions of a mass analyzer. Such collisional cooling devices are also increasingly used as 'linear', two-dimensional (2D) ion traps for ion storage and accumulation to achieve improved sensitivity and dynamic range. We have used the effective potential approach to study m/z dependent distribution of ions in the devices. Relationships obtained for the ideal 2D multipole demonstrate that after cooling the ion cloud forms concentric cylindrical layers, each of them composed of ions having the same m/z ratio; the higher the m/z, the larger is the radial position occupied by the ions. This behavior results from the fact that the effective RF focusing is stronger for ions of lower m/z, pushing these ions closer to the axis. Radial boundaries of the layers are more distinct for multiply charged ions, compared to singly charged ions having the same m/z and charge density. In the case of sufficiently high ion density and low ion kinetic energy, we show that each m/z layer is separated from its nearest neighbor by a radial gap of low ion density. The radial gaps of low ion population between the layers are formed due to the space charge repulsion. Conditions for establishing the m/z stratified structure include sufficiently high charge density and adequate collisional relaxation. These conditions are likely to occur in collisional RF multipoles operated as ion guides or 2D ion traps for external ion accumulation. When linear ion density increases, the maximum ion cloud radius also increases, and outer layers of high m/z ions approach the multipole rods and may be ejected. This 'overfilling' of the multipole capacity results in a strong discrimination against high m/z ions. A relationship is reported for the maximum linear ion density of a multipole that is not overfilled.  相似文献

Monensin A was studied by electrospray ionisation sequential mass spectrometry (ESI-MSn) and all fragments were confirmed by accurate-mass measurements. Analyses were performed on both a quadrupole time-of-flight (Q-tof) and a Fourier-transform ion cyclotron resonance (FT-ICR) mass spectrometer. MSn analysis shows that depending on sample preparation the ion at m/z 671 consists of two different ions with the same accurate-mass. It is either the monensin protonated parent ion or a different ion structure derived from the loss of water from the water adduct of monensin. Both ions show different fragmentation patterns. Major fragment ions from the protonated parent ion were produced by Grob-Wharton type fragmentations in addition to various simple neutral losses. The fragmentation pathways of the two different m/z 671 ions are proposed.  相似文献

Gestrinone was studied by high performance liquid chromatography (HPLC) for screening and by gas chromatography/mass spectrometry (GC/MS) for confirmation. When the chromatograms of blank, spiked urine and dosed urine were compared by HPLC, two unknown metabolites were found and these were excreted as the conjugated forms. Metabolites 1 and 2 were tested by LC/MS and LC/MS/MS and both had parent ions at m/z 325. The fragment ion of metabolite 1 was at m/z 263 and ions for metabolite 2 were m/z 307 [MH - H(2)O](+), 289, 279 and 241. LC/MS/MS of m/z 263 as the parent ion of metabolite 1 gave fragment ions at m/z 245 and 217, which were assumed to be [263 - H(2)O](+) and [235 - H(2)O](+), respectively. The trimethylsilyl (TMS)-enol-TMS ether derivative of gestrinone displayed three peaks in its GC/MS chromatogram, formed by tautomerism.  相似文献

Several volatile compounds, including terpenoids, fatty alcohols, fatty acids and some of their esters, were identified from solvent extracts prepared from anal scent glands of nutria (a.k.a. coypu), a serious rodent pest ravaging wetlands in the USA. The major terpenoid constituents were identified as (E,E)-farnesol and its esters by a comparison of their gas chromatographic retention times, and electron-ionization (EI) and chemical-ionization (CI) mass spectra with those of authentic compounds. EI mass spectra of the four farnesol isomers are very similar, however, the ChemStation (Agilent) and GC-MS Solution (Shimadzu) software algorithms were able to identify the natural compound as the (E,E)-isomer, when a high-quality mass spectral library was compiled from reference samples and used for searching. Similarly, the esters were identified as those of (E,E)-farnesol. In contrast to EI spectra, the CI spectra of the (E,E)- and (E,Z)-isomers are distinctly different from those of the (Z,E)- and (Z,Z)-isomers. The intensities (I) of the peaks for the m/z 137 and 121 ions in the CI spectra offer a way of determining the configuration of the C-2 double bond of farnesols (for 2E isomers I(137)>I(121), whereas for 2Z isomers I(137)相似文献

Bis(2-hydroxy-3,5-dichlorophenyl)methane and its cyclized derivatives (6-substituted 12H-tetrachlorodibenzo[d,g] [1,3, 2]dioxaphosphorin-6-oxide and sulfides) were analyzed by EI-MS, and the mass spectra are discussed. It was shown that the mass spectra of the cyclized derivatives are rather different from that of the parent compound. The relative roles of the S and O atoms were very important, and led to special ions which could indicate certain moieties within the molecular structure. Thus, the fragment ions at m/z 63 and 333 indicate a cyclic structure with a (P=S) group, while m/z 397 indicates P(=S)S or P(=S)-Cl) moieties, and m/z 380 with high intensity indicates P(=O)O. The ionization and fragmentation behaviors of carbohydrate derivatives prepared by reaction with 2-aminobenzamide (AB), 1-phenyl-3-methyl-5-pyrazolone (PMP), and phenylhydrazine (PHN) were compared under identical mass spectrometric conditions. It has been shown that the intensities of signals in MS spectra depend on the kind of saccharides investigated and reducing end labels used. PMP sialyllactose, when ionized by ESI/MALDI, produced a mixture of [M + H]+, [M + Na]+, [M - H + 2Na]+ ions in the positive mode and [M - H]-, [M + Na - 2H]- ions in the negative mode. The AB and PHN derivatives formed abundant [M + H]+ and [M - H]- ions in ESI, and by matrix-assisted laser desorption/ionization (MALDI) produced abundant [M + Na]+ ions. PMP- and reduced AB-sialyllactose produced only Y-type fragment ions under both MS/MS sources. In the electrospray ionization (ESI)-MS/MS spectrum of PHN-sialyllactose, abundant ions corresponded to B, Z cleavages and in its MALDI-MS/MS spectrum, the abundant ions were consistent with Y glycosidic cleavages with the concurrence of B, C, and cross-ring fragment ions. In the MALDI-MS spectra of oligosaccharides acquired immediately after derivatization, it was possible to detect only PHN derivatives. After purification, spectra of all three types of derivatives showed high signal-to-noise ratios with the most abundant ions observed for AB reduced saccharides. [M + Na]+ ions were the dominant products and their fragmentation patterns were influenced by the type of the labeling and the kind of oligosaccharide considered. In the MALDI-PSD and -MS/MS spectra of AB-derivatized glycans, higher m/z fragment ions corresponded to B and Y cleavages and the loss of bisecting GlcNAc appeared as a weak signal or was not detected at all. Fragmentation patterns observed in the spectra of hybrid/complex PHN and PMP glycans were more comparable-higher m/z fragments corresponded to B and C glycosidic cleavages. For PHN glycans, the abundance of ions resulting from the loss of bisecting GlcNAc depended on the number of residues linked to the 6-positioned mannose. Also, PHN and PMP derivatives produced cross-ring cleavages with abundances higher than observed in the spectra of AB derivatized oligosaccharides. For high-mannose glycans, the most informative cleavages were provided by AB and PHN type of labeling. Here, PMP produced dominant Y-cleavages from the chitobiose while other ions produced weak signals.  相似文献

By the reaction of syn-isomers of 5-substituted 2-(4-chlorobutyryl)aminobenzophenones oximes with NaOH syn-isomers of 5-substituted 2-(2-oxopyrrolidin-1-yl)benzophenones oximes were obtained. Similarly the anti-isomers of 5-substituted 2-(4-chlorobutyryl)aminobenzophenones oximes treated with NaOH underwent cyclization into anti-isomers of 5-substituted 2-(2-oxopyrrolidin-1-yl)benzophenones oximes. Crystal and molecular structures were investigated of the syn-isomer of 5-methyl-2-(2-oxopyrrolidin-1-yl)benzophenone oxime, the anti-isomer of 5-bromo-2-(2-oxopyrrolidin-1-yl)benzophenone oxime, and the syn-isomer of 5-methyl-2-(4-chlorobutyryl)aminobenzo-phenone oxime. The fragmentation features under the electron impact of syn-and anti-isomers of 5-substituted 2-(2-oxopyrrolidin-1-yl)benzophenones oximes are discussed.  相似文献

High-performance liquid chromatography coupled to electrospray ionization mass spectrometry (HPLC/ESI-MS) can provide both elemental and molecular information and, therefore, is a very useful tool for the identification of arsenic compounds. When a method for the identification of four arsenosugars was employed in our laboratory with an HPLC/ESI-MS system equipped with a Whatman model 75-72 nitrogen generator, a signal at m/z 75 (As(+)) could not be observed. When the HPLC/ESI-MS system was operated with nitrogen 5.0 (nitrogen of a purity of at least 99.999%) all four arsenosugars gave a signal at m/z 75. Because of this observation the influence of the quality of the nitrogen drying gas on the fragmentation of the four arsenosugars was systematically investigated. Standard solutions containing the four arsenosugars (0.5 ng As each) were separated on an anion-exchange column and detected with ESI-MS in the positive ion mode by monitoring the signals for [M+H](+), m/z 237, 91, and 75. Nitrogen with defined oxygen concentrations was used as drying gas. The purity of the nitrogen ranged from 99 to 99.999% (10 400 to 10 ppm oxygen impurity). The nitrogen with 99% purity gave no signal at m/z 75, but signals were obtained at m/z 91, 237, and for [M+H](+). When higher purity nitrogen (99.9%) was used, a signal was obtained at m/z 75, which accounted for 0.8-1.1% (depending on the kind of arsenosugar) of the sum of the signals for m/z 75, 91, 237 and [M+H](+). As the level of oxygen in the nitrogen decreased, the m/z 75 signal increased to 2.0-3.1%. This was accompanied by a concomitant decrease in the m/z 91 signal from 5.2-6.6% to 0.7-1.5%, whereas the signals for [M+H](+) and m/z 237 were essentially unchanged. Signals at m/z 75 with intensities comparable with those observed for the 99.9% pure nitrogen were also obtained for all the arsenosugars when the HPLC/ESI-MS system was operated with a Domnick Hunter Nitrox UHPLCMS18 nitrogen generator. Dimethylarsinic acid, arsenobetaine, trimethylarsine oxide, arsenocholine and the tetramethylarsonium cation also gave no signal at m/z 75 when they were analyzed with the Whatman model 75-72 nitrogen generator, but clear signals at m/z 75 were observed with the Domnick Hunter Nitrox UHPLCMS18 nitrogen generator. A nitrogen quality of at least 99.9% is required to obtain elemental information (m/z 75) when arsenic compounds are investigated by HPLC/ESI-MS. Molecular and elemental information from one chromatographic run is a valuable tool for the characterization of unknown arsenic compounds.  相似文献

2-(1-Alkoxyimino-2,2,2-trifluoroethyl)-5-trimethylsilylfurans were synthesized by the condensation of 2-(trifluoroacetyl)-5-trimethylsilylfuran with alkoxyamines. According to 1H and 19F NMR spectroscopic data, the alkoxyimino group in the E-isomers descreens the H-3 and H-4 protons of the furan ring more strongly than in the Z-isomers, shifting their signals downfield. Two types of naphthalene trimers linked by 1,3,4-oxadiazole spacers were synthesized and investigated for their physical and electronic properties. 2,6- and 2,7-isomers on central naphthalene moieties were obtained in the forms of pale yellow solids and colorless crystals, respectively. The melting point of the 2,6-isomer was higher than that of the 2,7-isomer. An X-ray crystallographic analysis revealed a π-stacked column with a short intermolecular distance in the crystals of the 2,6-isomer. The absorption maximum of the 2,6-isomer was red-shifted as compared to that of the 2,7-isomer, indicating a π-conjugation between di-2-naphthyloxadiazole moieties in the 2,6-isomer. The quantum yields of the 2,6- and 2,7-isomers were measured to be 0.97 and 0.74, relative to that of 2,5-di-2-naphthyl-1,3,4-oxadiazole (0.85). Molecular orbital (MO) calculations demonstrated that the 2,6-isomer had a higher electron affinity than the 2,7-isomer. Thus, the crosslinking of building blocks is important for the design of functional materials.  相似文献

We have observed unusual mass spectra of chloramphenicol (CAP) in solutions of methanol or acetonitrile showing intense ions at m/z 297, m/z 311, m/z 325 and m/z 339. The observed ions were different from those which are traditionally observed in the full scan ESI mass spectra of CAP with ions of m/z 321, m/z 323 and m/z 325. We have evidence to show that this process starts with offline methylation of CAP in solutions of methanol or acetonitrile to give m/z 339. Investigations using nuclear magnetic resonance (NMR) spectroscopy showed that there is a methylene group somewhere within the CAP molecule but not attached to any of the carbon atoms when the CAP is dissolved in methanol or acetonitrile before infusion into the mass spectrometer. The possible locations of attachment were speculated to be the electronegative atoms apart from the chlorine atoms due to valence considerations. The methylene group is attached to the nitrogen atom and forms a bond as observed in the MS/MS spectra of m/z 297, m/z 311, m/z 325 and m/z 339 which give m/z 183 as the base peak in all cases. Further experiments showed that there is cleavage of the methylated CAP molecule followed by cluster ion formation involving addition of methylene groups to the CAP fragment with m/z 183 to produce ions of m/z including m/z 297, m/z 311, m/z 325 and m/z 339. This process occurs in the mass spectrometer in the region housing the tube lens and is triggered when the ions are accelerated through this region by application of a negative tube lens offset voltage. This region affords collision of the charged droplets with a collision gas in this case nitrogen to strip the droplets of their solvent molecules. Experiments to follow the intensities of m/z 183, m/z 311, m/z 321, m/z 323, m/z 325 and m/z 339 as the tube lens offset voltage was varied were done in which the intensities of m/z 311, m/z 325 and m/z 339 were observed to be at their peak when the tube lens offset voltage was set at -40 V. When the tube lens offset voltage is swung to +40 V, thus decelerating the ions through the capillary skimmer region via the tube lens, the traditionally observed spectra with m/z 321, m/z 323 and m/z 325 were observed.  相似文献

Chemical ionization of a mixture of nitrogen and carbon monoxide produces three stable isobaric species at m/z 56: OCCO, OCNN and NNNN radical cations. Separated at increased resolution, these ions are readily identified by collisional activation. Neutralization-reionization experiments performed on two different mass spectrometers have not allowed the detection of any recovery signals for the corresponding neutrals.  相似文献

Complications with the gas chromatographic analysis of steroids prompted the use of alternative techniques for their identification. High-performance liquid chromatography/mass spectrometry with atmospheric pressure ionization allowed the collection of data for structural identification of these compounds. The objective of this study was to investigate the up-front collision-induced dissociation (UFCID) electrospray ionization (ESI) mass spectra of testosterone and monohydroxylated testosterones. The positive ion UFCID ESI mass spectrum of testosterone showed three significant ions at m/z 97, 109 and 123. The relative abundance of these ions in the UFCID ESI mass spectra of monohydroxylated testosterones varied with the position of the hydroxy group. Statistical data allowed the prediction of hydroxy group position on testosterone by evaluation of the relative abundance of the m/z 97, 109, 121 and 123 ions. Data from the ESI mass spectral analysis of testosterone in a deuterated solvent and from the analysis of cholestenone and 4-androstene-3 beta, 17 beta-diol indicated that the initial ionization of testosterone occurred at the 3-one position. CID parent ion monitoring analyses of the m/z 97, 109 and 123 ions indicated that each resulted from different fragmentation mechanisms and originated directly from the [M + H]+ parent ion. The elemental composition of these fragment ions is proposed based on evidence gathered from the CID analysis of the pseudo-molecular ions of [1,2-2H2]-, [2,2,4,6,6-2H5]-, [6,7-2H2]-, [7-2H]-, [19,19,19-2H3]- and [3,4-13C2]testosterone. The structure and a possible mechanism of formation of the m/z 109 and 123 ions is presented. The results of this study advance the understanding of the mechanisms of collision-induced fragmentation of ions.  相似文献

The concept of using a short ionisation event, in this case a pulsed corona discharge, in conjunction with programmed gate delay is described. This technique is proposed for the selective study of different ionisation processes within the reaction region of an ion mobility spectrometer. The utility of such an approach was tested in a study of the ionisation of dipropylene-glycol-monomethyl-ether (DPM); a compound commonly used to test the operation of ion mobility spectrometers. Dipropylene-glycol-monomethyl-ether at a concentration of 113 microg m(-3) in air, with a water level of 75 mg m(-3) in air, was analysed using a switchable, high resolution ion mobility spectrometer, operating in the positive mode at 40 degrees C at ambient pressure. The ion mobility spectrometer was fitted with a pulsed corona discharge ionisation source, doped with ammonia at a concentration of 1.3 mg m(-3) in the reaction region, and interfaced to a mass spectrometer. Synchronisation of the ionisation event to the operation of the shutter grids for the drift region enabled different parts of the product ion population to be injected into the drift tube, and programming the gate delays produced a map of the gate delay verses drift time response surface. Ammonium bound dipropylene-glycol-monomethyl-ether was observed, [(DPM)NH4]+ (m/z 166) as well as the ammonium bound dimer [(DPM)2NH4]+ (m/z 314), the same as those observed with a 63Ni source. Two other species were also observed, but their molecular identity was not elucidated. One of them m/z 146, also observed with 63Ni, formed ammonium bound ions [(m/z 146)NH4]+ (K0= 1.49 cm2 V(-1) s(-1)), ammonium bound dimer ions [(m/z 146)2NH4]+(K0= 1.18 cm2 V(-1) s(-1)) and a mixed cluster ion with DPM [(m/z 146)(DPM)NH4]+(K0= 1.18 cm2 V(-1) s(-1)); while the other, m/z 88 a decomposition product, formed ammonium bound monomer [(m/z 88)NH4]+(K0= 1.68 cm2 V(-1) s(-1)), dimer ions [(m/z 88)2NH4]+(K0= 1.40 cm2 V(-1) s(-1)) and a mixed cluster ion containing DPM and ammonium, [(DPM)(m/z 88)2NH4]+(K0= 1.40 cm2 V(-1) s(-1)). The assignment of responses to these ions required the additional dimensionality in the data provided from the gate delay studies. The relationships evident in the programmable gate delay data enabled these ions to be differentiated from alternative assignments of possible nitrogen clusters, formed at the interface of the mass spectrometer.  相似文献

A conventional fast-atom bombardment (FAB) ion source was used to achieve matrix-assisted laser desorption (MALD) in a high-mass, double-focusing, magnetic mass spectrometer. The pulsed ion signals generated by irradiation of a mixture of sample and matrix (2,5-dihydroxybenzoic acid) with either a XeF excimer laser (353 nm) or a nitrogen laser (337 nm) were recorded with a focal-plane detector. A resolution (full-width at half maximum) of 4500 was achieved at m/z 1347.7 (the peptide substance P), 2500 for CsI cluster ions at m/z 10,005.7, and 1250 for the isotope cluster of the small protein cytochrome c (horse) [M+H]+ = m/z 12,360 (average). Sensitivity is demonstrated with 11 fmol of substance P. A survey scan is taken to locate the m/z of the sample molecular ion. The segment that contains the sample can then be integrated for a longer time to produce a better signal-to-noise ratio. In addition to higher sensitivity and lower matrix interference, the advantage of MALD over FAB is the former's lower susceptibility to the presence of salts, and competition between hydrophobic and hydrophilic components of a mixture. This feature is demonstrated by the complete MALD spectrum of a crude partial tryptic digest of sperm-whale apomyoglobin, containing 24 peptides, representing the entire sequence of this protein.  相似文献

The fragment ion formation characteristics of the radical anions generated from hexahydro-1,3,5-trinitrotriazine (RDX) and its three nitroso metabolites were studied using GC/MS with negative chemical ionization (NCI) to understand the fragmentation mechanisms responsible for the formation of the most abundant ions observed in their NCI mass spectra. Ab initio and density functional theory calculations were used to calculate relative free energies for different fragment ion structures suggested by the m/z values of the most abundant ions observed in the NCI mass spectra. The NCI mass spectra of the four nitramines are dominated by ions formed by the cleavage of nitrogen-nitrogen and carbon-nitrogen bonds in the atrazine ring. The most abundant anions in the NCI mass spectra of these nitramines have the general formulas C(2)H(4)N(3)O (m/z 86) and C(2)H(4)N(3)O(2) (m/z 102). The analyses of isotope-labeled standards indicate that these two ions are formed by neutral losses that include two exocylic nitrogens and one atrazine ring nitrogen. Our calculations and observations of the nitramine mass spectra suggest that the m/z 86 and m/z 102 ions are formed from either the (M--NO)(-) or (M--NO(2))(-) fragment anions by a single fragmentation reaction producing neutral losses of CH(2)N(2)O or CH(2)N(2)O(2) rather than a set of sequential reactions involving neutral losses of HNO(2) or HNO and HCN.  相似文献

采用中性解吸电喷雾萃取电离质谱( ND-EESI-MS)技术,在无需样品预处理的条件下,建立了对蜂蜜中敌敌畏直接快速检测的方法。在正离子模式下,敌敌畏质子化离子峰位于 m/z 223,二级特征离子为m/z 109和127。在优化的条件下,以m/z 127的信号强度为定量指标,建立了蜂蜜中敌敌畏残留的定量检测方法。结果表明,在蜂蜜基质中,敌敌畏在5~1000 ng/mL浓度范围内与m/z 127的信号强度线性关系良好,相关系数为0.998,检出限为1.0 ng/mL(S/N=3);蜂蜜中3个加标水平(10,30和400 ng/mL)的敌敌畏的回收率为93.0%~103.0%,精密度(RSDs)小于4.4%。同时采用气相色谱(火焰光度检测器)方法作为对照方法,检测敌敌畏加标蜂蜜样品,结果表明,加标蜂蜜在5~1000 ng/mL浓度范围内与峰面积线性关系良好,相关系数为0.999,检出限为1.6 ng/mL;10,30和400 ng/mL 3个水平加标蜂蜜的回收率为94.9%~110.3%,精密度小于7.6%。  相似文献
