Xanthohumol, isoxanthohumol, and 8‐prenylnaringenin in beer, hop and hop pellet samples were analyzed by HPLC using an InertSustain phenyl column and the mobile phase containing 40% methanol and 12% 2‐propanol. Fractions of isoxanthohumol and 8‐prenylnaringenin obtained by the above HPLC were separately collected. Isoxanthohumol and 8‐prenylnaringenin were enantioseparated by HPLC using a Chiralcel OD‐H column with a mobile phase composed of hexane–ethanol (90:10, v/v) and a Chiralpak AD‐RH column with a mobile phase composed of methanol–2‐propanol–water (40:20:40, v/v/v), respectively. Isoxanthohumol and 8‐prenylnaringenin from beer, hop and hop pellet samples were found to be present in a racemic mixture. This can be explained by the fact that the two analytes were produced by a nonenzymatic process. The effects of boiling conditions on the conversion of xanthohumol into isoxanthohumol were also studied. A higher concentration of ethanol in heating solvent resulted in a decrease in the conversion ratio and the conversion was stopped by addition of ethanol at >50% (v/v). The isomerization was significantly affected pH (2−10) and the boiling medium at pH 5 was minimum for the conversion. Therefore, it was suggested that xanthohumol was relatively difficult to convert to isoxanthohumol in wort (pH 5−5.5) during boiling.  相似文献

Given the health-beneficial properties of compounds from hop, there is still a growing trend towards developing successful extraction methods with the highest yield and also receiving the products with high added value. The aim of this study was to develop efficient extraction method for isolation of bioactive compounds from the Polish “Marynka” hop variety. The modified two-step supercritical fluid extraction allowed to obtain two hop samples, namely crude extract (E1), composed of α-acids, β-acids, and terpene derivatives, as well as pure xanthohumol with higher yield than that of other available methods. The post-extraction residues (R1) were re-extracted in order to obtain extract E2 enriched in xanthohumol. Then, both samples were subjected to investigation of their antibacterial (anti-acne, anti-caries), cytotoxic, and anti-proliferative activities in vitro. It was demonstrated that extract (E1) possessed more beneficial biological properties than xanthohumol. It exhibited not only better antibacterial activity against Gram-positive bacteria strains (MIC, MBC) but also possessed a higher synergistic effect with commercial antibiotics when compared to xanthohumol. Moreover, cell culture experiments revealed that crude extract neither inhibited viability nor divisions of normal skin fibroblasts as strongly as xanthohumol. In turn, calculated selectivity indexes showed that the crude extract had from slightly to significantly better selective anti-proliferative activity towards cancer cells in comparison with xanthohumol.  相似文献

In this study, silica-coated magnetic nanoparticles (Fe(3)O(4)/SiO(2) NPs) modified by cetyltrimethylammonium bromide (CTAB) were synthesized. They were successfully applied for extraction of xanthohumol in beer based on magnetic mixed hemimicelles solid-phase extraction (MMHSPE) coupled with high-performance liquid chromatography-ultraviolet determination. The main factors influencing the extraction efficiency including the surfactant amount, the beer pH, the extraction time, the desorption condition and the maximum extraction beer volume were optimized. Under the optimized conditions, a concentration factor of 60 was achieved by extracting 120 mL beer sample using MMHSPE and the detection limit of xanthohumol is 0.0006 mg/L. The proposed method was successfully applied for determination of xanthohumol in various beer samples with the xanthohumol contents in the range of 0.031-0.567 mg/L. The satisfactory recoveries (90-103%) were obtained in analyzing spiked beer samples.  相似文献

In the present study, the composition of the hop strobilus extract by using different extraction methods under different solvent conditions was analysed and compared. Several separation methods were applied to obtaining detailed information about the hop extract: capillary zone electrophoresis (CZE), high-performance liquid chromatography-mass-spectrometry (HPLC-MS/MS) and gas chromatography-mass-spectrometry (GC-MS). A preparative supercritical fluid extraction system is described and was used with supercritical carbon dioxide to extract the active insecticide components pyrethrin I (PI) and pyrethrin II (PII) successfully from pyrethrum flower. A high-performance liquid chromatography method was developed and was used to separate and analyze the supercritical carbon dioxide extracts. Extraction efficiencies under several different extraction conditions were examined. Under the conditions examined, the most effective extractions of PI and PII (140 +/- 18 mg and 55 +/- 9 mg per 100 g of dry pyrethrum flower powder) were performed at 40 degrees C and 1200 psi. The results showed that extraction efficiencies of supercritical carbon dioxide are much better than those of n-hexane for pyrethrins I and II. During the extraction process, the most efficient extraction period was the first 3 h of the experiment.  相似文献

Xanthohumol was isolated from supercritical carbon dioxide-spent hop and transformed into isoxanthohumol. The demethylation of isoxanthohumol with the best yield 93% occurred when MgI2 etherate in anhydrous THF was applied. Salts such as MgBr2, MgCl2, CaI2, Mg(OAc)2, Mg(OMe)2 were also investigated. A convenient method for the xanthohumol isolation from supercritical carbon dioxide-spent hop is also described.  相似文献

Evodiamine and rutaecarpine have been intensively studied due to their pharmacological actions and clinical applications. In this report, supercritical fluid was used to extract evodiamine and rutaecarpine from the unripe fruit of Evodia rutaecarpa. Response surface methodology using Box-Behnken experimental design was utilized to optimize parameters for supercritical carbon dioxide extraction with methanol as co-solvent. The effect of various values of dynamic extraction time (30-90min), temperature (50-70°C) and pressure (200-400bar) on extraction yields of the two compounds was evaluated. Determinations of the extracts were performed by high-performance liquid chromatography. The experimental data obtained were fitted to second-order polynomial equations and analyzed by analysis of variance. The highest yields predicted were 1.217mg/g for evodiamine and 0.969mg/g for rutaecarpine at the optimal values (time 78min, temperature 62°C, pressure 280bar and co-solvent flow rate 0.4mL/min), based on the selected range of experimental conditions.  相似文献

Two less laborious extraction methods, viz. (i) a simplified liquid extraction using light petroleum or (ii) microwave-assisted solvent extraction (MASE), for the analysis of polycyclic aromatic hydrocarbons (PAHs) in samples of the compost worm Eisenia andrei, were compared with a reference method. After extraction and concentration, analytical methodology consisted of a cleanup of (part) of the extract with high-performance gel permeation chromatography (HPGPC) and instrumental analysis of 15 PAHs with reversed-phase liquid chromatography with fluorescence detection (RPLC-FLD). Comparison of the methods was done by analysing samples with incurred residues (n=15, each method) originating from an experiment in which worms were exposed to a soil contaminated with PAHs. Simultaneously, the performance of the total lipid determination of each method was established. Evaluation of the data by means of principal component analysis (PCA) and analysis of variance (ANOVA) revealed that the performance of the light petroleum method for both the extraction of PAHs (concentration range 1-30 ng/g) and lipid content corresponds very well with the reference method. Compared to the reference method, the MASE method yielded somewhat lower concentrations for the less volatile PAHs, e.g., dibenzo[ah]anthracene and benzo[ghi]perylene and provided a significant higher amount of co-extracted material.  相似文献

The concentration of endogenously formed tryptamine in central nervous system tissue was determined after extraction into ethyl acetate, purification on a weak cation-exchange resin and analysis using high-performance liquid chromatography with fluorometric detection. Final chromatographic separation of this indoleamine was achieved using a muBondapak C18 reversed-phase column under isocratic conditions. Using this method, the concentration of tryptamine in the whole brain of normal rats was found to be 0.60 +/- 0.06 ng/g of tissue, while pretreatment with dl-p-chlorophenylalanine, tranylcypromine and l-tryptophan increased the concentration to 96.7 +/- 21.9 ng/g.  相似文献

This work describes the development, validation, and application of a novel methodology for the determination of testosterone and methenolone in urine matrices by stir bar sorptive extraction using polyurethane foams [SBSE(PU)] followed by liquid desorption and high-performance liquid chromatography with diode array detection. The methodology was optimized in terms of extraction time, agitation speed, pH, ionic strength and organic modifier, as well as back-extraction solvent and desorption time. Under optimized experimental conditions, convenient accuracy were achieved with average recoveries of 49.7 8.6% for testosterone and 54.2 ± 4.7% for methenolone. Additionally, the methodology showed good precision (<9%), excellent linear dynamic ranges (>0.9963) and convenient detection limits (0.2-0.3 μg/L). When comparing the efficiency obtained by SBSE(PU) and with the conventional polydimethylsiloxane phase [SBSE(PDMS)], yields up to four-fold higher are attained for the former, under the same experimental conditions. The application of the proposed methodology for the analysis of testosterone and methenolone in urine matrices showed negligible matrix effects and good analytical performance.  相似文献

A rapid and sensitive high-performance liquid chromatography (LC) method was developed for the simultaneous determination of metronidazole (MNZ), dimetridazole (DMZ), ronidazole (RNZ), tinidazole (TNZ), and 2-hydroxymethyl-1-methyl-5-nitroimidazole (HMMNI) in honey. After extraction with ethyl acetate and evaporation, the residue containing the nitroimidazoles was dissolved in ethyl acetate-hexane and subjected to solid-phase extraction cleanup by amine extraction columns. The effluent was evaporated to dryness, and residues were dissolved and determined by LC with an ultraviolet detector set at 315 nm. The limits of detection were 1.0-2.0 ng/g for MNZ, DMZ, RNZ, TNZ, and HMMNI in honey. Average recoveries ranged from 71.5-101.4% in honey fortified at 10, 20, 50, and 100 ng/g. The method was validated for the analysis of real honey samples.  相似文献

采用Waters Sep-Pak C18固相萃取小柱对啤酒样品进行分离纯化,建立了啤酒中黄腐酚的固相萃取-高效液相色谱检测方法。选用色谱柱Zorbax Eclipse XDB-C18柱(250 mm×4.6 mm,5 μm),以甲醇和0.1%甲酸水溶液为流动相进行梯度洗脱,柱温25 ℃,流速0.4 mL/min,检测波长370 nm。在此条件下,黄腐酚分离良好且无杂质峰干扰,在0.5~500 μg/L的范围内线性关系良好(r21),在高、中、低浓度下的加标回收率为91.21%~95.58%,相对标准偏差小于2%。方法的检出限为0.24 μg/L,定量限为0.80 μg/L。该方法简便快速、结果准确、重现性好,是检测啤酒中黄腐酚含量的有效方法。  相似文献

A fast ultra-performance liquid chromatography method was developed for the separation and quantification of doxycycline, 4-epidoxycycline, and 6-epidoxycycline in the muscles of channel catfish, shrimp, eel and turtle. The analytes were separated by comprehensive optimization of the chromatographic conditions. The extraction method was performed using a modified QuEChERS methodology. 2?g of muscle sample were mixed with 5?mL Mcllvaine buffer (pH 4), and 4?g of sodium sulfate and 2?g of sodium chloride were added for liquid–liquid partitioning. The mixture was agitated for 30?s and extracted with 10?mL of acidified acetonitrile. The resulting extract was purified using 200?mg of a C18-silica adsorbent. The samples were analyzed using ultra-performance liquid chromatography with ultraviolet detection using a BEH C18 column (2.1?mm ×100?mm, 1.7?μm). The limits of detection were less than 25?μg/kg and the limits of quantitation were less than 50?μg/kg for all analytes. The recoveries of the analytes were from 62.3% to 89.0% with relative standard derivations below 10.0%. The method provided efficient extraction and purification that enabled rugged and low-cost determination of the analytes.  相似文献

Determination of small amounts of vitamin K1 (0.8 microg/g) in nutritional supplements with high fat content (20 mg/g) was performed by solid-phase extraction and high-performance liquid chromatography (HPLC) with fluorescence detection after reduction on a platinum oxide catalyst. The concentration ratio of plant oils to vitamin K1 (0.8 microg/g) was about 25,000:1. A sample solution was applied to a solid-phase extraction cartridge and vitamin K1 was eluted with ethanol, followed by HPLC. The proposed method was simple, rapid (analysis time: ca. 12 min), sensitive [detection limit: ca. 0.1 pg per injection (100 microl) at a signal-to-noise ratio of 3:1], highly selective and reproducible [relative standard deviation: ca. 1.3%. (n=5)]. The calibration graph of vitamin K1 was linear in the range of 0-2 pg per injection (100 microl). Recovery of vitamin K1 was over 90% by the standard addition method.  相似文献

Stir bar sorptive extraction with polyurethane (PU) and polydimethylsiloxane (PDMS) polymeric phases followed by high-performance liquid chromatography with diode array detection [SBSE(PU or PDMS)/HPLC-DAD] was studied for the determination of six acidic pharmaceuticals [o-acetylsalicylic acid (ACA), ibuprofen (IBU), diclofenac sodium (DIC), naproxen (NAP), mefenamic acid (MEF) and gemfibrozil (GEM)], selected as non-steroidal acidic anti-inflammatory drugs and lipid regulators model compounds in environmental water matrices. The main parameters affecting the efficiency of the proposed methodology are fully discussed. Assays performed on 25 mL of water samples spiked at the 10 microg L(-1) level under optimized experimental conditions, yielded recoveries ranging from 45.3+/-9.0% (ACA) to 90.6+/-7.2% (IBU) by SBSE(PU) and 9.8+/-1.6% (NAP) to 73.4+/-5.0% (GEM) by SBSE(PDMS), where the former polymeric phase presented a better affinity to extract these target analytes from water matrices at the trace level. The methodology showed also excellent linear dynamic ranges for the six acidic pharmaceuticals studied, with correlation coefficients higher than 0.9976, limits of detection and quantification between 0.40-1.7 microg L(-1) and 1.5-5.8 microg L(-1), respectively, and suitable precision (RSD <15%). Moreover, the developed methodology was applied for the determination of these target analytes in several environmental matrices, including river, sea and wastewater samples, having achieved good performance and moderate matrix effects. In short, the PU foams demonstrated to be an excellent alternative for the enrichment of the more polar metabolites from water matrices by SBSE, overcoming the limitations of the conventional PDMS phase.  相似文献

Supported liquid membrane microextraction (SLMME) with high-performance liquid chromatography (HPLC)-UV detection has been developed for the extraction, preconcentration, and determination of all the nine haloacetic acids (HAAs) in water. The HAAs are extracted into a supported liquid membrane, and then back-extracted into few microliters of an acceptor solution. The extract was directly analyzed by HPLC-UV with a 15-min run. Enrichment factors in the range of 300-3000 were obtained in a 60-min extraction, and detection limits were at low to sub-microg/L level with R.S.D. values between 1.5 and 10.8%. The parameters that affected analyte enrichment were studied. This approach offers an attractive alternative to the current US Environmental Protection Agency standard methods for HAA analysis, which require complex sample preparation and derivatization prior to analysis by gas chromatography. SLMME can also be used in conjunction with other analytical schemes, such as, ion chromatography and capillary electrophoresis.  相似文献

A simple and reliable reversed-phase high-performance liquid chromatography method was developed and validated for the determination of DHP-014, a niguldipine analogue with potent P-glycoprotein inhibitory and negligible calcium channel blocking properties, in rat plasma. DHP-014 and niguldipine hydrochloride (the internal standard) were extracted from rat plasma by liquid extraction using hexane. DHP-014 was then separated by HPLC on a C18 column and quantified by ultraviolet detection at 238 nm. The mobile phase consisted of acetonitrile-aqueous 5 mM phosphate buffer (65:35, v/v) containing 0.4% (v/v) triethylamine adjusted to pH 7.0. The mean extraction efficiency of DHP-014 was 109.0 +/- 12.9, 97.7 +/- 8.0 and 102.9 +/- 7.5% for DHP-014 concentrations of 10, 50 and 100 nM, respectively (n = 5). The method was linear over the concentration range 2.5-200 nM with a regression coefficient of 0.998. The limit of detection of DHP-014 in rat plasma was 1.0 nM. The intra- and inter-day coefficients of variation for DHP-014 in rat plasma were 4.7-7.9 and 6.9-9.9%, respectively. The intra- and inter-day accuracy was 98.2-99.5 and 97.9-103%, respectively. The bioanalytical technique was used to determine DHP-014 in plasma samples in a pharmacokinetic study of DHP-014 administered to female Sprague-Dawley rats.  相似文献

A high-performance liquid chromatography (HPLC) system with fluorescence detection and an automated on-line solid-phase extraction procedure for fumonisins B1 and B2 in corn and corn-based products is described. Different amounts of strong anion-exchange, C18 and end-capped C18 (C(18 ec)) silicas were tested for sample clean-up. Various HPLC parameters were analyzed. The best methodology was found to be extraction with acetonitrile-water and clean up on C(18 ec) disposable extraction cartridges. The system has the advantage of running in an unattended mode of operation and allows processing of 40 samples without system refuel, performing clean-up, o-phthaldialdehyde derivatization, injection and fumonisin detection by fluorescence detection linked to a computer integrator for automated data processing. Recoveries were performed with corn and corn-based feed samples (n=3) spiked with 0.1, 0.5, 1.0, 5.0 and 10 microg/g. Average recoveries for corn and corn-based feed were, respectively, 92.6 and 88.3% with relative standard deviations (RSDs) of 5.04 and 6.22%, for fumonisin B1 and 91.2 and 89.0% with RSDs of 5.84 and 7.88% for fumonisin B2. Detection limits (S/N=3) for corn and corn-based feed were approximately 0.03 microg/g for fumonisin B1 and 0.05 microg/g for fumonisin B2  相似文献

A high-performance liquid chromatographic (HPLC) method for determining glycarbylamide (GB) in chicken tissue was developed. GB was extracted with acetonitrile, followed by solid-phase extraction cleanup using a Bond Elut cartridge column with neutral alumina. After the extract had been evaporated to dryness, the residue was dissolved in 1.0 mL 0.1 N sodium hydroxide. Then 1.0 mL 0.1 M potassium dihydrogen phosphate solution was added to it. HPLC separation was done on a 250 x 4.6 mm id TSK-GEL ODS 80 column with 0.05M potassium dihydrogen phosphate as the mobile phase. Ultraviolet detection was done at a wavelength of 260 nm. The calibration curve of standard GB solutions was linear between 0.16 and 3 micrograms/mL (correlation coefficient, r = 0.999). The recovery of GB from chicken muscle spiked at 0.8 microgram/g was 88.6 +/- 2.3% (mean +/- standard deviation, n = 5), and the lower limit of determination was 0.05 microgram/g in chicken muscle.  相似文献
