四氢异喹啉衍生物的合成研究

以间甲氧基苯乙胺和氯甲酸乙酯为原料,通过亲核取代、关环、铝锂氢还原、脱甲基以及在N位上Boc保护及脱保护等9步反应得到一系列重要的1,2,3,4-四氢异喹啉及其衍生物.产物的结构经核磁共振氢谱、红外光谱及元素分析确证.与传统四氢异喹啉衍生物的合成工艺相比,该工艺反应条件温和、简便、经济、产率高且对环境友好.     

Synthesis and biological activity of 1-(aryloxy)-3-(6-chloro-4,4-dimethyl-1,2,3,4-tetrahydroisoquinolin-2-yl)propan-2-ols

Treatment of 1-(4-chlorobenzylamino)-2-methylpropan-2-ol with concentrated sulfuric acid at 0°C gave 6-chloro-4,4-dimethyl-1,2,3,4-tetrahydroisoquinoline which reacted with (aryloxymethyl)oxiranes to afford new propan-2-ol derivatives of the tetrahydroisoquinoline series, 1-(aryloxy)-3-(6-chloro-4,4-dimethyl-1,2,3,4-tetrahydroisoquinolin-2-yl)propan-2-ols. Some of the synthesized compounds or their hydrochlorides showed moderate adrenergic blocking and sympatholytic activities.     

Cleavage of 2-(1,2,3,4-tetrahydroisoquinolin-1-yl)-2-phenyl-1,3-dithianes with HgO/BF3

Summary The title compounds (5) were prepared by addition of 2-phenyl-1,3-dithiane anion (2-lithiated10) to adequately substituted N-alkyl-3,4-dihydroisoquinolinium salts (7a–7g). Cleavage of compounds5 with HgO/BF₃ affords S-benzoyl-1,3-propanedithiol (4a) and the corresponding disulfide4c, benzaldehyde, and Hg(I) ions. In contrast to the title compounds, 2-(-dialkylaminobenzyl)-2-phenyl-1,3-dithianes (6) yield benzil under these conditions.In commemoration of the late Prof. Dr. Dr. h.c. mult.Horst Böhme, Marburg/Germany, an outstanding representative of Pharmaceutical Chemistry     

Synthesis and X-ray diffraction study of 5-(8-methoxy-2-methyl-6,7-methylenedioxy-1,2,3,4-tetrahydroisoquinolin-1-yl)-1,3-dimethylbarbituric acid

The reaction of cotarnine with 1,3-dimethylbarbituric acid afforded 5-(8-methoxy-2-methyl-6,7-methylenedioxy-1,2,3,4-tetrahydroisoquinolin-1-yl)-1,3-dimethylbarbituric acid. Its crystal structure was established by X-ray diffraction analysis and the structure in solution was studied by ¹H NMR spectroscopy. This compound has a zwitterionic structure. In the crystal, the molecules are linked in dimers by intermolecular hydrogen bonds, and in solutions, the dimers occur in equilibrium with the monomers.     

Synthesis of 6-Methyl-4-(1-methyl-2-buten-1-yl)-2-(2-cyclohexen-1-yl)- and 6-Methyl-4-(1-methyl-2-buten-1-yl)-2-(1-cyclohexen-1-yl)anilines

1-(Cyano[benzimidazole-2-yl])methylene-3,3-Dimethyl-1,2,3,4-Tetrahydroisoquinoline: Synthesis,Structure, Spectral Parameters

1-(cyano[benzimidazole-2-yl])methylene-3,3-dimethyl-1,2,3,4-tetrahydroisoquinoline was synthesized and studied by X-ray diffraction analysis and IR and electronic absorption spectroscopy. The model quantum-chemical calculations were performed.     

Synthesis of Derivatives of 2-(Indol-1-yl)propionic Acids

We have developed a method for synthesis of N-substituted derivatives of indole from N-phenylalanine, including Fischer cyclization of the corresponding arylalkylhydrazones. The starting hydrazines were obtained by reduction of the corresponding nitrosamines. We have established that the optimum reduction method is to use metallic zinc in a methanol-hydrochloric acid system at low temperatures.

Synthesis of 3-(Benzotriazol-1-yl)naphthoquinone Derivatives

2,3-Bis(benzotriazol-1-yl)-1,4-naphthoquinone reacts with aliphatic and aromatic amines to give the corresponding 2-amino-3-(benzotriazol-1yl)-1,4-naphthoquinones, and its reaction with alkali gives 2-hydroxy-3-(benzotriazol-1-yl)-1,4-naphthoquinone.     

2-(1H-唑基)-1-(2,3,4-三甲氧基)苯乙酮衍生物的合成与结构表征

首先以2－溴－1－（2，3，4－三甲氧基）苯乙酮，四丁基铵－1H－1，2，4－三唑或咪唑为原料，分别合成了2－（1H－1，2，4－三唑－1－基）－1－（2，3，4－三甲氧基）苯乙酮（Ia)和2－（1H－咪唑0－1－基）－1－（2，3，4－三甲氧基）苯乙酮（Ib)与盐酸羟胺反应生成2－（1H－1，2，4－三唑－1－基）－1－（2，3，4－三甲氧基）苯乙酮肟和1－（1H－咪唑－1－基）－1－（2，3，4三甲氧基）苯乙酮肟，产率为63％－72％，并经元素分析，IR和1H NMR进行了表征。     

Short Synthesis of 1-(3-tert-Butyl-1-phenyl-1H-pyrazol-5-yl)-3-(5-(2-morpholinoethoxy)-2H-chromen-8-yl) Urea Derivatives

A short and efficient synthesis of 1-(3-tert-butyl-1-phenyl-1H-pyrazol-5-yl)-3-(5-(2-morpholinoethoxy)-2H-chromen-8-yl) urea derivatives (1a–c), a novel type of p38 MAPK inhibitors, is described. The Claisen thermal rearrangement of arylpropargyl ethers was employd as a key step to synthesize the chromene core. The solvent effect on the ratio of the resultant two isomers of Claisen thermal rearrangement, namely 2-methylbenzofuran and 2H-chromen, was also investigated.     

Functiaonalization of 2-(1-Cyclohexen-1-yl)aniline Derivatives

The reaction of 2-(1-cyclohexen-1-yl)aniline and -6-methylaniline with phthalic anhydride has afforded 2-(2-cyclohex-1-en-1-ylphenyl)- and 2-(2-cyclohex-1-en-1-ylphenyl)-6-methylphenyl)-1H-isoindole-1,3(2H)-diones. The reaction of the obtained isoindole-1,3-diones with bromine in dichloromethane in the presence of sodium bicarbonate has led to the formation of the product of pseudo-allylic halogenation. Replacement of the halogen atom by methoxy group has been performed by keeping 2-[2-(6-bromocyclohex-1-en-1-ylphenyl)-6-methylphenyl)]-1H-isoindole-1,3(2H)-dione in a methanolic solution in the presence of NaHCO₃. The reaction of 2-(2-cyclohex-1-en-1-yl-6-methylphenyl)-1H-isoindole-1,3(2H)-dione with molecular bromine in the presence of methanol has given a co-halogenation product, whereas the dibromination product has been obtained in the presence of octyl alcohol.

新型2-(吡咯烷-3-基)-噁唑衍生物的合成

根据亚结构链接法,以氯甲基三甲基硅烷和苄胺为原料,依次经酯化、缩合、环合、水解、成盐等12步反应制得5个新型的(4-甲基氨基甲酰基-2-吡咯烷-5-甲基噁唑) 胺基甲酸叔丁酯衍生物(15a~15e); 15a与醛(酮)或乙酰氯反应,合成了4个新型的5-胺基甲基-2-(1-甲基-吡咯烷)-噁唑-N-甲基甲酰胺盐酸盐衍生物,其结构经¹H NMR和ESI-MS表征。     

Synthesis of 3-(2-(4,5-Dihydro-3,5-diphenylpyrazol-1-yl)thiazol-4-yl)-2H-chromen-2-one Derivatives via Multicomponent Approach

An efficient synthesis of 3-(2-(4,5-dihydro-3,5-diphenylpyrazol-1-yl)thiazol-4-yl)-2H-chromen-2-one derivatives by a simple, atom-economical, and multicomponent reaction of chalcones, thiosemicarbazide, and 3-(2-bromoacetyl) coumarins in the presence of aqueous sodium hydroxide in refluxing ethanol is reported. The structures of newly prepared compounds have been confirmed by their analytical and spectral (IR, ¹HNMR, ¹³CNMR and mass) data.

Facile and Simple Synthesis of Novel 1-Methyl-2-(2-substituted-oxazol-4-yl)-1H-benzimidazole Derivatives

Novel 1-methyl-2-(2-substituted-oxazol-4-yl)-1H-benzimidazole derivatives were obtained in good yields and purity by treating corresponding 2-benzimidazolyl esters with acetamide in the presence of BF₃-etherate, a Lewis acid.
