Summary: Novel temperature‐responsive poly(N‐isopropylacrylamide) (PNIPAAm) nanoparticle‐containing PNIPAAm hydrogels were prepared by radical polymerization. In comparison with conventional PNIPAAm hydrogels, the PNIPAAm gels thus prepared exhibit much faster response rates as the temperature is raised above the lower critical solution temperature. The improved properties are a result of the incorporation of PNIPAAm particles, which first shrink and then generate pores for water molecules to be quickly squeezed out of the bulky PNIPAAm gels.

Thermosensitive graphene‐polymer composites have been prepared by attaching poly(N‐isopropylacrylamide) (PNIPAAm) onto the basal plane of graphene sheets via π‐π stacking. Pyrene‐terminated PNIPAAm was synthesized using reversible addition fragmentation chain transfer (RAFT) polymerization via a pyrene‐functional RAFT agent. Aqueous solutions of the graphene‐polymer composites were stable and thermosensitive. The lower critical solution temperature (LCST) of pyrene‐terminated PNIPAAm was measured to be 33 °C. When the pyrene‐functional polymer was attached to graphene the resultant composites were also thermosensitive in aqueous solutions exhibiting a reversible suspension behavior at 24 °C. Atomic force microscopy (AFM) analysis revealed that the thickness of a graphene‐PNIPAAm (Mn: 10,000 and PDI: 1.1) sheet was ~5.0 nm. The surface coverage of polymer chains on the graphene basal plane was calculated to be 7.2 × 10?11 mol cm?2. The graphene‐PNIPAAm composite material was successfully characterized using X‐ray photoelectron spectroscopy (XPS), attenuated total reflection infrared (ATR‐IR) spectroscopy, and thermogravimetric analysis (TGA). © 2009 Wiley Periodicals, Inc. In this work, poly(N‐isopropylacrylamide‐co‐acrylic acid) (poly(NIPAAm‐AA)) copolymer latex particles (microgels) were synthesized by the method of soapless emulsion polymerization. Poly(NIPAAm‐AA) copolymer microgels have the property of being thermosensitive. The concentration of acrylic acid (AA) and crosslinking agent N,N′‐methylenebisacrylamide were important factors to influence the lower critical solution temperature (LCST) of poly(NIPAAm‐AA) microgels. The effects of AA and crosslinking agent on the swelling behavior of poly(NIPAAm‐AA) microgels were also studied. The poly(NIPAAm‐AA) copolymer microgels were then used as a thermosensitive drug carrier to load caffeine. The effects of concentration of AA and crosslinking agent on the control release of caffeine were investigated. How the AA content and crosslinking agent influenced the morphology and LCST of the microgels was discussed in detail. The relationship of morphology, swelling, and control release behavior of these thermosensitive microgels was established. Novel thermo‐responsive poly(N‐isopropylacrylamide)‐block‐poly(l ‐lactide)‐block‐poly(N‐isopropylacylamide) (PNIPAAm‐b‐PLLA‐b‐PNIPAAm) triblock copolymers were successfully prepared by atom transfer radical polymerization of NIPAAm with Br‐PLLA‐Br macroinitiator, using a CuCl/tris(2‐dimethylaminoethyl) amine (Me6TREN) complex as catalyst at 25 °C in a N,N‐dimethylformamide/water mixture. The molecular weight of the copolymers ranges from 18,000 to 38,000 g mol?1, and the dispersity from 1.10 to 1.28. Micelles are formed by self‐assembly of copolymers in aqueous medium at room temperature, as evidenced by 1H NMR, dynamic light scattering (DLS) and transmission electron microscopy (TEM). The critical micelle concentration determined by fluorescence spectroscopy ranges from 0.0077 to 0.016 mg mL?1. 1H NMR analysis in selective solvents confirmed the core‐shell structure of micelles. The copolymers exhibit a lower critical solution temperature (LCST) between 32.1 and 32.8 °C. The micelles are spherical in shape with a mean diameter between 31.4 and 83.3 nm, as determined by TEM and DLS. When the temperature is raised above the LCST, micelle size increases at high copolymer concentrations due to aggregation. In this paper, self‐assembled polymeric toroids formed by a temperature‐driven process are reported. Rhodamine B (RhB) end‐capped poly(N‐isopropylacrylamide) (PNIPAAm) demonstrating a lower critical solution temperature (LCST) is prepared. In a two‐phase system, the polymer in the aqueous phase could move to the chloroform phase on raising the temperature above its LCST. This temperature‐driven process results in the formation of polymeric toroids in the chloroform phase, and the strategy affords a new pathway to toroidal self‐assembly of polymers. Moreover, the photoluminescent behavior of the RhB end‐capped PNIPAAm species formed by the process is also studied and discussed.

This work focused on surface modification of magnetite nanoparticle (MNP) with poly(poly(ethylene glycol) monomethyl ether methacylate)‐b‐(poly(N‐isopropylacrylamide)‐st‐poly(thiolactone acrylamide)), PPEGMA‐b‐(PNIPAAm‐st‐PTlaAm), diblock copolymer, synthesized via reversible addition‐fragmentation chain transfer (RAFT) polymerization to obtain the particles having good water dispersible PPEGMA brushes, thermo‐responsive PNIPAAm, and reactive thiolactone groups of PTlaAm. The thiolactone moiety in the copolymer can readily react with amino groups grafted on MNP surface and essentially induced the formation of MNP nanocluster. According to transmission electron microscopy (TEM), the size of the nanocluster ranged between 200 and 500 nm per cluster with 8 to 10 nm in diameter for each particle. Hydrodynamic diameter of the nanocluster significantly decreased as the dispersion temperature increased from 25°C to 45°C due to the shrinkage of thermo‐responsive PNIPAAm when crossing its lower critical solution temperature (LCST). This stable nanocluster might be potentially used as a magnetic carrier for control release of entrapped entities with a thermally triggering mechanism.  相似文献

Environmental switches may be fabricated for the controlled release of pharmaceutical drug using a thermally responsive polymer with the intrinsic chemical and physical nature of stimuli‐sensitive smart materials. Particularly, much attention has been paid to the biomedical applications of poly(N‐isopropyl acrylamide) (PNIPAAm) because of its unique reversible transition at a specific lower critical solution temperature (LCST).Thermally sensitive block copolymers, poly(N‐isopropyl acrylamide‐b‐poly(L ‐lactide‐co‐glycolide) (PNIPAAm‐b‐PLGA), and polyethylene glycol‐poly (lactide‐co‐glycolide) (PEG‐PLGA) triblock copolymers with different compositions and length of PLGA block were synthesized via ring‐opening polymerization of lactide and glycolide in the presence of OH‐terminated PNIPAAm or PEG. The composition and structure of the polymer were determined by NMR and FTIR. The effect of important factors, such as ionic strength, pH, and polymer concentration on the phase transition behavior of temperature‐sensitive polymers, were investigated by cloud point measurements. The resulting thermosensitive polymers were used for the entrapment of a narcotic antagonist drug, naltrexone, as the model drug. The loading efficiency and drug release behavior of naltrexone‐loaded hydrogels were investigated. The naltrexone loaded thermosensitive polymers were able to sustain the release of naltrexone for different periods of time, depending on the polymer composition, and concentration. In vitro release studies showed that these thermosensitive polymers are able to deliver naltrexone in biologically active forms at a controlled rate for 3–8 weeks. A novel double brush‐shaped copolymer with amphiphilic polyacrylate‐b‐poly(ethylene glycol)‐b‐poly acrylate copolymer (PA‐b‐PEG‐b‐PA) as a backbone and thermosensitive poly(N‐isopropylacrylamide) (PNIPAM) long side chains at both ends of the PEG was synthesized via an atom transfer radical polymerization (ATRP) route, and the structure was confirmed by FTIR, 1H NMR, and SEC. The thermosensitive self‐assembly behavior was examined via UV‐vis, TEM, DLS, and surface tension measurements, etc. The self‐assembled micelles, with low critical solution temperatures (LCST) of 34–38 °C, form irregular fusiform and/or spherical morphologies with single, double, and petaling cores in aqueous solution at room temperature, while above the LCST the micelles took on more regular and smooth spherical shapes with diameter ranges from 45 to 100 nm. The micelle exhibits high stabilities even in simulated physiological media, with low critical micellization concentration (CMC) up to 5.50, 4.89, and 5.05 mg L?1 in aqueous solution, pH 1.4 and 7.4 PBS solutions, respectively. The TEM and DLS determination reveled that the copolymer micelle had broad size distribution below its LCST while it produces narrow and homogeneous size above the LCST. The cytotoxicity was investigated by MTT assays to elucidate the application potential of the as‐prepared block polymer brushes as drug controlled release vehicles. © 2012 Wiley Periodicals, Inc. Exfoliated montmorillonite (MMT)/poly(N‐isopropylacrylamide) (PNIPAAm) and MMT/poly(N‐isopropylacrylamide‐co‐acrylamide) [P(NIPAAm‐co‐AAm)] nanocomposites were fabricated by soap‐free emulsion polymerization. Interestingly, as the content of MMT was increased from 0 to 10 wt %, the glass transition temperature of MMT/PNIPAAm was decreased from 145 to 122 °C, whereas that of the MMT/P(NIPAAm‐co‐AAm) increased from 95 to 153 °C. Although the lower critical solution temperature (LCST) of 32 °C for the MMT/PNIPAAm nanocomposites in aqueous solutions was slightly increased with the content of MMT, that of the MMT/P(NIPAAm‐co‐AAm) was decreased from 70 to 65 °C. A mechanism that the hydrogen bonds between the amide groups of PNIPAAm were interfered by the exfoliated MMT nano‐platelets for the MMT/PNIPAAm nanocomposites and the preferred absorption of acrylamide units to the MMT nanoplatelets rather than N‐isopropylacrylamide in the MMT/P(NIPAAm‐co‐AAm) nanocomposites was suggested to interpret these unusual transition behavior. © 2009 Wiley Periodicals, Inc. Poly(styrene) nanospheres having poly(N-vinylisobutyramide)s (PNVIBA)s, which are structurally the same composition as well-known thermosensitive poly(N-isopropylacrylamide)s (PNIPAAm)s and show the thermosensitive property as well, on their surfaces were synthesized by the free radical polymerization of hydrophilic PNVIBA macromonomers and hydrophobic styrene with AIBN as a radical initiator in ethanol as a polar solvent and were characterized in regard to their thermosensitive properties. Both the NVIBA oligomers and PNVIBA macromonomers that we synthesized showed a lower critical solution temperature (LCST) at around 40°C, as was predicted by our previous research. The nanospheres were spherical in form and have a narrow size distribution. Their sizes could be controlled by varying the molecular weight of the macromonomers and the amount of it in feed. The size in the nanosphere became small above the LCST of the corresponding macromonomer, possibly due to thermosensitive shrinking of the PNVIBA on the nanosphere surface, while transmittance of its dispersion did not change at all at studied temperature range. The nanospheres having the PNVIBA on their surfaces, which response sharply to atmospheres such as dispersion temperature, can be significant and useful materials in technological and medical fields. © 1998 John Wiley & Sons, Inc. A novel amphiphilic thermosensitive poly(ethylene glycol)45b‐poly(methyl methacrylate46co‐3‐(trimethoxysilyl)propyl methacrylate)2b‐poly(N‐isopropylacrylamide)429 (PEG45b‐P(MMA46co‐MPMA2)‐b‐PNIPAAm429) triblock copolymer was synthesized via consecutive atom transfer radical polymerization techniques. The thermoinduced association behavior of the resulting triblock copolymers in aqueous medium was further investigated in detail by 1H NMR, transmission electron microscopy, and dynamic light scattering. The results showed that at the temperature (25 °C) below the LCST, PEG45b‐P(MMA46co‐MPMA2)‐b‐PNIPAAm429 triblock copolymers self‐assembled into the core crosslinked micelles with the hydrophobic P(MMA‐co‐MPMA) block constructing a dense core, protected by the mixed soluble PEG and PNIPAAm chains acting as a hydrophilic shell simultaneously. With an increase in temperature, the resulting core‐shell micelles converted into a new type of micelles with the hydrophilic PEG chains stretching out from the hydrophobic core through the collapsed PNIPAAm shell. On the other hand, at the temperature (40 °C) above the LCST, such triblock copolymers formed the crosslinked vesicles with the hydrophobic PNIPAAm and P(MMA‐co‐MPMA) blocks constructing a membrane core and the soluble PEG chains building the hydrophilic lumen and the shell. On further decreasing the temperature, the resulting vesicles underwent transformation from the shrunken to the expanded status, leading to the formation of swollen vesicles with enlarged size. 以AgNO3为金属源,通过乙醇将与聚N-异丙基丙烯酰胺接枝聚丙烯腈/聚苯乙烯(PNIPAAm-g-PAN/PSt)聚合物微球表面酰胺基团配位的银离子(Ag+)还原,一步法制备了PNIPAAm-g-PAN/PSt载银复合微球。通过傅立叶变换红外(FTIR)和紫外-可见光光谱表征发现,由Ag+还原所得的Ag纳米颗粒被成功地固载在PNIPAAm-g-PAN/PSt 微球上;用透射电子显微镜(TEM)对载银微球的大小和形态进行了表征;热重分析(TGA)结果表明,固载在微球表面的银纳米颗粒的含量(质量分数)为12%;抗菌实验结果表明,所制备的载银微球具有抗革兰氏阴性菌的活性。  相似文献

A new atom transfer radical polymerization (ATRP) initiator, namely, 2‐(1‐(2‐azidoethoxy)ethoxy)ethyl 2‐bromo‐2‐methylpropanoate containing both “cleavable” acetal linkage and “clickable” azido group was synthesized. Well‐defined azido‐terminated poly(N‐isopropylacrylamide)s (PNIPAAm‐N3)s with molecular weights and dispersity in the range 11,000–19,000 g mol?1 and 1.20–1.28, respectively, were synthesized employing the initiator by ATRP. Acetal containing PCL‐b‐PNIPAAm block copolymer was obtained by alkyne–azide click reaction of azido‐terminated PNIPAAm‐N3 with propargyl‐terminated PCL. Critical aggregation concentration (CAC) of PCL‐b‐PNIPAAm copolymer in aqueous solution was found to be 8.99 × 10?6 M. Lower critical solution temperature (LCST) of PCL‐b‐PNIPAAm copolymer was found to be 32 °C which was lower than that of the precursor PNIPAAm‐N3 (36.4 °C). The effect of dual stimuli viz . temperature and pH on size and morphology of the assemblies of PCL‐b‐PNIPAAm block copolymer revealed that the copolymer below LCST assembled in spherical micelles which subsequently transformed to unstable vesicles above the LCST. Heating these assemblies above 40 °C led to the precipitation of PCL‐b‐PNIPAAm block copolymer. Whereas, at decreased pH, micelles of PCL‐b‐PNIPAAm copolymer disintegrate due to the cleavage of acetal linkage and precipitation of hydrophobic hydroxyl‐terminated PCL. The encapsulated pyrene release kinetics from the micelles of synthesized PCL‐b‐PNIPAAm copolymer was found to be faster at higher temperature and at lower pH. © 2016 Wiley Periodicals, Inc. J. The well‐defined, thermosensitive and biodegradable graft copolymers, poly(N‐isopropylacrylamide)‐b‐[2‐hydroxyethyl methacrylate‐poly(ε‐caprolactone)]n (PNIPAAm‐b‐(HEMA‐PCL)n) (n = 3 or 9), were synthesized by combining reversible addition‐fragmentation chain transfer polymerization and macromonomer method. The copolymers were able to self‐assemble into micelles in water with low critical micellar concentration and demonstrated temperature sensitivity with a lower critical solution temperature at around 36 °C. Transmission electron microscopy shows that the micelles exhibit a nanosized spherical morphology within a size range of 30–100 nm. The PNIPAAm‐b‐(HEMA‐PCL)3 copolymer exhibited biodegradation and low cytotoxicity. The paclitaxel‐loaded PNIPAAm‐b‐(HEMA‐PCL)3 micelles displayed thermosensitive controlled release behavior, which indicates potential as drug carriers. © 2007 Wiley Periodicals, Inc. Summary: A novel thermosensitive amphiphilic copolymer of PNIPAm grafted polyphosphazene (PNIPAm‐g‐PPP) has been synthesized through a co‐substitution reaction of poly(dichlorophosphazene) with amino terminated NIPAm oligomer and glycine ethyl ester (GlyEt). The polymer obtained had a molar ratio of 1:5.25 PNIPAm to GlyEt and had a lower critical solution temperature (LCST) near 30 °C. The formation of polymeric micelles was confirmed by fluorescence, dynamic light scattering and transmission electron microscopy measurements.

The influence of small alkylamines with increasing carbon chain length (≤5) on the temperature‐induced precipitation of two N‐alkylacrylamide oligomers, poly‐N‐isopropylacrylamide [PNIPAAm; weight‐average molecular weight (Mw ) = 1600 g/mol] and poly‐N,N‐diethylacrylamide (PDEAAm; Mw = 4000 g/mol), from an aqueous solution was investigated. The alkylamines in question were too small to form micelles in the classical sense but were capable of premicellar aggregation. PNIPAAm was prepared by radical polymerization in the presence of a chain‐transfer agent and, therefore, carried a carboxylic acid end group. The structure was heterotactic. PDEAAm was prepared by anionic polymerization and, therefore, carried a butyl end group. The structure was predominately isotactic. The solubility of the oligomers was investigated by cloud‐point measurements and differential scanning calorimetry. In addition, pyrene was used as a fluorescent polarity probe. Alkylamines up to C2 depressed the lower critical solution temperature (LCST) of PNIPAAm, whereas higher alkylamines first depressed the LCST and at higher concentrations elevated it. The LCST minimum showed a clear dependence on the alkyl chain length and structure. For PDEAAm, only pentylamine addition resulted in an LCST minimum. Otherwise, the LCST was raised. When the critical self‐association concentration (CSAC) of the alkylamines in water was compared to the critical association concentration (CAC) in aqueous oligomer solutions, PDEAAm, but not PNIPAAm, stabilized mixed aggregates (CAC < CSAC). The transition enthalpy of PNIPAAm decreased with an increasing alkylamine concentration and became 0 above the CAC. In this research, thermo‐ and pH‐responsive chitosan‐based porous nanoparticles were prepared by the temperature‐dependent self assembly method. The chitosan‐graft‐poly(N‐isopropylacrylamide) (CS‐g‐PNIPAAm) copolymer solution was prepared through polymerization of N‐isopropylacrylamide (NIPAAm) monomer in the presence of chitosan (CS) solution using cerium ammounium nitrate as the initiator. Then, CS‐g‐PNIPAAm solution was diluted by deionized water and heated to 40 °C for CS‐g‐PNIPAAm self‐assembly. After that, CS‐g‐PNIPAAm assembled to form micelles in which shell layer was CS. Crosslinking agent was used to reinforce the micelle structure to form nanoparticle. The molar ratio of CS/NIPAAm in the feed mixture was changed to investigate its effect on structure, morphology, thermal‐ and pH‐responsive properties of the nanoparticles. TEM images showed that a porous structure of nanoparticles was developed. The synthesized nanoparticles carried positive charges on the surface and exhibited stimuli‐responsive properties, and their mean diameter thus could be manipulated by changing pH value and temperature of the environment. The nanoparticles showed a continuous release of the encapsulated doxycycline hyclate up to 10 days during an in‐vitro release experiment. These porous particles with environmentally sensitive properties are expected to be utilized in hydrophilic drug delivery system. © 2009 Wiley Periodicals, Inc. Temperature‐responsive diblock copolymers made of poly(N‐isopropylacrylamide) (PNIPAAm) generated by reversible addition‐fragmentation chain transfer (RAFT) polymerization and a single‐stranded DNA (ssDNA) self‐assembled into polymer micelles. The micelles consisted of the PNIPAAm core surrounded by the ssDNA corona with a hydrodynamic diameter up to 300 nm in an aqueous medium above the lower critical solution temperature. In a medium of high ionic strength, the formation of the fully matched duplex with the complementary ssDNA on the surface of the polymer micelles induced rapid and spontaneous aggregation. By contrast, the micelles remained dispersed under the identical conditions when single‐base‐substituted ssDNA was added to form the corresponding terminal‐mismatched duplex on the micellar surface. This highly sequence‐selective process took place irrespective of the size of the PNIPAAm core.  相似文献

In this work, the poly(methacrylic acid‐coN‐isopropylacrylamide) thermosensitive composite hollow latex particles was synthesized by a three‐step reaction. The first step was to synthesize the poly(methyl methacrylate‐co‐methacrylic acid) (poly(MMA‐MAA)) copolymer latex particles by the method of soapless emulsion polymerization. The second step was to polymerize methacrylic acid (MAA), N‐isopropylacrylamide (NIPAAm), and N,N′‐methylenebisacrylamide in the presence of poly(MMA‐MAA) latex particles to form the linear poly(methyl methacrylate‐co‐methacrylic acid)/crosslinking poly(methacrylic acid‐coN‐isopropylacrylamide) (poly(MMA‐MAA)/poly(MAA‐NIPAAm)) core–shell latex particles. In the third step, the core–shell latex particles were heated in the presence of ammonia solution to form the crosslinking poly(MAA‐NIPAAm) thermosensitive hollow latex particles. The morphologies of poly(MMA‐MAA)/poly(MAA‐NIPAAm) core–shell latex particles and poly(MAA‐NIPAAm) hollow latex particles were observed. The influences of crosslinking agent and shell composition on the lower critical solution temperature of poly(MMA‐MAA)/poly(MAA‐NIPAAm) core–shell latex particles and poly(MAA‐NIPAAm) hollow latex particles were, respectively, studied. Besides, the poly(MAA‐NIPAAm) thermosensitive hollow latex particles were used as carriers to load with the model drug, caffeine. Summary: A series of thermally responsive dendritic core-shell polymers were prepared based upon dendritic polyamidoamine (PAMAM), modified with carboxyl end-capped linear poly(N-isopropylacrylamide) (PNIPAAm-COOH) in different ratios via an esterification process to obtain PNIPAAm-g-PAMAM. The graft ratio of PNIPAAm could be adjusted by changing the feed ratio of PAMAM-OH to PNIPAAm-COOH and was verified by 1H NMR and gel penetration chromatography (GPC). The lower critical solution temperature (LCST) of PNIPAAm-g-PAMAM evaluated by UV-vis spectrophotometer was about 32 °C. Indomethacin (IMC) as a model drug was loaded in the thermosensitive polymer-grafted dendrimer derivative and its release behavior was studied below and above its LCST (27 °C vs 37 °C). Results showed that the LCST of PNIPAAm-g-PAMAM was around 32 °C compared with that of the pure PNIPAAm. The release behavior of the indomethacin entrapped in the internal cavities of the PNIPAAm-g-PAMAM showed that almost 77% of the drug was cumulatively released at 27 °C after 10 hours, whereas only 20% was released at 37 °C. The release rate of IMC from the IMC/PNIPAAm-g-PAMAM complex at 37 °C is significantly slower than that at 27 °C, which indicates that the PNIPAAm chains grafted on the surface of PAMAM dendrimer could act as a channel switching on-off button through expending or contracting in response to the temperature variation and could control the drug release by varying the surrounding temperature.  相似文献
