建立了中空纤维液相微萃取(HFLPME)耦合高效液相色谱法(HPLC)用于测定血浆和尿液中大黄素及其代谢物的浓度,比较了中药有效成分大黄素在不同性别大鼠体内的吸收和代谢能力,阐述了大黄素在体内的代谢和转化过程。本实验以聚偏氟乙烯纤维为溶剂载体,正辛醇为萃取溶剂,对血浆和尿液样品进行HFLPME处理,萃取后挥干有机溶剂,用50 μL甲醇溶解,进行HPLC测定。在优化的微萃取条件下,血浆和尿液样品中大黄素及其代谢物标准曲线线性良好(相关系数(r)大于0.9960);检出限为0.1~3.0 μg/L;富集倍数为12.2~26.3;日内、日间精密度(以相对标准偏差(RSD)计)小于11.0%;血浆和尿液中代谢物的平均回收率为97.9%~103%。HFLPME操作简单,富集倍数高,能有效去除生物样品中复杂基体的干扰,适用于复杂样品中微量、痕量成分分析物的分析测定。  相似文献

建立了利用中空纤维三相液相微萃取-高效液相色谱联用技术(HF-LPME-HPLC)同时测定环境水中痕量麦草畏(dicamba)、氟草烟(fluroxypyr)、4-氯苯氧乙酸(4-CPA)、2甲4氯(MCPA)、2,4-滴(2,4-D)、2,4-滴苯氧丁酸(2,4-DB)和2甲4氯苯氧丁酸(MCPB)等7种苯氧羧酸类除草剂的分析方法。考察了萃取剂﹑接受相和给出相pH值、萃取时间﹑搅拌速度和盐效应等对检测的影响,通过正交试验优化萃取条件,得到的最佳萃取条件为正辛醇作萃取剂,给出相pH为3,接受相pH为12,萃取30 min,搅拌速度400 r/min。结果表明7种除草剂在较宽的线性范围内线性良好,相关系数为0.9953~0.9988,检出限(信噪比为3)为0.2~1.0 μg/L,富集倍数为76.7~121,加标回收率为68%~104%,相对标准偏差为3.2%~8.1%。该法灵敏度高、操作简单、检测快速、有机溶剂消耗少,为环境水样中痕量苯氧羧酸类除草剂残留的分析提供了有益的参考。  相似文献

建立了中空纤维液相微萃取-高效液相色谱法测定纺织品中10种含氯苯酚类化合物的方法。系统地优化了影响萃取效率的因素,得到的最佳萃取条件为:萃取溶剂为正己烷,接受相NaOH溶液的浓度为0.10 mol/L,萃取时间为60 min,搅拌速度为600 r/min。在最佳萃取条件下,10种含氯苯酚在0.01~1.00 mg/L范围内线性关系良好(r>0.999),10种含氯苯酚的检出限(信噪比为3)为0.01 mg/kg,富集倍数为95~101。在空白样品中添加0.01、0.05和0.1 mg/kg 3个不同水平的10种含氯苯酚类化合物,其平均回收率为78.8%~105.1%,相对标准偏差为0.3%~7.3%。研究结果表明该方法灵敏度高、简便、准确,可用于纺织品中含氯苯酚类化合物的测定。  相似文献

In the present work, a novel sample pre-treatment technique for the determination of trace concentrations of some insecticide compounds in aqueous samples has been developed and applied to the determination of the selected analytes in environmental water samples. The extraction procedure is based on coupling polypropylene hollow fiber liquid phase microextraction (HF-LPME) with gas chromatography by flame thermionic detection (GC-FTD). For the development of the method, seven organophosphorous insecticides (dichlorvos, mevinphos-cis, ethoprophos, chlorpyrifos methyl, phenthoate, methidathion and carbofenothion) and one carbamate (carbofuran) were considered as target analytes. Several factors that influence the efficiency of HF-LPME were investigated and optimized including agitation, organic solvent, sample volume, exposure time, salt additives and pH. The optimized methodology exhibited good linearity with correlation coefficient = 0.990. The analytical precision for the target analytes ranged from 4.3 to 11.1 for within-day variation and 4.6 to 12.0% for between-day variation. The detection limits for all analytes were found in the range from 0.001 to 0.072 microg/L, well below the limits established by the EC Drinking Water Directive (EEC 80/778). Relative recoveries obtained by the proposed method from drinking and river water samples ranged from 80 to 104% with coefficient of variations ranging from 4.5 to 10.7%. The present methodology is easy, rapid, sensitive and requires small sample volumes to screen environmental water samples for insecticide residues.  相似文献

Two extraction procedures, matrix solid‐phase dispersion and hollow fiber liquid‐phase microextraction, were combined and applied to determine triazine herbicides in peanut samples. The results showed that the established method has high extraction efficiency and could greatly eliminate the interferences from complex matrix samples. A series of important experimental parameters were all investigated in detail. Under the optimal conditions, the developed method has the limits of detection for triazine herbicides in the range of 0.05 to 1.71 μg/kg. Moreover, it has the recovery in the range of 80.4–120.0% with relative standard deviations of equal or lower than 8.9%. The established method may have a great potential in separation, enrichment, and purification of triazines from complex fatty solid samples.  相似文献

Ion-pair based hollow fiber liquid phase microextraction (IP-HFLPME) coupled with high performance liquid chromatography-ultraviolet detection was applied for the preconcentration and determination of methimazole in biological samples and animal feed. Optimization of the conditions for the high extraction efficiency was studied simultaneously using the experimental design. For the first step, the Plackett-Burman design was applied to screen the significant factors on the extraction efficiency. Central composite design (CCD) was then used for the optimization of important factors and the response surface equations were obtained. The optimum experimental conditions were donor phase pH, 12.2; extraction temperature, 45°C; extraction time, 50 min; sodium perchlorate concentration, 1.5 M; cetyltrimethylammonium bromide concentration, 0.65 mM, and without salt addition in donor phase. The limit of detection and the dynamic linear range were in the range of 0.1-0.7 μg L(-1) and 0.5-1000 μg L(-1) , respectively. Preconcentration factors were obtained in the range of 93-155 in different matrices. Finally, the performance of the proposed method was tested for the determination of trace amounts of methimazole in plasma, urine, bovine milk, and animal feed samples, and satisfactory results were obtained (RSDs < 7.1%).  相似文献

A simple hollow fiber liquid‐phase microextraction method for the determination of synthetic pyrethroid metabolites, 3‐phenoxybenzoic acid and 4‐hydroxy‐3‐phenoxybenzoic acid, in human and rat urine was developed and validated. A polypropylene hollow fiber tightly fitted onto a Nylon rod and impregnated with organic solvent served as a disposable extraction device. Desorption of analytes was carried out in NaOH solution, analyzed further by gradient HPLC and diode array detection method. Important factors were identified using Taguchi OA16 (45) orthogonal array design and further optimized using univariate approach. The optimum method performance was observed when 1 mL of urine hydrolyzed with 0.2 mL of concentrated HCl was further supplemented with 100 mg of NaCl and extracted for 120 min into dihexyl ether immobilized in the pores of the hollow fiber. Metabolites were desorbed into 0.1 mL of 0.1 M NaOH for another 120 min. Limits of detection and quantitation of 15 and 50 ng/mL were obtained for both analytes. Relative standard deviations of 1.6–12.6% over the linear range (50–10,000 ng/mL, r > 0.9906) were observed. Intra‐ and inter‐day accuracies of the method ranged from 98.3 to 109.5% and from 93.3 to 110.9%, respectively. This paper describes an extraction method using a polypropylene membrane supporting dihexyl ether (three-phase hollow fiber-based liquid phase microextraction (HF-LPME)) for the analysis of several pharmaceuticals (salicylic acid (SAC), ibuprofen (IBU) and diclofenac (DIC)) followed by a HPLC determination using a monolithic silica type HPLC column, that allows lower retention times than the usual packed columns with adequate resolution. Detection was realized by means of a coupled in series diode array (DAD) and fluorescence (FLD) detectors. HF-LPME is a relatively new technique employed in analytical chemistry for sample pretreatment which offers more selectivity and sensitivity than any traditional extraction technique. Detection limits by DAD are 12, 53 and 40 ng mL−1 for salicylic acid, diclofenac and ibuprofen, respectively and by FLD 7 and 2 ng mL−1 for salicylic acid, and ibuprofen. The method has been successfully applied to their direct determination in human urine and the results obtained demonstrated that could be also applied to the determination of the corresponding metabolites.  相似文献

A new and fast hollow fiber based liquid phase microextraction (HF-LPME) method using volatile organic solvents coupled with high-performance liquid chromatography (HPLC) was developed for determination of aromatic amines in the environmental water samples. Analytes including 3-nitroaniline, 3-chloroaniline and 4-bromoaniline were extracted from 6 mL basic aqueous sample solution (donor phase, NaOH 1 mol L−1) into the thin film of organic solvent that surrounded and impregnated the pores of the polypropylene hollow fiber wall (toluene, 20 μL), then back-extracted into the 6 μL acidified aqueous solution (acceptor phase, HCl 0.5 mol L−1) in the lumen of the two-end sealed hollow fiber. After the extraction, 5 μL of the acceptor phase was withdrawn into the syringe and injected directly into the HPLC system for the analysis. The parameters influencing the extraction efficiency including the kind of organic solvent and its volume, composition of donor and acceptor phases and the volume ratio between them, extraction time, stirring rate, salt addition and the effect of the analyte complexation with 18-crown-6 ether were investigated and optimized. Under the optimal conditions (donor phase: 6 mL of 1 mol L−1 NaOH with 10% NaCl; organic phase: 20 μL of toluene; acceptor phase: 6 μL of 0.5 mol L−1 HCl and 600 m mol L−1 18-crown-6 ether; pre-extraction and back-extraction times: 75 s and 10 min, respectively; stirring rate: 800 rpm), the obtained EFs were between 259 and 674, dynamic linear ranges were 0.1-1000 μg L−1 (R > 0.9991), and also the limits of detection were in the range of 0.01-0.1 μg L−1. The proposed procedure worked very well for real environmental water samples with microgram per liter level of the analytes, and good relative recoveries (91-102%) were obtained for the spiked sample solutions.  相似文献

A simple and efficient method based on hollow fiber protected headspace liquid-phase in conjunction with high performance liquid chromatography has been introduced for extraction and determination of three residual monomers (2-ethylhexyl acrylate (EHA), vinyl acetate (VA), glycidyl methacrylate (GM)) in polymer latex. Using this methodology, the analytes of interest extracted from a sample are led into organic solvent located inside the porous hollow fiber membrane. Initially, several experimental parameters were controlled and optimized and the optimum conditions were reached with 8 cm neatly cut hollow fibers containing heptanol, which were exposed to the headspace of a 12 mL sample solution containing 20% (w/v) NaCl thermostated at 110 °C and stirred at 800 rpm for 20 min. Finally, 20 μL of the extraction solution was withdrawn into a syringe and injected into HPLC for analysis. The calibration curves were linear (r2 ≥ 0.994) over the concentration range of 0.05-10 mg L−1 for VA and 0.02-10 mg L−1 for other analytes. The relative standard deviation (RSD%) for three-replicate extractions and measurements was below 8.6%. The limits of detection of this method for quantitative determination of the analytes were found within the range of 0.005 to 0.011 mg kg−1 with the enrichment factors within the 5-164 range. The method was successfully applied for determination of residual monomers in polymer latex.  相似文献

The analysis of plant growth regulators presents a challenge due to their trace quantities and complex matrices. A novel, simple, and effective analytical method for the determination of three trace acidic plant growth regulators in Anoectochilus roxburghii (Wall.) Lindl was developed to address this issue. Three‐phase hollow fiber liquid‐phase microextraction combined with high‐performance liquid chromatography was applied for the enrichment, purification, and determination of three acidic plant growth regulators, namely, indole‐3‐acetic‐acid, indole‐3‐butyric‐acid, and (+)‐abscisic acid. The factors affecting extraction performance, including extractant species, pH of donor and acceptor phases, salt addition dosage, extraction time, temperature, and stirring rate, were investigated and optimized. Under optimum conditions, the proposed method provided good linearity (R2, 0.9994–0.9999), low limit of detection (0.038–0.12 ng/mL), and acceptable relative recoveries (56.7–117.6%). The enrichment factors were between 153 and 328. The developed method was successfully applied to the enrichment and determination of plant growth regulators in Anoectochilus roxburghii (Wall.) Lindl and exhibited increased purification capacity, higher sensitivity, and decreased organic solvent consumption compared with conventional sample preparation methods. This method may provide a testing platform for the monitoring of plant growth regulator residues, ensuring the safe and effective use of traditional Chinese medicine.  相似文献

As the drive towards green extraction methods has gained momentum in recent years, it has not always been possible to eliminate organic solvents completely. However, the volumes employed have been reduced remarkably, so that a single microdrop is sufficient in some cases. This effort has led to the development of various liquid phase microextractions namely single drop microextraction (SDME), hollow fiber liquid phase microextraction (HF-LPME), dispersive liquid-liquid microextraction (DLLME) and solidified floating organic drop microextraction (SFODME). In this review, the historical development and overview of these miniaturized liquid phase extraction methodologies have briefly been discussed and a comprehensive collection of application of the these methods in combination with different analytical techniques for preconcentration and determination of ultra trace amounts of metals and organometal ions in various matrices have been summarized.  相似文献

Sample pretreatment techniques or preconcentration constitute a very important step before the analysis of environmental, clinical, pharmaceutical, and other complex samples. Thanks to extraction techniques it is possible to achieve higher method sensitivities and selectivities. Miniaturization microextraction methods make them more environmentally friendly and only small amounts of samples are required. In the past 30 years, a number of microextraction methods have been developed and used and are documented in thousands of articles. Many reviews have been written focusing on their use in specified professional fields or on the latest trends. Unfortunately, no uniform nomenclature has been introduced for these methods. Therefore, this review attempts to classify all the essential microextraction techniques and describes their advantages, disadvantages, and the latest innovations. The methods are divided into two main groups: single drop and sorbent‐based techniques according to the type of extraction phase.  相似文献

A novel three‐phase hollow fiber liquid‐phase microextraction was developed based on reverse micelle as extraction solvent and acceptor phase, and compared with conventional two‐phase hollow fiber liquid‐phase microextraction. Both procedures were used in the extraction and concentration of four cinnamic acids (caffeic acid, p‐hydroxycinnamic acid, ferulic acid, and cinnamic acid) in traditional Chinese medicines prior to high‐performance liquid chromatography analysis. Parameters affecting the two procedures were investigated and optimized to obtain the optimum enrichment factors. The mechanism of the developed procedure was explored and elucidated by comparison with conventional two‐phase hollow fiber liquid‐phase microextraction. Under the optimized conditions, the analytes’ enrichment factors were between 50 and 118 for the proposed procedure, and 31–96 for conventional two‐phase mode. Satisfactory linear ranges (r2 ≥ 0.99), detection limits (0.1–0.6 ng/mL), precisions (<9.2%), and accuracies (recoveries: 80–123.1%) were observed for the two procedures. The results showed that the enrichment capacity of the proposed procedure for the cinnamic acids is better than that of conventional two‐phase procedure, and both are eco‐friendly, simple, and effective for the enrichment and detection of cinnamic acids in traditional Chinese medicines.  相似文献

For the first time, an ionic liquid based three-phase liquid-liquid-liquid solvent bar microextraction (IL-LLL-SBME) was developed for the analysis of phenols in seawater samples. The ionic liquid, 1-butyl-3-methylimidazolium hexafluorophosphate ([BMIM][PF(6)]), was used as the intermediary solvent for LLL-SBME, enhancing the extraction efficiency for polar analytes. In the procedure, the analytes were extracted from the aqueous sample into the ionic liquid intermediary and finally, back-extracted into an aqueous acceptor solution in the lumen of the hollow fiber. The porous polypropylene membrane acted as a filter to prevent potential interfering materials from being extracted, and no additional cleanup was required. After extraction, the acceptor solution could be directly injected into a high-performance liquid chromatographic system for analysis. Six phenols, 2-nitrophenol, 4-chlorophenol, 2,3-dichlorophenol, 2,4-dichlorophenol, 2,4,6-trichlorophenol and pentachlorophenol were selected here as model compounds for developing and evaluating the method. The most influential extraction parameters were evaluated, including the ionic liquid, the composition of donor solution and acceptor solution, the extraction time and the extraction temperature, the effect of ionic strength, and the agitation speed. Under the most favorable extraction parameters, the method showed good linearity (from 0.05-50 to 0.5-50 μg/L, depending on the analytes) and repeatability of extractions (RSD below 8.3%, n=5). The proposed method was compared to conventional three-phase LLL-SBME and ionic liquid supported hollow fiber protected three-phase liquid-liquid-liquid microextraction, and showed higher extraction efficiency. The proposed method was demonstrated to be a simple, fast, and efficient method for the analysis of phenols from environmental water samples.  相似文献

A simple and efficient hollow fiber liquid‐phase microextraction (HF‐LPME) technique in conjunction with high‐performance liquid chromatography is presented for extraction and quantitative determination of aristolochic acid I in human urine samples. Several parameters influencing the efficiency of HF‐LPME were investigated and optimized, including extraction solvent, stirring rate, extraction time, pH of donor phase and acceptor phase. Excellent sample clean‐up was observed and good linearity with coefficient of 0.9999 was obtained in the range of 15.4–960 µg/L. This method provided a 230‐fold enrichment factor and good repeatability with relative standard deviations (RSD) lower than 6.0%. The limit of detection value for the analyte in urine sample was 0.01 µg/L at a signal‐to‐noise ratio of 3. The extraction recovery from urine samples was 61.8% with an RSD of 9.71%. Hollow fiber cell fishing (HFCF) and hollow fiber liquid phase microextraction (HF-LPME) with high performance liquid chromatography were utilized to research the anti-hepatoma HepG-2 or anti-renal tubular ACHN cells coumarins from Radix Angelicae dahuricae (RAD) in vitro and vivo. Before application, the coumarins of psoralen, bergapten, oxypeucedanin, imperatorin and isoimperatorin were selected as model compounds, the experiment conditions and methodology parameters of both HFCF and HF-LPME were investigated. Under the optimized conditions, cell growth state and survival rate, cell specific binding, repeatability for HFCF, and linear ranges, limits of quantification (1.3–21.0?ng/mL), precisions (<15.2%), recoveries (80.4%–109.3%) and stability for HF-LPME were achieved. Results showed that the five model compounds and xanthotoxol, xanthotoxin were screened in RAD extraction; xanthotoxin and xanthotoxol were fished by HepG-2 cell from both plasma and liver, xanthotoxol, xanthotoxin, bergapten and imperatorin were fished by ACHN cell from plasma and kidney; and these coumarins maintained a certain concentration in vivo. The study results provide experimental basis for researching the active anticancer components and expound the medicinal material base in RAD. The combination of HFCF and HF-LPME is simple and effective in studying bioactive anticancer compounds in traditional Chinese medicine in vitro and in vivo.  相似文献

A headspace single-drop microextraction (HS-SDME) procedure using room temperature ionic liquid and coupled to high-performance liquid chromatography capable of quantifying trace amounts of chlorobenzenes in environmental water samples is proposed. A Plackett-Burman design for screening was carried out in order to determine the significant experimental conditions affecting the HS-SDME process (namely drop volume, aqueous sample volume, stirring speed, ionic strength, extraction time and temperature), and then a central composite design was used to optimize the significant conditions. The optimum experimental conditions found from this statistical evaluation were: a 5 μL microdrop of 1-butyl-3-methylimidazolium hexafluorophosphate, exposed for 37 min to the headspace of a 10 mL aqueous sample placed in a 15 mL vial, stirred at 1580 rpm at room temperature and containing 30% (w/v) NaCl. The calculated calibration curves gave a high level of linearity for all target analytes with correlation coefficients ranging between 0.9981 and 0.9997. The repeatability of the proposed method, expressed as relative standard deviation, varied between 1.6 and 5.1% (n = 5). The limits of detection ranged between 0.102 and 0.203 μg L−1. Matrix effects upon extraction were evaluated by analysing spiked tap and river water as well as effluent water samples originating from a municipal wastewater treatment plant.  相似文献

In this paper two applications of three‐phase HF‐LPME for the determination of pharmaceuticals in human urine are proposed: a capillary electrophoresis with a photodiode array detection method for the analysis of seven nonsteroidal anti‐inflammatory drugs (NSAIDs) and a high‐performance liquid chromatographic with photo diode array and fluorescence detection method for the determination of four sulfonamides and their corresponding N4‐acetyl‐metabolites. Q3/2 Accurel® polypropylene hollow fibers were used for both procedures. Dihexyl ether was used as the supported liquid membrane for the determination of anti‐inflammatories and 1‐octanol for sulfonamides. An aqueous solution (pH 12) was used in both procedures as the acceptor phase and as the donor phase an aqueous solution (pH 2), and a 2 m Na2SO4 aqueous solution (pH 4) was used for the determination of the anti‐inflammatories and sulfonamides. The detection limits obtained were between 0.25 (naproxen) and 0.86 ng/mL (aceclofenac) for the determination of anti‐inflammatories and 7 × 10?4 (sulfamethoxazole) and 0.048 ng/mL (N4‐acetyl‐sulfamethazine) for sulfonamides.
