Seven new phloroglucinol derivatives, myrtucommunins A–D (14), 6-methylisomyrtucommulone B (5), 4-methylmyrtucommulone B (6), and 2-isobutyryl-4-methylphloroglucinol 1-O-β-d-glucopyranoside (7), and one new chromone derivative, undulatoside A 6′-O-gallate (8), were isolated from the leaves of Myrtus communis (Myrtaceae). Myrtucommunins A–D (14) were conjugates of polymethylated acylphloroglucinol and flavonol rhamnoside. The absolute configurations of the rhamnosyl moieties for 14 were confirmed to be l in each case by HPLC analyses, while those of the aglycones were assigned by comparisons of the experimental and TDDFT calculated ECD spectra. 6-Methylisomyrtucommulone B (5) and 4-methylmyrtucommulone B (6) were assigned as 6/6/6 tricyclic acylphloroglucinol derivatives with a racemic nature. Antimicrobial activities of 18 and related known compounds were evaluated.  相似文献

Three new azaphilones, eupenicilazaphilones A–C (13), and one new isocoumarin, eupenicillin A (4), as well as five known azaphilones were isolated from the sponge-derived fungus Eupenicillium sp. 6A-9. Their structures were elucidated by detailed spectroscopic analysis, comparison with literature data, and 13C NMR chemical shifts and TDDFT/ECD calculations. The antibacterial activity against Staphylococcus aureus ATCC25923, methicillin-resistant Staphylococcus aureus ATCC4330, and Acinetobacter baumanii ATCC19606 as well as the cytotoxic activity towards human cancer lines MCF-7 and A549 were examined.  相似文献

Nine new mycotoxins; five xanthones 15, hydroxanthone 6, and three anthraquinones 79, together with nine known compounds; sterigmatocystin (10), demethylsterigmatocystin (11), dihydrodemethylsterigmatocystin (12), sterigmatin (13), austocystin F (14), averufin (15), aflatoxin B1, paeciloquinone A, and zeorin, were isolated from the scale insect fungus Aschersonia coffeae Henn. BCC 28712. The structures of these compounds were elucidated using NMR spectroscopic and MS spectrometric analyses. Compounds 13 and 69 displayed cytotoxic activity while the xanthone 2 and anthraquinones 8 and 9 also showed antimalarial activity.  相似文献

Six novel quinolinone alkaloids bearing a phenylpropanoid or a coumarin moiety, named melicodenines C–H (16), two new cinnamyl alcohol derivatives, named melicodins A (7) and B (8), and a new coumarinolignan, named melicodin C (9), were isolated from the leaves of Melicope denhamii (Seem.) T. G. Hartley. Their structures were established by spectroscopic analyses including extensive 2D-NMR experiments. Compounds 15 and 9 possess a cyclobutane ring and are hypothetically produced by a [2+2] cycloaddition, whereas compound 6 is presumed to form through a Diels–Alder cycloaddition followed by the elimination of an acetone molecule. Ten quinolinone alkaloids (16, 1013) and a coumarinolignan (9) were tested for anti-proliferative activity against DLD-1 human colon cancer cells. Melicodenine G (5) showed the most potent inhibitory activity, causing the induction of apoptosis with an IC50 value of 9.4±0.3 μM.  相似文献

Chemical investigation of a methanolic extract of leaves from Araucaria bidwillii (Araucariaceae) from Egypt afforded four new labdane diterpenoidal metabolites (14) together with one known diterpene, 7-oxocallitrisic acid (5), two triterpenoidal metabolites, 2-O-acetyl-11-keto-boswellic acid (6) and β-sitosterol-3-O-glucopyranoside (7), phloretic acid (8), and two methylated bisflavonoids, agathisflavone-4′,7,7″-trimethyl ether (9) and cupressuflavone-4′,7,7″-trimethyl ether (10). The new metabolites 14 were unambiguously identified by applying extensive 1D and 2D NMR spectroscopic studies as well as HRESIMS. The relative and absolute configurations of 14 were determined using ROESY and the modified Mosher's method, respectively. All isolated compounds were assessed for their antimicrobial, antitubercular and cytotoxic activities. Among the tested compounds, the new labdane diterpenes 14 revealed significant cytotoxic activity against mouse lymphoma L5178Y cell line with IC50 values ranging from 1.4 to 12.9 μM, respectively.  相似文献

Three new polyketides including one new diphenyl ether, penicillither (1), one new anthraquinone, penicilliquinone (2), and one new xanthone, penicillixanthone (3), together with twelve known compounds were isolated from the soil fungus Penicillium sp. PSU-RSPG99. Their structures were identified by spectroscopic evidence. In addition, the position of a chlorine atom in 1 was confirmed by the plausible biosynthetic pathway from the isolated chlorobenzophenone. Known GKK1032B displayed mild antimycobacterial and moderate cytotoxic (against human oral carcinoma, KB, human breast cancer, MCF-7, and noncancerous Vero cell lines) activities.  相似文献

Nine new highly oxygenated nortriterpenoids, pre-schisanartanins E–J (16) and sphenadilactones D–F (79), together with 17 known ones (1026), have been isolated from the acetone extract of the roots and stems of Schisandra sphenanthera. The structures of the new metabolites were characterized on the basis of extensive spectroscopic analyses including 1D and 2D NMR experiments. These compounds were all evaluated for their cytotoxicity against HL-60, SMMC-7721, A549, MCF-7, and SW480 tumor cell lines.  相似文献

Seven new indole alkaloids including a new indoloazepinone derivative (2), two new quinazolinone derivatives (7, 8) and four new pyrazinoquinazolinone derivatives: epi-fiscalin C (10), epi-fiscalin A (12), neofiscalin A (13), epi-neofiscalin A (14) were isolated, together with 4-dihydroxy-3-methylacetophenone (1), tryptoquivaline (3), tryptoquivalines L (4), H (5), F (6) as well as fiscalins A (11) and C (9), from the culture of the fungus Neosartorya siamensis (KUFC 6349). The absolute configuration of the stereogenic carbons of the previously reported tryptoquivalines F, H, L was revised using the data obtained from an X-ray analysis of tryptoquivaline l and the NOESY correlations. The structures of the new pyrazinoquinazolinone derivatives (10, 12, 13, 14) were established based on 1D and 2D NMR spectral analysis, and the absolute configuration of their stereogenic carbons was determined by an X-ray crystallographic analysis of fiscalin C (9) and a new compound epi-fiscalin C (10), in conjunction with the correlations observed in their NOESY and long range COSY spectra. Compounds 28 and 12 were evaluated for their in vitro growth inhibitory activity on the human U373 and Hs683 glioblastoma, the A549 non-small cell lung cancer, the MCF-7 breast cancer and the SKMEL-28 melanoma cell lines by MTT colorimetric assay.  相似文献

The phytochemical investigation of the leaf extract of Uvaria cherrevensis (Annonaceae) yielded three new cyclohexene (9Z)-octadec-9-enyl ethers, cherrevenols M-O (13), and a known fatty ester derivative (4). The structures of the isolated compounds were elucidated by spectroscopic and computer-aided molecular modelling methods. Ozone Induced Dissociation (OzID) mass spectrometry was employed to determine the C-9 position of the side chain olefinic double bonds, while 13C NMR spectroscopy indicated their (Z)-configurations. All isolated compounds were evaluated for their antimalarial and cytotoxic activities; all were inactive.  相似文献

Three new isocoumarins and two new phthalides, tubakialactones A–E (15), together with three known polyketides including (R)-3,4-dihydro-4-hydroxyl-6,8-dimethoxy-4-methyl-3-methylene-1H-2-benzopyran-1-one (6), (?)-5,7-dimethoxy-3-methyl-1(3H)-isobenzofuranone (7), and isosclerone (8) were found in the endophytic fungus Tubakia sp. ECN-111 isolated from the leaves of Houttuynia cordata. The chemical structures of the new compounds were determined by spectroscopic analyses, including extensive 2D-NMR experiments. The absolute configuration of 3 and 7 was determined by optical rotation and circular dichroism.  相似文献

Twelve new compounds, including nine tropolones, nemanolones A?I (19), three 7-isochromenones, nemanecins A?C (1012), and a new naturally isolated 4-isochromanone (13), along with two known compounds, 7,8-dihydroxy-3-methyl isochroman-4-one (XJP), and chaetoquadrin F, were isolated from culture broth of the fungus Nemania sp. BCC 30850. Structures of these compounds were elucidated by NMR and MS spectroscopic analyses. Nemanolones exhibited cytotoxic activities and two of them, compounds 1 and 2, also showed antibacterial activity against Bacillus cereus and antifungal activity against Candida albicans.  相似文献

New indole alkaloids streptoprenylindoles A–C (13) and diterpenoids 18-acetyl-cyclooctatin (8), 5,18-dedihydroxy-cyclooctatin (9), and 5-dehydroxy-cyclooctatin (10) were isolated from the culture of marine-derived Streptomyces sp. ZZ820, along with known 3-cyanomethyl-6-[3-methyl-2-butenyl]indole (4), N-(2-(1H-indol-3-yl)ethylacetamide (5), 1-acetyl-β-carboline (6), indole-3-methylethanoate (7), cyclooctatin (11), and chromomycin A3 (12). Their structures were elucidated by a combination of extensive spectroscopic analyses, ECD calculation, and the Mosher's method. Streptoprenylindoles A (1) and B (2) are enantiomers that were separated through the preparation of their Mosher esters. Three new diterpenoids (810) showed antibacterial activities against methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli with MIC values of 24.11–55.12?μM, while chromomycin A3 (12) showed potent antibacterial activities against MRSA (MIC: 0.59?μM) and E. coli (MIC 0.04?μM).  相似文献

The concomitant addition of a histone deacetylase inhibitor, suberoyl bis-hydroxamic acid, and a DNA methyltransferase inhibitor, RG-108, to the culture medium of Gibellula formosana, an entomopathogenic fungus, induced a significant increase in diversity of secondary metabolites. From the culture media were isolated two new highly oxidized ergosterols, formosterols A (1) and B (2), and five new isariotin analogs, 12′-O-acetylisariotin A (4), 1-epi-isariotin A (5), and isariotins K–M (68), together with 22,23-epoxy-3,12,14,16-tetrahydroxyergosta-5,7-dien-11-one (named formosterol C) (3), isariotins A (9), C (10), and E (11), TK-57-164A (12), and beauvericin (13). The NMR spectra, X-ray single crystallographic diffraction, and chemical transformations revealed the structures of the two new formosterols and five new isariotins. The stereochemistry of formosterol C (3) was deduced from its spectroscopic data. The side chains of formosterols A–C (13) contained cis-22,23-epoxide, which is rarely present in naturally occurring sterols and triterpenes.  相似文献

A strain of Chaetomium globosum (DAOM 240359) was isolated from an indoor air sample in Ottawa, Ontario, Canada. When fermented in liquid culture, this strain produced a number of known metabolites including chaetoglobosins A (6), C, and F (7), chaetomugilin D (5), chaetoviridin A (4), and three new nitrogenous azaphilones; 4′-epi-N-2-hydroxyethyl-azachaetoviridin A (1), N-2-butyric-azochaetoviridin E (2), and isochromophilone XIII (3). The structures were elucidated by spectroscopic analysis including; HRMS, 1D and 2D NMR, UV, and ORD. Compounds 27 were antimicrobial when tested using quantitative growth inhibition assays.  相似文献

Spongiains A-C (13), the first examples of spongian diterpenes bearing a pentacyclic skeleton composed of a fused 5/5/6/6/5 ring system through ring A rearrangement, together with four new spongian diterpenes, spongiains D-G (47), were isolated from the marine sponge Spongia sp.. The structures with absolute configurations of these compounds were elucidated by the methods of NMR, X-ray diffraction and quantum chemical approaches. The hypothetical biogenetic pathways as well as cytotoxic activities of 17 were also discussed.  相似文献

Four new aristolane sesquiterpenes named nambinones A–C (13) and 1-epi-nambinone B (4), a new sesquiterpene, nambinone D (5), a known compound, aurisin A (6) as well as a new dimeric sesquiterpene, aurisin K (7), were isolated from two isolates of luminescent mushroom, Neonothopanus nambi, PW1 and PW2. These structures were established on the basis of spectroscopic evidence. The relative configuration of 6 was determined by X-ray crystallographic analysis. Compounds 6 and 7 exhibited antimalarial activity against Plasmodium falciparum and antimycobacterial activity against Mycobacterium tuberculosis. Compounds 3, 6, and 7 showed cytotoxicity against NCI-H187 cancer cell lines. In addition, 6 and 7 showed cytotoxicity against the cholangiocarcinoma cell lines.  相似文献

Alokicenones A-H (1–8), eight new tetrahydroanthracenes and one known okicenone (9) were identified from the secondary metabolites of mangrove-derived Streptomyces sp. HN-A101. Their structures were elucidated by HRESIMS and NMR spectroscopic data. The absolute configurations of them were determined by the calculated and experimental ECD curves. Compounds 1–2 and 9 showed moderate cytotoxicity against HCT116 and SW620 cancer cell lines with IC50 values from 0.63 to 7.73?μM. In addition, compounds 13 and 79 also exhibited inhibitory activities against ROCK2 or BRD4.  相似文献

One new sesquiterpene with a novel carbon skeleton, flammulinol A (1), one new isolactarane sesquiterpene and six isolactarane-related norsesquiterpenes, flammulinolides A–G (28), as well as sterpuric acid, were isolated from the solid culture of Flammulina velutipes. The structures of the new compounds were elucidated by spectroscopic methods. The absolute configuration of C-3 in 1 was determined via the circular dichroism data analysis based on the octane rule of cyclopentenone, whereas the absolute configurations of compounds 2, and 58 were assigned using the circular dichroism data of the [Rh2(OCOCF3)4] complex. Compounds 2, 3, and 7 showed strong cytotoxicity against KB cell line with the IC50 of 3.9, 3.6, and 4.7 μM, respectively. Compound 4 showed strong cytotoxicity against Hela cell line with the IC50 of 3.0 μM. The plausible biosynthetic pathway for 29 in F. velutipes was discussed.  相似文献

Detailed investigation of the Antarctic nudibranch Austrodoris kerguelenensis has resulted in the isolation of a diverse suite of new diterpenoid glyceride esters (116) related to the palmadorins (1719), including one (palmadorin L (9)) that is the first halogenated diterpene from this well-studied nudibranch. Previous collections of A. kerguelenensis from McMurdo Sound, the Weddell Sea and the Western Antarctic Peninsula have afforded related diterpene glycerides, a natural product class implicated as a chemical defense in nudibranchs. In this paper we describe the isolation, structure elucidation, and stereochemical analysis of sixteen new palmadorins using a combination of one- and two-dimensional NMR techniques. Palmadorin A (17), B (18), D (1), M (10), N (11), and O (12) inhibit human erythroleukemia (HEL) cells with low micromolar IC50's, and palmadorin M inhibits Jak2, STAT5, and Erk1/2 activation in HEL cells and causes apoptosis, at 5 μM.  相似文献

Seven new prenylated phenols, vaccinols A–G (17), and one new naphthalene derivative, vaccinal A (8), together with five known phenol derivatives (913) were isolated from Pestalotiopsis vaccinii (cgmcc3.9199) endogenous with the mangrove plant Kandelia candel (L.) Druce (Rhizophoraceae). The new structures were established on the basis of spectroscopic analysis, optical rotation, CD spectra, quantum ECD calculations. Furthermore, the absolute configuration of compound 1 was unambiguously elucidated by single-crystal X-ray diffraction analysis using the Flack parameter. Most new compounds were tested for their antiviral (EV71), cytotoxic, antituberculosis, and anti-inflammatory effects. Among them, compound 8 exhibited in vitro anti-enterovirus 71 (EV71) with IC50 value of 19.2 μM. In addition, 8 showed potent COX-2 inhibitory activity with IC50 value of 1.8 μM.  相似文献
