A general chiral building block containing the 1R,2R-trans-diol moiety was constructed utilizing the stereoselective Shi-epoxidation reaction on a tetralone scaffold assembled by a Negishi cross-coupling on N,N-diethylbenzamide. Further elaboration of this chiral building block into polycyclic aromatic compounds was demonstrated with the total synthesis of the precursor for the most carcinogenic metabolite of chrysene, (−)-(1R,2R)-1,2-dihydrochrysene-1,2-diol in 87% ee.  相似文献

We have developed an efficient method for selective monobenzoylation of 1,2- and 1,3-diols in water catalyzed by Me(2)SnCl(2). Treatment of 1,2- and 1,3-diols with benzoyl chlorides, DMT-MM, and potassium carbonate in the presence of a catalytic amount of Me(2)SnCl(2) and DMAP in water at room temperature gave monobenzoates in up to 97% yield.  相似文献

An enantioselective synthesis of sterically congested 1,2-di-tert-butyl and 1,2-di-(1-adamantyl)ethylenediamines has been developed. Thus, diastereomerically pure trans-1-apocamphanecarbonyl-4,5-dimethoxy-2-imidazolidinones 6 and 7 were successfully prepared by optical resolution of (±)-trans-4,5-dimethoxy-2-imidazolidinone using apocamphanecarbonyl chloride (MAC-Cl) followed by stereospecific and stepwise substitution of the dimethoxyl groups using tert-butyl or 1-adamantyl cuprates to provide (4S,5S)-4,5-di-tert-butyl and (4R,5R)-4,5-di-(1-adamantyl)-2-imidazolidinones 12 and 15, respectively. Furthermore, N-acetyl 4,5-di-tert-butyl and 4,5-di-(1-adamantyl)-2-imidazolidinones 16a,b were enantioselectively deacetylated using a catalytic oxazaborolidine system to provide enantiopure 1-p-tolylsulfonyl-4,5-di-tert-butyl-2-imidazolidinones 12 and 19 and 1-p-tolylsulfonyl-4,5-di-(1-adamantyl)-2-imidazolidinones 18 and 20, respectively. Finally, N-p-tolylsulfonyl-2-imidazolidinones 12 and 15 were treated with 30 equiv of Ba(OH)2·8H2O to achieve ring cleavage and to provide (1S,2S)-1,2-di-tert-butylethylenediamine 3 and (1R,2R)-1,2-di-(1-adamantyl)ethylenediamine 4.  相似文献

The electronic structures of four members of the electron-transfer series [Fe2(1L)4]n (n = 2-, 1-, 0, 1+) have been elucidated in some detail by electronic absorption, IR, X-band electron paramagnetic resonance (EPR), and M?ssbauer spectroscopies where (1L)(2-) represents the ligand 1,2-bis(4-tert-butylphenyl)-1,2-ethylenedithiolate(2-) and (1L*)- is its pi-radical monoanion. It is conclusively shown that all redox processes are ligand-centered and that high-valent iron(IV) is not accessible. The following complexes have been synthesized: [FeIII2(1L*)2(1L)2]0 (1), [FeIII2(2L*)2(2L)2].2CH2Cl2 (1') where (2L)(2-) is 1,2-bis(p-tolyl)-1,2-ethylenedithiolate(2-) and (2L*)- represents its pi-radical monoanion, [Cp2Co][FeIII2(1L*))(1L)3].4(toluene).0.5Et2O (2), and [Cp2Co]2[FeIII2(1L)4].2(toluene) (3). The crystal structures of 1' and 2 have been determined by single-crystal X-ray crystallography at 100 K. The ground states of complexes have been determined by temperature-dependent magnetic susceptibility measurements and EPR spectroscopy: 1' and 1 are diamagnetic (S(t) = 0); 2 (S(t) = 1/2); 3 (S(t) = 0); the monocation [Fe(III)2(1L*)3(1L)]+ possesses an S(t) = 1/2 ground state (S(t) = total spin ground state of dinuclear species). All species contain pairs of intermediate-spin ferric ions (S(Fe) = 3/2), which are strongly antiferromagnetically coupled (H = -2JS(1).S(2), where S1 = S2 = 3/2 and J = approximately -250 cm(-1)).  相似文献

Methodology directed at the preparation of (Z)-1,2-difluorostilbenes has been evaluated. For symmetrical (Z)-1,2-difluorostilbenes, photochemical isomerization of the isomeric (E)-1,2-difluorostilbenes, and HPLC separation of the mixture of stilbene isomers is a reasonable route to a particular (Z)-stilbene. An alternative approach to both symmetrical and/or unsymmetrical (Z)-1,2-difluorostilbenes has been developed via stereospecific Pd(0) coupling of (E)-1,2-difluoro-aryl-ethenyltributylstannanes under Stille-Liebiskind conditions with aryl idodies. The requisite arylstannanes can be obtained via the reported route developed by Davis or via (E)-1,2-difluorovinyltributylstannane - a new route described in this work. The methodology tolerates almost any functionality in the aryl ring, is easily carried out, is stereospecific and provides the first general route to (Z)-1,2-difluorostilbenes.  相似文献

A new chiral benzimidazolide is developed as a nonenzymatic acylating agent for enantioselective benzoylation of racemic alpha-amino esters. The process is highly efficient, which exhibits uniformly high enantioselectivity for alpha-amino esters with or without aryl substituents under mild reaction conditions. [formula: see text] The bakers' yeast reduction of 3-(1,3-benzodioxol-5-yl)-6-propoxy-1H-inden-1-one 4 has been shown to give (S)-3-(1,3-benzodioxol-5-yl)-2,3-dihydro-6-propoxy-1H-indan-1-one 6 in 65% yield with high enantioselectivity (> 99.0% ee), a key intermediate for the synthesis of the endothelin receptor antagonist SB 217242. In addition, the substituted 3-arylinden-1-ones 10a-e gave equally high enantioselectivity for the 3-arylindan-1-one products 13a-e. Mechanistic studies of the reaction indicate the operative pathway to be an asymmetric conjugate reduction, wherein the hydride transfer from NAD(P)H occurs from the Re-face of the indenone substrate.  相似文献

A phosphinite derivative that can be easily prepared in two steps from commercially available aminoindanol was found to be an effective catalyst for enantioselective acylation of diols. For the asymmetric desymmetrization of meso-1,2-diols, the corresponding monoester was obtained in up to 95% ee from the reaction in the presence of 5 mol % catalyst.  相似文献

Conclusions A seven-stage synthesis has given optically active (2R/S, 3S, 7R/S)-2-acetoxy-3,7-dimethylpentadecane from the readily available (S)-(+)-3,7-dimethyl-1,6-octadiene, in an overall yield of 36%. Reactions of the title meso forms, (1R,2S)-1,2-di(2-furyl)-1,2-di(3-guaiazulenyl)ethane (1) and (1R,2S)-1,2-di(3-guaiazulenyl)-1,2-di(2-thienyl)ethane (2), with a two molar amount of TCNE in benzene at 25 °C for 5 h (for 1) and 48 h (for 2) under oxygen give new compounds, 2,2,3,3-tetracyano-4-(2-furyl)-8-isopropyl-6-methyl-1,4-dihydrocyclohepta[c,d]azulene (3) and 2,2,3,3-tetracyano-8-isopropyl-6-methyl-4-(2-thienyl)-1,4-dihydrocyclohepta[c,d]azulene (4), respectively, in 74 and 21% isolated yields. Comparative studies on the above reactions as well as the spectroscopic properties of the unique products 3 and 4, possessing interesting molecular structures, are reported and, further, a plausible reaction pathway for the formation of these products is described.  相似文献

The title chiral amino alcohol 6 was prepared from (s)-3-(2,5-dimethylphenyl)-alanine methyl ester hydrochloride by its reaction with phenyl magnesium bromide. (S)-trans-4-Phenylbut-3-en-2-ol has been obtained in excellent yields and with high enantiomeric excess in the reduction of trans-4-phenylbut-3-en-2-one using the comminuted roots of carrot (Daucus carota L.), celeriac (Apium graveolens L. var. rapaceum), and beetroot (Beta vulgaris L. subsp. The microbial oxidation of racemic 2-phenyl-1-propanol by Gluconobacter oxydans DSM 50049 was investigated. Whole bacterial cells were used to produce (S)-(+)-2-phenylpropanoic acid with high enantiomeric excess (E>200). A simplex sequential method was employed as an experimental design to guide the optimization process. Temperature of 26–28°C, pH 6.0–6.2, substrate concentration of 20–25 mM and agitation of 150 rpm have been found the best conditions to achieve the highest reaction rates and enantioselectivities.  相似文献

Forty one microorganisms belonging to different taxonomical groups were used to carry out the enantioselective reduction of methyl benzoylformate to afford the corresponding (R)-methyl mandelate, with moderate to high ee. In contrast, the monooxygenase enzyme in Helminthosporium sp. CIOC3.3316 catalyzed the hydroxylation of methyl 2-phenylacetate to (S)-methyl mandelate. This combination of oxidation and reduction biotransformations thus provides a method for preparing the enantiomers of chiral α-hydroxy acid derivatives.  相似文献
