A cobalt(salen) complex has been shown to catalyse the asymmetric Darzens condensation between α-haloamides and aldehydes, allowing both the relative and absolute stereochemistry of the epoxy-amide products to be controlled. Under optimal conditions, cis-epoxides can be obtained diastereoselectively with up to 50% enantiomeric excess, whilst by changing the leaving group and base, trans-epoxides can be produced diastereoselectively with up to 43% enantiomeric excess.  相似文献

Summary Optically activeMichael adducts were synthesized by addition of 1,3-dicarbonyl compounds to conjugated nitroalkenes. Good chemical yields were obtained for nitroalkenes stabilized by an aromatic substituent without any further substituents at the double bond. Acetylacetone and methyl-2,3-dihydro-1-oxo-1H-indene-2-carboxylate were used asMichael donors and four cinchona alkaloids as chiral base catalysts. Enantiomeric excess determinations were performed by1H NMR spectroscopy in the presence of thePirkle alcohol and by HPLC on chiral stationary phases. A correlation between the relative configuration of the prevailing isomer of theMichael adduct and the catalysts was established.

Die Bestimmung des Enantiomerenüberschusses wird mittels1H-NMR-Spektroskopie in Gegenwart vonPirkle-Alkohol und HPLC an chiralen stationären Phasen durchgeführt. Eine Korrelation zwischen der relativen Konfiguration der Vorzugsisomeren derMichael-Addukte und den Katalysatoren wurde hergestellt.

The first asymmetric phase transfer catalyzed alkylation reaction of a range of carbon acids with N-sulfonyl aziridines is reported. When 10 mol % of a cinchona derived quaternary ammonium salt was employed as the catalyst under mildly basic conditions, N-o-(trifluoromethane)benzenesulfonyl aziridine was efficiently ring-opened to afford the amino ethylene products in consistently high yields and high enantioselectivities (up to 97% ee). By employing substituted aziridines in single enantiomeric form, the corresponding enantiopure alkylation products could be obtained with a range of pronucleophiles in high yields and moderate to high diastereoselectivities (up to 30:1 dr).  相似文献

A convenient and novel one-pot organocatalytic methodology for the stereoselective synthesis of highly functionalized nitrocyclopropanes is reported. The addition of dimethyl chloromalonate to a variety of nitroolefins catalyzed by tertiary amines leads to a Michael adduct which cyclizes to form the cyclopropane in the presence of DBU under carefully controlled reaction conditions with outstanding diastereoselectivity.  相似文献

Asymmetric conjugate addition of ethyl 4-chloro-3-oxobutanoate to nitroalkenes and subsequent intramolecular cyclization had been developed. This one-pot reaction provided tetronic acid derivatives in good yields and with excellent enantioselectivities. 6′-Demethyl quinine was found to be the best catalyst for the conjugate addition and AcOLi was identified as the best base for the intramolecular cyclization. Various β-aryl, heteroaryl, and alkyl nitroalkenes are generally applicable in the reaction.  相似文献

The conjugate addition of thioacetic acid to methacrylamides with chiral C2-symmetric trans-2,5-disubstituted pyrrolidines afforded the addition products in excellent stereoselectivities (>99% de) and good yields (80–90%). The high selectivity was attributed mainly to the steric effect of the chiral auxiliaries. The cyclic nature of the chiral auxiliaries seemed also important for both the stereoselectivity and the reaction rate. Acidic hydrolysis of the adduct containing (2R,5R)-bis(methoxymethyl)pyrrolidine gave (S)-3-mercapto-2-methylpropanoic acid, a key intermediate for captopril, in 98% ee and 96% yield. The chiral auxiliary was recovered in the demethylated form of N-Boc-(2R,3R)-bis(hydroxymethyl)pyrrolidine in 90% yield.  相似文献

Chiral (S)-pyrrolidine trifluoromethanesulfonamide has been shown to serve as an effective catalyst for direct Michael addition reactions of aldehydes and ketones with nitroolefins. A wide range of aldehydes and ketones as Michael donors and nitroolefins as acceptors participate in the process, which proceeds with high levels of enantioselectivity (up to 99 % ee) and diastereoselectivity (up to 50:1 d.r.). The methodology has been employed successfully in an efficient synthesis of the potent H(3) agonist Sch 50917. In addition, a practical three-step procedure for the preparation of (S)-pyrrolidine trifluoromethanesulfonamide has been developed. The high levels of stereochemical control attending Michael addition reactions catalyzed by this pyrrolidine sulfonamide, have been investigated by using ab initio and density functional methods. Transition state structures for the rate-limiting C--C bond-forming step, corresponding to re- and si-face addition to the reactive conformation of the key enamine intermediates have been calculated. Analysis of these structures indicates that hydrogen bonding plays an important role in catalysis and that the energy barrier for si-face attack in reactions of aldehydes to form 2R,3S products is lower than that for the re-face attack leading to 2S,3R products. In contrast, the energy barrier for re-face addition is lower than that for si-face addition in reactions of ketones. The computational results, which are in good agreement with the experimental observations, are discussed in the context of the stereochemical course of these Michael addition reactions.  相似文献
