Reactions of aryl trifluoromethyl ketone imines with 2-sulfanylacetic and 3-sulfanylpropanoic acids afforded 2-aryl-2-trifluoromethyl-1,3-thiazolidin-4-ones and 2-aryl-2-trifluoromethyltetrahydro-4H-1,3-thiazin-4-ones, respectively. Their subsequent oxidation with hydrogen peroxide gave the corresponding 2-aryl-2-trifluoromethyl-1,3-thiazolidin-4-one 1-oxides and 2-aryl-2-trifluoromethyltetrahydro-4H-1,3-thiazin-4-one 1-oxides and 1,1-dioxides.  相似文献

The nitrosation of the oximes of 4-phenyl-3-buten-2-one and 1,3-diphenyl-2-propen-1-one under oxygen has been reinvestigated. In addition to 4-oxo- and 4-oximino-4H-pyrazole 1,2-dioxides previously reported, the reactions give 4-nitro-1-hydroxypyrazole 2-oxides. In the case of 1,3-diphenyl-2-propen-1-one oxime the nitrosation reaction also gives 3,5-diphenyl-4-nitrato-4,5-dihydroisoxazole. Evidence is presented suggesting that the nitrate ester is formed through the rearrangement of a peroxynitrite intermediate.  相似文献

The reactions of 3-hydroxy-2-(2-hydroxyalkyl)- [or (2-hydroxyaryl)]-1,2-dihydro-quinazolin-4-ones with formaldehyde or acetaldehyde afford 1,3-oxazino[3,4-a]quinazolin-4-one derivatives. The reactions with other aldehydes RCHO and 3-hydroxy-2-R′-1,2-dihydroquinazolin-4-ones can give 3-hydroxy-2-R-1,2-dihydroquinazolin-4-ones, 2-substituted quinazolin-4-ones, or dianthranilide. Nitrosation of the oximes of 3-bromo-3-penten-2-one, 3-bromo-4-phenyl-3-buten-2-one, and 2-bromo-1,3-diphenyl-2-propen-1-one using sodium nitrite in acetic acid gave low yields of 4-pyrazolone 1,2-dioxides. Nitrosation using butyl nitrite in the presence of copper(II) sulfate and pyridine in aqueous ethanol produced insoluble copper complexes from which 3,5-dimethyl-, 3-methyl-5-phenyl-, and 3,5-diphenyl-4-bromo-1-hydroxypyrazole 2-oxides could be liberated by treatment with dilute potassium hydroxide, filtration, and acidification of the filtrate. High yields were obtained with the first two oximes, but, presumably due to unfavorable stereochemistry of the oxime, the diphenyl derivative gave a lower yield of the complex, accompanied by 4-bromo- and 4-nitro-3,5-diphenylisoxazole and 4-oximino-3,5-diphenyl-4,5-dihydroisoxazole.  相似文献

Methyl- and phenylhydrazines react with 2-(diphenylacetyl)-1,3-indandione ( 1 ) to yield respectively the 1-(methylhydrazone) and the 1-(phenylhydrazone) of 2-(diphenylacetyl)-1,3-indandione ( 2a and 2b ). In comparison, acetic and benzoic acid hydrazides react with 1 to give respectively the α-(acetylhydrazone) and the α-(benzoylhydrazone) of 2-(diphenylacetyl)-1,3-indandione ( 3a and 3b ). Cyclization of 2a and 2b gives 2,3-disubstituted indeno[1,2-c]pyrazol-4(2H)-ones ( 7a and 7b ). Cyclization of 3a and 3b , followed by methylation, gives 1-methyl- and 2-methyl-3-(diphenylmethyl)indeno[1,2-c]pyrazol-4(1 and 2H)-ones ( 9a and 9b ). 2-Isovaleryl-1,3-indandione reacts with phenylhydrazine to give directly 3-isobutyl-1-phenylindeno[1,2-c]-pyrazol-4(1H)-one ( 10 ).  相似文献

Methyl 2-acetyl-3-{[2-(dimethylamino)-1-(methoxycarbonyl)ethenyl]amino}prop-2-enoate ( 4 ) and phenyl-methyl 2-acetyl-3-{[2-(dimethylamino)-1(methoxycarbonyl)ethenyl]amino}prop-2-enoate ( 5 ) were prepared in three steps from the corresponding acetoacetic esters, and used as reagents for the preparation of N3-protected 3-amino-4H-pyrido[1,2-a]pyrimidin-4-ones 10 – 12 , 5H-thiazolo[3,2-a]pyrimidin-5-one 13 , 4H-pyrido[1,2-a]-pyridin-4-one 19 and 2H-1-benzopyran-2-ones 20 – 23 . Free 3-amino-4H-pyrido[1,2-a]pyrimidin-4-ones 24 – 26 were prepared from 10 – 12 by removal of the 2-(methoxycarbonyl)-3-oxobut-1-enyl or 3-oxo-2-[(phenyl-methoxy)carbonyl]but-1-envl as N-protecting group by various methods.  相似文献

Oxidation of 2H, 6H-thiin-3-ones 1a – c with 3-chloroperbenzoic acid affords the corresponding 1-oxides 2a – c . On irradiation (350 nm) in either benzene or MeCN, these cyclic sulfoxides 2 isomerize to 3H, 7H-1,2-oxathiepin-4-ones 3 . The tetramethyl derivative 3a is isolated by flash chromatography at ?10°, but, at higher temperatures, it undergoes ring contraction and H2O elimination to give 4,4-dimethyl-2(2-methylprop-2enylidene)thietan-3-one ( 4 ). Diemthyloxathiepinones 3b and 3c undergo ring contraction in MeOH to afford 1-(4-methylthiophen-2-yl)ethanone ( 5 ) and two diastereoisomeric 4,4-dimethyl-2-methoxy-2-(1-methoxyethyl)thietan-3-ones ( 6 and 7 , respectively).  相似文献

8-Substituted 2-morpholin-4-yl-quinolin-4-ones and 9-substituted 2-morpholin-4-yl-pyrido[1,2-a]pyrimidin-4-ones with selected aryl and heteroaryl groups as the substituent have been synthesised as potential inhibitors of DNA-dependent protein kinase. A multiple-parallel approach, employing Suzuki cross-coupling methodology, was utilised in the preparation of 8-substituted 2-morpholin-4-yl-quinolin-4-ones. For this purpose 8-bromo-2-morpholin-4-yl-quinolin-4-one was required as an intermediate. This compound was obtained by adapting a literature route in which thermal cyclocondensation of (2-bromoanilino)-morpholin-4-yl-5-methylene-2,2-dimethyl[1,3]dioxane-4,6-dione afforded 8-bromo-2-morpholin-4-yl-quinolin-4-one. A multiple-parallel approach, employing Suzuki cross-coupling methodology, was also utilised to prepare 9-substituted 2-morpholin-4-yl-pyrido[1,2-a]pyrimidin-4-ones using 9-hydroxy-2-morpholin-4-yl-pyrido[1,2-a]pyrimidin-4-one O-trifluoromethanesulfonate as an intermediate. 8-Substituted 2-morpholin-4-yl-quinolin-4-ones and 9-substituted 2-morpholin-4-yl-pyrido[1,2-a]pyrimidin-4-ones were both inhibitors of DNA-dependent protein kinase. When the substituent was dibenzothiophen-4-yl, dibenzofuran-4-yl or biphen-3-yl, IC50 values in the low nanomolar range were observed. Interestingly, the pyridopyrimidinones and quinolinones were essentially equipotent with the corresponding 8-substituted 2-morpholin-4-yl-chromen-4-ones previously reported (I. R. Hardcastle, X. Iodine catalyzed one-pot reactions of salicylaldehyde and dimolecular 1 H-indene-1,3(2H)-dione,barbituric acids,4-hydroxycoumarin, or 4-hydroxy-6-methylpyran-2-one were performed and provided a rapid,convenient and general approach to synthesize the chromene derivatives.2-(11-Oxo-10,11-dihydroindeno[l,2-b]chromen-10-yl)-1H-indene-1,3(2H)-diones P1-P4 and 10-(4-hydroxy-6-methyl-2-oxo-2H-pyran-3-yl)-3-methylpyrano[4,3-6]chromen-1(10H)-ones P8-P9 were unprecedentedly prepared and structurally identified by NMR and Mass.The confirmation of structure by single crystal X-ray crystallography is reported for P3.  相似文献

4-Hydroxy-4,5,5-trimethyl-3-[2-(2-methyl-7-R-1H-indol-3-yl)ethyl]-1,3-oxazolidin-2-ones (R = H or Alk), which were synthesized from 4-methylidene-1,3-dioxolan-2-one and substituted 2-methyltryptamines, undergo intramolecular amidoalkylation when treated with polyphosphoric acid. The scope of this reaction is exemplified and the proposed mechanism is also implicated in another degradation process. For the first time 5,7-di-tert-butyl-1,3-dimethyl-3a,9a-diphenyl-3,3a-dihydro-1H-benzo[5,6][1,4]dioxino[2,3-d]imidazol-2(9aH)-one 13 and complex 9 of 4,6-di-tert-butyl-3-nitrobenzene-1,2-diol with 1,3-dimethyl-4,5-diphenyl-1H-imidazol-2(3H)-one 10a were prepared by the reactions of 3-alkylthio-5,7-dimethyl-4a,7a-diphenyl-4a,5,7,7a-tetrahydro-1H-imidazo[4,5-e]-1,2,4-triazin-6(4H)-ones with 3,5-di-tert-butyl-1,2-benzoquinone 1 and 4,6-di-tert-butyl-3-nitro-1,2-benzoquinone 2, respectively. Photochemical transformations of compounds 9 and 10a as well as products of its photooxygenation involving singlet oxygen under UV irradiation: urea 16, isomeric 1,3-dimethyl-4,5-diphenylimidazolidin-2-ones 17 and 17′, and compound 18 were studied by the spectral-kinetic method. Data on the absorption and fluorescence properties of synthesized compounds and their photoproducts were obtained.  相似文献

An efficient and robust solid-phase synthesis of 6-substituted-2,5,6,8-tetrahydro-3H-imidazo[1,2-a]pyrimidin-7-ones, 3-substituted-1,3,4,6,7,8-hexahydro-pyrimido[1,2-a]pyrimidin-2-ones and 3-substituted-3,4,6,7,8,9-hexahydro-1H-pyrimido[1,2-a][1,3] diazepin-2-ones from the acetates of Baylis-Hillman adducts employing Michael addition of diamines followed by intramolecular cyclization with cyanogen bromide and, finally, base promoted cyclative cleavage has been developed. The procedure is validated through an automated parallel synthesis of a small library of fourteen compounds of the 3H-imidazo[1,2-a]pyrimidin-7-one series.  相似文献

Tetrazolo[1,5-a]pyridines/2-azidopyridines 1 undergo photochemical nitrogen elimination and ring expansion to 1,3-diazacyclohepta-1,2,4,6-tetraenes 3, which react with alcohols to afford 2-alkoxy-1H-1,3-diazepines 4 (5), with secondary amines to 2-dialkylamino-5H-1,3-diazepines 16, sometimes via isolable 2-dialkylamino-1H-1,3-diazepines 15, and with water to 1,3-diazepin-2-ones 19. The latter are also obtained by elimination of isobutene or propene from 2-tert-butoxy- or 2-isopropoxy-1H-1,3-diazepines 4 or 5. 1,3-Diazepin-2-one 22B and 1,3-diazepin-4-one 24 were obtained from hydrolysis of the corresponding 4-chlorodiazepines. Diazepinones 19 undergo photochemical ring closure to diazabicycloheptenones 25 in high yields. The 2-alkoxy-1H-1,3-diazepines 4 and 5 interconvert by rapid proton exchange between positions N1 and N3. The free energies of activation for the proton exchange were measured by the Forsén-Hoffman method as DeltaG([double dagger])298= 16.2 +/- 0.6 kcal mol(-1) as an average for 4a-c in CD2Cl2, acetone-d6, and methanol-d4, and 14.1 +/- 0.6 kcal mol(-1) for in 4c acetone/D2O. The structures of 2-methoxy-5,6-bis(trifluoromethyl)-1H-1,3-diazepine 4k, 1,2-dihydro-4-diethylamino-5H-1,3-diazepin-2-one 22bB, and diazabicycloheptanone were 26 determined by X-ray crystallography. The former represents the first reported X-ray crystal structure of any monocyclic N-unsubstituted 1H-azepine.  相似文献

Alkaline hydrolysis of 1,3-disubstituted 6-(2-dimethylaminovinyl)uracils 2 induced a novel ring transformation giving 4-alkylaminopyridin-2-ones 3 via ring-opening and ring-closure processes. The 4-methylamino-3-nitropyridin-2-one ( 3a ) thus obtained was employed for the synthesis of 3-deazahypoxanthine derivative 8. 4-Alkylamino-3-cyanopyridin-2-ones 11 , ricinine analogs, were also prepared by the reaction of 4-chloro-3-cyano-1-methylpyridin-2-one ( 10 ) with amines.  相似文献
