Some new mono-, bis-spiro- and dispiro-β-lactams have been synthesized from imines derived from 9H-fluoren-9-one and a ketene derived from 9H-xanthene-9-carboxylic acid or phenoxyacetic acid by a [2+2] cycloaddition reaction in good to excellent yields varying from 45 to 83%. The biological activity of these monocyclic β-lactams was successfully investigated against Plasmodium falciparum K14 resistant strain with excellent EC50 values up to 5 μM.  相似文献

The synthesis of two series of β-amino ketones containing a p-aminobenzoic acid moiety (TM-1 and TM-2) using a modified protocol of the Mannich reaction is reported. The molecular structures of a total of tweenty three new target compounds were characterized by 1H NMR, 13C NMR, ESI-MS and HR-MS. Subsequently, their antidiabetic activities were screened in vitro. The α-glucodase inhibition (α-GI) activity of compound 1e reached a remarkable level of 66.50%. The peroxisome proliferatoractivated receptor (PPAR) relative activation activities of six compounds are above 80%, and in particular 2i displays an unprecedentedly high PPAR of 130.91%. The structure-activity relationships of the compounds were established. 2i is also subject to further in-depth investigation.  相似文献

Endoperoxides belonging to the G-factor family, containing a spiroalkane moiety in the α position to the O-O bond, have been synthesized via an autoxidation reaction on the corresponding dienol precursors. Methylated derivatives in the peroxyhemiketal position have also been prepared. The in vitro antimalarial activities are reported. A five-step synthesis of novel tricyclic β-lactams has been exploited by stereoselective intramolecular cycloadditions of appropriate nitrilimines 6 and 12. These labile intermediates were generated in situ from the corresponding hydrazonoyl chlorides giving rise to the hitherto unknown azeto[3′,4′:1,2]hexano[3,4-c]pyrazole and azeto[3′,4′:1,2]heptano[3,4-c]pyrazole skeletons.  相似文献

As an aspect of our ongoing research on developing novel antiproliferative agents, 31 new triazole–isoindoline hybrids bearing 3,4,5-trimethoxyphenyl moiety were synthesized and evaluated for their antiproliferative activity against four cancer cell lines (HepG2, HeLa, PC-3, and HCT116). Some compounds showed excellent potency, and compared to fluorouracil, the most promising compound 6s exhibited 5.8-, 4.3-, and 1.3- fold increase in activities against HeLa, HepG2, and PC-3 cell lines with IC50 values of 9.7, 10.7, and 16.8 μM, respectively. Moreover, structure–activity relationship studies indicated that a much shorter amide linkage and electron-withdrawing groups at phenyl ring of the acetamide fragment contribute to the antitumour activity.  相似文献

N-Thiolated β-lactams are an interesting new family of bioactive agents having antibacterial, antifungal, and anticancer capabilities. Our interest in these β-lactams stems from their highly unusual structure–activity profiles and selectivity toward only a few genera of pathogenic bacteria, including Staphylococcus aureus, Bacillus anthracis, and Micrococcus. Here, we provide a complete outline on our work in the discovery and ongoing development of these compounds, starting from our initial, unexpected finding of antimicrobial activity for one of the lead compounds, to a more complete understanding of their chemical and biological mode of action and potential utility as antibacterial compounds.  相似文献

Based on the original structure of harmine, several novel 1,2,3,4-tetrahydro-β-carboline, β-carboline and 1-substituted-β-carboline derivatives bearing a substituted carbohydrazide group at C-3 were designed and synthesized to investigate the structure-activity relationship of their analogues. All of the compounds were characterized by infrared (IR), proton and carbon nuclear magnetic resonance (1H-NMR, 13C-NMR), and mass spectroscopy (MS). The bioassay tests showed that N'-benzylidene-1-phenyl-β-carboline-3-carbohydrazide (C(25)H(18)N(4)O, m.w. 390.4) (c2) and N'-(4-trifluoromethyl-benzylidene)-1-phenyl-β-carboline-3-carbohydrazide (C(26)H(17)N(4)OF(3), m.w. 458) (d2) exhibited good inhibitory activity against dicotyledonous and monocotyledonous weeds, with EC(50) values of 4.83 μM and 14.25 μM, respectively.  相似文献

A series of 1-indolyl substituted β-carbolines including the natural products hyrtiosulawesine, pityriacitrin and pityriacitrin B were prepared via Pictet–Spengler condensation—oxidation strategy from the corresponding indolyl-acetaldehydes and substituted tryptamines. Efforts to prepare the C-1 methylene-linked β-carboline analogues for structure–activity relationship studies were unsuccessful. Biological evaluation revealed two analogues (5 and 41) to exhibit weak inhibition of phospholipase A2 (IC50 171 and 131 μM, respectively), two to act as antioxidants (3 and 43), and 12 analogues with activity towards a chloroquine-resistant strain (FcB1) of Plasmodium falciparum (IC50 1.0–23 μM). Testing against a panel of 60 human tumour cell lines revealed a general lack of cytotoxic effect for most of the compounds with the exception of β-carboline 42 exhibiting modest antileukaemic activity towards the HL-60(TB) cell line (LC50 4.2 μM). In addition, two novel structures (30 and 32) resulting from aldol condensation followed by Pictet–Spengler cyclisation displayed cytotoxicity with pronounced subpanel specificities towards colon cancer (COLO 205 and HCC-2998) cell lines.  相似文献

Several 6,8-dichloroquinolone analogues were synthesized from the key intermediate compound of 2,3,4,5-tetrachlorobenzene carbonyl chloride,which was obtained from the starting material of tetrachlorophthalic anhydride.Their in vitro antibacterial activities were evaluated.As a result of this study,compounds 21c and 21d were twofold more potent than ciprofloxacin (CPFX) and norfloxacin (NFLX) against Staphylococcus aureus-9,and with the same potent as CPFX and NFLX while against Escherichia coli-2,but were less potent than references in against Pseudomonas aeruginosa-17.  相似文献

The reaction of chiral (2S)-enolates of dioxolan-4-ones, derived from lactic and mandelic acids, with (SR)-tert-butyl sulfinyl ketimines, derived from butan-2-one, pentan-2-one, and decan-2-one, afforded conformationally restrained β2,2,3,3-isoserines bearing two adjacent quaternary stereogenic centers in the form of N-sulfinyl protected 1′-amino-dioxolan-4-ones. The selective acid-induced removal of the sulfinyl protecting group provided the corresponding 1′-aminodioxolanones, whose base-induced cyclization afforded the corresponding chiral tetra-substituted 3-hydroxy-β-lactams. The synthesis of a dipeptide by reaction coupling between the 1′-aminodioxolanone (2S,5R,1′R)-19 and N,N-dimethylglycine was successfully achieved.  相似文献

A novel fluorescence dye based on pyrazine-boron complexes bearing a β-iminoketone ligand has been synthesized by using a simple two-step reaction. Synthesized complexes exhibited fluorescence in solution (F(max): 472-604 nm) and in the solid state (F(max): 496-624 nm). These complexes showed a larger Stokes shift (3690-4900 cm(-1)) than well-known boron dipyrromethene dyes (400-600 cm(-1), in most cases).  相似文献

Pseudo-C-nucleosides have potential biological activity, and an efficient synthesis of new pseudo-C-nucleosides has been developed via the reaction of a β-formyl-α,β-unsaturated ester bearing a β-sugar moiety with hydrazines in neutral and acidic conditions. The preparation of the β-formyl-α,β-unsaturated ester was accomplished by oxidation of the secondary hydroxyl group of 3-O-benzyl-1,2-O-isopropylidene-α-d-glucofuranose, followed by elongation of its carbon chain with (ethoxycarbonylmethylene)triphenylphosphorane and oxidation of the hydroxymethyl group.  相似文献

A facile synthesis of novel α-aminophosphonates 5a-j was accomplished by condensation of imines (3a-j) with diethyl phosphite (4) in ethanol at 50–60°C using easily recoverable and reusable catalyst, tetramethyl guanidine (TMG) via pudovik reaction in high yields. All the title compounds were characterized by spectral and elemental analysis. They were further screened for their abilities towards in vitro antibacterial, antifungal and antioxidant activities. The compounds 5g, 5d, 5f exhibited good antibacterial and antifungal activities compared to the standard bactericide, Penicillin and fungicide, Griseofulvin, respectively. The compounds 5h, 5i, 5g, and 5c exhibited good antioxidant activity compared to the standard ascorbic acid.  相似文献

Starting from cinnamates 9 , obtained by Wittig reaction or Heck coupling, the diols 17 were prepared by asymmetric dihydroxylation. This was followed by a regioselective substitution of the 3‐OH group with hydrazoic acid under Mitsunobu conditions. Methylation of the 2‐OH group and reduction of the azide group led to the β‐tyrosine derivatives 8 . Condensation with the dipeptide acid 6 furnished the tripeptide part of the chondramides. The derived acids 21 were combined with the hydroxy ester 7 to the esters 22 . Cleavage of the tert‐butyl groups and intramolecular lactam formation gave rise to the chondramide A analogues 2 b – k . Growth inhibition assays showed most of the analogues to be biologically active. Some of them even reach the activity of jasplakinolide. It can be concluded that the 4‐position of the aryl ring in the β‐tyrosine of chondramide A tolerates structural modifications quite well.  相似文献

Several new monocyclic β-lactam monomers bearing the NO2 group 2ag were synthesized via a [2 + 2] ketene–imine cycloaddition reaction (Staudinger reaction). Calculation of coupling constant of H-3 and H-4, and the X-ray crystallography of β-lactam 2e confirmed the cis stereochemistry of these β-lactams. Then aminophenyl β-lactams were synthesized by the reduction of NO2 to NH2 group in the presence of Raney Ni and hydrazine hydrate. Treatment of these aminophenyl β-lactams with acryloyl chloride and sodium bicarbonate (NaHCO3), afforded the monomers bearing the NHCOCH=CH2 group. These acrylated β-lactam monomers were dissolved in a warm mixture of butyl acrylate and styrene and then were converted to the corresponding polyacrylate nano β-lactams by emulsion polymerization in water. A unique feature of this methodology is the ability to incorporate water-insoluble compounds directly into the nanoparticle framework. Structures of the synthesized compounds were confirmed by physical and spectral analyses. Dynamic light scattering analysis and transmission electron microscopy of the final emulsions show that the nanoparticles were about 50–70 nm in diameter.  相似文献

This paper describes the synthesis of some new β-lactam derivatives containing the 1,2,3-triazole moiety. All the compounds were evaluated for their in vitro antimicrobial and antimalarial activities. Moderate to excellent antimalarial activities were encountered. The results showed that compounds 5a, 5c, 5f, 5i exhibited the most potent antimalarial activity with IC50 values of 0.85, < 0.97, < 0.97, 1.81 µM against chloroquine-resistant P. falciparum K1 strain. A QSAR study highlights the structure–activity relationships that correlate the observed antimalarial activities with changes in the compounds’ structural features.  相似文献

β-Lactams are crucial structural feature of β-lactam antibiotics and important intermediates in synthetic and pharmaceutical chemistry. Synthetic methods for β-lactams with π electron-withdrawing substituents, such as formyl (carbaldehyde), acyl, imino, carboxylic acids, carboxylates, carboxamides, cyano (carbonitriles), and nitro groups, on their 3- and/or 4-position(s) are presented in this review. The methods are divided mainly into intramolecular cyclizations, cycloaddition, and other methods, for example, Ugi-type reaction of β-keto acids, amines, and isonitriles, and modification of β-lactam derivatives. Cyclizations include cyclization of haloacetamidoacetates(malonates), intramolecular carbene insertion of α-diazoalkanamides, ring-opening cyclization of α,β-epoxyalkanamidoacetates, oxidative coupling of 3-oxoalkanamidoacetates, oxidative cyclization of N-p-hydroxyphenyl β-oxoalkanamides, intramolecular cyclization of aspartic acid derivatives or β-hydroxyalkanamides, and radical cyclization of N-vinyl β-oxoalkanamides. Cycloadditions incorporate Staudinger cycloaddition of ketenes and imines, cycloaddition of nitrones and alkynes, cycloaddition of nitrones and alkylidenecycopropanes and subsequent acidic rearrangement, and condensation of imines and enolates (ethers) of esters. The scope, limitation, and stereoselectivity are also discussed for some methods. Most of the β-lactam derivatives are key intermediates or precursors for preparation of β-lactam antibiotics and their analogs.  相似文献

Novel chiral norephedrine-based β-amino alcohol ligands containing a thiophene ring were prepared from norephedrine and substituted furan carbaldehydes (methyl- or ethyl-substituted) and used in combination with VO(acac)2 for the asymmetric oxidation of aryl methyl sulfides using H2O2 as an oxidant. Amino alcohol derived Schiff bases 4,5ab gave higher enantiomeric excesses than amino alcohol-derived reduced Schiff based ligands 6,7ab. Of these chiral ligands, (1S,2R)-5b and (1S,2R)-7b gave high yields (90%) with moderate to high enantioselectivities (78%, 96% ee, respectively). The oxidation of other aryl methyl sulfides with (1S,2R)-5b and (1S,2R)-7b as ligands afforded the corresponding sulfoxides in 60–89% yields and with 92–99% ee.  相似文献
