Abstract

A simple and sensitive extractive spectrophotometric method has been described for the determination of Ketotifen. The developed method is based on the formation of a colored ion‐pair complex (1∶1 drug/dye) of Ketotifen and bromocresol green in buffer pH 3 and extraction in chloroform. The extracted complex shows absorbance maxima at 423 nm. Beer's law is obeyed in the concentration range of 5.15–61.91 µg/ml. The proposed method has been applied successfully for the determination of drug in commercial tablets and syrup dosage form. No significant interference was observed from the excipients commonly used as pharmaceutical aids with the assay procedure.  相似文献

A new formulation of nifedipine tablets was prepared. The tablets were conditioned in amber-colored glass containers and placed in a climatized room at 40 °C and relative humidity of 75% for 180 days. Differential scanning calorimetry (DSC) and Thermogravimetry (TG) were used in order to evaluate the thermal properties of nifedipine, the excipients and two well-known nifedipine degradation products. There is no evidence of interaction between nifedipine and excipients or degradation products. High performance liquid chromatography (HPLC) was used in the dosage of nifedipine tablets before and after acclimatized exposure. Results show that DSC and TG offer important data for a more detailed assessment of the stability of a pharmaceutical formulation.  相似文献

First-derivative spectrophotometry, applying the peak-zero method, was developed for the determination of rosiglitazone (RSG) in coated tablets. The solutions of standard and sample were prepared in ethanol. Quantitative determination of the drug was performed at 331.4 nm (N = 4; delta lambda = 3.2 nm) and was evaluated for the parameters specificity, linearity, precision, and accuracy. The specificity test showed that there was no interference from excipients commonly found in the commercial pharmaceutical formulation at 331.4 nm. The standard curve showed a correlation coefficient of 0.9997. Precision was demonstrated by a relative standard deviation value of 0.50%. The recovery test resulted in an average of 100.06%, which confirmed the accuracy of the method. The results for first-derivative spectrophotometry (D(1)), liquid chromatography, and micellar electrokinetic chromatography were compared by analysis of variance (ANOVA), and there were no significant differences among these methods. Therefore, D(1) can be easily and directly applied to analyze RSG in coated tablets.  相似文献

Abstract

Azithromycin (AZT), an antibiotic belonging to the family of macrolides, can be analyzed by a new spectrophotometric method based on the formation of an ion pair between this drug and an inorganic complex of (Mo(V)–thiocyanate) followed by its extraction with dichloroethane. This ion‐association complex shows an orange color and exhibits a maximum absorbance at 469 nm. The experimental conditions of the reaction were studied and optimized. The calibration graph was linear (r=0.9996) over the range 10?6 M–10?5 M of AZT. This simple and validated method has been successfully applied to the determination of azithromycin in pharmaceutical formulations with a mean relative standard deviation of 1.07% and mean recovery of 99.66%. The common excipients present in azithromycin formulations did not interfere in its determination. This new spectophotometric method has been applied successfully to illustrate the dissolution profiles of original tablets and generic compounds; hence, it could be employed in routine quality control of azithromycin in pharmaceutical dosage forms.  相似文献

A simple and sensitive fluorometric method for determination of ketorolac tromethamine was studied. The method depends on oxidation of the drug with cerium(IV) and subsequent monitoring of the fluorescence of the induced cerium(III) at lambda(em) 365 nm after excitation at 255 nm. Different variables affecting the reaction conditions, such as the concentrations of cerium(IV), sulfuric acid concentration, reaction time, and temperature, were carefully studied and optimized. Under the optimum conditions, a linear relationship was found between the relative fluorescence intensity and the concentration of the investigated drug in the range of 0.1-0.8 microg/mL. No interferences could be observed from the excipients commonly present in dosage forms. The proposed method was successfully applied to the analysis of the investigated drug in its pure form, pharmaceutical preparations, and biological fluids with good accuracy and precision. The recoveries for pharmaceutical formulations ranged from 99.8-101.0 +/- 0.6% for tablets, 98.5-101.0 +/- 1.0% for ampoules, and 99.0-100.5 +/- 0.7% for eye drops. The results obtained by the proposed method were satisfactory compared with those obtained by the official method. The recoveries for biological fluids were 99.1-100.4 +/- 0.7 and 99.0-100.0 +/- 0.5% for plasma and urine, respectively.  相似文献

Reflectance near-IR (RNIR) spectroscopy was used for the simultaneous determination of chondroitin (CH), glucosamine (GO), and methyl sulfonyl methane (MSM) in tablets. Simple sample preparation was done by grinding, sieving, and compression of the tablets for improving RNIR spectra. Principal component regression and partial least squares (PLS-1 and PLS-2) were successfully applied to quantify the three components in the studied mixture using information included in RNIR spectra in the range of 4350-9100 cm(-1). The calibration model was developed with drug concentration ranges of 14.5-44.2% (w/w) for CH, 18.4-55.3% (w/w) for GO, and 6-18.6% (w/w) for MSM with addition of tablet excipients to the calibration set in the same ratio as in the tested tablets. The calibration models were evaluated by internal validation, cross-validation, and external validation using synthetic and pharmaceutical preparations. The proposed method was applied for analysis of six batches of the pharmaceutical product. The results of the proposed method were compared with the results of the pharmacopoeial method for the same batch of the pharmaceutical product. No significant differences between the results were found. The RNIR method is accurate and precise, and can be used for QC of pharmaceutical products.  相似文献

A rapid and simple method has been developed for determination of inorganic elements in airborne particulate matter (PM10) by using acidified subcritical water and ICP–OES. Elements such as Al, As, B, Ba, Cd, Cu, Fe, Mn, Pb, Se, and Zn were rapidly and efficiently extracted from PM10 samples with a solution of 0.1 mol L–1 HNO3 under subcritical conditions. The method requires approximately 5% of the amount of acid used in the standard microwave extraction procedure. The material selected for the subcritical extraction manifold was poly ether ether ketone (PEEK), to avoid sample contamination with elements present in previously reported stainless-steel manifolds. The extraction temperature, time of static and dynamic extraction, and flow rate of acidified water were studied keeping the pressure controlled at about 1,500 psig. The efficiency of extraction of most of the analytes increased with temperature, tending to quantitative extraction at temperatures near 150°C. After the extraction process the analytes were determined directly in the extract by ICP–OES. When the method was compared with the USEPA counterpart, the results indicate that under optimized conditions (static extraction time: 15 min, dynamic extraction time: 30 min, flow rate: 2 mL min–1) the analytes were extracted with recoveries between 73 and 158%. Alternatively, by using an extraction time of 15 min, the method could be used to screen for all the elements, with recoveries over 50%. The developed method was applied to the determination of inorganic elements in airborne particulate matter in the atmosphere of Santiago, Chile.  相似文献

Melatonin was determined in pharmaceutical preparations by means of two simple and reliable analytical methods based on micellar electrokinetic chromatography (MEKC) and spectrofluorimetry. The fluorescence emission values were measured at λ=350 nm when exciting at λ=275 nm. The MEKC analysis was achieved using a system consisting of 40 mM SDS in phosphate buffer (20 mM, pH 7.5). The extraction of melatonin from the tablets was achieved by means of a simple one-step dissolution with methanol/water. Both methods were applied for the determination of melatonin in commercial formulations and galenic preparations. The MEKC procedure allows the quantitative determination of melatonin in all pharmaceutical preparations tested. On the contrary, the spectrofluorimetric method is not suitable for tablets which also contain tryptophan; this interference can be eliminated by a suitable liquid-liquid extraction procedure. The results obtained with the two methods are in good agreement and satisfactory in terms of precision and accuracy.  相似文献

The differential pulse polarographic determination of Corticosteroids in single-component tablets is described. The active compounds are extracted from the excipients with a solvent with similar lipophilicity as the polarographic solvents, with a yield of about 100%. The corticosteroid esters and alcohols are determined by standard addition methods. The standard deviations range from 0.5 to 2.5% depending on the excipients and on the method of extraction.  相似文献

Two spectrophotometric methods were developed for the determination of ranitidine. The first method was a kinetic spectrophotometric method based on the catalytic effect of ranitidine on the reaction between sodium azide and iodine in an aqueous solution. The calibration graph was linear from 4–24 μg/mL. The drug was determined by measuring the decrease in the absorbance of iodine at 348 nm using a fixed time method. The decrease in the absorbance after 1 minute from the initiation of the reaction was related to the concentration of drug. The detection limit of the procedure was 0.76 μg/mL. The proposed procedure was successfully utilized in the determination of the drug in pharmaceutical preparations with mean recovery in the range of 99.83 ? 101.16%. The second method is a colorimetric method, which depends on the measurement of absorbances of tris (o‐phenanthroline) iron(II) [method 2A] and tris (bipyridyl) iron(II) [method 2B] complexes at 512 nm. The complexes obeyed Beer's law over the concentration range of 2–16 μg/mL and 4–40 μg/mL for methods 2A and 2B, respectively. The developed method has been successfully applied for the determination of ranitidine in bulk drugs and pharmaceutical formulations. The common excipients and additives did not interfere in its determination.  相似文献

A simple, accurate, precise and fully automated method for the simultaneous determination of abacavir, lamivudine and zidovudine in pharmaceutical tablets, human serum samples and drug dissolution studies has been developed. Separation was performed on a 5 μm Zorbax® C18 column (150 × 4.6 mm ID) with methanol:water:phosphate buffer at pH 5.65 (80:10:10; v/v/v) isocratic elution in less than 7 min with a flow rate of 0.6 mL min?1.Good sensitivity for all analytes was observed with UV detection at 275 nm. The method allowed quantitation over the 500–3,000 ng mL?1 range for abacavir and 500–5,000 ng mL?1 range for lamivudine and zidovudine. The method has been applied, without any interference from excipients or endogenous substances, for the simultaneous determination of these three compounds in tablets. A simple, precise and stability-indicating reversed-phase liquid chromatography method was developed and validated for the determination of vildagliptin (VLG) in pharmaceutical dosage form. The chromatographic separation was obtained within 6 min and was linear in the range of 20-80 μg/mL (r(2) = 0.9999). Limit of detection and limit of quantitation were 0.63 and 2.82 μg/mL, respectively. The method was validated in accordance with International Conference on Harmonization acceptance criteria for specificity, linearity, precision, accuracy, robustness and system suitability. Stress studies were carried out and no interference of the degradation products was observed. The excipients did not interfere in the determination of VLG. Furthermore, the main degradation product obtained from the stress studies (thermal, oxidative and alkaline hydrolysis) was evaluated for mass spectrometry and its molecular structure was predicted. The proposed method was successfully applied for the quantitative analysis of VLG in tablet dosage form, which will help to improve quality control and contribute to stability studies of pharmaceutical tablets containing this drug.  相似文献

A sensitive spectrofluorimetric procedure for the determination of paroxetine-HCl in pharmaceutical formulations and human plasma has been described. The native fluorescence of the drug has been studied under different conditions. Maximum fluorescence intensity was obtained in methanol at 340 nm using 290 nm for excitation. Different surfactants showed negative effect on the fluorescence intensity of paroxetine-HCl. Regression analysis of Beer's plot showed good correlation (r=0.9999) between fluorescence intensity and concentration over the range of 0.05-0.40 microg ml-1 with lower limit of detection (LOD) of 0.015 microg ml-1. The drug was successfully determined in its tablets with average % recovery of 98.00+/-0.99% which was in accordance with those given by a compendial method. The method was also applied to the determination of paroxetine-HCl in spiked human plasma with average recovery of 77.70+/-1.06%.  相似文献

建立了亚临界水萃取及气相色谱-串联质谱(GC-MS/MS)检测红茶中21种有机氯和拟除虫菊酯农药残留的方法。在萃取压力为5 MPa条件下,样品经150 ℃的亚临界水提取15 min后,将目标物转移至丙酮-正己烷(1:1, v/v)中,经ENVI-Carb固相萃取净化小柱净化,DB-5毛细管气相色谱柱分离,在多反应监测(MRM)模式下进行MS/MS检测,基质匹配溶液内标法定量。各目标物在5.0~320.0 μg/L范围内线性关系良好,相关系数均大于0.99,其定量限(信噪比(S/N)>10)为50 ng/g,检出限(S/N>3)为10 ng/g。茶叶基质中添加50、100和200 ng/g的标准品时,21种农药的回收率为70.18%~119.98%,相对标准偏差(RSD)为5.01%~11.76%。该方法的灵敏度、准确度和精密度均符合农药残留测定的技术要求,适用于红茶中有机氯和拟除虫菊酯农药残留的检测。  相似文献

This paper reports an indirect flow-injection (FI) method for the determination of the anti-hyperthyroid drugs methimazole and carbimazole in pharmaceuticals. The method was based on the inhibition that these thioimidazole drugs caused on the Cu(II)-catalysed chemiluminescence (CL) reaction between luminol and H2O2. The CL reaction was induced on-line and injection of the sample produced negative peaks as a result of the Cu(II) complexation by the analytes. The height of the FI peaks was proportional to the drug concentration in the sample. The linear range was 2-100 and 3-120 mg l−1 for methimazole and carbimazole, respectively. The relative standard deviation was 1.9% for methimazole and 2.1% for carbimazole at the 50 mg l−1 level (n=10). Common excipients present in pharmaceutical tablets were found not to interfere with the analysis. The method was applied to the determination of methimazole and carbimazole in pharmaceutical formulations with recoveries in the range 100±4%.  相似文献

A simple potentiometric method is described for the rapid determination of nafronyl-drugs in pharmaceutical preparations such as tablets. Nafronyl ion-selective membrane electrodes with either the nafronyl-dipicrylamine ion-pair complex in 1,2-dichloroethane or the nafronyl-dinonylnaphthalenesulphonic acid ion-pair complex in a PVC matrix as electroactive materials were used. Both electrodes exhibit near-Nernstian responses to protonated-nafronyl activity from 10(-2) to about 10(-5)M, in pH ranges that depend on the nature of the electroactive material used in the membrane. Nafronyl in the mg-range can be determined by potentiometric titration with sodium tetraphenylborate solution, with a relative standard deviation of less than 2.0%. No interference from any excipients in the tablets was observed.  相似文献

In the present study, a reverse-phase high performance liquid chromatography method was developed, validated and applied for the simultaneous determination of gliquidone, pioglitazone hydrochloride and verapamil in tablets and human serum. Chromatographic separation was achieved on a C18 column (5 μm, 25 × 0.46 cm) with a mobile phase consisting of methanol-water-acetonitrile (80:10:10 v/v/v) with a flow rate of 0.7 mL/min and pH adjusted to 3.50 with phosphoric acid at 230 nm. Glibenclamide was used as internal standard. The experimentally derived limit of detection and limit of quantitation were determined to be 0.24, 0.93, 0.40, and 0.80, 3.11, 1.36 μg/mL for gliquidone, pioglitazone, and verapamil, respectively. There were no interfering peaks due to the excipients present in the pharmaceutical tablets. Thus, the proposed method is simple and suitable for the simultaneous analysis of active ingredients in dosage forms and human serum.  相似文献

A stability-indicating liquid chromatographic method was developed and validated for quantitative determination of olmesartan medoxomil (OLM) in coated tablets in the presence of degradation products generated under stress conditions. An isocratic LC separation was performed using a Phenomenex RP-18 column using a mobile phase consisting of water:triethylamine:acetonitrile (60:0.3:40 v/v/v, pH adjusted to 6.3 with phosphoric acid). The flow rate was 1.2 mL min−1 and the detection was achieved with a photodiode array detector set at 257 nm. The response was linear over a range of 10.0 to 30.0 μg mL−1 (r = 0.9999). The specificity and stability-indicating capability of the method was verified subjecting the reference substance and drug product to hydrolytic, oxidative, photolytic, and thermal stress conditions. The method showed a good and consistent recovery (100.2%) with low intra- and inter-day relative standard deviation (RSD) (≤1.0%). A considerable degradation occurred in all stress conditions and the degradation product was well resolved from the main peak. There was no interference of the excipients in the determination of the active pharmaceutical ingredient. Thus, the proposed method was found to be stability-indicating and can be used for routine analysis for quantitative determination of OLM in coated tablets without the interference of major degradation products.
