Known synthetic precursors of strobilurins A and X, i.e., methyl (3Z,5E)-6-aryl-3-methylhexa-3,5-dienoates (aryl is phenyl, 4-methoxyphenyl), were synthesized by highly stereospecific reactions from 2-(2-tert-butyldimethylsilyloxyethyl)- and 2-[2-(4-methoxybenzyloxy)-ethyl]-5-arylpenta-2E,4E-dien-1-ols. These dienols were efficiently dehydroxylated to (1E,3Z)-4-methyl-6-(4-methoxybenzyloxy)hexa-1,3-dienylarenes with their subsequent demethoxyben-zylation to (3Z,5E)-6-aryl-3-methylhexa-3,5-dien-1-ols. The latter through the step of corresponding aryldienals and aryldienoic acids were transformed to the target methyl (3Z,5E)-6-aryl-3-methylhexa-3,5-dienoates, which completes a formal synthesis of strobilurins A and X. Configuration of the C=C bonds of the conjugated aryldiene system is preserved in the considered transformations by 95–97%.  相似文献

The sponges Raspailia pumila and ramosa (Demospongiae, Tetractinomorpha, Axinellida) from the North-East Atlantic are shown to contain a series of novel long-chain enol ethers of glycerol where the enol ether C?C bond is conjugated, in sequence, to both an acetylenic and an olefinic bond. Polar extracts give raspailynes hydroxylated at their (1Z5Z)-1,5-alkadien-3-ynyl chain, like raspailyne Al ( = (+)-(S)-3-[((1Z,5Z)-16-hydroxy-hexadeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; (+ 2 ) and isoraspailyne A ( = (+)-3-[((1Z,5Z)-17-hydroxyocta-deca-1,5-dien-3-ynyl)oxy]-1,2-[propanediol; (+)- 3 ). Less polar extracts give 3 different types of raspailynes not hydroxylated at the chain. Raspailynes of the first type have either the (1Z,5Z)-configuration in a linear chain such as raspailyne B2 (( = (?)-(s)-3-[((1Z,5Z)-trideca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; (?)-4), raspailyne Bl ( = (?)-3-[((1Z,5Z)-tetradeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol;(?)- 5 ), and raspailyne B ( = 3-[((1Z,5Z)-pentadeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; 6 ) or the (1Z,5Z)-pentadeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; 6 )or the (1Z,5Z)-configuration in a chain ending with an isopropyl group, like isoraspailyne Bl ( = 3-[((1Z,5Z)-12-methyltrideca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; 7 ) and isoraspailyne B ( = 3-[((1Z,5Z)-13-methyltetradeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; 8 ). Raspailynes of the second type have the (1Z,5E)-configuration, like isoraspailyne Bla ( =3-[((1Z,5E)-tetradeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; 9 ) and isoraspailyne Ba ( = 3-[((1Z,5E)-13-methyltetradeca-1,5-dien-3-ynyl)oxy]-1,2-propanediol; 10 ). Raspailynes of the third type have the (1E,5Z)-configuration, like isoraspailyne Blb ( = 3-[((1E,5Z)-tetradeca-1,5-dien-3-ynyl)oxy]-1,2,-propanediol; 11 ). The (S)-configuration for (+)- 1 ,((+)- 2 , and (?)- 4 is derived from chemical correlations.  相似文献

The green seaweed Caulerpa taxifolia (VAHL ) C. AGARDH (Caulerpales), which, after its recent accidental introduction, is growing in the region of Cap Martin much more vigorously than in the tropics, is shown to contain the known sesquiterpenic toxins caulerpenyne ( 1 ) – in larger amounts than in tropical Caulerpales – and oxytoxin 1 ( 2 ). Novel, potentially toxic products isolated in small amounts from this seaweed include the sesquiterpenes taxifolial A ( = (5E)-6,10-dimethyl-2-[(E)2-oxoethylidene]undeca-5,9-dien-7- yne-1,3-diyl diacetate; 3 ), taxifolial B (= (1E,6E,10E)-3-[( Z )-acetoxymethylidene]-7, 11-dimethyl-12-oxododeca-1,6,10-trien-8-yne-1,4-diyl diacetate; 4 ), 10,11-epoxycaulerpenyne ( = (1E,6E)-3-[(Z)-acetoxymethylidene]-10,11-epoxy-7, 11-dimethyldodeca-1,6-dien-8-yne-1,4-diyl diacetate; 1:1 diastereoisomer mixture; 5 ), and taxifolial C ( = (2Z,6E)-3-formyl-7,11-dimethyldodeca-2,6,10-trien-8-yne-1,1, 4-triyl triacetate; 6 ), besides, as the first example of a monoterpene from the Caulerpales, taxifolial D ( = (2Z)-3,7-dimethylocta-2, 6-dien-4-ynal; 7 ).  相似文献

(4R,2E)-4-Benzyloxy-octa-2,7-dienyl(tributyl)stannane was transmetalated by tin(IV) chloride to generate an allyltin trichloride, which reacted with aldehydes to give (3Z)-1,5-syn-5-benzyloxynona-3,8-dien-1-ols with useful 1,5-stereocontrol. O-Benzylation, hydroboration and oxidation of the terminal double-bond of the product from 2-methylpropanal gave (5R,9S,6Z)-5,9-dibenzyloxy-10-methylundec-6-enal. Further reactions with 4-alkoxyalk-2-enylstannanes proceeded with useful 1,5-stereocontrol to give open-chain products with hydroxy or benzyloxy substituents stereoselectively disposed at remote positions along the chain.  相似文献

[RuCl2(p-cymene)]2 efficiently catalyzes the rearrangement of 2-aryl-2-(phenylthio)penta-3,4-dienyl 2,2,2-trichloroacetimidate to afford (Z)-2,2,2-trichloro-N-(4-aryl-1-(phenylthio)penta-2,4-dien-2-yl)acetamide. Ru carbene is assumed as the reactive intermediate in this rearrangement.  相似文献

Caulerpenyne ( 1 ), the most abundant of the ecotoxicologically relevant sesquiterpenoids of the Mediterranean-adapted tropical green seaweed Caulerpa taxifolia, was found to react with Et3N or pyridine in MeOH by initial deprotection of C(1)HO to give oxytoxin 1 ( 2a ), previously isolated from the sacoglossan mollusc Oxynoe olivacea. With BuNH2, without any precaution to exclude light, 1 gave the series of racemic 3 and 4 , and achiral (4E,6E)- 5 , (4E,6Z)- 5 , (4Z,6E)- 5 , and (4Z,6Z)- 5 pyrrole compounds, corresponding to formal C(4) substitution, 4,5-β-elimination, and (E/Z)-isomerization at the C(4)?C(5) and C(6)?C(7) bonds. Changing to CDCl3 as solvent in the dark, 1 gave cleanly, via 2a as an intermediate, 3 and (4E,6E)- 5 . The latter proved to be prone to (E/Z)-photoisomerization. Under standard acetylation conditions, 3 gave (4E,6E)- 5 via acetamide 7 as an intermediate. Particular notice is warranted by selective deprotection of 1 at C(1), mimicking enzyme reactions, and unprecedented formation of pyrrole compounds from freely-rotating, protected 1,4-dialdehyde systems.  相似文献

(1S,2S)-1-Aryl-2-tozylaminopropan-1-ols were synthesized by cyclization of 1-aryl-2-aminopropane-1,3-diol to aryl(1-tosylaziridin-2-yl)methanols, followed by hydride reduction of the latter. Reduction of the aza-Payne rearrangement products of intermediate aryl(1-tosylaziridin-2-yl)methanols gave (1S)-1-aryl-3-tosylaminopropan-1-ols. On triplet excitation (E)- 2 isomerizes to (Z)- 2 and reacts by cleavage of the C(γ), O-bond to isomeric δ-ketoester compounds ( 3 and 4 ) and 2,5-dihydrofuran compounds ( 5 and 19 , s. Scheme 1). - On singulet excitation (E)- 2 gives mainly isomers formed by cleavage of the C(γ), C(δ)-bond ( 6–14 , s. Scheme 1). However, the products 3–5 of the triplet induced cleavage of the C(γ), O-bond are obtained in small amounts, too. The conversion of (E)- 2 to an intermediate ketonium-ylide b (s. Scheme 5) is proven by the isolation of its cyclization product 13 and of the acetals 16 and 17 , the products of solvent addition to b . - Excitation (λ = 254 nm) of the enol ether (E/Z)- 6 yields the isomeric α, β-unsaturated ε-ketoesters (E/Z)- 8 and 9 , which undergo photodeconjugation to give the isomeric γ, δ-unsaturated ε-ketoesters (E/Z)- 10 . - On treatment with BF3O(C2H5)2 (E)- 2 isomerizes by cleavage of the C(δ), O-bond to the γ-ketoester (E)- 20 (s. Scheme 2). Conversion of (Z)- 2 with FeCl3 gives the isomeric furan compound 21 exclusively.  相似文献

Stereoselective syntheses on a gram scale of (3Z,6E,8E)-, (3Z,6E,8Z)-and (3Z,6Z,8Z)-3,6,8-dodecatrien-1-ol, 8, 9 and 10, respectively, are described. A key step of the synthesis of 8 consisted of a copper-mediated coupling reaction between 4-(2-tetrahydropyranyloxy)-1-butynylmagnesium bromide (15) and the mesyl ester of (2E,4E)-2,4-octadien-1-ol (14). A similar copper-mediated reaction between 15 and the mesyl ester of (E)-2-octen-4-yn-1-ol (19) was used to construct the C-12 carbon skeleton of 9. On the other hand, the synthesis of 10 was based on a palladium-promoted reaction between (Z)-1-bromo-1-pentene (23) and the organozinc bromide derived from 3,6-heptadiyn-1-yl acetate (27).  相似文献

Synthesis of Enantiomerically Pure Apoviolaxanthinoic Acids, Apoviolaxanthinols, and Apoviolaxanthinals (Including Persicaxanthin, Sinensiaxanthin, and β-Citraurin Epoxide) and of their Furanoid Rearrangement Products Starting from (1′S,2′R,4′S,2E,4E)-5-(1′,2′-epoxy-4′-hydroxy-2′,6′,6′-trimethylcyclohexy1)-3-methy1-2,4-pentadienal ( 3 ), a recently described synthon [6], a full range of C20-, C25-, C27-, and C30-polyenic acids, alcohols, and aldehydes and their (8R)- and (8S)-diastereoisomeric furanoid rearrangement products was prepared. The synthetic C25-alcohols proved to be identical with persicaxanthin (= 12′-apoviolaxanthin-12′-ol) and perisicachromes (= 12′-apoauroxanthin-12′-ols) and the C27-alcohols analogously with sinensiaxanthin and sinensiachromes. A correlation between the sign of the Cotton effects in the CD spectra of 5,6-and 5,8-epoxides and their configuration at C(6) and C(8), respectively, was established.  相似文献

In order to characterize the structural elements that might play a role in non-covalent DNA binding by the antitumor antibiotic leinamycin, we have solved the crystal structures of the two leinamycin analogs, methyl (R)-5-{2-[1-(tert-butoxy­carbonyl­amino)­ethyl]­thia­zol-4-yl}penta-(E,E)-2,4-dienoate, C16H22N2O4S, (II), and 2-methyl-8-oxa-16-thia-3,17-di­aza­bicyclo­[12.2.1]­heptadeca-(Z,E)-1(17),10,12,14-tetraene-4,9-di­one, C14H16N2O3S, (III). The penta-2,4-dienone moiety in both of these analogs adopts a conformation close to planarity, with the thia­zole ring twisted out of the plane by 12.9 (2)° in (II) and by 21.4 (4)° in (III).  相似文献

The Photoinduced Cleavage of Conjugated γ, δ-Epoxyenones: UV.-Irradiation of 5,6-Epoxy-3, 4-didehydro-5,6-dihydro-β-ionone On 1n, π*-excitation (λ ≥ 347 nm) in pentane or CClF2CFCl2 (E)- 1 is isomerized to the dihydrofurane (E/Z)- 2 as well to the ethers 3 and 5. Besides these products the isomeric cyclopropane derivative (E)- 4 and the acetal 6 are obtained in methanol. The detection of 6 indicates the formation of an intermediate ketoniumylide a which may give 6 by addition of methanol. ? On 1π, π*-excitation (λ=254 nm) in acetonitrile-d3, CClF2CFCl2 or pentane (E)- 1 yields almost exclusively (E)- 2. In methanol 6 is obtained in addition to (E/Z)- 2 , but no (E)- 4 and 5 is formed.  相似文献

A new germacranolide, ajanolide A, was isolated from aerial parts ofAjania fruticulosa by means of extraction with CHCl3 and adsorption chromatography. This compound was identified as (1(10)E,3S,4Z,6R,7S,11R)-3-acetoxygermacra-1(10),4-dien-12,6-olide ((1S,7S,10R,13R)-7-acetoxy-4,8,13-trimethyl-11-oxabicyclo[8.3.0]trideca-4(E),8(Z)-dien-12-one) by X-ray diffraction analysis. 2D1H−1H (COSY) and13C−1H (COSY) NMR spectroscopy was used for assigning the1H and13C NMR signals in the spectra of ajanolide A. The photoisomerization behavior of three mono[(E)-3-(4-hydroxyphenyl)prop-2-enoyl]spermidines, 1, 2 , and 3 , and three bis[(E)-3-(4-hydroxyphenyl)prop-2-enoyl]spermidines, 4, 5 , and 6 , are investigated. The synthetic product (E)- 1 could be almost quantitatively (> 96%) converted into its isomer (Z)- 1 under UV light irradiation. In the cases of (E)- 2 and (E)- 3 , a mixture of (E)/(Z) ca. 1:2 was obtained, when the same conditions were applied. The comparison of their UV spectra provides the possible explanation for these different behaviors. Furthermore, it was noticed that the (Z) → (E) isomerization of the C?C bond took place during the purification by reverse-phase high-performance liquid chromatography (RP-HPLC), and the (E)/(Z)-mixture is thus inseparable. The same feature could be observed during the isolation of the (Z,Z)-N,N′-bis[3-(4-hydroxyphenyl)prop-2-enoyl]-spermidines, (Z,Z)- 4 , (Z,Z)- 5 , and (Z,Z)- 6 . Nevertheless, the fractions of (Z,Z)- 5 and (Z,Z)- 6 were in almost pure state collected, and their 1-NMR spectra are presented.  相似文献

(E)-(1,2-Difluoro-1,2-ethenediyl)bis[tributylstannane], 3, readily undergoes a Pd(PPh3)4/CuI-catalyzed cross-coupling reaction with iodotrifluoroethene to yield (E)-octafluoro-1,3,5-hexatriene, 4, in high isomeric purity. (1Z,3E,5Z)-(1,2,3,4,5,6-Hexafluoro-1,3,5-hexenetriyl)bis[tributylstannane], 7, was sequentially prepared from (1Z,3E,5Z)-(1,2,3,4,5,6-hexafluoro-1,3,5-hexenetriyl)bis[triethylsilane], 5, which was prepared via a Pd(PPh3)4/CuI-catalyzed cross-coupling reaction of 3 with (E)-1,2-difluoro-1-iodo-2-triethylsilylethene, 6. Reduction of (E)-3-aryl-2-(4-arylthiazol-2-yl)acrylonitriles with lithium aluminum hydride in dry ether afforded (Z)-1-amino-3-aryl-2-(thiazol-2-yl)prop-1-ene derivatives in 15 to 40% yields. The structure of (Z)-1-amino-3-(2-chlorophenyl)-2-[4-(4-methylphenyl)thiazol-2-yl]prop-1-ene was confirmed by X-ray diffraction analysis. Dedicated to Academician N. K. 5-Arylfuran-2,3-diones react with (Z)-2-[3,3-dimethyl-3,4-dihydroisoquinolin-1(2H)-ylidene]acetamides to give (3E,5Z)-5-(2-aryl-2-oxoethylidene)-3-[3,3-dimethyl-3,4-dihydroisoquinolin-1(2H)-ylidene]-pyrrolidine-2,4-diones. A new skeleton bisabolane-type sesquiterpene curcuminoid,bisabocurcumin(1),along with 5 known compounds,curcumin(2), demethoxycurcumin(3),bidemethoxycurcumin(4),(1E,4E)-1,5-bis(4-hydroxy-3-methoxyphenyl)-penta-1,4-dien-3-one(5),and (1E,4E)-1-(4-hydroxy-3-methoxyphenyl)-5-(4-hydroxy phenyl-)-penta-1,4-dien-3-one(6)were isolated from the rhizomes of Curcuma longa L.Their structures were determined on the basis of spectroscopic analysis.Bisabocurcumin(1) is firstly obtained from nature with a new skeleton combined by a bisabolane-type sesquiterpene and a 1,7-diphenylheptanoid through a C-C bond.  相似文献

A similar hydrogenation of Z-5-arylcarbarnoyl)methylene-5,6-dihydro-3-phenyl-4H-1,2,4-oxadiazine ( 1d-f ) gives E-4-(arylcarbamoyl)methylene-2-phenyl-2-oxazoline ( 5 ), 4-(arylcarbamoyl)methyl-2-phenyloxazole ( 6 ), and Z-4-(aryl-carbamoyl)methylene-2-phenyl-2-oxazoline ( 7 ).  相似文献

Aroylketenes generated by thermolysis of 6-aryl-2,2-dimethyl-4H-1,3-dioxin-4-ones reacted with 1,3,3-trimethyl-2-methylidene-1,3-dihydro-2H-indole (Fischer’s base) to produce (1E,3Z)-4-aryl-4-hydroxy-1-(1,3,3-trimethyl-1,3-dihydro-2H-indol-2-ylidene)but-3-en-2-ones. The crystalline and molecular structures of (1E,3Z)-4-(4-chlorophenyl)-4-hydroxy-1-(1,3,3-trimethyl-2,3-dihydro-1H-indol-2-ylidene)but-3-en-2-one were determined by X-ray analysis.  相似文献
