抗癌性吲哚喹唑啉衍生物的定量构效关系   总被引:10,自引:0,他引:10  
用量子化学密度泛函理论(DFT)、分子力学(MM+)及回归分析方法,对一系列抗癌性吲哚喹唑啉衍生物进行了定量构效关系(QSAR)的研究.通过回归分析,筛选了影响抗癌活性的主要因素,建立了定量构效关系方程.结果表明,化合物的最低未占据分子轨道(LUMO)与最高占据分子轨道(HOMO)之间的能量差(ΔεL-H)、分子的疏水性(lgP)以及环D上的总电荷(ΣQD)和环D上R1取代基的第一个原子的净电荷(QFR1)是影响化合物抗癌活性的主要因素.所得模型对化合物抗癌活性有较好的预测效果. 同时, 与ΔεL-H密切相关的LUMO轨道能量及共轭平面面积对药物的DNA-结合及其活性起着十分重要的作用,可通过选取具有较强的拉电子性质同时又能与本系列化合物的骨架形成更大共轭体系的取代基R1,设计抗癌活性较高的化合物.  相似文献

Indolo[1,2-b]quinazoline derivatives have recently been reported as a type of compound with potential anticancer activity. On the basis of our the published two-dimensional quantitative structure-activity relationship (2D-QSAR) of these compounds, a further study on the three-dimensional quantitative structure-activity relationship (3D-QSAR) was carried out using the method of comparative molecular field analysis (CoMFA). A reasonable, receivable, and an effective 3D-QSAR model has been established, in which the correlation coefficient (r2) and cross-validation coefficient (q2) values are 0.986 and 0.695, respectively, the statistical squared deviation ratio (F) is 114.6, and the standard deviation (SD) is 0.084. The results suggest that the electrostatic effect of substituent R1 and steric effect of substituent R2 play a very important role in the improvement of the anticancer activity of these compounds. In this article, some significant conclusions, which are in good agreement with the conclusions obtained using 2D-QSAR, were drawn as follows. (1) The electrostatic effect in the substituent R1 part plays a major role, and it is very important to make the first atom of R1 carrying more positive charges in order to improve the anticancer activity of the compounds. (2) The steric effect plays a major role in the substituent R2 part, and the volume of R2 should be moderate. Based on the above conclusions, three new molecules of Indolo[1,2-b]quinazoline derivatives with higher anticancer activity have been theoretically designed and are waiting for support from experiment. The QSAR results can offer a theoretical reference for the pharmaceutical synthesis.  相似文献

The synthesis and antimicrobial activity of 2,4-thiazolidinedione derivatives 5a–g and 6a–g are described. The structures of the newly synthesized compounds were confirmed by IR, NMR, mass and elemental analyses. All compounds were evaluated for their preliminary in vitro antibacterial and antifungal activity. The results revealed that most of the compounds showed high or moderate biological activity against tested microorganisms.  相似文献

吲哚喹唑啉衍生物是近年来发现的一类具有良好抗癌活性的化合物. 作者在最近报道的二维定量构效关系(2D-QSAR)的基础上, 采用比较分子力场方法(CoMFA)进一步对该系列化合物进行三维定量构效关系(3D-QSAR)研究, 建立了3D-QSAR的CoMFA模型, 其非交叉验证相关系数r2=0.986, 标准偏差SD=0.084, 统计方差比F=114.6, 交叉验证相关系数q2=0.695, 表明该模型合理、可信, 并具有良好的预测能力. 研究结果表明: (1) 取代基R1的部位上静电效应起主要作用, 并且确保取代基R1的第一个原子具有较大的净正电荷, 对提高化合物的抗癌活性十分重要. 这与2D-QSAR研究结果相一致. (2) 取代基R2的部位上立体效应起主要作用, R2的体积大小要适中. 应用这些规律进行了分子设计, 在理论上获得了一些具有较高抗癌活性的新的吲哚喹唑啉衍生物, 并期待实验证实. 该QSAR的研究结果可为实验工作者合成新药提供理论参考.  相似文献

This work focuses the light on some remarkable achievements in clean and efficient green experimental synthesis, characterization and evaluation of the pharmaceutical and biochemical importance of new series of pyrazolo[1,2-b]phthalazinediones which synthesized through one-pot three-component condensation reaction of the appropriate of 1,2,3-triazolyl-pyrazole-carbaldehydes with active methylene compounds (as malononitriles or ethyl cyanoacetate) and 6-nitrophthalhydrazide using the grinding method in the presence of sodium hydroxide under solvent-free condition at room temperature, in very excellent yields. All the newly synthesized compounds were characterized by physical and chemical tools (FT-IR, 1H NMR and mass spectrometry). In addition, all the new synthesized derivatives were screened for their anticancer activity against hepatic cancer cell lines to evaluate their pharmaceutical importance.  相似文献

In our attempt towards the synthesis and development of effective antimicrobial, anticancer and antioxidant agents, a novel series of 2,3-dihydropyrido[2,3-d]pyrimidin-4-one 7a–e and pyrrolo[2,1-b][1,3]benzothiazoles 9a–e were synthesized. The synthesis of 2-(1,3-benzo thiazol-2-yl)-3-(aryl)prop-2-enenitrile (5a–e) as the key intermediate was accomplished by a microwave efficient method. Via a new variety oriented synthetic microwave pathway, these highly functionalized building blocks allowed access to numerous fused heteroaromatic such as 7-amino-6-(1,3-benzo thiazol-2-yl)-5-(aryl)-2-thioxo-2,3dihydropyrido [2,3-d]pyrimidin-4(1H)-one 7a–e and 1-amino-2-(aryl)pyrrolo[2,1-b][1,3]benzothiazole-3-carbonitrile derivatives 9a–e in order to study their antimicrobial and anticancer activity. The present investigation offers effective and rapid new procedures for the synthesis of the newly polycondensed heterocyclic ring systems. All the newly synthesized compounds were evaluated for antimicrobial, anticancer and antioxidant activity. Compounds 7a,d, and 9a,d showed higher antimicrobial activity than cefotaxime and fluconazole while the remaining compounds exhibited good to moderate activity against bacteria and fungi. An anticancer evaluation of the newly synthesized compounds against the three tumor cell lines (lung cell NCI-H460, liver cancer HepG2 and colon cancer HCT-116) exhibited that compounds 7a, d, and 9a,d have higher cytotoxicity against the three human cell lines compared to doxorubicin as a reference drug. These compounds also exhibited higher antioxidant activity and a great ability to protect DNA from damage induced by bleomycin.  相似文献

The synthesis has been developed for novel potential anticancer agents whose structures contain an aziridine amidoxime group. The cytotoxic activity of all of the target compounds on different cell lines are given together with the in vitro IC50 and LD50 values found. The most promising anticancer agents were found to be the methyl 4-[1-aziridinyl(hydroxyimino)methyl]benzoate, 1-aziridinyl(2-naphthyl)-methanone oxime, and 1-aziridinyl(2-quinolyl)methanone oxime. Summary Sodium 2-aminothiophenoxide (1) reacts with ethyl 2-bromoalkanoates (2) under direct cyclization to form 2-alkyl-2H-1,4-benzothiazin-3(4H)-ones (3). Reaction of the sodium salt of 2-mercaptobenzimidazole (4) with2 or 2-bromoalkanoic acids (5) affords only S-alkylated products (6 or7, respectively). The cyclization products — 2-alkylbenzo[d]imidazolo[2,1-b]thiazolidin-3-ones (8) — can be obtained only from the corresponding 2-(2-benzimidazolylthio)alkanoic acids (7) by the action of acetic anhydride. Both compounds3 and8 exhibit only moderate antimicrobial activity against some gram-positive bacteria.
Synthese und antimikrobielle Wirkung von einigen 2-Alkyl-2H-1,4-benzothiazin-3(4H)-onen und 2-Alkylbenzo[d]imidazolo[2,1-b]thiazolidin-3-onen
Zusammenfassung Bei der Reaktion von Natrium-2-aminothiophenolat mit 2-Bromoalkansäure-ethylestern (2) entstehen als Cyclisierungsprodukte 2-Alkyl-2H-1,4-benzothiazin-3(4H)-one (3). Die Umsetzung von Natriumbenzimidazol-2-thiolat mit2 oder mit 2-Bromoalkansäuren (5) liefert nur S-Alkylierungsprodukte (6 oder7). Die Cyclisierungsprodukte — 2-Alkylbenzo[d]imidazolo[2,1-b]thiazolidin-3-one (8) — sind nur durch Umsetzung von entsprechenden 2-(2-Benzimidazolylthio)-alkansäuren (7) mit Acetanhydrid erhältlich. Die Verbindungen3 und8 weisen nur mäßige antimikrobielle Wirkung gegen einige gram-positive Bakterien aus.

An efficient and green method has been developed for the synthesis of 2H-indazolo[2,1-b]phthalazinetriones derivatives by employing 15 mol%β-cyclodextrinvia a one-pot multicomponent reaction of aldehyde,dimedone,hydrazine hydrate with succinic anhydride/phthalic anhydride in water at 80 ℃ for first time.The catalyst could be recovered and reused for four consecutive cycles without appreciable loss in catalytic activity and evaluated for in vitro antimicrobial activity against different Gram-positive and Gram-negative bacterial strains.The outcome of the screening study showed that compound 6d,6f and7 n exhibited excellent activity against E.coil.Whereas,compound 6f and 6h exhibited excellent activity against P.aeurginosa,and compound 6c,and 6e displayed again excellent activity against Staphylococcus aureus whereas compound 7o shows excellent activity against S.aureus and B.subtilis when compared with Ampicillin(standard control).The results indicated that maximum compounds are moderately effective against bacterial growth and their effectiveness is highest against standard drugs.  相似文献

5,10,15,20‐Tetrakis[4‐(triorganostannyloxy)phenyl]porphyrins, (R3SnO)4TPP [2, R = Cy (a), Ph (b), PhC(CH3)2CH2 (c)], have been synthesized by the condensation of 4‐(triorganostannyloxy)benzaldehyde, 4‐(R3SnO)C6H4CHO (1), with pyrrole in the presence of BF3 followed by oxidation by p‐chloranil and characterized by means of elemental analysis, IR, UV–visible and NMR (1H, 13C and 119Sn) spectra. The results of X‐ray single‐crystal diffraction show that 1a and 1b possess a trans‐C3SnO2 trigonal bipyramidal geometry with the axial positions occupied by the phenolate oxygen and formyl group oxygen of an adjacent molecule and form a one‐dimensional zigzag chain. In 2a, the macrocyclic core of the porphyrin is coplanar and each tin atom possesses a distorted tetrahedral geometry. These compounds (1 and 2) have potent in vitro cytotoxic activity against two human tumor cell lines – CoLo205 and MCF‐7 – and the activity decreases in the order Ph > Cy > PhC(CH3)2CH2 for the R group bound to tin. A series of novel triazolothione, thiadiazole, triazole-functionalized furo/thieno[2,3-b]pyridine derivatives 9a–l, respectively, were prepared starting from 2 (1H) pyridone 3 through selective O/S-alkylation followed by Thorpe–Ziegler cyclization to form furo/thieno[2,3-b]pyridine derivatives 6. Compounds 6 on reaction with hydrazine hydrate resulted carbohydrazide derivatives 7 and further reacted with diverse substituted phenyl isothiocyanates to form phenyl hydrazine carbothiamide derivatives 8. Each compound 8 is independently reacted in presence of NaOH, H2SO4, and N2H4.H2O to form triazolothione, thiadiazole, triazole-functionalized furo/thieno[2,3-b]pyridine derivatives 9a–l, respectively. All the products 9a–l were screened against Gram +ve, Gram –ve bacteria and fungal strains. Compounds 9c–h showed high activity against Bacillus subtilis microbial-type culture collection (MTCC) 121 at <8.0 micromolar concentration. Promising compounds further screened for minimum bactericidal concentration against B. subtilis MTCC 121 using ciprofloxacin as standard and found to show very good activity. These compounds also screened for biofilm inhibition activity against B. subtilis MTCC 121 using erythromycin as standard and confirmed the high activity.  相似文献

The docking study on a series of furo[1]benzofuran derivatives with ERα has been demonstrated. The synthesis and characterization of a series of furo[1]benzofuran derivatives were described. All the target compounds were conducted to in vitro for the inhibitory activities against human breast cancer strains T-47D, MCF-7 and toxicity against human liver normal cell strains HL7702 via MTT assay. Most of the target compounds possessed anti-estrogen receptor-dependent breast cancer activities with weak toxicity to healthy cell strains. The preliminary structure–activity relationships were discussed.  相似文献

A new series of vitamin E/selenated pyridine, vitamin E/selenated pyridazine, vitamin E/selenated coumarine and vitamin E/selenated nicotine moieties were synthesized and their cytotoxic activity is investigated using the human breast cancer cell line. The newly synthesized compounds were characterized using spectroscopic tools (IR, 1H NMR, 13C NMR, and mass spectroscopy) as well as microanalysis. Our study reveals that compound vitamin E/selenated nicotine moiety has the highest cytotoxic effect than the other synthesized compounds.  相似文献
