荧光光谱法研究葫芦[7]脲与6-巯嘌呤和腺嘌呤的包结作用   总被引:1,自引:0,他引:1  
采用荧光光谱法分别研究了葫芦[7]脲(CB[7])对6-巯嘌呤(6-MP)和腺嘌呤(ADP)的包结作用。实验考察了时间、pH值以及温度对荧光强度和包结作用的影响,利用Benesi-Hildebrand方程分别计算出6-MP和ADP与CB[7]的包结常数。结果表明:酸度对体系的包结有明显的影响。在pH值为8.0和2.0左右时,6-MP和ADP分别具有稳定和最佳激发和发射波长,随着CB[7]浓度的增大,体系的荧光强度都有明显增强,包结作用迅速(小于5 min)。实验得出CB[7]与6-MP和ADP的包结比均为1∶1,在298 K时的包结常数分别为3.6797×102L·mol-1和2.2033×102L·mol-1。通过热力学参数的变化,探讨了维系包结物稳定性的主要作用力。CB[7]是葫芦脲家族中水溶性最强的主体分子,作为一种安全低毒的药物载体极具潜力。  相似文献

利用1H NMR以及荧光技术研究了六、七、八元瓜环与2-苯基喹啉、N-正丙基溴化异喹啉、3-氨基喹啉及7,8-苯并喹啉的相互作用.两种方法的考察结果均表明,2-苯基喹啉能与这3种瓜环发生相互作用,其中六、七元瓜环与2-苯基喹啉形成1∶1的稳定包结配合物,包结常数分别为1.6×104和3.2×103L/mol.八元瓜环能与2-苯基喹啉形成1∶2包结物.1H NMR结果还表明,3种瓜环均能与N-正丙基溴化异喹啉相互作用,其化学计量比均为1∶2;七元瓜环与7,8-苯并喹啉相互作用,化学计量比约为1∶1.荧光法也表明八元瓜环能与N-正丙基溴化异喹啉、3-氨基喹啉及7,8-苯并喹啉发生相互作用,并且荧光强度随瓜环浓度增加而下降,其化学计量比为1∶2.同时,讨论了上述主客体包结配合物的作用模式.  相似文献

Molecular dynamics (MD) simulations were carried out to study the host–guest complexation in aqueous solution between cucurbit[7]uril (CB7) and the neutral and protonated forms of benzimidazole derivatives. Complexation occurs via encapsulation of the hydrophobic part (benzene ring) of the guest within the CB7 hydrophobic cavity, and the interactions of the amine group(s) of the imidazole ring of the guest with the CB7 carbonyl portals. The molecular mechanics Poisson–Boltzmann surface area (MM-PBSA) method is used to estimate the host–guest Gibbs energy of binding. The results indicate that CB7 binds the protonated form more strongly than the neutral one, and that the dominant contribution to the Gibbs energy of complexation for the neutral and protonated guests is associated, respectively, with the host–guest van der Waals and electrostatic interactions. Quantum chemical calculations using dispersion-corrected density functional theory (DFT) are used to calculate the binding affinities and to predict the pKa values of the free and complexed guests. The calculated pKa values for the free guests reveal excellent agreement with the experimental values, while for the complexed guests, general trends are obtained.  相似文献

利用密度泛函理论对葫芦脲[7]和核黄素包结物的可能结构进行了理论计算.从5种可能结构分析主客体在包结物中的作用得出:核黄素作为客体分子与主体葫芦脲[7]分子通过不同官能团作用可形成稳定性不同的包结物,其中以核糖醇的羟基插入到葫芦脲[7]分子的腔体与其端羰基氧原子形成较多的氢键最稳定.在理论上证明了此包结反应为一放热反应的实验事实,并进一步说明了包结物分子的相对稳定性受主客体分子的几何畸变能和主客体之间作用能的影响,说明了成键能和稳定性之间的区别.在此基础上利用时间相关的密度泛函理论进行了不同构型包结物的激发态计算并对光谱性质进行了理论预测.比较了包结物中核黄素分子的跃迁轨道的组成变化,分析得出:葫芦脲[7]的包结作用改变了核黄素分子的电子跃迁性质,使其吸收光谱发生红移.并预测出其荧光猝灭的微观机理,此理论计算结果很好地解释了相应的实验现象.  相似文献

利用核磁共振(NMR)技术研究了瓜环[n](n=7,8)对外消旋盐酸哌甲酯的手性识别。研究结果显示,盐酸哌甲酯对映异构体与瓜环形成包合物后,其质子的化学位移移动方向和幅度不同。同时,手性碳附近的某些原子所对应的吸收峰裂分为两组信号,这两组信号分别对应盐酸哌甲酯的两个对映体。说明两个对映体与瓜环形成了作用模式不同的超分子包结物,从而使对映体能够区分开来,即非手性的瓜环将手性盐酸哌甲酯区别开来,实现了瓜环对苏式哌甲酯两个对映体的快速手性识别。  相似文献

The structures and optical properties of host–guest complexes produced from cucurbit[n = 6–8]urils and some benzimidazole derivatives have been investigated by 1H NMR spectroscopy, electronic absorption spectroscopy and fluorescence spectroscopy. The experimental results reveal that calculations of A∼NQ[n]/Nguest and If∼NQ[n]/Nguest for the same association complex both support a good fit to an identical binding model. In particular, the A∼NQ[n]/Nguest, If∼NQ[n]/Nguest calculations and the 1H NMR determinations for three Q[6]–ge(1∼3) complexes and three Q[8]–ge(1∼3) complexes all support a binding model of 1:1 and 1:2 respectively.  相似文献

Using bis-cyanopyridyl alkane compounds as guests, three cucurbit[6]uril-based new pseudorotaxanes (2a, 2b and 2c) were synthesised and characterised. The crystal structures for 2a and 2c have been determined and discussed. 本文采用核磁共振技术研究瓜环与哌嗪衍生物主客体配合物的结构特征。1H核磁共振谱证实八元瓜环能与一些哌嗪衍生物形成稳定的双客体主客体配合物,六元瓜环与一些哌嗪衍生物形成稳定的1∶1主客体配合物。而五、七元瓜环和哌嗪衍生物则未表现出明显地相互作用。形成的自组装主客体配合物得到质谱的进一步证实。  相似文献

Alkyl amines are able to form complexes with either crown ethers or cyclodextrins or cucurbit[6]uril. The same is known for polyamines such as spermidine and spermine. However, the simultaneous formation of such polyamines with crown ethers and cucurbit[6]uril has not been studied. The ability of polyamines such as spermidine and spermine to form mixed complexes with different ligands, e.g. crown ethers and cucurbit[6]uril has been studied in aqueous solution using pH-metric and calorimetric titrations. The thermodynamic data of reaction between crown ethers with spermidine, spermine and their cucurbit[6]uril complexes have been determined. The presence of cucurbit[6]uril on the polyamines has no important influence upon the reaction of these amines with crown ethers. The reactions between polyamines, cucurbit[6]uril and crown ethers are simple examples for the self organization of molecules due to specific interactions. 利用紫外吸收光谱、荧光光谱以及1H NMR方法详细考察了六、七、八元瓜环(Q[6], Q[7], Q[8])与苯胺系列衍生物客体的相互作用和体系pH对其作用的影响. 实验结果表明, 3种瓜环与苯胺系列衍生物客体的相互作用强弱、作用比例以及作用模式与体系的酸度密切相关: 在“高”或“低”pH条件下, 未观察到瓜环与这些客体的明显作用; 在介于“高”与“较高”或“低”与“较低”的pH范围, 瓜环与这些客体发生相互作用, 形成1∶1的包结配合物; 而在介于“较高”与“较低”的pH范围, 瓜环与这些客体发生相互作用, 可形成1∶2的包结配合物. 对于不同的瓜环-客体作用体系, 相应的pH范围各不相同. 本文利用简便的实验方法, 测试了这些pH值及其范围. 根据测定的结果, 结合瓜环以及客体的结构特征, 对体系主客体在不同的酸度区域表现出的不同作用模式进行了探讨.  相似文献

Summary The reaction of 3,4-methylenedioxycinnamic acid (1) with thionyl chloride resulted in the formation of 7-chlorothieno[2,3-f]-1,3-benzodioxole-6-carbonyl chloride (2) and cinnamoyl chloride (3). Subsequent reaction of the former withp-substituted anilines led to the formation of 7-chloro-N-(p-substituted phenyl)-thieno[2,3-f]-1,3-benzodioxole-6-carboxamides (4a–c) which on photocyclization afforded 2-substituted [1,3]dioxolo[5,6][1]benzothieno[2,3-c]quinolin-6(5H)-ones (5a–c) in fairly good yields and high purity. 用核磁共振氢谱和紫外-可见光谱滴定法考察了葫[n]环联脲(Cucurbit[n]uril,n=5,6,7,8)与对甲苯重氮氟硼酸盐和4,4′-联苯二重氮氟硼酸盐的配位情况,并用曲线拟合求得形成的包结配合物的稳定常数.结果表明,不同空腔的葫[n]环联脲对不同尺寸的重氮氟硼酸盐具有很显著的选择性包结作用.在相同条件下,与葫[6]环联脲相比,葫[7]环联脲更易于容纳苯环.同时,随着酸性的增强,葫[n]环联脲上的脲羰基质子化程度加大,使得其配位能力有所减弱.  相似文献

The reaction of 1,2-diamino-4,5-diphenylimidazole with 1,3-diarylpropenones and 1,3-diaryl-2,3-dibromopropenones gives dihydro- and heteroaromatic derivatives of 6-hydroxyimidazopyrimidines and also imidazopyrimidines which do not contain the 6-hydroxy group. The intramolecular hydrogen bond between the phenolic hydroxyl and the pyrimidine nitrogen atom in the title compounds exerts a destabilizing effect on the tetrazole ring and shifts the azide-tetrazole equilibrium toward the azide form, especially in the case of tetrazolo[c]pyrimidine and -[c]quinazoline.
