Five α-pyrone meroterpenoids, including one new 3-epiarigsugacin E (1) and four known compounds, arisugacin D (2), arisugacin B (3), territrem C (4) and terreulactone C (5) were obtained from the marine fungus Penicillium sp. SK5GW1L. Their structures were identified by MS and NMR experiments, and the absolute configuration of compound 1 was further confirmed by low temperature (150 K) single crystal X-ray diffraction with Cu Kα radiation. Compounds 3, 4 and 5 showed strong inhibitory activities against acetylcholinesterase (AchE) with IC50 values of 3.03, 0.23 and 0.028 μM, respectively.  相似文献

Abstract

To discover novel structural compounds which are producted by endophytic fungi, a primary chemical profiling of Camptotheca acuminata Decne derived endophytic fungus Penicillum polonicum had been taken. Two new compounds β-lactone polonicin A (1) and enoic acid polonicin B (2) together with seven known compounds 3-9 were isolated from Penicillum polonicum obtained from C. acuminata. The structures of the new compounds 1 and 2 were identified by modern spectrum technology including detailed 1D, 2D NMR and MS data analyses. When tested against HepG2 hepatocellular carcinoma (HCC) cell lines, compounds 4-8 showed moderate anti-HCC activity. In addition, compound 1-3 have effects on increasing GLUT4 translocation and glucose uptake in vitro. Compound 1 showed the strongest glucose uptake and GLUT4 translocation activities in rat skeleton (L6) myoblast cell line with enhancements of 1.8 and 2.1 folds respectively compared to the control.  相似文献

Three new caryophyllene sesquiterpenes, pestalotiopsins I–K, one new chromone, pseudopestalone, and one new 10-membered macrolide, decarestrictine Q, were isolated from the fermentation broth of the marine-derived fungus Pseudopestalotiopsis sp. PSU-AMF45 along with twelve known compounds. Their structures were identified by spectroscopic data including 1D and 2D NMR. The absolute configuration of pestalotiopsin I was assigned based on single-crystal X-ray diffraction crystallography whereas that of the secondary alcohol moiety in pestalotiopsin J was established by Mosher's method. For pseudopestalone, the absolute configuration was determined by ECD spectrum associated with TD-DFT calculation. The antimicrobial activity of the isolated compounds was evaluated.  相似文献

A new polyoxygenated dihydropyrano[2,3-c]pyrrole-4,5-dione derivative, pyranonigrin F (1), together with a related known compound, pyranonigrin A (2), were isolated and identified from Penicillium brocae MA-231, an endophytic fungus obtained from the fresh tissue of the marine mangrove plant Avicennia marina. The structures of these metabolites were determined based on comprehensive spectral interpretation and the absolute configuration of compound 1 was established by X-ray crystallographic analysis. Compounds 1, and 2 showed potent activity against a broad spectrum of human-, aqua-, and plant-pathogens.  相似文献

Cultivation of the fungus Penicillium spp. HS‐3, an endophytic fungus from the stems of Huperzia serrata, led to the isolation of a new diketopiperazine alkaloid, named as tryhistatin ( 1 ), together with three known ones, 2 – 4 , all of which share a similar backbone. The structure of 1 was established by detailed interpretation of the 1D‐ and 2D‐NMR spectra, and HR‐ESI‐MS data. Furthermore, 1D‐ and 2D‐NMR experiments were employed to elucidate the structure of 2 which was deduced previously only on the basis of the LC/MS method. In this article, we confirmed the correctness of the previously reported structure and made a complete assignment of the NMR signals of 2 for the first time.  相似文献

Two new polyketides, penicilloxalones A (1) and B (2), together with 13 known compounds (315), were isolated from the ethyl acetate extract of the solid substrate fermentation cultures of the fungus Penicillium oxalicum MHZ153. The structures of the isolates were determined by spectroscopic analysis and comparison of their spectroscopic and physicochemical data with the literature values. Compounds 7 and 11 showed inhibition of nitric oxide production in lipopolysaccharide-stimulated BV-2 microglial cells with IC50 values of 0.9 ± 0.2 μM and 87.9 ± 0.7 μM, respectively.  相似文献

Three novel carbon-bridged citrinin dimers, dicitrinones A-C (1-3), were isolated from a volcano ash-derived fungus Penicillium citrinum. Their structures were elucidated by spectroscopic methods. Dicitrinone A (1) and B (2) are two atropisomeric mixtures, which were verified by both NMR and quantum chemical calculations. Their cytotoxicity against four tumor cell lines (HL-60, MOLT-4, A-549, and BEL-7402) was evaluated by SRB and MTT methods. Dicitrinone B (2) arrested the HL-60 cell cycle at the G2/M phase, which was analyzed using flow cytometry.  相似文献

Abstract

The pigment was extracted from Penicillium aculeatum, purified and characterized as Ankaflavin by spectroscopic analysis. The stability of the pigment was determined under various conditions and was found to possess high stability. The cytotoxicity property of the purified pigment was determined by MTT assay in MCF-7, HCT116 and PC-3 and the studies were compared with its activity in CHOK1 cells. In MCF-7 and in CHOK 1 cells, the pigment exhibited very less toxicity. However, significant cytotoxicity was observed in HCT116 and PC-3 cells with IC50 of 162?μg mL?1 and 85?μg mL?1 for HCT116 and PC-3 cells respectively. In vitro toxicity was tested by haemolysis assay and MTT assay in HEK 293 cells. The pigment showed least cytotoxicity (<5%) at 160 and 320?μg mL?1 concentrations HEK 293 cells and negligible (<5%) toxicity on human erythrocytes at 160 and 320?μg mL?1, the highest concentrations tested.  相似文献

A new compound 6-formamide-chetomin (1) together with chetomin (2) was isolated from solid fermentation products of the endophytic fungus Chaetomium sp. M336, which were elucidated by extensive spectroscopic analyses, including 1D and 2D NMR, and HR-ESI-MS experiments. The bioassay result showed that compound 1 had strong antibacterial activity against Escherichia coli, Staphylococcus aureus, Salmonella typhimurium ATCC 6539 and Enterococcus faecalis with minimum inhibitory concentration (MIC) value of 0.78 μg/mL; meanwhile it exhibited strong cytotoxicity with IC50 values of 21.6–27.1 nM against cell lines HeLa, SGC-7901 and A549.  相似文献

Abstract

Mycophenolic acid (MPA) is a group of metabolite derived from several species of Penicillium, which shows potent bioactivity. In this study, a new derivative of MPA compound named penicacid D (1), was isolated from the marine derived fungus Penicillium sp. SCSIO sof101, along with seven known compounds (2-8). Their structures were elucidated based on the HR-ESI-MS and NMR data. Moreover, the 1H and 13C NMR data of compound 2 and the 13C NMR data of compound 3 are reported. Compounds 1, 4 and 6 exhibited weak activities against Escherichia coli (clinical isolation number 100385570) and Acinetobacter baumannii (clinical isolation number 100069).  相似文献

Two new butyrolactone derivatives, terrelactones A and B (1 and 2), together with four known butyrolactone derivatives (36), were isolated from the fermentation products of the fungus Penicillium oxalicum 0403. Their structures were elucidated by spectroscopic methods, including extensive 1D and 2D NMR techniques. The anti-tobacco mosaic virus (anti-TMV) activities of 1 and 2 were evaluated. The results showed that compounds 1 and 2 exhibited high anti-TMV activity with inhibition rates of 25.5% and 27.4%, respectively. A new polyene compound (1) and a new diketopiperazine (2), as well as three known compounds (35), were isolated from the Antarctic marine-derived fungus Penicillium crustosum HDN153086. The structures of 15 were deduced based on MS, NMR and TD-DFT calculations of specific ECD spectra. These compounds were evaluated for their cytotoxic activities against K562 cell line and only compound 2 exhibited cytotoxicity against K562 cell, with IC50 value of 12.7 μM.  相似文献

Chemical epigenetic manipulation was applied to explore secondary metabolite of an endophytic fungus Penicillium herquei, which was obtained from the fruiting body of Cordyceps sinensis, and three previously undescribed polyketides with pyran-2-one scaffold were isolated from its fermentation broth containing 10?mg/L 5-aza-2-deoxycytidine (a frequently-used DNA methyltransferase inhibitor). A novel flavone, penimethavone A (1), possessing a rare unique methyl group at ring-B, was isolated from the fungus Penicillium chrysogenum cultured from a gorgonian Carijoa sp. collected from the South China Sea. The structure was elucidated by extensive spectroscopic analysis and by comparison with related known compound. One new benzopyran derivative (2R*,4R*)-3,4-dihydro-5-methoxy-2-methyl-2H-1-benzopyran-4-ol (1), together with five known compounds (2?6), were obtained from the EtOAc extract of the endophytic fungus Penicillium citrinum HL-5126 isolated from the mangrove Brguiera sexangula var. rhynchopetala collected in the South China Sea. Their structures were elucidated by the detailed analysis of comprehensive spectroscopic data. All compounds were evaluated for their antibacterial activities. Compound 6 exhibited potent inhibitory activity against Bacillus subtilis, Bacillus cereus and Micrococcus tetragenus with the same MIC values of 6.94 μM.  相似文献

Seven polyketides, including two new isochromanes, penisochromanes A and B (1 and 2), as well as five known compounds were obtained from an ascidian-derived fungus Penicillium sp. 4829. Their structures were identified by extensive spectroscopic analyses. The structures of compounds 1 and 3 were further determined by the X-ray crystallography. Compounds 1 and 2 were the first example of isochromane with three adjacent oxy substituents in natural source. Compound 4 exhibited selective activities against two Gram-positive bacteria Staphylococcus aureus and Bacillus subtilis, with MIC values 6.25 and 12.5 μM, respectively.  相似文献
