FtsZ is the key protein in cell division in bacteria. We have proposed that lipid domains in the cytoplasmic membrane play a role in the localisation of FtsZ. In order to test this hypothesis, we used a model system based on Langmuir films to simulate the bacterial membrane. In this simple system we used a single phospholipid, dipalmytoylphosphatidylethanolamine, which is the major constituent of the inner membrane in Escherichia coli. The first results show clearly the importance of the GTP-controlled assembly process in the appearance of circular or fibrillar structures.  相似文献

设计合成了含偶氮苯基团的糖脂Gal-azo-Cn,通过Langmuir Blodgett(LB)膜、原子力显微镜技术对该糖脂在气液界面上的光异构化行为进行研究。实验表明,气液界面上的纯偶氮苯糖脂及与磷脂混合形成单分子膜的偶氮苯糖脂均具有反-顺、顺-反异构化行为。紫外光引起的异构化过程导致表面压力上升,但是由于二维界面上膜压的存在对其反-顺异构化行为有抑制作用,膜压越大,压力上升越小,黑暗环境中顺-反异构化过程则越快。对比纯偶氮苯糖脂的界面异构化行为,混合体系中磷脂分子与偶氮苯糖脂分子间的相互作用,对二维界面上糖脂的反-顺异构化有促进作用,提高了偶氮苯糖脂的反-顺异构化程度。磷脂-糖脂混合体系中,混合组分中含不饱和碳链的磷脂含量越高,膜的流动性越强,稳定性越差,对偶氮苯糖脂异构化行为的促进作用越弱。  相似文献

Dipalmitoylphosphatidylethanolamine (DDPE) Langmuir films at the air/water interface have been studied. These films exhibit high stability. The resulting films transferred on muscovite have been studied by scanning force microscopy with the contact mode. At the microscopic scale, DDPE Langmuir-Blodgett films appear densely packed with few defects. A hydrotalcite clay/dendrimer nanocomposite prepared by the ionic exchange process was adsorbed from suspension of the nanocomposite on a Langmuir monolayer of arachidic acid at the air/water interface, followed by compressing and transferring onto an arachidic acid monolayer Langmuir-Blodgett (LB) film on mica. For comparison, the hydrotalcite-adsorbed hybrid film was also prepared. The morphology of hydrotalcite and the nanocomposite studied by transmission electron microscopy indicated the layered structures with respectively 1.2 +/- 0.3 and 3.2 +/- 0.5 nm repeating distances. The hybrid Langmuir films displayed the occupied surface area of 0.24 nm2 for both hydrotalcite and the nanocomposite. The formation of hybrid Langmuir films was confirmed by Brewster angle microscopy. Atomic force microscopic images of hybrid LB films revealed the formation of plateau domains with the height difference of 6 nm for hydrotalcite and 12 nm for the nanocomposite and the presence of dendrimers adsorbed on the clay surface of the nanocomposite.  相似文献

Dielectric properties of E. coli cells before and after Cu2+ incubation were investigated by using the dielectric spectroscopic technique. The dielectric spectra are analyzed theoretically by means of the extended three-shell ellipsoidal model, which can reflect the complicated morphological structure of E. coli cell including the outer membrane, the periplasmic space, the inner membrane and the cytoplasm. The results showed that dielectric properties of these cellular components were changed with Cu2+ treatment in a time- and concentration-dependent way. The permttivity of the outer membrane increased with the incubation time and concentration of Cu2+, possibly because polarizability of the outer leaflet of lipopolysaccharides was affected by Cu2+. The conductivity of the periplasmic space decreased with the incubation time and concentration of Cu2+, possibly due to the damage of peptidoglycan. The decreased permittivity of the inner membrane may be caused by disturbance of the lipid bilayer structure produced by Cu2+ incubation. The decreased cytoplasmic conductivity may be the consequence of the leakage of K+ from it. The cytoplasmic permittivity decreased with Cu2+ treatment probably because of the leakage of its some components.  相似文献

E. coli has two-component systems composed of histidine kinase proteins and response regulator proteins. For a given extracellular stimulus, a histidine kinase senses the stimulus, autophosphorylates and then passes the phosphates to the cognate response regulators. The histidine kinase in an orthodox two-component system has only one histidine domain where the autophosphorylation occurs, but a histidine kinase in some unusual two-component systems (unorthodox two-component systems) has two histidine domains and one aspartate domain. So, the unorthodox two-component systems have more complex phosphorelay mechanisms than orthodox two-component systems. In general, the two-component systems are required to promptly respond to external stimuli for survival of E. coli. In this respect, the complex multi-step phosphorelay mechanism seems to be disadvantageous, but there are several unorthodox two-component systems in E. coli. In this paper, we investigate the reason why such unorthodox two-component systems are present in E. coli. For this purpose, we have developed simplified mathematical models of both orthodox and unorthodox two-component systems and analyzed their dynamical characteristics through extensive computer simulations. We have finally revealed that the unorthodox two-component systems realize ultrasensitive responses to external stimuli and also more robust responses to noises than the orthodox two-component systems.  相似文献

Microfiltration (MF) of a fermentation broth containing Escherichia coli is reported in this article. We used a ceramic membrane filter (zirconia on sintered carbon) having a nominal pore size of 0.2 μm. Our results indicate that the filtration resistance was mainly caused by the cake formed on the membrane surface. Both transmembrane pressure (TMP) and fluid sweeping velocity influenced this cake resistance. Resistances due to membrane itself and due to internal pore blockage by E. coli were less important and insensitive to both TMP and fluid sweeping velocity. Preliminary results also showed that the cell density could be significantly increased when we connected such a ceramic filter on-line with our fermentation system. In particular it was found that the gas bubbles entrained in the broth side of the filter could increase the filtration flux by as much as 80%.  相似文献

Knowledge of the mesoscopic morphology of condensed phase domains formed after the main phase transition in the two-phase coexistence region of Langmuir monolayers progressed rapidly with the development of the highly-sensitive imaging techniques, particularly by Brewster angle microscopy (BAM). Latest developments of commercial BAM instruments have been developed to a high technical level and allow upgrading to imaging ellipsometers which combine optical microscopy and ellipsometry and make the assessment of small layered structures or patterned thin films possible. A large variety of condensed phase domains different in mesoscopic sizes and shapes as well as their textural features has been observed which depend sensitively on the chemical structure of the amphiphilic monolayer and the system conditions, such as surface pressure and temperature. This unsuspected morphological variety of condensed phase domains has been proven not only in Langmuir monolayers but also in adsorbed monolayers (Gibbs monolayers), in Langmuir monolayers penetrated by dissolved surfactants or in adequate molecular recognition systems. The inner textures of domains can be explained on the basis of their geometry and the two-dimensional lattice in dependence of the tilt angle of the alkyl chains and gave rise to the development of a geometric concept on the basis of the molecular packing. New knowledge has been gained about non-equilibrium structures and their transition kinetics into the equilibrium state. Combined results obtained recently by BAM have enhanced the understanding of molecular organization in phase diagrams and binary mixtures. Recent advances in model studies about chiral discrimination effects and of the highly specific structural changes of host-monolayers by recognition of non-surface active guest-components have made progress. Semi-empirical quantum chemical methods have been used to gain insight into the role of different types of interactions involved in the main characteristics of mesoscopic length scale aggregates of mimetic systems.  相似文献

The Langmuir films of two liquid crystal materials, 4-octyl-4'-cyanobiphenyl (8CB) and 4-pentyl-4"-cyano-p-terphenyl (5CT), and of their mixtures have been studied by recording surface pressure-area isotherms and Brewster angle microscopy (BAM) images. The pure liquid crystals revealed very different characters of the surface pressure-area isotherms indicating different organization of the molecules and different molecular interactions in the monolayer at the water-air interface. The surface pressure-area isotherms of Langmuir films formed from 8CB/5CT mixtures give evidence for phase separation of the components over the whole range of molar fractions. Similar conclusions have been drawn on the basis of BAM image analysis.  相似文献

In order to establish ELISA (enzyme-linked immunosorbent assay) method to detect Total E. coli in water environment, E. coli multi-characters antigens in water environment were prepared according to the characters of kinds of E. coli serotypes, including antigen of whole cell, antigen of disrupted whole cell, somatic antigen, flagellar antigen and fimbrial antigen. Total E. coli polyclonal antibodies were obtained from the New Zealand rabbits immunized with these five antigens, respectively. Antibodies generated in this research are with high titers and good purity, can conjugate with antigens, specifically, stably and strongly. Indirect ELISA shows the titers of antibody of whole cell and antibody of disrupted whole cell are both over 1 × 105. The cross-reactivity of the antibody is from 12 to 30% which indicate the specificity of the antibody against Total E. coli. Based on these antibodies, we established indirect ELISA method to detect Total E. coli in water environment. The matrix effects were studied and the results show that there is no significant influence by all the factors. The ELISA result shows that the detection limitation could be 104 CFU (colony forming units) L−1. The indirect ELISA method developed in this study is well suited for Total E. coli analysis in real water samples as a rapid screen method.  相似文献

The organization of bacteriorhodopsin (bR) within reconstituted purple membranes (RPM) was examined using atomic force microscopy (AFM). Five reconstituted species were examined: RPM 3 (bR/native polar lipids/dimyristoylphosphatidylcholine (DMPC) in a 1:9:14 molar ratio), RPM 4 (bR/native polar lipids in a 1:7 molar ratio), RPM 5 (bR/native polar lipids/1,2-di-O-phytanyl-sn-glycerol in a 1:3.5:6.1 molar ratio), RPM 6 (bR/native polar lipids/1,2-di-O-phytanyl-sn-glycero-3-phosphocholine in a 1:3.5:4.9 molar ratio), and RPM 7 (bR/native polar lipids/1,2-diphytanoyl-sn-glycero-3-[phospho-l-serine] in a 1:3.5:4.6 molar ratio). RPM 3 patches adsorbed onto mica exhibit domains of crystallized bR trimers arranged in a hexagonal packing structure, similar to those found in native purple membrane (NPM). These domains are enclosed by DMPC-rich regions. RPM 4 patches were observed to have larger domains of crystallized bR, with trimer orientation 30° different from that found in NPM. The bR-rich domains are enclosed by a large, protein-free, lipid-rich region. The topography of RPM 5 was difficult to resolve as the surface had no discernable patterns or structure. The topographies of RPM 6 and 7 were similar to that found in RPM 3 in that higher domains were formed within the patch adsorbed onto mica. They may contain protein-rich regions, but clear images of protein arrangement could not be obtained using AFM. This may be a result of imaging limitations or of the lack of organization of bR within these domains.  相似文献

The aim of this work was to get insight into cholesterol distribution between two leaflets of a phospholipids bilayer. In this order, the thermodynamic analysis of the interactions between membrane lipids in binary (cholesterol/phospholipid) and ternary (phospholipid/ phospholipid/cholesterol) mixed Langmuir monolayers has been performed. For our investigation, phosphatidylcholine and phosphatidylethanolamine, which are the main types of phospholipids determining the distribution of cholesterol in membrane leaflets, were chosen and mixed in proportions corresponding to their molar ratios in the inner and outer layers of the natural human erythrocyte membrane. Into these mixed systems, various amount of cholesterol were incorporated. It has been found that despite strong differences in the phospholipid composition of both investigated ternary mixed systems, the influence of cholesterol is very similar, which indicates that cholesterol is symmetrically distributed between the inner and outer leaflets of the human erythrocytes membrane.  相似文献

The structure and the dynamic organization of a mixed Langmuir film of glucose oxidase and stearylamine at the air–water interface have been studied. The film has been first characterized at the air–water interface by surface pressure/area isotherms. The dynamics of the mixed film was studied by following the evolution of the film area at a constant pressure and the evolution of the pressure at a constant area. After transfer of the films on solid substrates, the chemical composition of the mixed film has been quantified by UV–vis and IR spectroscopies. These characterizations were carried out in order to study the incorporation of glucose oxidase into the stearylamine film, and its influence on the structural evolution of the film. From these results, the dynamic organization of this mixed film may be described. For short times, glucose oxidase molecules interact with stearylamine molecules in solution or at the interface; these interactions would lead to the formation of a complex between stearylamine and glucose oxidase molecules. For long times (at least 3 h), a homogeneous mixed film constituted essentially of this complex is obtained at the air–water interface. A detailed analysis by atomic force microscopy allowed us to support this model and the existence of the glucose oxidase/stearylamine complex.  相似文献

Langmuir films of members of two homologous series, the 4‐n‐alkyl‐4′‐cyanobiphenyls (nCB) for n = 2–14 and trans‐4‐n‐alkyl(4′‐cyanophenyl)cyclohexanes (PCHn) for n = 2–12, have been studied by recording surface pressure/area isotherms and by Brewster angle microscopy. It has been found that the compounds with very short chains (n3) and very long chains (n>12 for nCB, n>10 for PCHn) are unable to form compressible monolayers at the air–water interface. Other members of both series can form stable Langmuir films, but both their rigidity and stability as well as the molecular packing vary with the alkyl chain length. The isotherms and BAM images imply that the organization of the liquid crystal molecules in the films is to some extent correlated with their ability to form corresponding mesophase in the bulk: nematogenic compounds tend to form rounded droplet‐like domains, whereas smectogenic compounds tend to form flat domains.  相似文献

Low density lipoproteins (LDL) from egg yolk have a classical structure of lipoprotein with a core of neutral lipids surrounded by a monolayer of apoproteins and phospholipids. This structure collapses during adsorption and all constituents spread at the interface. To understand better the nature of the interactions between apoproteins and lipids at the interface, we have deposited LDL at an air-water interface and analysed the isotherms during their compression on a Langmuir trough. Then, these LDL films were studied by atomic force microscopy (AFM) imaging. To identify the protein and lipid structures, we imaged films before and after lipid solubilisation by butanol. To study the interactions in the LDL films, we have varied the pH, ionic strength and used simplified model systems. We also studied the correlation between observed structures and interfacial rheology of the film. The isotherms of interfacial LDL films were similar for pH 3 and 7, but their structures observed in AFM were different. At surface pressures below the transition corresponding to the demixion of apoprotein-neutral lipid complexes, the LDL film structure was not governed by electrostatic interactions. However, above this surface pressure transition (45mN/m), there was an effect of charge on this structure. Around the transition zone, the rheological properties of LDL films at pH 3 were different as a function of pH (viscous at pH 3 and visco-elastic at pH 7). So, the rheological properties of LDL films could be linked to the structures formed by apoproteins and observed in AFM.  相似文献

Fabrication of Langmuir films at the air-water interface of four linear-dendritic block copolymers (LDBCs) is described. The LDBCs are composed of a linear hydrophilic chain of poly(ethylene glycol) (PEG) and the first four generations of hydrophobic aliphatic polyester dendrons functionalized at the periphery with cyanoazobenzene chromophores. Langmuir films of the LDBCs, coded as PEG-AZOn (n indicates the number of cyanoazobenzene units at the periphery of the dendritic block), have been characterized by a combination of surface pressure versus area per molecule isotherms, UV-vis reflection spectroscopy and Brewster angle microscopy. The observed PEG-AZOn Langmuir film behavior depends strongly on the hydrophilic/hydrophobic ratio. A typical transition, related to PEG chains desorption from the air-water interface into the water subphase is observed for all the LDBCs, except for PEG-AZO16. In addition, PEG-AZO2 and PEG-AZO4 show a second transition whose nature has been studied in detail. Azobenzene chromophore interactions have been shown to be relevant in the organization of PEG-AZOn (n=4, 8 and 16) Langmuir films. Moreover, for PEG-AZO16 the orientation of the azobenzene units has been determined, revealing the formation of a well organized structure of azobenzene moieties at the air-water interface.  相似文献

One-pot self-assembled hybrid films were synthesized by the cohydrolysis of methyltriethoxysilane and tetraethoxysilane and deposited via dip-coating. The films show a high "defect-free" mesophase organization that extends throughout the film thickness and for domains of a micrometer scale, as shown by scanning transmission electron microscopy. We have defined these films defect-free to describe the high degree of order that is achieved without defects in the pore organization, such as dislocations of pores or stacking faults. A novel mesophase, which is tetragonal I4/mmm (space group), is observed in the films. This phase evolves but retains the same symmetry throughout a wide range of temperatures of calcination. The thermal stability and the structural changes as a function of the calcination temperature have been studied by small-angle X-ray scattering, scanning transmission electron microscopy, and Fourier transform infrared spectroscopy. In situ Fourier transform infrared spectroscopy employing synchrotron radiation has been used to study the kinetics of film formation during the deposition. The experiments have shown that the slower kinetics of silica species can explain the high degree of organization of the mesostructure.  相似文献

The presence of microdomains, called lipid rafts, in biological membranes is usually explained by lateral segregation between specific lipids and proteins. These rafts present similarities with the membrane domains isolated by their non-ionic detergent-resistance at 4 degrees C. They are enriched in sphingomyelin and cholesterol as compared with the outer leaflet of eukaryotic cell membranes. To understand the role played by the lipids enriched in rafts in their resistance to solubilization by detergents, the interactions between these lipids and the non-ionic detergent Triton X-100 were studied by using different lipid monolayers at the air-water interface. The influence of Triton X-100 on the Langmuir isotherms (i.e. surface pressure/area isotherms) of monolayers containing sphingomyelin and cholesterol at different mole ratios was analyzed and the results were compared with the influence of Triton X-100 on monolayers containing a phosphatidylcholine bearing a saturated and an unsaturated fatty acid (i.e. palmitoyloleylphosphatidylcholine) and cholesterol. This phosphatidylcholine was chosen since the phosphatidylcholines present in rafts isolated from bovine kidney could contain about 50% of saturated fatty acids. Triton X-100 induces an increase in the condensing effect observed as compared with ideal mixture of phospholipid/cholesterol. Triton X-100-induced changes in the morphology of the monolayers were visualized by Brewster angle microscopy, which confirmed the differences of behavior observed by analyzing the isotherms.  相似文献

Mixed Langmuir monolayers and Langmuir-Schaefer (LS) films containing the cationic metallosurfactant bis(2-phenylpyridine)(4,4'-diheptadecyl-2,2'-bipyridine)-iridium(III) chloride (Ir-complex) and the anionic tetrakis(4-sulfonatophenyl)porphyrin (TSPP) in 4:1 molar ratio have been successfully prepared by the co-spreading method at the air-water interface. The presence of both luminescent species at the interface, as well as the organization of the TSPP underneath the Ir-complex matrix in Langmuir and LS films, is inferred by surface techniques such as π-A isotherms, reflection spectroscopy, Brewster angle microscopy (BAM) and UV-visible absorption spectroscopy. A red-shift in the absorption band of the porphyrin under the compression of the mixed monolayer suggests the J-aggregation of the TSPP under the Ir-complex matrix. To date, this is the first report of Langmuir and/or LS films containing these two types of species together. Furthermore, the intermolecular energy transfer between Ir-complex and TSPP molecules in solution and in transferred mixed films is investigated through steady-state fluorescence and lifetime measurements. These results indicate that effective intermolecular energy transfer occurs from the Ir-complex to the TSPP molecules in LS films. The influence of the spatial proximity of donor and acceptor molecules has been studied by the insertion of lipid interlayers among them.  相似文献
