The total synthesis of the natural product cytostatin is described which inhibits protein phosphatase 2A. Cytostatin has anti-metastatic properties and induces apoptosis. On the basis of this synthesis the relative and absolute configuration of cytostatin could be assigned. Key structural elements of cytostatin are an alpha,beta-unsaturated lactone group and a side chain embodying a phosphate and a rather unstable (Z,Z,E)-triene subunit. In addition, the natural product carries six stereocenters. For the construction of the stereocenters reagent-controlled transformations were used in order to ensure maximum stereochemical flexibility. The Evans syn-aldol reaction was chosen to establish the stereochemistry at C-4, C-5, C-9 and C-10; C-6 was introduced by means of the Evans asymmetric alkylation. In all cases the same chiral auxiliary was employed as stereodirecting group. The stereocenter at C-11 was established by an asymmetric reduction using CBS-oxazaborolidine. Temporary protection of the phosphate group was achieved best by using the base-labile 9-fluorenylmethyl group, which could be cleanly cleaved by an excess of triethylamine; this reaction yielded analytically pure phosphates after a simple aqueous work-up. The (Z,Z,E)-triene embodied in cytostatin was synthesized by means of a Stille coupling as key transformation. The synthesis sequence established in this way readily gave access to a series of analogues with simplified structure. Initial biological testing of these analogues proved that the alpha,beta-unsaturated lactone, the C-11-hydroxy group and a fully deprotected phosphate moiety at C-9 are essential for the PP2A-inhibitory activity of cytostatin. The rather unstable triene moiety in the side chain can be replaced by other lipophilic residues with only moderate decrease of biological activity. Other phosphatases, that is, PP1, VHR, PTP1B, CD45, were not inhibited by cytostatin or any of the analogues, demonstrating the high selectivity of this compound. These findings will be useful for the design and synthesis of cytostatin-derived chemical tools for the study of biological processes influenced by PP2A.  相似文献

A novel class of fulgimide, (Z)-4-oxazolylfulgimide ((Z)-1-benzyl-4-isopropylidene-3-[1-(2-aryl-5-methyloxazolyl)ethylidene]tetrahydropyrrole-2,5-dione), was synthesized by the reaction of (Z)-4-oxazolyl fulgide (4-isopropylidene-3-[1-(2-aryl-5-methyloxazolyl) ethylidene]tetrahydrofuran-2,5-dione) with benzyl amine. Photochromic property of (Z)-4-oxazolylfulgimide was studied. Compared with (Z)-4-oxazolylfulgide, the absorption maximum of the colored form of (Z)-4-oxazolylfulgimide is bathochromic-shifted. Substituents on the aryl ring affect the absorption maximum of the open form and the colored form of (Z)-4-oxazolylfulgimide.  相似文献

An unusual 3D porous network assembled by tridentate ligand Tbmiz [1,3,5-tris(benzimidazol-1-ylmethyl)-2,4,6-trimethylbenzene] and Cu-I cluster was synthesized by means of hydrothermal process. It crystallizes in the trigonal system, space group P-3c1 with α= 14.2362(8), b = 14.2362(8), c = 36.4017(35) A, γ = 120.00°, V= 6389.1(8), D_c = 1.976 g/cm~3, Z = 2, μ(MoKα) = 0.71073 and F(000) = 3692.0. Of the 40832 total reflections, 3948 were unique (R_(int)=0.0366). The final R = 0.0460 and wR = 0.1335 for 3948 observed reflections with I > 2σ(I).  相似文献

(Z)-α-Selanyl alkenyl Grignard reagents 2 were prepared conveniently by treatment of (-bromovinylselenides 1 with magnesium filings in THF. Intermediates 2 were reacted with alkyl iodides in the presence of CuI or Pd(PPh3)4 to afford (Z)-1,2-disubstituted vinylselenides 3, which were cross-coupled with Grignard reagents in the presence of (PPh3)2NiCl2 to give trisubstituted alkenes 4 stereoselectively in good yields.  相似文献

Four new (Z)-2-phenoxymethylene-4-pentanolide derivatives and four (Z)-2-phenoxymethyl-2penten-γ-lactone derivatives were prepared via Pd-catalyzed carbon monoxide insertion and intramolecular cyclization. A copper(Ⅱ) complex [CuL2] [L=2-(1H-1,2,4-triazol-1-yl)acetic acid] has been synthesized by means of hydrothermal method, and characterized by elemental analysis, IR spectra and X-ray diffraction. Crystal data for this complex: monoclinic, space group P21/n, a=0.481 3(2) nm, b=1.151 1(6) nm, c=0.993 9(5) nm, β=100.528(8)°, V=0.541 4(5) nm3, Z=2, F(000)=318, Dc=1.937 g·cm-3, Final GooF=1.109, R1=0.027 6, wR2=0.075 6. The crystal structure shows the copper(Ⅱ) atom is six-coordinated by two triazole nitrogen atom and four carboxylate group oxygen atom from four distinct L- ligand, thereby forming a 2D network structure. On the basis of the Ag(I)-promoted coupling reaction of zinc(II)-5,15-bis(3,5-dioctyloxyphenyl)porphyrin Z1, chain elongation has been attempted by using a stepwise doubling approach, which provides Z2, Z4, Z8, Z16, Z32, Z64, Z128, Z256, Z384, and Z512. The porphyrin arrays up to Z128 are sufficiently soluble in CHCl3 and THF despite their very long molecular lengths and rodlike structures, while the arrays over Z128 show a significant drop in solubility and stability. The discrete porphyrin arrays thus isolated were characterized by means of (1)H NMR spectroscopy, matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry, UV/Vis spectroscopy, gel-permeation chromatography (GPC), cyclic voltammetry (CV), single-crystal X-ray crystallography, scanning tunneling microscopy (STM), and atomic force microscopy (AFM). Contrary to expected linear conformations of the arrays Z n (where n is the number of porphyrins), the single molecular images of Z128, Z256, and Z512 revealed largely bent structures; this finding indicates the substantial conformational flexibility of Z n. We also exploited an effective synthetic route by means of which Z n can be fabricated with a thiol-protected aryl group to provide Z n S(2) through Z n Br(2), by bromination with N-bromosuccinimide and subsequent Pd-catalyzed Suzuki-Miyaura arylation. Finally, the reaction of Z256 provided Z512, Z768, and Z1024. Collectively, this work provides an important milestone in the preparation of sub-microscale discrete organic molecules and the fabrication of molecular-based materials, hence significantly contributing to device applications.  相似文献

A novel (Z)-nitromethylene neonicotinoid analogue (C23H27Cl2N5O4·2H2O) (II) with optical activity has been synthesized, the structure was characterized by elemental analysis, IR and 1H NMR spectra, and the (Z)-configuration was confirmed by single-crystal X-ray diffraction. The crystal belongs to triclinic, space group P1 with a = 7.4638(3), b = 12.6232(5), c = 15.2990(6), α = 71.907(1), β = 89.397(2), γ = 80.314(1)°, V = 1349.28(9)3, Z = 2, Dc = 1.340 g/cm3, μ = 0.286 mm-1, Mr = 544.43, F(000) = 572, S = 1.056, R = 0.0801 and wR = 0.2366 for 4998 unique reflections with 3012 observed ones (I > 2σ(I)). In the crystal, the dihedral angle between the pyridine and 4-Cl-phenyl rings is 58.13°. Intermolecular O–H···O, C–H···O and C–H···Cl hydrogen bonds involving water molecules stabilize the crystal structure.  相似文献

The topological resonance energy (TRE) was conceived in the 1970s. From the very beginning, it was known that TRE is equal to the collective energy-effect of all cycles present in a conjugated molecule. Also in the 1970s a theory of cyclic conjugation was elaborated, by means of which it was possible to compute the energy-effect ef(Z) of each individual cycle Z present in a conjugated molecule. Yet, the connection between TRE and the ef(Z)-values was, until now, not studied. We now show that TRE and the sum of the ef(Z)-values are closely correlated but that a certain correction needs to be made by taking into account the energy-effects of pairs, triplets, quartets, etc. of cycles.  相似文献
