One‐pot synthesis of 2‐aryltetrahydrofurans was achieved by a coupling reaction between arylmagnesium bromides prepared in situ and tetrahydrofuran under mild conditions. The reaction between ArBr and n ‐BuLi gave unexpected butylbenzene derivatives in moderate yields.  相似文献

The study toward the total synthesis of (R)‐(+)‐harmicine is reported in this paper. The enantioselective synthesis of pyrrolidinone, the main backbone of of (R)‐(+)‐harmicine, has been completed by the methodology based on photo‐induced Wolff rearrangement of α‐diazo‐β‐carbonyl compounds.  相似文献

An efficient stereoselective total synthesis of (3R,5R)‐1‐(4‐hydroxyphenyl)‐7‐phenylheptane‐3,5‐diol ( 1 ) is reported based on the Mukaiyama aldol reaction. The total synthesis of compound 1 was accomplished with 30% overall yield in simple eight steps from commercially available trans‐cinnamaldehyde.  相似文献

An efficient synthesis of polycyclic indole derivatives is achieved via domino Knoevenagel–hetero‐Diels–Alder reaction of O‐acrylated salicylaldehyde derivatives with dihydroindole‐2‐thiones in H2O as solvent. The products are formed in good‐to‐excellent yields with high regio‐ and stereoselectivity.  相似文献

A racemic cis/trans mixture of 5‐hexyl‐4‐methyl‐γ‐butyrolactone was easily synthesized from 1‐iodoheptane in four steps with inexpensive and readily available reagents. Our new synthesis method can be potentially employed for mass production of the 4‐methyl‐5‐hexyl‐γ‐butyrolactone as well as other poly‐alkyl substituted γ‐butyrolactones.  相似文献

A sequential one‐pot two‐step reaction for efficient synthesis of pyrrolo[2,1‐a]isoquinoline and pyrrolo[1,2‐a]quinoline derivatives in good yields has been successfully developed. The reaction included firstly Cu‐catalyzed three‐component reaction of isoquinoline (quinoline), acetylenedicarboxylate and alkynylbenzene and then Pd‐catalyzed intramolecular C(sp)‐C(sp2) coupling reaction of initially formed 1‐alkenyl‐2‐alkynyl‐1,2‐dihydroisoquinoline (1,2‐dihydroquinoline).  相似文献

15‐Cyano‐12‐oxopentadecano‐15‐lactone was synthesized in 59% total yield starting from 2‐nitrocyclododecanone by Michael addition to acrylaldehyde, followed by reaction with trimethylsilylcyanide, hydrolysis, ring‐expansion, and Nef reaction. A two‐step, one‐pot synthesis of intermediate 2‐hydroxy‐4‐(1‐nitro‐2‐oxycyclododecyl)butanenitrile from 3‐(1‐nitro‐2‐oxocyclododecyl)propanal was developed and the conditions for the Nef reaction were studied. 15‐Cyano‐12‐oxopentadecano‐15‐lactam was synthesized in 40% total yield starting from 2‐nitrocyclododecanone by Michael addition to acrylaldehyde, followed by Strecker reaction, ring‐expansion, and Nef reaction. The conditions for the Strecker and Nef reactions were studied. The structures of the target compounds, intermediates, and by‐product were characterized by IR, 1H‐ and 13C‐NMR, and elemental analysis or MS.  相似文献

An efficient and concise synthesis of the biologically interesting (+)‐machaeriol B ( 2 ) and its enantiomer 5 was accomplished from O‐phenylhydroxylamine ( 7 ) in four steps (Scheme 2). In addition, the first total synthesis of natural (+)‐machaeriol C ( 3 ) and its enantiomer 6 was achieved from the readily available ester 15 in eight steps (Scheme 4). The key strategies in the syntheses of 2 and 5 involved benzofuran formation through a [3,3]‐sigmatropic rearrangement and trans‐hexahydrodibenzopyran formation by a domino aldol‐type/hetero‐Diels–Alder reaction. In the case of 3 and 6 , the key steps were stilbene formation by a Horner–Wadsworth–Emmons reaction and trans‐hexahydrodibenzopyran formation by domino reactions.  相似文献

A direct three‐component approach has been developed for the synthesis of α‐cyano acrylates starting from aldehydes, alcohols and α‐cyano acetamide by employing cyanuric chloride as an organocatalyst. An efficient method for the synthesis of 2‐aryl‐2,3‐dihydro‐3‐sulfanyl‐1H‐isoindol‐1‐ones 1 via Pummerer‐type cyclization of N‐aryl‐2‐(sulfinylmethyl)benzamides 2 is described. Thus, treatment of these sulfinyl‐benzamides 2 , easily prepared from 2‐(bromomethyl)benzoates 3 in three steps, with Ac2O at ca. 100° resulted in the formation of the desired isoindolones 1 in generally good yields.  相似文献

A short, efficient synthesis of 2‐methoxy‐6‐alkyl‐1,4‐benzoquinones is described. Ultrasound‐assisted Wittig reaction of alkyltriphenyl phosphonium bromides with o‐vanillin in basic aqueous conditions followed by reduction with Na/n‐BuOH gave 2‐methoxy‐6‐alkylphenols. Oxidation of 2‐methoxy‐6‐alkylphenols with Fremy's salt produced the title compounds.  相似文献

A palladium‐catalyzed cascade protocol has been established for the synthesis of 4‐methyl‐1‐(1H‐pyrrolo[2,3‐b]‐quinoxalin‐2‐yl)cyclohexanols and 2‐phenyl‐1‐(1H‐pyrrolo[2,3‐b]quinoxalin‐2‐yl)propan‐1‐ols through the reaction of N‐alkyl(aryl)‐3‐chloroquinoxalin‐2‐amines with calcium carbide and cyclohexanones or 2‐phenylpropanal. This one‐pot process, carried out without any copper salt in the key step of the Sonogashira coupling reaction, provides an efficient method for the synthesis of 2,3‐disubstituted pyrrolo[2,3‐b]quinoxalines in the presence of catalytic amounts of Pd(PPh3)2Cl2 in DMSO/H2O with high yields. The benefit of this strategy is the use of a commercially available, inexpensive, and less hazardous primary chemical feedstock, calcium carbide, as an acetylene source in a wet solvent.  相似文献

A study on the synthesis of the novel N‐(cyclic phosphonate)‐substituted phosphoramidothioates, i.e., O,O‐diethyl N‐[(trans‐4‐aryl‐5,5‐dimethyl‐2‐oxido‐2λ5‐1,3,2‐dioxaphosphorinan‐2‐yl)methyl]phosphoramidothioates 4a – l , from O,O‐diethyl phosphoramidothioate ( 1 ), a benzaldehyde or ketone 2 , and a 1,3,2‐dioxaphosphorinane 2‐oxide 3 was carried out (Scheme 1 and Table 1). Some of their stereoisomers were isolated, and their structure was established. The presence of acetyl chloride was essential for this reaction and accelerated the process of intramolecular dehydration of intermediate 5 forming the corresponding Schiff base 7 (Scheme 2).  相似文献

A simple, one‐step, stereoconservative synthesis of diamine‐based peptidomimetics is described, by split‐Ugi multicomponent reaction, involving chiral N‐protected amino acids and α‐substituted isocyanoacetate. In particular, piperazine and bispidine (3,7‐diazabicyclo[3.3.1]nonane) are exploited as diamine components, bispidine being the first example of a sterically demanding bicyclic system employed in a split‐Ugi reaction.  相似文献

A facile and benign synthetic strategy is proposed for the synthesis of 2‐arylpyrroloacridin‐1(2H )‐ones via a lactic acid‐catalyzed three‐component reaction of dimedone, various anilines, and isatins under solvent‐free conditions. Avoidance of hazardous organic solvents, the use of a one‐pot multicomponent procedure for the synthesis of 2‐arylpyrroloacridin‐1(2H )‐ones, operational simplicity, no need for column chromatography, lactic acid utilization as a bio‐based organic compound, reusability, homogeneity, and commercial availability of the catalyst, and superior synthetic performance are some important aspects of this methodology to access a series of pyrroloacridine motifs with potentially biological scaffolds.  相似文献
