Iminofuran chemistry: VII. Intramolecular cyclization of 2-N-aryl-substituted derivatives of 2-amino-4-aryl-4-oxobut-2-enoic and 2-amino-5,5-dimethyl4-oxohex-2-enoic acids

The cyclization at the treatment of acetic anhydride of 4-aryl-2-arylamino-4-oxobut-2-enoic and 2-arylamino-5,5-dimethyl-4-oxohex-2-enoic acids was investigated furnishing derivatives of 5-aryl-3-aryl-imino-3H-furan-2-ones and 4-arylamino-2-tert-butyl-2,5-dihydro-5-oxofuran-2-yl acetate respectively. 2-N²-Methylenesubstituted 4-aryl-2-hydrazino-4-oxobut-2-enoic and 5,5-dimethyl-2-hydrazino-4-oxohex-2-enoic acids cleanly underwent cyclization under the effect of acetic anhydride into 5-aryl-3-hydrazono-3H-furan-2-ones and 5-tert-butyl-3-hydrazono-3H-furan-2-ones.     

Iminofurans chemistry: IV. Synthesis and structure of 2-<Emphasis Type="Italic">N</Emphasis>-aryl-substituted derivatives of 2-amino-4-aryl-4-oxobut-2-enoic and 2-amino-5,5-dimethyl-4-oxohex-2-enoic acids

Reactions of arylamines with 4-aryl-2-hydroxy-4-oxobut-2-enoic and 2-hydroxy-5,5-dimethyl-4-oxohex-2-enoic acids gave rise to 4-aryl-2-arylamino-4-oxobut-2-enoic and 2-aryl-amino-5,5-dimethyl-4-oxohex-2-enoic acids that existed in solutions as Z- and E-isomers or in a ring form as 3-arylamino-5-tert-butyl-5-hydroxyfuran-2(5H)-ones. The probable cyclization mechanism of these compounds into 5-R-3-arylimino-3H-furan-2-one derivatives was considered.     

Chemistry of iminofurans: XI. Synthesis,structure, and cyclization of 4-substituted 2-(aroylhydrazinylidene)-4-oxobutanoic acids

Aromatic hydrazides reacted with 4-aryl-2-hydroxy-4-oxobut-2-enoic and 2-hydroxy-5,5-dimethyl- 4-oxohex-2-enoic acids to give 4-aryl-2-(2-aroylhydrazinylidene)-4-oxobutanoic and 5,5-dimethyl-2-(aroylhydrazinylidene)- 4-oxohexanoic acids. The products were found to exist in solution as mixtures of Z/E-hydrazinylidene and cyclic dihydropyrazole tautomers, and they underwent intramolecular cyclization to 5-aryland 5-tert-butyl-3-(aroylhydrazinylidene)furan-2(3H)-ones by the action of acetic anhydride.

Chemistry of iminofurans: V. Synthesis,structure, and cyclization of 4-R-4-oxo-2-[2-(2-oxo-1,2-diphenylethylidene)hydrazino]but-2-enoic acids

Reactions of 1,2-diphenylethane-1,2-dione hydrazone with 4-aryl-2-hydroxy-4-oxobut-2-enoic and 2-hydroxy-5,5-dimethyl-4-oxohex-2-enoic acids provided 4-aryl-4-oxo-2-[2-(2-oxo-1,2-diphenylethylidene) hydrazino]but-2-enoic and 5,5-dimethyl-4-oxo-2-[2-(2-oxo-1,2-diphenylethylidene)hydrazino]hex-2-enoic acids. The acids derivatives can exist in solutions as Z- and β-enehydrazino-or β-ketohydrazone forms, and under the treatment with acetic anhydride they undergo cyclization into 5-aryl- and 5-tert-butyl-3-[2-(2-oxo-1,2-diphenylethylidene)hydrazono]-2,3-furandiones.     

Indolyl substituted 4-oxobut-2-enoic acids. Synthesis and aza-Michael additions

The synthesis of three new substituted 4-hetereoaryl-4-oxobut-2-enoic acids with indole ring substitutedin positions 3-, 5- and 7- is described. The addition of these Michael acceptors to 4-(1-phenylsulphonylpyrrol-3-yl)-4-oxobut-2-enoic acid in conjugate addition was explored using both racemic and chiral amines. In tandem with the crystallization-induced asymmetric transformation (CIAT) protocol the effective methodology for the synthesis of enantiomerically highly enriched substituted 2-amino-4-heteroaryl-4-oxobutanoic acids as multifunctional homotryptophan analogues was developed.      

Synthesis of Alkyl 4-(1-Alkyl-2-aryl-2-oxoethyl)-5,5-dimethyl-2-oxotetrahydrofuran-3-carboxylates

Zinc enolates derived from 1-aryl-2-bromoalkanones react with alkyl 5,5-dimethyl-2-oxo-2,5-dihydrofuran-3-carboxylates to give alkyl 4-(1-alkyl-2-aryl-2-oxoethyl)-5,5-dimethyl-2-oxotetrahydrofuran-3-carboxylates. Reactions of the latter with amines, such as p-toluidine, cyclohexylamine, and piperidine, lead to the corresponding carboxamides.     

Synthesis, characterization of the luminescent lanthanide complexes with (Z)-4-(4-methoxyphenoxy)-4-oxobut-2-enoic acid

(Z)-4-(4-Methoxyphenoxy)-4-oxobut-2-enoic acid and its solid rare earth complexes LnL3.2H2O (Ln=La, Eu, Tb) were synthesized and characterized by means of MS, elemental analysis, FTIR, 13C NMR and TG-DTA. The IR and 13C NMR results show that the carboxylic groups in the complexes coordinated to the rare earth ions in the form of a bidentate ligand, but the ester carboxylic groups have not taken part in the coordination. The luminescence spectra of Eu(III) and Tb(III) complexes in solid state were also studied. The strong luminescence emitting peaks at 616nm for Eu(III) and 547nm for Tb(III) can be observed, which could be attributed to the ligand has an enhanced effect to the luminescence intensity of the Eu and Tb.     

Synthesis of methyl 4-aryl-2-{[4-(carbamimidoylsulfamoyl)- phenyl]amino}-4-oxobut-2-enoates

Methyl aroylpyruvates reacted with 4-amino-N-carbamimidoylbenzene-1-sulfonamide in acetic acid–ethanol (1: 1) to give methyl 4-aryl-2-{[4-(carbamimidoylsulfamoyl)phenyl]amino}-4-oxobut-2-enoates which were found to exist in solution as mixtures of Z and E isomers.

4-aryl-2-hydroxy-4-oxobut-2-enoic acids <Emphasis Type="Italic">N</Emphasis>-(2-pyridyl)amides in reactions with diazo compounds

Reactions of 4-aryl-2-hydroxy-4-oxobut-2-enoic acids N-(2-pyridyl)amides with diazomethane, diazoethane, diaryldiazomethanes, and diazofluorene lead to the formation of 2-alkoxy-4-aryl-4-oxobut-2-enoic acids N-(2-pyridyl)amides, 3-aroyl-5-methylpyrazole-4-carboxylic acids N-(2-pyridyl)amides, and 3-alkoxy-3-(2-aryl-2-oxoethyl)-2,3-dihydro-2-oxoimidazo[1,2-a]pyridines. The composition and structure of compounds obtained depend on the nucleophilic nature of the diazo compound and on the character of substituents in the aryl and pyridine parts of the substrate.     

2-氧代-2-[1-(3-吡啶甲基氨基)-1''''-芳基]甲基-4-芳基-5,5-二甲基-1,3,2-二氧磷杂环己烷的合成与生物活性

采用环状亚磷酸酯与亚胺的加成反应, 合成了9个未见文献报道的含吡啶基的α-氨基环状膦酸酯化合物. 其结构经1H NMR, 31P NMR, MS 和元素分析确证, 同时采用X射线单晶衍射对化合物3i的立体构型进行了进一步确证. 测定了该系列化合物的杀虫、杀菌和除草活性, 结果表明, 化合物的杀虫活性虽然较差, 但所有化合物在药液质量浓度为1.0×10-4 g/mL时都具有较好的除草活性, 部分化合物在5.0×10-5 g/mL时显示出良好的杀菌活性.     

2－甲基－3－芳基－7－（5，5－二甲基－3－酮－1－环己烯－1－基）甲酸酯－4（3H）－喹唑啉酮的合成

以2－氨基对苯二甲酸（1）为起始原料，与乙酸酐缩合生成7－羧基乙酰苯邻 甲内酰胺（2）；2和芳胺缩合产生7－羧基－2－甲基－3－芳基－4（3H）－喹唑啉 酮（3）；3和N，N－双环己基碳双亚胺（DCC）加成得到中间体4，4在4－二甲氨基 吡啶（DMAP）催化下和5，5－二甲基－1，3－环己二酮（5）缩合得到目标产物2－ 甲基－3－芳基－7－（5，5－二甲基－3－酮－1－环己烯－1－基）甲酸酯－4（ 3H）－喹唑啉酮（6）。所得15个新型化合物的结构均经^1H NMR、元素分析确证， 部分化合物经IC／MSD确证。

Chemistry of iminofurans: IX. Synthesis and cyclization of (2<Emphasis Type="Italic">Z</Emphasis>)-2-{(2<Emphasis Type="Italic">Z</Emphasis>)-2-[2-(3-R-adamantan-1-yl)-2-oxoethylidene]hydrazinyl}-4-(het)aryl-4-oxobut-2-enoic acids

1-(3-R-adamantan-1-yl)-2-[(triphenyl-λ⁵-phosphanylidene)hydrazinylidene]ethanone reacted with 4-aryl(hetaryl)-2,4-dioxobutanoic acids to give 2-{2-[2-(3-R-adamantan-1-yl)-2-oxoethylidene]hydrazinyl}-4-aryl(hetaryl)-4-oxobut-2-enoic acids which were shown to exist in solution as mixtures of Z- and E-isomeric enehydrazine tautomers. The products underwent cyclization to 3-{[2-(3-R-adamantan-1-yl)-2-oxoethylidene]- hydrazinylidene}-5-aryl(hetaryl)furan-2(3H)-ones.     

Synthesis and search for compounds with hemostatic activity in the series of 4-(het)aryl-4-oxobut-2-enoic acid derivatives

New 4-aryl-2-(4-arylamino)-1-(piperazin-1-yl)but-2-ene-1,4-diones and 2-{[2-(3-R-adamantan-1-yl)-2-oxoethylidene]hydrazinyl}-4-(het)aryl-4-oxobut-2-enoic acids were synthesized and used to obtain water-soluble compounds. Their influence on the blood coagulation system was studied, which revealed compounds with a high hemostatic activity and a low acute toxicity. A relationship between structure and pharmacological effect of the synthesized compounds was established.     

Synthesis of 4-substituted 1-methyl(benzyl)-2,5-dimethyl-4-R-aminopiperidines

Schiff base derivatives of 1-methyl(benzyl)-2,5-dimethylpiperidin-4-ones with phenyl, -pyridyl, benzyl, and -hydroxyethyl substituents attached to the imine nitrogen atom react with organometallic compounds to give analogously substituted piperidine bases with methyl, allyl, phenyl, and benzyl substituents in the 4-position.
