HBV X蛋白与宿主APOBEC3G蛋白之间的相互作用

摘    要:将原核重组质粒双酶切后释放的χ基因与真核表达载体连接,构建真核重组质粒pcDNA3.1-X,然后与真核重组质粒pcDNA3.1-APOBEC3G共转染HepG2细胞,提取蛋白进行免疫共沉淀并用Western blot分析结果。免疫共沉淀后,HA-APOBEC3G融合蛋白能够在抗HBx的免疫沉淀物中检测出来,在抗HA标签的免疫沉淀物中也能检测到HBx。揭示了这两种蛋白能在受体细胞内形成复合物。结果表明乙肝病毒X蛋白与APOBEC3G蛋白在细胞内能发生相互作用,提示APOBEC3G抗HBV作用可能与乙肝病毒X蛋白有关。

Abstract:To investigate the interaction between HBV X protein and Human APOBEC3G protein, then to discover the mechanism of its antiviral activity in another perspective. The X gene was digested from prokaryotic expressing plasmid, and ligated with eukaryotic expressing vector. Then HepG2 cells cotransfected with recombinant plasmid pcDNA3.1-X and pcDNA3.1-APOBEC3G. The interaction between HBx and APOBEC3G protein was detected by co-immunoprecipitation. We construct eukaryotic expressing vector with X gene successfully. The HBx and HA-APOBEC3G fusion protein were detected respectively in the immunoprecipitation by anti HBx and anti HA-tag. It indicates that the two proteins can form compounds in HepG2 cells. X protein can combine with APOBEC3G protein in vivo. It suggests HBx plays a role in the effect of anti-human hepatitis B virus of APOBEC3G.
