摘    要:建立小鼠急性高尿酸血症模型,通过灌胃给药考察车前草的降血尿酸作用.采用正交试验设计,以车前草所含大车前苷为指标,考察乙醇浓度、提取时间、提取次数和乙醇用量4种因素对提取结果的影响.大车前苷和血尿酸的含量均用HPLC法测定.结果表明,车前草醇提组的小鼠血清尿酸水平明显低于模型组(P0.05).乙醇浓度、提取时间、提取次数和乙醇用量4种因素对车前草乙醇提取物中的大车前苷含量均有显著影响.最佳提取工艺为用10倍药材量的60%乙醇提取2次,每次2 h.

Abstract:The extracts of Plantain were investigated in the mice model of hyperuricemia.Orthogonal test design was utilized to optimize the extraction conditions including the concentration of ethanol,extraction duration per time,extraction times and amount of ethanol when plantamajoside was selected as the evaluation index.Both plantamajoside and serum uric acid levels were measured by using HPLC.The results indicated that the extracts of Plantain could significantly decrease the serum uric acid level of the hyperuricemic mice(P0.05).All of the concentration of ethanol,extraction duration per time,extraction times and amount of ethanol had significant effects on the content of plantamajoside extracted from Plantain.The optimum extraction process was to extract two times using tenfold 60% ethanol and 2 h per time.
