嗜酸乳杆菌发酵产亚油酸异构酶培养基条件的研究

摘    要:以产亚油酸异构酶为指标,鉴定从泡菜中分离的嗜酸乳杆菌L1,对产酶培养基的碳源、氮源、无机盐及其配比和表面活性剂等方面进行了优化,最后得到了优化后培养基,其组成为:蛋白胨10g,酵母提取物5g,牛肉膏15g,葡萄糖20g,无水乙酸钠2.5g,柠檬酸二铵3g,Tween-801.5mL,MgSO40.14g,MnSO4.H2O0.3g,K2HPO4.3H2O1.31g.经发酵验证:培养基经优化后产酶是未优化前的1.32倍。

Abstract:A type of Lactobacillus acidophilus L1,isolated from pickled vegetable,was used as the benchmark for linoleic acid isomerase production.The medium components and concentration such as carbon source,nitro source,inorganic salt and surfactant,etc.were tested with linoleic acid isomerase as an indicator.At last,the optimum medium as well as its concentration was obtained,which has the formulation of(per liter) peptone 10 g,yeast extract 5 g,beef extract 15 g,glucose 20 g,sodium acetate anhydrous 2.5 g,ammonium citrate dibasic 3 g,Tween-80 1.5 mL,MgSO4 0.14 g,MnSO4.H2O 0.3 g and K2HPO4.3H2O 1.31 g.The enzymatic activity unit of linoleic acid isomerase is found to be 1.32 times those values without optimization.
