核酸适配体功能化纳米界面用于循环肿瘤细胞捕获与释放

摘    要:循环肿瘤细胞(CTCs)是肿瘤研究和临床癌症诊断中的重要对象,也是"液体活检"的重要标志物。CTCs携带着肿瘤组织的遗传和表型信息,有助于肿瘤的早期诊断、个体化治疗和预后监测。然而,CTCs是一种极其罕见的细胞群体,在癌症患者外周血中十分稀少,这对从患者血液中分离CTCs并无损释放进行下游分析提出了挑战。目前,基于CTCs的物理和生物学特性已经发展了许多分离和富集CTCs的策略。为了克服目前CTCs捕获的常规界面局限性,具有独特性能的纳米界面应运而生,特别是利用多功能的"化学抗体"(核酸适配体)作为细胞识别元件。本文综述了适配体功能化的多维(从零维到三维)纳米结构界面分离和释放CTCs的重要进展,并讨论了纳米界面与微流控相结合的优势。

Abstract:Circulating tumor cells(CTCs)are not only the important topics in tumor research and clinical diagnosis,but also important markers of‘liquid biopsy’.CTCs carry genetic and phenotypic information of tumor tissues,which is helpful for early diagnosis,individualized treatment and prognosis monitoring of tumor.However,CTC is an extremely rare cell population in the peripheral blood of cancer patients,which poses a challenge to isolate CTCs from patients’blood with no-damage release for downstream analysis.Currently,many strategies for isolating and enriching CTCs have been developed based on the physical and biological characteristics of CTCs.To overcome the limitations of the conventional macroscale interface,the nanostructured interface with unique properties has been deployed in CTCs capture and release,particularly the use of multifunctional‘chemical-antibodies’(aptamers)as cell-recognition elements.This review summarizes the progress in the separation and release of CTCs based on nanostructured interface with nano-architectures from the zero dimensional(0 D)to 3 D,and discusses the advantages of the combination of nanointerface and microfluidics.
