首次报道了应用ＰｅｒｆｕｓｉｏｎＧ蛋白亲和色谱法快速测定人体生长激素与抗体免疫反应活性和摩尔配比的方法。这一新方法与传统的免疫测试技术相比较，具有快速、易于自动化操作和低劳动强度的优点。

Characterization of Immunochemical Reaction for Monoclonal Antibody to Human Growth Hormone by Perfusion Protein G Affinity Chromatography

n extremely rapid assay technique for antibody-antigen interaction has been developed utilizing ProteinG bound to perfusion chromatography matrices. The antibody and antigen were mixed and incubated at differ-ent molar ratios by keeping concentration of antibody at constant. Then the mixture solution of antibody andantigen can be injected on the Protein G column. The complex of antibody and antigen formed in sample solu-tion can be retained on tbe Protein G column as free antibody does. The peak area and height of the retainedcomplex and antibody peak linearly increase with the molar concentration of antigen when it is not stoichio-metric ally in excess to the corresponding antibody.However,those of the retained peak keep constant whenthe concentration of antigen is in excess. The method developed can be used to determine not only the biologi-cal activity between antibody and antigen very quickly, but also the immunological reation between antibodyand antigen when the complex of them forms.
