摘    要:建立了采用超高效液相色谱测定婴幼儿配方奶粉中叶黄素的检测方法。样品经丙酮溶液提取,离心分层,冷冻离心去脂,YMC Carotenoid C30色谱柱(150 mm×4.6 mm,3 μm)分离。以甲醇-甲基叔丁基醚(70:30,v/v)为流动相等度洗脱,流速为0.5 mL/min,进样量5 μL,柱温25 ℃,二极管阵列检测器检测,检测波长445 nm。方法在20~500 μg/L范围内线性关系良好;相关系数为0.999 9;定量限为20 μg/L。添加量在50、250、2000 μg/kg时,叶黄素的回收率为97.9%~104.4%。本方法操作简便,结果准确,灵敏度高,适用于婴幼儿奶粉中叶黄素的测定。

Abstract:An ultra-high performance liquid chromatography (U-HPLC) method for the determination of lutein in the infant formula milk powder was developed. The sample was extracted with acetone and defatted using freezing centrifugation method. The U-HPLC separation was achieved using a YMC Carotenoid C30 column (150 mm×4.6 mm, 3 μm) with the mixture of methanol/methyl tert-butyl ether (70:30, v/v) as the mobile phase under isocratic elution. The flow rate was 0.5 mL/min and the column oven temperature was 25 ℃. The injection volume was 5 μL. It was detected on a photodiode array detector at a wavelength of 445 nm. The results showed that the linear range was 20-500 μg/L (r=0.999 9), and the limit of quantification was 20 μg/L. The mean recoveries of lutein varied from 97.9% to 104.4% spiked at 50, 250 and 2000 μg/kg. The established method is simple, accurate and sensitive for the rapid determination of lutein in infant formula milk powder.
