摘    要:本实验采用非线性色谱的展开方式之一-超载洗脱, 在普通的分析型离子交换柱TSK Gel-5PW (Φ7.5×75mm)上, 一次进样蛋白质混合样150mg, 成功地进行了溶菌酶的分离纯化, 回收率达90%。收集的馏份经透析和冰冻干燥后,通过高效毛细管电泳(HPCE)测定纯度, 得到了满意的结果, 活性回收率达88%。

Abstract:The separation and purification of lysozyme from 150mg protein mixture on TSK Gel CM-5PW, an analytical cation exchange column (Φ7.5 ×75mm), by overloading elution mode of nonlinear chromatography was reported in this paper. It has been found that more than 90% of recovery and a satisfactory purity of lysozyme fraction, which was checked with standard sample by high performance capillary electrophoresis and UV spectrophotometry, was achieved.
