摘    要:在稀硫酸介质中,蛋白质可灵敏地阻抑溴酸钾氧化二甲酚橙褪色的兰多尔特(Landolt)反应,据此建立了阻抑动力学光度法测定痕量蛋白质的新方法.在优化实验条件下,于430 nm波长处进行测定,蛋白质含量在0.2~2.0 mg/L范围内与吸光度呈线性关系,检出限为0.141 mg/L,标准加入回收结果满意.

Abstract:In this paper, the discoloration of Xylenol orange by potassium bromate in H_2SO_4 medium-Landolt reaction was studied.Based on this fact, a new method for the determination of trace BSA was established by inhibition kinetic spectrophotometry.Under the optimum condition, the linear range is 0.2~2.0 μg/mL, and the detection limit of the method was 0.141 mg/L.The developed method was applied to the determination of BSA with satisfactory results.
