摘    要:用紫外分光光度法和荧光光谱法研究了多柔比星( Adriamycin,ADM)稀土金属离子配合物(ADM-M)与DNA的相互作用.结果发现,在pH=7.0时,ADM与Eu3+、yb3+能形成稳定配合物,该配合物可使DNA的最大吸收产生明显的减色效应及红移,并能够竞争置换溴化乙锭(EB)与DNA的结合点.KI猝灭试验发现D...

Abstract:The interaction of adriamycin rare earth metal complexes with DNA have been studied using UV spectra and fluorescence spectra.The results show that adriamycin can form stable complexes with rare earth metal Eu3+and Yb3+in pH=7.0solution.Extensive hypochromism and red shifts in the absorption spectra were observed when ADM-M bound to DNA and there was a strong competition for DNA binding between ADM-M and EB in the presence of the complexes.Iodide quenching experiment revealed obvious protection of DNA to ADM-M.The fluorescence intensity of ADM-M-DNA increased with the change of ionic strength.Thermal denaturation experiments exhibited that the quenching of fluorescence from ADM-M by single strand(ssDNA)was smaller than by double strand(dsDNA).The results suggested that the ADM-M complexes interact with DNA mainly by inserting binding and electrostatic binding.
