摘    要:在 p H6 .0 0— 7.0 0和离子强度低于 0 .0 5 0 mol/ L的条件下 ,研究了百里酚蓝 (TB) -十六烷基三甲基溴化铵 (CTMAB) -核酸体系的共振光散射光谱 (RLS) ,影响因素及最佳反应条件。在最佳条件下 ,体系的ΔIRL S与 y DNA、ct DNA和 fs DNA在一定的浓度范围内呈线性关系。检出限可达 1.30 ng/ m L。该方法简便、快速 ,具有较高的灵敏度和准确度 ,应用于合成样品中核酸的测定 ,结果令人满意

Abstract:In hexamethylene tetramine(HMTA) buffer(pH=7.60),the anionic dye thymol blue (TB) reacts with nucleic acid and cationic surfactant cetyltrimethylammonium bromide (CTMAB) to form a kind of large particles of complex,which results in significant enhancement of RLS intensity.The spectral characteristics of RLS,the effective factors and optimum conditions of the reaction were investigated.The enhanced RLS comes from the aggregation of TB on DNA through the bridged and synergistic effect of CTMAB.Under the optimum conditions, the enhanced RLS intensity is proportional to the concentration of nucleic acids in the appropriate range.The detection limit is 1.30ng/mL.This method is simple,rapid and was applied satisfactorily to the determination of DNA in synthetic samples.
